[Classification of the site of the conduction disturbance in right bundle branch block using body surface mapping].

We can estimate the conduction disturbance in right bundle branch block (RBBB) using body surface mapping. Characteristics of body surface mapping in the cases with RBBB are classified into three types using the appearance time and the site of the breakthrough. In the first type, breakthrough appeared in the center of the anterior chest within the normal time. It shows the peripheral block of right bundle branch. In the second type, breakthrough appeared in the center of the anterior chest later than the normal time. It shows the conduction delay of the main trunk of right bundle branch. In the third type, breakthrough appeared at the left of the anterior chest later than the normal time. It suggests the complete block of the main trunk of right bundle branch.

[Characteristics of body surface mapping in the cases of idiopathic ventricular fibrillation].

Body surface mapping was recorded in three patients with idiopathic ventricular fibrillation (VF). A late r' wave appeared and disappeared in the precordial leads in all three patients. In one case, the late r' wave appeared with epithides of VF. In the presence of the late r' wave, the QRS duration was extremely prolonged. However, the appearance time of the breakthrough in the QRS isopotential map was not significantly prolonged. The time of the peaks of the r' wave was elongated in the mid anterior chest. These findings suggest that peripheral conduction disturbances in the right ventricle are related to the occurrence of VF in patients with idiopathic VF.

L protein, encoded by psbL, restores normal functioning of the primary quinone acceptor, QA, in isolated D1/D2/CP47/Cytb-559/I photosystem II reaction center core complex.

Plastoquinone-9 (PQ-9)-depleted PSII reaction center core complex, consisting of CP47/D1/D2/Cytb-559/I, was isolated from spinach PSII particles. PQ-9, lipids and several proteins were extracted from the original PSII particles and separated by several steps of chromatography to be reconstituted into the isolated complex. PQ-9 reconstituted in the complex with the help of thylakoid lipids (digalactosyldiglyceride) did not function as QA by itself. However, PQ-9 simultaneously reconstituted with L protein and the thylakoid lipids successfully functioned as QA in the complex. Other proteins of PSII origin, such as CP43, H, K, nuclear encoded 4.1 and 5.0 kDa proteins, are unable to restore the QA activity in the complex.

[Effect of catechin on the ultrastructure of Trichophyton mentagrophytes].

The effects of (-) epigallocatechin gallate (EGCg) on the morphology of hyphae and conidia of Trichophyton mentagrophytes were examined using scanning and transmission electron microscopy. EGCg inhibited the germination of conidia and subsequent hyphal growth. After being treated with EGCg for 3 days, conidia showed morphological changes such as deformation, swelling, wrinkles, exfoliation of the surface layer and accumulation of granular, amorphous materials on the conidia and short hyphae surfaces. These morphological changes became apparent day by day. After 5 days, most of the ungerminating conidia were broken down. The hyphal cell walls were also exfoliated and the cytoplasm in most of the cells disappeared completely. Inner organellas were variable in size and shape. Many high electron dense particles, greatly enlarged vacuoles appeared in the cytoplasm. The degradation of cells occurred. These observations indicates that EGCg causes lysis of the conidia and hyphae. These findings suggest that EGCg may be used as an antidermatophytic agent against T. mentagrophytes infection.

Effect of dimethyl sulfoxide pretreatment on activities of lipid peroxide formation, superoxide dismutase and glutathione peroxidase in the mouse liver after whole-body irradiation.

We investigated the effects of dimethyl sulfoxide (DMSO) on radiation damage in the mouse. DMSO (i.p. 0.11 g/mouse) administered 30 min before exposure protected the mice from the gamma-whole body irradiation: the 30 days lethality was significantly decreased from 44% to 16% (P < 0.05). The contents of thiobarbituric acid reactive substances(TBA-RS) in the mouse liver increased linearly between days 2 and 10 after 9 Gy gamma ray irradiation. The TBA-RS contents in the liver on days 2 to 10 after irradiation were reduced by DMSO pretreatment. The irradiation decreased superoxide dismutase (SOD) activity in the liver on day 10. Decrease in SOD activity was prevented by DMSO pretreatment. In the electron microscopic study, the mitochondria in the irradiated mouse liver were swollen, but we could observe no change after DMSO pretreatment. The results suggest that DMSO has radioprotective effects, probably due to inhibition of lipid peroxidation.

Dynamical behavior of psb gene transcripts in greening wheat seedlings. I. Time course of accumulation of the pshA through psbN gene transcripts during light-induced greening.

