Evaluation of the anti-tumor and anti-angiogenic effect of paclitaxel and thalidomide on the xenotransplanted oral squamous cell carcinoma.

Angiogenesis is an essential process for the growth and invasion of cancer. However, it is uncertain that anti-angiogenic effects can be a major treatment strategy of oral cancer. The aim of this study was to investigate whether thalidomide and paclitaxel, which are known to be potent inhibitors of angiogenesis, have inhibitory effects on the growth of oral squamous cell carcinoma (OSCC) xenotransplanted into nude mice and whether anti-angiogenesis can be included as a major treatment strategy of oral cancer. After human OSCC cell line, KB, was subcutaneously inoculated into 32 nude mice, the volume of tumor was measured every 3 days. When the tumor mass reached 300-500 mm3, thalidomide (200 mg/kg) and paclitaxel (13 mg/kg) were administered into the animals and tumor volume change was checked. The excised tumor masses on the 30th day after administration were frozen and processed for immunohistochemistry using vascular endothelial growth factor (VEGF) and CD31, and for real-time reverse transcription-polymerase chain reaction (RT-PCR). We evaluated VEGF expression and the expression of its mRNA and CD31 for vessel density. Paclitaxel showed an inhibitory effect on the growth of transplanted human OSCC and reduced the immunohistochemical expression of VEGF and CD31 and VEGF mRNA (P<0.01). Thalidomide also lowered remarkably VEGF expression (P<0.01) and CD31 (P<0.01) as well as VEGF mRNA (P<0.05), but it did not show statistically significant inhibitory effect on the tumor growth. These results suggest that the growth of human OSCC is not simply dependent on VEGF-induced angiogenesis and that anti-angiogenic therapy alone is not likely to be effective for the treatment of OSCC, but might be regarded as adjuvant chemotherapeutic strategy.

Effect of cataract extraction on blue-on-yellow visual field.

PURPOSE: It is assumed that cataract can influence the results of blue-on-yellow perimetry. However, actual sensitivity change in blue-on-yellow perimetry before and after cataract surgery has not been fully demonstrated. METHODS: Prospective. SETTING: Institutional. STUDY POPULATION: Twenty-two eyes of 22 consecutive patients without ocular pathology, other than cataract, known to influence visual field undergoing cataract surgery. OBSERVATION PROCEDURES: Both white-on-white (W-W) and blue-on-yellow (B-Y) perimetries. MAIN OUTCOME MEASURES: Global indexes including mean deviation and other pertinent data of white-on-white and blue-on-yellow perimetries were compared before and after cataract surgery. RESULTS: Mean deviation was improved after cataract surgery in both white-on-white perimetry (from -6.88 dB to -3.36 dB, P <.0001) and blue-on-yellow perimetry (from -12.22 dB to -3.64 dB, P <.0001, paired t test). However, the mean difference between preoperative and postoperative mean deviation in blue-on-yellow perimetry (8.58 +/- 3.96 dB) was significantly higher than that of white-on-white perimetry (3.52 +/- 2.69 dB; P <.0001, unpaired t test). That is, the change in mean deviation was greater in blue-on-yellow perimetry than in white-on-white perimetry by a factor of 2.4 times. CONCLUSION: Cataract causes predominantly a general reduction of sensitivity in both blue-on-yellow and white-on-white perimetries, and the general reduction of blue-on-yellow sensitivity is far greater than that of white-on-white sensitivity.

Differences in sensitization rates to outdoor aeroallergens, especially citrus red mite (Panonychus citri), between urban and rural children.

