A case of tick bite from Amblyomma testudinarium on the glans penis.

A tick which bit the glans penis of a 40-year-old Japanese man was surgically excised together with the skin where the bite occurred. It was identified as Amblyomma testudinarium through the taxonomical investigation of its morphological characteristics. To our best knowledge, our patient is the sixth tick bite from this species of tick recorded in the Chugoku District of Japan.

Latex agglutination test for canine parvovirus.

Canine parvovirus (CPV) was detected in faeces from dogs with diarrhoea by a specific slide agglutination test using latex particles coated with anti-CPV monoclonal antibody (LA-anti-CPV). The agglutination of LA-anti-CPV with CPV on a glass slide was evident macroscopically within 2 min. The sensitivity of the latex agglutination (LA) test was similar to that of the hemagglutination test. The LA test is available for the rapid diagnosis of CPV infection at an animal hospital.

Area dorsalis pars lateralis of the telencephalon in a teleost (Sebastiscus marmoratus) can be divided into dorsal and ventral regions.

Area dorsalis pars lateralis was shown to be subdivided into a dorsal region and a ventral region by a shallow groove on the telencephalic surface. In the dorsal region, three layers were distinguished by Nissl staining; a surface layer I, a middle layer II, and a deep layer III. Although a three-layered structure was also observed in the ventral region, it was not as clear as that in the dorsal region. Cells in area dorsalis pars lateralis were classified by Golgi-Cox staining into 5 types; small cells, horizontal cells, pyriform cells, inverted cells, and multipolar cells. The distribution of zinc was examined by the Neo-Timm method and found to be present in layers I and II and weakly in layer III of the dorsal region but not in layers I and II of the ventral region. The presence of zinc in synaptic terminals in the dorsal region was verified by analytical electron microscopy. Following separate and localized injections of horseradish peroxidase into dorsal and ventral regions, some different fiber connections were revealed. Both dorsal and ventral regions had reciprocal connections with ipsilateral area dorsalis pars centralis, area ventralis pars supracommissuralis, area ventralis pars dorsalis, area ventralis pars posterior, nucleus prethalamicus, and had afferent connections from area ventralis pars lateralis, area ventralis pars intermedia, raphe nuclei, and locus ceruleus. Only the dorsal region, however, received projections from contralateral area dorsalis pars centralis, ipsilateral area dorsalis pars dorsalis, area dorsalis pars medialis, and area dorsalis pars posterior and sent fibers to the inferior lobe, nucleus paracommissuralis, and optic tectum. Although area dorsalis pars lateralis had been thought to be a homogeneous region of the primary visual area of the telencephalon, histochemistry of zinc and fiber connections of the region indicates that the area is subdivided into two regions: dorsal and ventral.

[Density distribution of dopaminergic neurons in the retina of a marine teleost, Thamnoconus (Navodon) modestus].

By means of tyrosine hydroxylase immnocytochemical detection, regional densities of dopaminergic (DAergic) neurons were measured in 6 whole-mounted retinas of a marine teleost, Thamnoconus (Navodon) modestus. Isodensity lines of DAergic cells in the right retina were drawn to make a contour map. Depending upon the range of cell densities, the retinal field was divided into 3 regions: zone A, containing more than 300 per mm square; zone B, from 200 to 299; and zone C, below 200. Zone A was located in the small temporal (A1) and nasal (A2) areas as well as in the peripheral margin (A3). Zone B formed a horizontal streak-like band between regions A1 and A2. Low density areas (C) were located dorsally or ventrally to zone B. Where the densities were higher, soma sizes appeared to be smaller. DAergic neurons in the peripheral zone A3 were bipolar and the dendrites ran parallel to the retinal margin, while DAergic neurons in the other zones were multipolar and the dendrites ran in various directions. The presence of the small temporal (A1) and nasal areas (A2) with high densities of DAergic neurons, as well as the streak-like area (B) with a relatively high density, may correspond to similar areas in the isodensity contour map of ganglion cells.

Detection of antibodies against porcine reproductive and respiratory syndrome (PRRS) virus in swine sera by enzyme-linked immunosorbent assay.

We developed an enzyme-linked immunosorbent assay (ELISA) to detect antibodies against PRRS virus in swine sera. The ELISA antigen was prepared from MARC-145 cells infected with PRRS virus. The results from serial serum samples from experimentally infected and random swine indicated that the ELISA was more sensitive than indirect fluorescent and immunoperoxidase monolayer assays. Since the ELISA enables many sera to be simultaneously and rapidly tested, it was useful for detection antibodies against PRRS virus in swine sera.

Growth and metastasis of two canine mast cell tumors in SCID mice.