The time course of the accumulation of the transcripts from 13 psb genes encoding a major part of the proteins composing photosystem II during light-induced greening of dark-grown wheat seedlings was examined focusing on early stages of plastid development (0.5 h through 72 h). The 13 genes can be divided into three groups. (1) The psbA gene is transcribed as a single transcript of 1.3 kb in the dark-grown seedlings, but its level increases 5- to 7-fold in response to light due to selective increase in RNA stability as well as in transcription activity. (2) The psbE-F-L-J operon, psbM and psbN genes are transcribed as a single transcript of 1.1 kb, two transcripts of 0.5 and 0.7 kb and a single transcript of 0.3 kb, respectively, in the dark-grown seedlings. The levels of accumulation of every transcript remain unchanged or rather decrease during plastid development under illumination. (3) The psbK-I-D-C gene cluster and psbB-H operon exhibit fairly complicated northern hybridization patterns during the greening process. When a psbC or psbD gene probe was used for northern hybridization, five transcripts differing in length were detected in the etioplasts from 5-day old dark-grown seedlings. After 2 h illumination, two new transcripts of different length appeared. Light induction of new transcripts was also observed in the psbB-H operon.

Promotion of lymphocyte blastogenesis by hemodialysate of chronic renal failure.

Effects of hemodialysate of patients with chronic renal failure (CRF) on blastogenesis of human peripheral blood lymphocytes in the presence of concanavalin A or phytohemagglutinin was investigated. Hemodialysates from 11 CRF patients significantly promoted lymphocyte blastogenesis when compared with control dialysis fluids (p less than 0.01). The strongest activity (39% promotion of the lymphocyte blastogenesis) was observed with a hemodialysate obtained at 0.5 h after beginning dialysis. The activity decreased thereafter. On the contrary, the blastogenesis-promoting activity in plasma decreased significantly after hemodialysis (p less than 0.01, n = 11). To further confirm the presence of low-molecular-weight factor(s) in CRF, ultrafiltration of plasma obtained from CRF and healthy subjects was conducted using a membrane filter with a molecular cutoff of 3,000 D. The filtrate of CRF plasma significantly promoted lymphocyte blastogenesis when compared to that of healthy subjects (p less than 0.01). Heat treatment (100 degrees C, 40 min) did not abolish the activity of the hemodialysate. None of the drugs taken by the patients nor creatinine accumulated in CRF promoted the lymphocyte blastogenesis. Chromatographic analysis of a hemodialysate demonstrated several peaks which were absent in the control dialysis fluid. These results showed the presence of a novel lymphocyte-stimulating factor(s) in CRF, which is heat stable and has a low molecular weight (less than 3,000 D).

Inhibition of monoamine oxidase by two substrate-analogues, with different preferences for 5-hydroxytryptamine neurons.

Various intraperitoneal doses of 5-fluoro-alpha-methyltryptamine (5-FMT), given to mice, dose-dependently inhibited only MAOA activity, with similar degrees of inhibition in the striatum, hypothalamus and the rest of the forebrain. The activity inhibited in these regions, completely recovered to control levels within 24 hr after the injection. In contrast, p-chloro-beta-methylphenethylamine (p-CMP), selectively inhibited MAOB activity, with complete recovery within 45 min after the injection. Regardless of the differences in time interval and degree of inhibition of MAOA by 5-FMT or MAOB by p-CMP, both kinds of inhibition were competitive, with respect to oxidation of the respective substrate. 5-Fluoro-alpha-methyltryptamine markedly protected only MAOA against inhibition by phenelzine, without protecting MAOB. Also, 5-FMT greatly increased one kind of animal behaviour, the head-twitch and this behaviour was greatly reduced by treatment with fluoxetine, but increased by reserpine. The results indicate that p-CMP is a short-acting, probably reversible, MAOB-selective inhibitor and 5-FMT has the same characteristics of selectivity for MAOA in central serotonergic neurons.

[Experimental study of bone resorption after tooth extraction with implanted hydroxyapatite tooth root substitute].

Hydroxyapatite (HAP) tooth root substitute implantation have been applied to extraction sockets in many studies for purpose of preservating alveolar bone. Dimensional changes were measured with sectioned study cast of cephalometric X-ray photo. In this study, dimensional changes were determined by newly developed evaluation system which utilizing image analyzer (Immunomedica Co., Ltd.), in a view of conserving alveolar ridges beneath the denture base. Cone shaped HAP tooth roots (5 or 6 mm in length) were implanted to one subject and disk-like shaped HAP tooth roots (2 mm in length) were also implanted to the other subject. Preservation of alveolar bone was observed in both shaped HAP tooth roots implanted site compared to control site. Bony ankylosis between HAP tooth root and alveolar bone, observed on 1 month after histological specimens, may contribute to this result. Smaller changes of blood flow compared to controls were obtained on implanted site, which were measured with Laser Doppler blood flow meter (Perimed KB; Sweden) on buccal mucosa. We consider that HAP shows good biocompatibility and HAP tooth roots implantation decrease the wound volume consequently.