BACKGROUND: A recent investigation has suggested that citrus red mite (Panonychus citri, CRM) is the most important allergen in citrus-cultivating farmers with asthma and allergic rhinitis. OBJECTIVE: A cross-sectional survey was performed to evaluate the prevalence of asthma and chronic rhinitis symptoms and sensitization to common indoor and outdoor aeroallergens, including CRM and Japanese cedar pollen, in rural and urban Korean children. METHODS: A total of 2,055 children (1,055 subjects living in rural areas with citrus farms and 1,000 controls in urban areas without citrus farms) were enrolled. They were evaluated by a questionnaire, and by skin prick tests with 13 common indoor and outdoor aeroallergens, including CRM and Japanese cedar pollen. RESULTS: The prevalence of wheezing and chronic rhinitis symptoms during the last 12 months was 8.3% and 35.7% in the rural children and 10.5% and 22.4% in the control group. The most common sensitizing allergens in order of decreasing frequency were Dermatophagoides pteronyssinus (26.6%), Dermatoplagoides farinae (22.7%), CRM (14.2%), cockroach (11.3%), and Japanese cedar pollen (9.7%) among the rural children, but the sensitization rates to CRM and Japanese cedar pollen were 1.3% and 0.2% among the control children, respectively. The prevalence of wheeze during the last 12 months was not different between rural children with sensitization to CRM or Japanese cedar pollen and those without sensitization (5.4% vs 6.1%; 6.9% vs 5.9%). However, the prevalence of chronic rhinitis during the last 12 months was higher among those with sensitization to CRM or to Japanese cedar pollen than among those without sensitization (40.8% vs 34.4%; 51.5% vs 33.5%). CONCLUSIONS: CRM is a common sensitizing allergen in rural children, and the sensitization rates to outdoor aeroallergens, especially CRM and Japanese cedar pollen, are very different between children from rural and urban areas in Korea.

Repeated exposure to low levels of sulfur dioxide (SO2) enhances the development of ovalbumin-induced asthmatic reactions in guinea pigs.

BACKGROUND: Sulfur dioxide (SO2) is one of the major air pollutants. It is known to aggravate asthma symptoms in human beings, but few studies have focused on the effects of SO2 upon the development of bronchial asthma in animal models. OBJECTIVE: This study was undertaken to evaluate the role of SO2 upon the development of ovalbumin (OA)-induced asthmatic reactions in guinea pigs. METHODS: Guinea pigs were divided into four groups: (1) OA- and SO2-exposed group (n = 12), (2) SO2-exposed group (n = 12), (3) OA-exposed group (n = 11), and (4) saline-exposed group (n = 7). Guinea pigs of the first and second groups were exposed to 0.1 ppm SO2 for 5 hours a day on 5 consecutive days. Guinea pigs in the first and third groups inhaled 0.1% OA aerosols for 45 minutes a day on days 3, 4, and 5. One week after the sensitization procedure, all the guinea pigs underwent bronchial challenge with 1.0% OA aerosols, using unrestricted whole-body plethysmography. Bronchoalveolar lavage and histopathologic examination were performed 24 hours after the bronchial challenge. RESULTS: Increases in enhanced pause (Penh), as an index of airway obstruction, after the bronchial challenge was significantly higher in OA- and SO2-exposed group (group 1) than the other groups (P < .05, respectively). Eosinophil counts in bronchoalveolar lavage fluids were also significantly higher in group 1 than in the other groups (P < .05, respectively). Histopathologic findings of bronchial and lung tissue in the group 1 showed an infiltration of inflammatory cells, bronchiolar epithelial damage, and mucus and cell plug in the lumen, but no significant abnormalities were observed in the other groups. CONCLUSIONS: These results indicate that repeated exposure to low levels of sulfur dioxide may enhance the development of ovalbumin-induced asthmatic reactions in guinea pigs.

Outdoor air pollutants derived from industrial processes may be causally related to the development of asthma in children.

BACKGROUND: There is no consistent evidence that outdoor air pollutants are involved in the development of asthma. OBJECTIVE: The aim of this study was to determine whether outdoor air pollutants derived from industrial processes were related to the prevalence of asthma, bronchial hyperresponsiveness, and atopy in exposed children. METHODS: A total of 7,511 children from 7 to 12 years of age were recruited in the study. Eight hundred eighty-eight of 1,009 (88%) of the subjects living around industrial factories and 5,998 of 6,502 (92%) living in a less polluted neighboring area responded to the ISAAC questionnaire. A total of 1,492 subjects 8 to 9 years old underwent skin prick tests for 11 common aeroallergens. A total of 732 of the subjects from 8 to 9 years of age underwent bronchial provocation tests with hypertonic saline (BPT-HS). RESULTS: The prevalence of ever experiencing wheezing and wheeze during the last 12 months was 25.6%, and 18.8% among the children living in the more polluted area. This was significantly higher than for those living in the less polluted area (14.2% and 9.0%, respectively). The positive rate of BPT-HS (the provocative dose of hypertonic saline causing 20% fall of FEV, < or = 23 mL) was higher among those in the more polluted area than in children living in the less polluted area (12.2% vs 7.5%). However, the rate of atopy based on the skin tests was the same in the two groups (28.3% vs 30.6%). CONCLUSIONS: The prevalence of asthma and bronchial hyperresponsiveness was higher among children living around the heavily industrial area compared with those living in the less polluted area, despite similar atopic sensitization.