Intermediately differentiated mast cell tumors in two dogs were subcutaneously xenotransplanted into severe combined immunodeficiency (SCID) mice. Both tumors primarily grew and were serially transplantable in SCID mice. The histological features of the xenografts were similar to those of original tumors in dogs. Both of these subcutaneous tumors were judged as connective tissue mast cells by toluidine blue stain. One of the two xenografts metastasized to the tracheobronchial lymph nodes, omentum, mesentery, subpleural region and retroperitoneum of the SCID mouse. These canine mast cell tumor xenografts in SCID mice may be valuable tools for investigating the growth and metastatic behaviors of the tumor.

The use of epoxy patch grafts for the repair of experimentally-created diaphragmatic defects in dogs.

Canine pericardium which had been treated with polyepoxy compounds (Denacol EX-313) was used as a patch graft for the correction of experimentally-created diaphragmatic defects in five dogs belonging to the same litter. Clinical, macroscopic and histological examinations were conducted every month up to five months after suturing of the patch graft. Clinical examination of the patch graft showed no apparent abnormalities. Macroscopic examination conducted during autopsy showed that the patch graft maintained adequate elasticity for five months after suturing, the surface of the patch graft was covered with a thin membrane and neovascularization was observed. Histological examination showed that the surface of the patch graft was covered with a thin membrane. Inflammatory tissue reactions were observed at one month, but gradually decreased from the second month onwards. In addition, the patch graft had excellent tissue affinity.

Evaluation of water durability at adhesion interfaces by peeling test of resin film.

The water durability at adhesion interfaces was investigated by measurement of the peeled area of thin resin films bonded with 4-META resin on metal surfaces after imposing thermal stress using liquid nitrogen. Thermal stress at the adhesion interface was calculated by a computer-aided finite element method. On 18-8 stainless-steel specimens which bond strongly with 4-META resin, total interface failure occurs on specimens with resin thicker than 0.55 mm in dry condition. A resin layer of 0.25 mm was chosen to study degradation of the adhesion interface by water. The shearing stress was calculated as 16 MPa for a 0.25 mm thick resin layer. On mild-steel adherent interface with 4-META resin which degrades rapidly by water molecule, the relationship between water immersion time and degradation at the adhesion interface was discussed together with the amount of water penetrated at the interface. The method proposed in the present study is effective as a quick evaluation method for water durability at the adhesion interface.

Linear immunoglobulin A (IgA) bullous dermatosis of childhood: identification of the target antigens and study of the cellular sources.

A 4-year-old girl developed numerous tense blisters on the body. The blisters healed without scarring. Histopathological and immunofluorescence studies showed findings consistent with linear immunoglobulin A (IgA) bullous dermatosis of childhood. Immunoelectron microscopy revealed deposition of IgA in the lamina lucida of the basement membrane zone of the dermal-epidermal junction. Circulating IgA autoantibody was positive at the titre of 1 : 128 and recognized the antigens located on epidermal sites of 1 mol/l NaCl-split skin. Immunofluorescence staining of cultured normal human fibroblasts and cultured DJM-1 cells (derived from human squamous cell carcinoma of skin) with the patient's sera demonstrated that both of these cells synthesize the antigens in vitro, although fibroblasts produce the antigens more abundantly. When DJM-1 cells were injected intracutaneously into nude mice, the antigens recognized by the sera were present mainly around the tumour cell islands in a linear pattern, while the dermal-epidermal junction of mouse skin was negative, suggesting that epidermal cells may contribute directly to synthesis and deposition of the antigens at the basement membrane. By immunoprecipitation using cultured normal human fibroblasts, the patient's sera could precipitate at least two specific molecules at 100-kDa and 145-kDa molecular weight.

[In situ detection of Epstein-Barr virus in the non-neoplastic gastric glands and infiltrated lymphocytes].

Recently, clonal EBV-DNA and/or EBV-encoded small RNA(EBER1) have been detected in some gastric carcinomas. We reported the first observation of EBV infection in gastric glands with intestinal metaplasia, and characterized the EBV-infected lymphocytes which infiltrated in gastric mucosa. To determine the cellular location of EBV, EBER1 in situ hybridization(ISH) with an EBER1 oligonucleotide probe was applied to paraffin sections of the non-neoplastic gastric mucosa in 80 cases of gastric carcinoma and 49 cases of gastric ulcer. Not only was EBER1 expression detected in the nucleus of gastric cancer cells(5 cases) but also in non-neoplastic gastric epithelial cells(3 cases) and in infiltrated lymphocytes(40 cases). A single or a few shedding non-neoplastic epithelial cells in 2 cases of EBV-associated gastric cancer showed EBER1 expression. In one case, EBER1 was observed in all of the epithelial cells of a few gastric glands with intestinal metaplasia, suggesting that the stem cells of the metaplastic gastric glands were infected with EBV. EBV DNA was also detected in the EBER1-positive metaplastic glands scratched from the paraffin section by a single cell PCR method with a BamHI W primer pair. However, immunohistochemical examination showed that these metaplastic glands lacked expression of EBNA2 and LMP-1. The observation of these rare EBV-infected metaplastic glands reinforces the pre-transformation EBV infection hypothesis for EBV-associated gastric carcinoma. The double staining using ISH and immunohistochemistry revealed that EBER1 positive lymphocytes showed a B cell marker of L26(< 50%) but not T cell markers of UCHL1 and OPD4.

