Immobilization stress and direct glucocorticoid effects on rat septohippocampus.

The rat septohippocampal cholinergic system to a large extent regulates the adaptive physiological and behavioral response to stress. The mesoseptal dopaminergic (DA) system, one of the converging inputs to the lateral septum, exerts a tonic inhibitory action on the septohippocampal cholinergic neurons. High concentrations of pituitary-adrenocortical hormones in plasma may activate the septohippocampal cholinergic system. We have sought to determine whether this mode of activation may be directly initiated by hormonal action on the cholinergic terminals, or indirectly induced through an alteration in the DA septal inputs. The results indicate that stress initiates rapid and transient changes in DA uptake by septal DA terminals, changes which probably contribute to the initial transient activation of the hippocampal cholinergic system. While the effects of glucocorticoids, observed in vitro, may mimic the enhanced ACh release in stress, they do not mimic the increased choline uptake. Nevertheless, high glucocorticoid concentrations may act directly on septal dopaminergic terminals to reduce their DA uptake capacity. These results imply that the septohippocampal cholinergic activity represents an integrative pathway for neuronal and hormonal signals of stress.

Sheep bronchoalveolar carcinoma: tissue associated protein complex (TAPC) in normal lung tissue and in the tumor differed quantitatively.

Sheep lungs experimentally and naturally affected by bronchoalveolar carcinoma were washed out exhaustively of soluble components by phosphate-buffered saline, pH 7.4 (PBS), followed by glycine buffer, pH 2.8 (GB), and then again by 1M KCl followed by PBS. The tissue matrix (TM) of the tumor-free region and the tumor-affected tissue were analysed separately by sodium dodecyl sulfate (SDS) polyacrylamide electrophoresis. Normal lung tissues obtained from normal sheep served as controls. Several protein fractions and fragments, identified in both the normal and the tumorous lung, have the molecular weight (MW) of 130,000-228,000, as compared with the major soluble tissue associated protein having MW of 70,000. Coomassie blue staining used in the SDS polyacrylamide system and alkaline phosphatase immunoreaction used in the Enzyme Linked Immunosorbant Assay (ELISA) showed tenfold increased concentration of the TAPC in the TM of the tumor tissue and in the blood of tumor-affected animals, respectively. Total concentration of the TAPC in the serum of tumor-affected animals was higher than in the normal. Immunofluorescent antibody test (IAT) detected the TAPC in the cytoplasm of tumor as well as in normal lung cells, and the study suggested that the TAPC reaches the peripheral blood during tissue destruction occurring at the tumor site, as observed by light and electron microscopy (LM and EM). The concentration of each of the TAPC fractions was higher in the tumor-affected sheep lung as compared with normal sheep lung. Antibodies prepared against the TAPC fractions were toxic to sheep lung cells in tissue culture. Tumor cells were more susceptible.

In vitro presynaptic modulation of cholinergic hippocampal activity by pituitary-adrenocortical hormones.

In vivo studies have shown that high blood concentrations of pituitary-adrenocortical hormones can activate the hippocampal cholinergic terminals. Incubation of hippocampal synaptosomal preparations with methylprednisolone, or with ACTH at concentrations comparable to stress-induced high concentrations in plasma, did not have any significant effects on the cholinergic parameters measured under unactivated conditions. In the presence of either high K+ or of ACh, choline uptake was decreased. This decrease was not affected by methylprednisolone. However, methylprednisolone did enhance ACh release both after a previous increase (induced by K+) or a decrease (induced by ACh) of ACh release. In contrast, ACTH had no direct effects on either unactivated or K+-stimulated synaptosomes. Thus, a differential effect was exerted by methylprednisolone on the two presynaptic regulatory mechanisms: choline uptake (no change) and ACh release (increase). We suggest that the activation, observed in vivo, resulted mainly from indirect action of the hormones on the hippocampal cholinergic terminals, in view of the fact that the direct effect in vitro was partial.

Iron, transferrin, and acid and alkaline phosphatase in healthy turkeys and in turkeys inoculated with the lymphoproliferative disease virus.

Presented are data on iron-binding capacity determinations in the serum of turkeys infected with lymphoproliferative disease (LPD) virus and in healthy males and females (laying eggs and nonlaying) from a breeding flock. Also presented are results of serum and tissue total acid and alkaline phosphatase determinations in turkey poults infected with LPD virus and their uninfected controls and of serum enzyme levels in healthy males and females from the breeding flock. There was no significant alteration in total iron binding capacity (transferrin level) in the serum of turkeys with LPD. Turkey poults inoculated with LPD virus showed a significant decrease in serum alkaline phosphatase activity 4 and 7 weeks postinfection (pi), and a decrease in serum acid phosphatase activity 7 weeks pi. Acid and alkaline phosphatase activity determined in the spleen and pancreas (organs with pronounced tumor involvement) 7 weeks pi did not differ significantly from that of healthy controls, although there was a tendency for both enzymes to decline in the pancreas of the infected turkeys. Healthy laying female turkeys demonstrated marked elevation in serum transferrin level and in acid and alkaline phosphatase activity, as compared with males of the same age. Serum alkaline phosphatase of turkey poults was markedly higher than that of adult turkeys.

The contribution of thermo-moxibustion to surgical treatment in transplanted mouse mammary carcinoma.