Neuroprotective constituents from Hedyotis diffusa.

In a bioassay-guided search for neuroprotective compounds from medicinal plants, a MeOH extract of whole plants of Hedoytis diffusa yielded five flavonol glycosides, kaempferol 3-O-[2-O-(6-O-E-feruloyl)-beta-D-glucopyranosyl]-beta-D-galactopyranoside (1), quercetin 3-O-[2-O-(6-O-E-feruloyl)-beta-D-glucopyranosyl]-beta-D-galactopyranoside (2), quercetin 3-O-[2-O-(6-O-E-feruloyl)-beta-D-glucopyranosyl]-beta-D-glucopyranoside (3), kaempferol 3-O-(2-O-beta-D-glucopyranosyl)-beta-D-galactopyranoside (4), and quercetin 3-O-(2-O-beta-D-glucopyranosyl)-beta-D-galactopyranoside (5), and four O-acylated iridoid glycosides (6-9). Compounds 1 and 2 are previously unreported natural products, and all nine compounds exhibited significant neuroprotective activity in primary cultures of rat cortical cells damaged by L-glutamate.

Citrus red mite (Panonychus citri) may be an important allergen in the development of asthma among exposed children.

BACKGROUND: Recent investigations have demonstrated that spider mites are important allergens in the development of asthma in fruit-cultivating farmers. OBJECTIVE: The aim of this study was to evaluate the sensitization rate to the citrus red mite (Panonychus citri) in children living in rural areas, and to determine the allergenic cross-reactivity with other mites. METHODS: A total of 7254 children (ages 7-15 years) living in rural areas were enrolled, and each subject was evaluated by a questionnaire and a skin prick test. Allergenic cross-reactivity was evaluated by ELISA inhibition tests. RESULTS: The most common sensitizing allergens were house dust mites, followed by citrus red mite and cockroach. High serum-specific IgE bindings to the citrus red mite were detected in 21 of 100 randomly selected subjects. The prevalence of asthma was higher among those with positive skin responses to the citrus red mite than with negative skin responses to this mite. ELISA inhibition tests showed that IgE bindings to this mite were minimally inhibited with additions of domestic mites. CONCLUSION: Spider mites such as the citrus red mite may be important outdoor allergens among children living in rural areas, and spider mite-derived allergens have unique allergenic determinants compared with domestic mites.

IgE-mediated occupational asthma induced by herbal medicine, Banha (Pinellia ternata).

There have been few reported cases of occupational asthma induced by Pinellia ternata (Banha), and the mechanism responsible for this type of asthma is still undetermined. We report a case of Banha-induced occupational asthma with IgE-mediated mechanism. The patient had positive skin responses to Banha extract and Banha-specific bronchial challenge elicited an early asthmatic response. The serum-specific IgE binding to Banha extract was detectable and completely inhibited with the additions of 0.1 microg/mL of Banha extract on ELISA inhibition. Seven IgE binding components to Banha extract (6.5, 22, 24, 32, 34, and 48 kDa) were detected using SDS-PAGE and immunoblot analysis. In conclusion, the results of this study suggest that P. ternata (Banha)-derived allergens are able to cause IgE-mediated bronchoconstriction in exposed workers.

Identification of IgE-binding components of citrus red mite in sera of patients with citrus red mite-induced asthma.