Radical formation site of cerebral complex I and Parkinson's disease.

Paraquat was reduced to the paraquat radical via complex I in bovine cerebral mitochondria and accelerated lipid peroxidation. Thirty-kilodalton subunit of complex I was considered to be the radical formation site, because of its marked destruction by the paraquat radical. The lipid peroxidation by the paraquat radical was suppressed not only by superoxide dismutase (SOD) but also by mannitol. The destruction of complex I subunits via lipid peroxidation must have been caused by the hydroxyl radical which was formed from the superoxide radical. The same phenomenon was observed by using 1-methylnicotinamide (MNA), which contains the same partial structure as paraquat in itself and is metabolized from nicotinamide in a living body. We observed NADH oxidation by MNA via cerebral complex I (Km = 26.3 mM), and MNA destroyed some complex I subunits, especially 30-kilodalton protein. Paraquat might be useful for studying the pathogenesis of Parkinson's disease (PD) in vitro, and MNA is expected to be one of the causal substances of PD from the viewpoint of the oxidative stress theory.

The fatty acids of each lipid fraction and their use in providing energy source of the plerocercoid of Spirometra erinacei.

The fatty acid concentration of each lipid fraction of plerocercoids of Spirometra erinacei and the host snake serum was investigated. The major fatty acids of phospholipid of the plerocercoids were C18:1, C18:0 and C16:0, and those of the host snake serum were C16:0, C18:1 and C18:0, in order of amount in both cases. The changes of the fatty acid composition of phospholipid of the plerocercoids when they were incubated in physiological saline at 18 degrees C and at 37 degrees C for 24 h were investigated in both cases. Polyunsaturated fatty acids increased at 18 degrees C, and saturated fatty acids increased at 37 degrees C. Michaelis constants (Km) of beta-hydroxyacyl-CoA dehydrogenase (HAD), NADH: ubiquinone oxidoreductase (complex I) (NADH: ferricyanide reaction) and complex I (NADH: ubiquinone reaction) for NADH were 20.6, 50 and 13.3 microM, respectively. The ATP production in mitochondria of the plerocercoids was accelerated by adding ADP and inhibited by adding such electron transport system inhibitors as rotenone, antimycin A and sodium cyanide. These results suggested that the fatty acids in the plerocercoids played an important role in regulating the fluidity of membrane by changing the composition in membrane lipid corresponding with the change of temperature circumstance. The NADH reduced by HAD might be accepted by the complex I in the electron transport system, and thus the parasites were capable of ATP production in a classical pathway of the oxidative phosphorylation system.

Malignant schwannoma arising in patients with von Recklinghausen's disease: report of two cases and the comparison of mast cells between benign and malignant portions.

We described two cases of malignant schwannoma arising in patients with von Recklinghausen's disease and examined the mast cells infiltrated into histologic sections. One of the two cases histologically revealed apparent mast cell infiltration in some areas of malignant schwannoma as well as in the benign neurofibroma. The malignant lesion demonstrated significantly increased percentages of degranulated mast cells over the benign lesion using FITC-avidin staining. In an electron microscopic study, mast cells in the malignant lesion displayed empty granules, piecemeal degranulation, and canaliculi structures suggesting activation. These findings were not observed in the benign lesion. The other patient histologically showed no mast cells in the malignant lesion, although the benign neurofibroma in the patient disclosed numerous mast cells. The first patient had neither recurrence nor distant metastasis. On the other hand, the second patient without mast cells in the histology had multiple distant metastases.

The vicious cycle of nonhealing neointima in fabric vascular prostheses.