BALB/C mice bearing progressive transplanted mammary carcinoma were treated by surgical removal of the tumor mass either with or without thermo-moxibustion, applied to the GV 14-point-equivalent. A surgical removal of the tumor, 14 days post-inoculation resulted in 61% of deaths compared with 90.0% of deaths in the sham operated control. By the addition of the thermo-moxibustion therapy, the mortality rate, in the surgically treated group was reduced to 37.5%. Surgical removal of the tumor mass at day 17 post-inoculation resulted in 70% death while surgery supported by thermo-moxibustion protected the animals to the range of 40% death. Surgical removal of the tumor mass followed by local heterologous immuno-therapy was very effective when applied 14 days post-inoculation (12.5% death only). However, the same procedures applied at day 17 post-inoculation were not effective (80% death). Surgery and moxibustion together were more effective than surgery alone. Surgery and immunotherapy were very effective when applied from day 14 post-inoculation. Thermo-moxibustion as the sole treatment was effective when applied either before or very close to the tumor cell inoculation (35% of death and 33% of death respectively, as compared to 61.7% death in the control). Thermo-moxibustion applied before and after tumor cell inoculation was more effective (15% death rate). Thermo-moxibustion either alone or in conjunction with surgery is a contribution to the protection of BALB/C mice against the early and the late development of mouse mammary carcinoma.

Heterotransplantation of experimentally induced sheep lung adenomatosis into nude mice.

Male ICR nude mice inoculated subcutaneously with minced tissue obtained from pulmonary adenomatosis of sheep which was induced by RNA type-C retrovirus developed a transplant which contained cysts lined by a proliferative epithelium. These cysts showed several characteristics of the epithelial tumour cells of origin, such as high glycogen content and secretion of mucin. Microinvasion of epithelial cells from the adenomatous cysts into the stroma was observed. The stromal tissue demonstrated islets of cartilage. It was concluded that virus-induced pulmonary carcinoma of sheep can be transplanted and grown in nude mice.

The effect of tiamulin administered by different routes and at different ages to turkeys receiving monensin in their feed.

Healthy turkeys receiving 80 ppm monensin in their feed were injected at 26, 40 and 61 days of age with tiamulin at dosages of 12.5 and 25 mg/kg body weight. The aim of the study was to develop a regime for medicating with tiamulin turkeys receiving monensin in their feed, and which would circumvent the known toxicity created by the simultaneous administration of the two drugs. One injection of 12.5 mg/kg tiamulin up to the age of 61 days or 2 injections of 12.5 mg/kg tiamulin up to 40 days of age caused no mortality or adverse reaction.

The effect of high doses of methyl-CCNU on male and female fertility in SJL/J mice.

Single IP injection of high dose (LD 10) of methyl-CCNU administered to sexually mature male mice resulted in severe inhibition of spermatogenesis and reduction in testicular wet weight, without significant changes in plasma testosterone levels, and with hyperplasia of the interstitum, including Leydig cells, in the testis. These effects were temporary, spermatogenesis and testiculer weight recovered 50 days after treatment. Mating of the treated males with normal females demonstrated absolute sterility at 20 days and full recovery at 50 days after treatment. Administration of a single lethal dose (38 mg/kg, LD70) of methyl-CCNU to immature (25 day-old) male mice also caused severe but temporary inhibition of spermatogenesis, and mating of mice which survived the treatment and reached sexual maturity, with normal females, resulted in 80 to 100% pregnancies at 40 and 70 days after treatment, respectively. Three repeated injections of 30 mg/kg (at 10 day intervals) of methyl-CCNU to 15 day-old male mice resulted in inhibition of spermatogenesis without alteration in plasma testosterone. Ninety days after this treatment, only 40% of matings with normal females resulted in pregnancies. Female mice treated once with 20 mg/kg of methyl-CCNU, 3 days before or 7 days after mating with normal males, showed complete failure to complete pregnancy-resorption of the embryos was demonstrated in those female mice treated 7 days after mating. After a second mating trial with these two treated groups, performed 40 to 55 and 42 days after the first trial, respectively, 80 and 86% of the females delivered offspring. However, average litter size was reduced to 4 in the second group, as compared to 8 in untreated mice. There were no apparent birth defects in the offspring of methyl-CCNU treated male or female mice.

Effect of tiamulin administered by various methods and dosages on turkeys at different ages.

Healthy male turkeys not receiving monensin in their feed were treated with tiamulin by various methods and at different ages. Nine cycles of treatments were performed at the ages of 26, 40, 61, 89, 103, 117, 131, 145, and 160 days. Intramuscular of subcutaneous injections of 12.5 mg/kg tiamulin up to 145 days did not result in any signs of toxicity or impair growth rate. Administration at a dose of up to 25 mg/kg, using the drinking-water-deprivation technique, caused no effect in turkeys up to 160 days (marketing age).

Early detection of tracheal damage in chickens by scanning electron microscopy.

Fifteen-day-old chickens were inoculated with effective doses of cultured Mycoplasma gallisepticum and treated with the antibiotic tiamulin or tylosin. Scanning electron microscopy was used to detect early and late damages in the tracheal mucosa of the affected birds. Edematous changes of various degrees were demonstrated as early as day 4 postinoculation. Catarrhal changes were observed at day 10 postinoculation, and obstruction of the tracheal lumen by mucofibrinous plugs was observed at day 10 or later postinoculation. Scanning electron microscopy was useful in early detection of inflammatory changes in the trachea of apparently healthy chicks.