BACKGROUND: Our previous investigations demonstrated that citrus red mite (CRM) antigen could cause IgE-mediated bronchoconstriction in exposed farmers working on citrus farms. OBJECTIVE: This study was performed to confirm IgE-binding components and major allergens within the CRM antigens. METHODS: Ten subjects who had been diagnosed as having CRM-induced asthma were enrolled. Serum-specific IgE antibodies to CRM antigens were measured by using an ELISA. To identify IgE-binding components and major allergens, SDS-PAGE, 2-dimensional PAGE, IgE-immunoblot analysis, and amino acid sequencing of major allergens were performed. RESULTS: All the asthmatic subjects had high specific IgE antibodies to CRMs. Twelve percent SDS-PAGE analysis showed more than 10 protein bands ranging from 6 to 64 kd. SDS-PAGE and IgE-immunoblot analysis with each individual serum showed 5 IgE-binding components (11, 24, 35, 40, and 64 kd), with 2 (24 and 35 kd) of them bound in more than 50% of the study subjects. Two-dimensional PAGE and IgE-immunoblot analysis demonstrated that the major allergen at 24 kd had 2 bands with different isoelectric points of 4.75 and 5.1. Thirty-five kilodaltons had one band with an isoelectric point of 4.75. All amino acid sequencing of the 2 major allergens was performed, which was not homologous with any previously characterized allergens. CONCLUSION: Five IgE-binding components and 2 major allergens (24 and 35 kd) were identified within the CRM antigen. The N-terminal amino acid sequence of the 2 major allergens (24 and 35 kd) was determined.

Comparative radiologic study of bone density and cortical thickness of donor bone used in mandibular reconstruction.

OBJECTIVE: The aim of this study was to compare the total cancellous bone density, bone-implant interface density, and cortical thickness of 6 donor bone types commonly used in oral and maxillofacial reconstruction. METHODS: A total of 120 bones from 20 Korean adults-including iliac bones, fibulas, cranial bones, scapulas, ribs, and clavicles-were selected. The implant recipient site was determined by the shape, contour, and anatomical limitations of the bones. The serial cross-sectional images of each bone were then acquired through computed tomography. Total cancellous bone density, bone-implant interface density around the imaginary implant fixture, and the cortical thickness along both sides of the imaginary fixture on each cross-sectional image were evaluated and compared. RESULTS: The cancellous bone density of each donor bone type had a statistically significant difference. The cranial bone showed the highest cancellous bone density, followed by the iliac bone, clavicle, scapula, rib, and fibula (P <.05). The bone-implant interface density of the cranial bone, clavicle, fibula, and scapula each belonged to the same Duncan's group, whereas the rib and iliac bone showed lower bone-implant interface density. In average cortical thickness, the scapula and fibula had a thicker cortex surrounding the imaginary implant than the other bones, and the rib had the thinnest cortex. CONCLUSION: Although more extensive testing is needed to explain the clinical implications of these results, the findings of this study may help clinicians choose the most appropriate donor bone.

Spatio-temporal pattern of EEG in young brain respiration-training children.

We have evaluated the effect of 'Brain Respiration' training on brain activity using Karhunen-Loeve (KL) decomposition as a method for spatio-temporal analysis of the electroencephalogram (EEG). BR training is a form of breath-work to optimize the function of the brain by concentrating Qi energy in the brain. Recently, BR-training has been reported to improve emotional maturity (i.e., EQ), short-term memory and intuition (Yoo et al., 1998). EEG data were taken during BR-training from 12 young BR-trainees (average age: 9.4 years) who had trained for 4 to 14 months, and during relaxation from age matched non-trained children. Spatio-temporal analysis showed a significant difference of EEG dynamics in right prefrontal, right inferior frontal, posterior temporal, parietal and occipital areas between BR-trainees and the control group. Amplitude of eigenvector components of BR-trainees in the areas of frontal, temporal and occipital cortex was larger than that of non-trained children (values were smaller in parietal cortex), with remarkably high amplitude alpha coherence all over the scalp. These results suggest that BR-training possibly activates brain function through changes in the activity of the frontal association area where higher mental integration and creative activities are mediated.

Comparative effects of high dietary levels of organic and inorganic selenium on selenium toxicity of growing-finishing pigs.