Delayed neointimal healing of a fabric vascular prosthesis was investigated in an animal study focusing on the relationship between red thrombus, fibrinolysis, and endothelialization on the luminal surface. Fabric vascular prostheses were implanted into the descending aortas of 72 dogs. Fifty-nine grafts were explanted from 1 h to 1,705 days after implantation. One hour after implantation, the graft wall was red in color due to fresh thrombus; however, at 1 day the luminal surface became white. Red thrombus reappeared at 1 week and remained present in the long-term. Microscopically the initial red thrombus contained numerous erythrocytes. The white thrombus at 1 day was composed of a dense fibrin network without erythrocytes. At 2 days numerous lacunae appeared in the fibrin layer, and at 3-5 days cavernae and low density fibrin areas were present secondary to fibrinolysis. These areas allowed the blood components to infiltrate into the fibrin layer, and as a result red thrombus reformed within it. The thrombi on the luminal surface in the long-term was always red in color and composed of complicated, multiple stages of thrombus formation, i.e., fresh thrombus with erythrocytes, dense fibrin without erythrocytes, low fibrin density areas, lacunae and cavernae in the fibrin layer, and blood component infiltration into these spaces. Thrombus was always newly formed and present, and involuted in parallel due to fibrinolysis, suggesting that these phenomena perpetuated in a vicious cycle. However, at the anastomoses fibrinolysis was present, but blood component infiltration was prevented by the endothelial cell lining. These results suggest that endothelialization may arrest the vicious cycle of nonhealing neointima in fabric vascular prostheses.

Transplantation of autologous tissue fragments into an e-PTFE graft with long fibrils.

An expanded polytetrafluoroethylene (e-PTFE) graft with long fibrils transplanted with bone marrow showed rapid and uniform neointima formation in a dog study. The e-PTFE grafts (fibril length, 90 microns; 6 mm internal diameter; length, 6-8 cm) transplanted with autologous bone marrow were implanted in the abdominal aortae of 10 dogs and retrieved at 3 weeks and 3 months after implantation. Control e-PTFE grafts without bone marrow treatment were also implanted in the same manner in 8 dogs. Macroscopically the treated graft wall appeared red in color; however, there was no thrombus deposition on the surface. Light microscopic observation revealed that the treated grafts were completely lined with endothelial cells at 3 weeks. The neointima was uniform without intimal hyperplasia at the anastomotic sites. Inside the graft wall many capillary blood vessels were observed. At 3 months moderate intimal hyperplasia throughout the graft with complete endothelialization was observed. In the control grafts, endothelialization was observed at the anastomotic sites; however, half of the other areas were covered with a fibrin layer devoid of endothelial cells even in the 3-month grafts. These results indicated that neointima formation was effectively accelerated with the autologous bone marrow transplantation, but moderate intimal hyperplasia throughout the graft was inevitable in e-PTFE grafts even after complete endothelialization.

A case of deoxycorticosterone-producing adrenal adenoma.

A 29-year-old woman with deoxycorticosterone (DOC)-producing adrenocortical adenoma had hypertension and hypokalemia but without Cushingoid features. Plasma renin activity and the aldosterone concentration were low, while the DOC concentration was high (6.10-10.3 ng/ml; normal range 0.03-0.33). Plasma cortisol, androgens, and estrogens as well as urinary 17-OHCS and 17-KS were within normal limits. Furosemide administration and two hours upright posture resulted in a 3-fold increase in plasma DOC, but the administration of ACTH, dexamethasone, or angiotensin III had no effect on plasma DOC. Following resection of a right adrenal tumor weighing 70 g, the hypertension and hypokalemia disappeared. DOC content in the tumor was high. On light microscopic examination, the tumor was encapsulated, composed of cells with clear cytoplasm and large nuclei and there were extensive areas of fibrosis and infiltration of lymphocytes. According to Weiss's criteria, the tumor was considered to be an adrenocortical adenoma. Immunohistochemically, P450scc, 3 beta HSD, P450C21 and P45011 beta were positive with heterogeneity of intra-tumoral expression. No immunoreactivity for P45017 alpha in this adenoma was detected. This is different from a previous report in which a relatively small number of cells in DOC-secreting adrenocortical carcinoma were positive for P45017 alpha.

[Left diaphragmatic hernia due to blunt chest trauma complicated with intrapleural gastric perforation].

A 17-year-old male was injured by traffic accidents. Multiple left rib fractures, radiopacity of left hemithorax and rightward mediastinal shift lead us to the diagnosis of traumatic hemothorax. But left tube thoracostomy could not obtain any fluid. After this finding we suspected diaphragmatic hernia. Neither inserted nasogastric tube nor abdominal echogram could not lead to definitive diagnosis. Chest CT could identify stomach and spleen in the thoracic cavity. Insufflation of 300 ml air through nasogastric tube could not expand the stomach. By insertion of another thoracic tube some coffee-ground like materials could be obtained. Laparotomy disclosed rupture of left hemidiaphragm and intrathoracic displacement of the stomach and spleen. After reposition of stomach, we discovered 5 cm fissure of the stomach and left thoracic cavity filled with leaked gastric contents. Though reports of combined diaphragmatic hernia and gastric rupture are rare, these combination could occur if a patient with diaphragmatic hernia encounters blunt chest trauma on a full stomach.