Studies on 5-fluoro-alpha-methyltryptamine and p-chloro-beta-methylphenethylamine: determination of the MAO-A or MAO-B selective inhibition in vitro.

To further clarify highly MAO-A- or -B-selective inhibitory properties of 5-fluoro-alpha-methyltryptamine (5-FMT) and p-chloro-beta-methylphenethylamine (p-CMPEA), we determined the types and K1 values of inhibition of rat brain MAO-A and -B activity in vitro. The kinetic data obtained showed that 5-FMT is a competitive MAO-A-selective inhibitor with about a 18,000-fold higher sensitivity than MAO-B. In contrast, p-CMPEA is a competitive MAO-B-selective inhibitor with about a 620-fold higher sensitivity. Based on the present findings of highly MAO-A- or -B-selective inhibition, these two compounds might prove to be of value in in vivo studies.

The ex vivo effect of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) on rat intra- and extraneuronal monoamine oxidase activity.

After i.p. injection of 30 mg/kg 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) daily for 4 days and sacrificing the rats 4 h after the last injection, striatal monoamine oxidase (MAO)-A and -B activities, assayed by conventional method with 5-hydroxytryptamine (5-HT) and benzylamine, were not changed. By an uptake technique, with dopamine as the substrate for both uptake and MAO, intrasynaptosomal MAO-A and -B activities were found to be greatly reduced with a greater MAO-A reduction. Intrasynaptosomal 5-HT oxidation by MAO-A was not changed in other forebrain regions treated with these MPTP doses. Similar results were also found with two brain preparations treated with single MPTP doses (30 mg/kg). This reduction in intrasynaptosomal MAO activity was completely absent after treatment with lower MPTP doses (15 mg/kg, daily for 5 days) and 5 days of a withdrawal period. The decrease in MAO activity might have been due to the decrease in DA transport into striatal synaptosomes during the enzyme assay and/or to reversible inhibition of intrasynaptosomal MAO by MPP+.

Studies of monoamine oxidase and semicarbazide-sensitive amine oxidase. II. Inhibition by alpha-methylated substrate-analogue monoamines, alpha-methyltryptamine, alpha-methylbenzylamine and two enantiomers of alpha-methylbenzylamine.

The alpha-methylated substrate-analogue monoamines, dl-alpha-methyltryptamine, dl-alpha-methylbenzylamine and two optical isomers of alpha-methylbenzylamine, were shown to be inhibitors of rat lung semicarbazide-sensitive amine oxidase (SSAO), with dl-alpha-methyltryptamine being the most potent and d-alpha-methylbenzylamine, the least. The three compounds, dl-alpha-methyltryptamine and the two isomers of alpha-methylbenzylamine also inhibited rat brain monoamine oxidase (MAO)-A and -B with a greater selectivity towards MAO-A. Preincubation of rat lung and brain homogenates with either of these compounds revealed that the inhibition of MAO and SSAO is reversible. The modes of inhibition of MAO-A and -B were competitive with the substrates tested. However, inhibition of SSAO by dl-alpha-methyltryptamine was found to be a mixed type (with a Ki value of 47 microM) and those by the racemic form and two isomers of alpha-methylbenzylamine were non-competitive (with Ki values of 90 microM for the racemic compound, 1070 microM for the d-isomer and 72 microM for the l-isomer). The present results indicate that SSAO can recognize optical isomers and that some alpha-methylated monoamines tested in the present study inhibit SSAO with properties different from those as MAO inhibitors.

Participation of brain monoamine oxidase B form in the neurotoxicity of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine: relationship between the enzyme inhibition and the neurotoxicity.

A neurotoxin for nigrostriatal dopaminergic neurons, 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) and its oxidized metabolite, 1-methyl-4-phenylpyridine (MPP+), both dose-dependently inhibited rat striatal and forebrain monoamine oxidase (MAO) activity with monoamine oxidase A (MAO-A) selectively reversible (competitive, Ki = 4.5 and 2.0 microM) inhibition. A comparison of the Ki values indicated the affinity of MPP+ for MAO-A to be greater than that of MPTP. MPTP inhibited monoamine oxidase B (MAO-B) with both a reversible (competitive, Ki = 116 microM) and an irreversible time-dependent component, but inhibition by MPP+ was reversible and competitive (Ki = 180 microM). These results, together with previous findings on metabolism of MPTP to MPP+ by brain MAO-B, suggest that MPP+ is a simple inhibitor of MAO-A and MAO-B, but MPTP might be a 'suicide substrate' inhibitor for MAO-B.