This experiment evaluated the effect of high dietary Se levels using organic or inorganic Se on the selenosis responses in growing-finishing swine. A 2 x 4 factorial arrangement of treatments in a randomized complete block design was conducted in two replicates. Sodium selenite or Se-enriched yeast was added at 5, 10, 15, or 20 ppm Se to corn-soybean meal diets. A basal diet without added Se was a ninth treatment group. Ninety crossbred barrows initially averaging 24.7 kg BW were allotted at five pigs per pen. Pigs were bled at 3-wk intervals and plasma Se, glutathione peroxidase (GSH-Px) activity, glutamic oxalacetic transaminase (PGOT), hemoglobin, packed cell volume, and blood cell Se concentration were measured. After 12 wk, pigs were killed and various tissues and bile were collected for Se analyses. Pig body weights, daily gains, and feed intakes were similar for both Se sources when provided at < or = 5 ppm Se, but each measurement declined in a different manner for each Se source as the dietary Se level increased. The decline was more rapid when the inorganic rather than organic Se source was fed, resulting in interaction responses (P < 0.01). Hair loss (alopecia) and separation of the hoof at the coronary band site occurred at > or = 10 ppm inorganic Se but at > or = 15 ppm organic Se level. Plasma GSH-Px activity increased (P < 0.01) when high dietary Se levels of either Se source was fed. Plasma and blood cell Se increased at each period as dietary Se level increased (P < 0.01) and was greater when organic Se was provided (P < 0.05). Blood cell Se concentration reached a plateau when inorganic Se, but not when organic Se, was fed and increased as the experiment progressed. This resulted in a three-way interaction (P < 0.01). Plasma GOT activity at the 12-wk period was elevated when inorganic Se was provided at > or = 15 ppm Se but not when organic Se was fed, resulting in an interaction (P < 0.05). Tissue Se concentrations increased as dietary Se level increased and when organic Se was provided, resulting in interaction responses (P < 0.05). Bile was a yellow color when the basal diet was fed but was dark brown at > 10 ppm inorganic Se and at 20 ppm when organic Se was provided. Bile Se increased as dietary Se level increased (P < 0.01). These results suggest that dietary Se from inorganic or organic sources was toxic at > or = 5 ppm Se, but subsequent selenosis effects were more severe and occurred sooner when sodium selenite was the Se source.

Effect of dietary selenium source, level, and pig hair color on various selenium indices.

The first experiment evaluated the effects of feeding various levels of Se, two Se sources, and hair color on selenosis responses in growing-finishing pigs. The study conducted in two replicates was a 2 x 6 x 2 factorial arrangement in a split-plot design. Sodium selenite and Se-enriched yeast added at 0.3, 1, 3, 5, 7, and 10 ppm Se served as the main plot and pig hair color as the subplot. A total of 96 crossbred pigs were allotted and fed their treatment diets for a 12-wk period. White and dark (red or black) hair samples were collected from the dorsal-midline at the 4-, 8-, and 12-wk periods from one pig of each hair color from each treatment pen. Lower pig weights (P < 0.10) and daily gains (P < 0.05) occurred as dietary Se level increased when pigs were fed either Se source. Selenosis responses were somewhat more severe, when the inorganic Se source was fed. Alopecia and hoof separation were encountered after the 8-wk period when pigs were fed inorganic rather than organic Se. Plasma Se increased as dietary level increased (P < 0.01), when organic Se was provided (P < 0.01), and was higher (P < 0.05) when pigs were white-haired. A time x hair color x dietary Se level interaction (P < 0.05) occurred, in which hair Se concentration was higher in dark- than in white-colored pigs and increased as dietary Se level increased as the experiment progressed. The correlation coefficient between dietary Se level and hair Se concentration averaged 0.90 (P < 0.01). Cysteine was the amino acid in the highest concentration in hair, but this and other amino acids were not affected by Se level, Se source, or hair color. A second experiment was a 3 x 6 factorial arrangement in a split-plot design with three 9-mo-old gilts from each of the Yorkshire, Duroc, and Hampshire breeds to determine whether hair Se concentration differed by body location and breed. Hair samples were collected from the shoulder, back, rump, front-leg, belly, and hind-leg areas. Hair Se concentration was higher in red- and white-haired pigs and lower in black-haired gilts (P < 0.01). Higher hair Se concentrations (P < 0.05) occurred from the lower than from the upper body areas. Our results suggest that selenosis occurs at dietary levels > 5 ppm and that white-haired pigs exhibit alopecia sooner than dark-haired pigs. No difference in hair Se concentration occurred when diets were < 1 ppm Se, but as dietary Se level increased dark-haired pigs retained more Se in their hair than white-haired pigs.

Prolonged feeding of high dietary levels of organic and inorganic selenium to gilts from 25 kg body weight through one parity.

An experiment evaluated the selenosis effects from feeding high dietary Se levels of organic or inorganic Se sources to growing gilts with the dietary treatments continued through a reproductive cycle. A total of 88 gilts were allotted at 25 kg BW to two replicates in a 2 x 4 factorial arrangement in a randomized complete block design. Inorganic Se (sodium selenite) or organic (Se-enriched yeast) Se were added to diets at 0.3, 3, 7, or 10 ppm Se. At 105 kg BW, four gilts per treatment were killed and livers collected for Se analysis. At 8 mo of age, three gilts from each treatment group were bred and fed their treatment diet, with subsequent reproductive performance and selenosis effects evaluated. Serum collected at various intervals in gilts, sows, and progeny measured glutathione peroxidase activity and Se concentrations. Sow colostrum and milk was analyzed for their Se concentrations. Three pigs per treatment were killed before colostrum consumption and at weaning (14 d) and tissue collected for Se analysis. Gilt gains (P < 0.01) and feed intakes (P < 0.05) declined during the grower period as dietary Se level increased for both Se sources. Serum and liver Se concentrations increased as dietary Se level increased and was higher when organic Se was fed (P < 0.01). Sows fed dietary Se levels at > 7 ppm had lower gestation weights (P < 0.05) and lower lactation feed intakes (P < 0.05). As Se level increased, sows fed organic Se had a lower number of live pigs born (P < 0.05) and weaned fewer pigs (P < 0.05) with lower litter gains (P < 0.05) than did sows fed inorganic Se. Colostrum and milk Se concentrations increased as dietary Se levels increased particularly when organic Se was fed (P < 0.01). Neonatal and weanling pig tissue Se and serum Se concentrations increased as dietary Se level increased and when organic Se was fed, resulting in interaction responses (P < 0.01). Pigs nursing sows fed > 7 ppm inorganic Se had hoof separation and alopecia, with the severity being greater when sows were fed the inorganic Se source. These results suggest that both the organic and inorganic Se sources were toxic when fed at 7 to 10 ppm for a prolonged period, but organic Se seemed to express the selenotic effects more on reproductive performance, whereas inorganic Se was more detrimental during lactation.

Malaria transmission potential by Anopheles sinensis in the Republic of Korea.

To evaluate the factors that determine the transmission level of vivax malaria using vectorial capacity, entomological surveys were conducted from June to August, 2000. From 6 nights of human-bait collection in Paju, the human biting rate (ma) was counted as 87.5 bites/man/night. The parity of Anopheles sinensis from human baiting collections fluctuated from 41% to 71% (average 48.8%) of which the rate gradually increased as time passed on: 35.2% in Jun.; 55.0% in July; 66.2% in Aug. From this proportion of parous, we could estimate the probability of daily survival rate of An. sinensis to be 0.79 assumed with 3 days gonotrophic cycle and the expectancy of infective life through 11 days could be defined as 0.073. Blood meal analysis was performed using ELISA to determine the blood meal source. Only 0.8% of blood meals were from human hosts. We could conclude that An. sinensis is highly zoophilic (cow 61.8%). Malaria is highly unstable (stability index < 0.5) in this area. From these data, vectorial capacity (VC) was determined to be 0.081. In spite of a high human biting rate (ma), malaria transmission potential is very low due to a low human blood index. Therefore, we could conclude that malaria transmission by An. sinensis is resulted by high population density, not by high transmission potential. For this reason, we need more effort to decrease vector population and vector-human contact to eradicate malaria in Korea.

Linkage between bronchial responsiveness to methacholine and gene markers of IL-4 cytokine gene cluster and T-cell receptor alpha/delta gene complex in Korean nuclear families.

BACKGROUND: Several candidate genes have been reported to be linked to intermediate phenotypes of asthma in Caucasian populations. OBJECTIVE: To evaluate linkage between phenotypes of asthma and gene markers of high affinity IgE receptor-beta gene (D11S97), IL-4 cytokine gene cluster (IL-4R1), and T-cell receptor alpha/delta gene complex (D14S50) in Korean nuclear families. METHODS: Nuclear families (127 probands and their 130 siblings) for the linkage analysis were ascertained through asthmatic children. Linkages between total serum IgE response, skin responses to common aeroallergens, and bronchial responsiveness to methacholine were performed using a sib-pair approach. RESULTS: The square difference of the slope of the dose-response curve (DRS) between sib-pairs with two IL-4R1 identical alleles was smaller than with one or with neither IL-4R1 identical allele (P = 0.004). As for D14S50, the differences of DRS between sib-pairs with two identical alleles and with one identical allele were smaller than with neither identical alleles (P = 0.01). As for D11S97, no significant differences were observed among the groups with identical alleles of two, one or zero. With regard to total serum IgE levels, no significant linkage was found between this phenotype and the above three gene markers. As for skin responses to common aeroallergens, significant evidence was obtained to establish a linkage between this phenotype and the marker IL-4R1 (P = 0.01). However, no significant linkage was found between this phenotype and the markers D11S97 and D14S50. CONCLUSION: The expression of bronchial responsiveness to methacholine may be influenced by genetic factors in the IL-4 cytokine gene cluster and/or T-cell receptor alpha/delta gene complex, but the genetic influence of the FcepsilonRI-beta gene may be minimal in the expression of bronchial responsiveness in Korean nuclear families.

HLA-DRB1*07 may have a susceptibility and DRB1*04 a protective effect upon the development of a sensitization to house dust mite Dermatophagoides pteronyssinus.

BACKGROUND: IgE responses to house dust mite-derived allergens seem to be the most important in the development of atopic asthma and rhinitis, but it has been difficult to demonstrate genetic control of the IgE response to the allergens. OBJECTIVE: This study was undertaken to investigate the association between sensitization to house dust mite, D. pteronyssinus (DP), and genotypes of HLA-DRB1 alleles. METHODS: DNAs were extracted from two groups of unrelated Koreans: (1) 178 with sensitization to DP; and (2) 99 age-matched non-atopic controls. Genotypes of the HLA-DRB1 alleles were determined using polymerase chain reaction (PCR)-based methods. RESULTS: The frequency of HLA-DRB1*07 was significantly increased in the DP-sensitive subjects compared with the controls (15.7% vs 4.0%, P = 0.009). Conversely, the frequencies of HLA-DRB1*04 and *14 were decreased in the DP-sensitive subjects compared with the controls (27.5% vs 45.5%, P = 0.002; 13.5% vs 24.2%, P = 0.02). Of the DRB1*04 alleles, DRB1*0403 was significantly decreased in the DP-sensitive subjects compared with the controls (3.9% vs 13.1%, P = 0.005). No significant differences were found in the distributions of the other HLA-DRB1 alleles between the two groups. CONCLUSION: HLA-DRB1*07 may have a susceptibility, and DRB1*04 and DRB1*14 may have a protective effect, upon the development of a sensitization to DP.

Oxidation of polycyclic aromatic hydrocarbons by ozone in the presence of sand.

A series of soil slurry experiments was performed to investigate the characteristics of PAHs removal by ozone in various conditions. Gaseous ozone was bottled into the aqueous phase in the presence of soil contaminated by PAHs. The effects of soil media, OH radical scavengers, ozone dosage, and humic acid were examined at the given experimental conditions. There exists a substantial difference in the removal of PAH according to the types of soil media tested. Baked sand showed the highest removal efficiency compared to the others. The descending order of removal rate was: BS > S > GB. This is considered to be due to the OH radical effect produced by catalytic reactions of ozone with the reactive site on the and. This is qualitatively proved by the experiment of scavenging OH radicals using tert-butanol. The comparison of half-lives of ozone in sand and glass bead columns further supports this hypothesis. It was found that about 22% of enhancement of phenanthrene destruction was accomplished by OH radicals produced by the catalytic ozone decomposition. The rate of ozone consumption for the phenanthrene oxidation was obtained as 1.88 mg/mgO3/min.
