Long-period accelerometer monitoring shows the role of physical activity in overweight and obesity.

CONTEXT: Physical activity (PA) plays an important role in obesity. A new accelerometer has been developed to assess total energy expenditure as well as PA. OBJECTIVE: To investigate the association of PA with overweight and obesity in Japanese men and women, a large cross-sectional study was performed using a single-axis accelerometer. DESIGN, SETTING AND PARTICIPANTS: Population-based cross-sectional study of Japanese 18-84 y of age. Height, body weight and PA were measured in 400 male and 388 female Japanese volunteers from 1999 to 2000. The outcome measurements were overweight and obesity, which are defined as a body mass index >/=25 kg/m(2). PA was measured for 1 to 4 weeks and was then categorized into three activity levels, which were defined as light, moderate and vigorous PA. RESULTS: Prevalence of overweight and obesity was 22.3%. Number of steps and time spent in moderate and vigorous PA per day were lower in overweight and obese individuals. No difference was found in time spent in light PA. Individuals who are in the 4th and 5th quintile of moderate and vigorous PA showed a significantly lower body mass index. When odd ratios (ORs) of overweight and obesity estimated by logistic regression were used as effect measures, overweight and obesity were negatively associated with vigorous PA (ORs=0.91). CONCLUSION: These results indicate that overweight and obese individuals have a lower step rate and are spending less time for moderate to vigorous PA. Participation in vigorous PA is an important predictor of overweight and obesity.

Electrophoretically separation of the synovial fluid proteins in rabbit temporomandibular arthritis induced by mechanical loading.

BACKGROUND: The concentration of protein in synovial fluid (SF) of temporomandibular joints (TMJs) with disorders tends to be increased. We investigated the protein composition of SF of rabbits in which arthritis of the TMJ was induced. METHOD: Arthritis was induced in six TMJs in six rabbits by exertion of a load for 4 weeks. Six non-loaded TMJs in six rabbits served as controls. The protein concentration and content in TMJ SF of the two groups were compared. RESULTS: The mean protein concentration was higher in the SF of the loaded group than in that of the non-loaded group (1824 microg/ml vs. 398 microg/ml, P = 0.002). Proteins with molecular weights of more than 95 kDa were abundant in the loaded group (P < 0.05). CONCLUSION: Temporomandibular arthritis induced by mechanical loading in rabbit is accompanied by an increase in the abundance of relatively high molecular weight proteins in SF.

Relationship of physical fitness to chewing in an 80-year-old population.

OBJECTIVE: Although tooth loss causes a decrease in masticatory ability, which may influence nutritional status, and impair an individual's general health including physical activity, little is known whether a decrease in chewing ability could result in deterioration in physical fitness in a very elderly population. Thus, the present study evaluated the relationship of chewing ability or teeth number with measures of physical fitness in a sample of 80-years-old in Japan. DESIGN: Cross-sectional survey. SUBJECTS AND METHODS: A total of 1282 people who were 80-years old and resided in the Fukuoka Prefecture were approached. A total of 697 individuals (277 male, 420 female) agreed to participate. RESULTS: Chewing food number and teeth number were related positively with physical fitness measurements of hand grip strength, leg extensor strength, leg extensor power, stepping rate, and one-leg standing time. However, the significant relationship between the number of teeth and physical ability disappeared after adjustment for various confounders, using multiple regression analysis or logistic regression analysis. On the other hand, the relationship of chewing ability with physical fitness measurements of leg extensor strength, one-leg standing time, or isokinetic leg extensor power remained significant even after adjustment for these confounders. CONCLUSION: There is a relationship between perceived chewing ability (number of foods considered chewable) and physical fitness in this 80-year-old population. Chewing ability may be an independent predictor of physical fitness, thus preventative dental care aimed at preserving chewing ability may be able to enhance activities of daily life and quality of life in very elderly individuals.

Expression of interleukin 6 in synovial tissues in patients with internal derangement of the temporomandibular joint.

Using an immunohistochemical technique, we examined synovial tissue from 46 temporomandibular joints (TMJ) with internal derangement in 44 patients. As controls, we examined synovial tissue specimens from 7 joints with habitual dislocation without pain. In synovial tissues from 21 of the 46 joints with internal derangement, interleukin 6 (IL-6) was expressed in the synovial lining cells and in the mononuclear cells infiltrating the periphery of the blood vessels. The density of IL-6-stained cells in specimens with internal derangement correlated significantly with the grade of joint effusion shown by magnetic resonance imaging (P=0.01, r=0.32).

Correlations of the expression of fibroblast growth factor-2, vascular endothelial growth factor, and their receptors with angiogenesis in synovial tissues from patients with internal derangement of the temporomandibular joint.

Synovitis in internal derangement of the temporomandibular joint (TMJ) is accompanied by the growth of new blood vessels. Fibroblast growth factor-2 (FGF-2) and vascular endothelial growth factor (VEGF) are well-characterized angiogenic factors. The objective of this study was to elucidate the correlation between the expression of FGF-2, VEGF, and their receptors-FGF receptor-1 (FGFR-1) and VEGF receptor-1 (Flt-1)-with microvessel density in synovial tissues of the TMJ. Using an immunohistochemical technique, we examined 47 joints (45 patients) with internal derangement. Individual microvessel density was evaluated by means of the CD34 antibody, a specific endothelial marker. The correlation between the percentage of immuno-positive cells and microvessel density was evaluated. In multiple logistic regression analysis, the correlation between the percentage of Flt-1-positive cells and microvessel density was significant [p = 0.005, odds ratio = 1.071, 95% confidence interval = 1.021-1.124]. These results suggest that the expression of the VEGF/Flt-1 system is involved in angiogenesis in inflamed synovial tissue in the TMJ.

The expression of fibroblast growth factor-2 and fibroblast growth factor receptor-1 in chondrocytes in synovial chondromatosis of the temporomandibular joint. report of two cases.

Synovial chondromatosis (SC) is a rare, benign condition characterized by the formation of metaplastic cartilaginous nodules. The expression of fibroblast growth factor-2 (FGF-2) and fibroblast growth factor receptor-1 (FGFR-1) in two cases of SC of the temporomandibular joint (TMJ) were immunohistochemically studied. The possible roles of FGF-2 and FGFR-1 in SC of the TMJ are discussed.

Cyclization of electron-deficient cyclopentadienone with 2-alkenyl and 2-alkynylamines via sequential pericyclic reaction pathway.

The pericyclic reactions of 2,5-bis(methoxycarbonyl)-3,4-diphenylcyclopentadienone (1a) with both allylic and propargylic amines have been investigated. The reaction proceeded via initial formation of the 1,4 adducts followed by the ene cyclization and/or sequential pericyclic reactions depending upon the structures of the amines. The reaction of 1a with diallylamine (2a) gave the tetracyclic compound (3a). On the other hand, the reaction of 1a with 2-propynylamine (2c) gave exclusively the bicyclic compound (5c). In the reactions with the secondary 2-propynylamines (2d,e), the tetracyclic compounds (3d,e) were exclusively formed. The reactions of 1a with alpha-branched primary 1,1-dialkyl-2-propynylamines (2f,g) gave mixtures of 3- and 5-type compounds. The tetracyclic compounds 3 were formed from the intramolecular [4+2]pi cycloadditions of the [1,5]-sigmatropic rearrangement products of the 1,4 adducts of 1a and 2, followed by the [1,5]-sigmatropic rearrangement of hydrogen and dehydrogenation. The bicyclic compounds 5 were derived from the [2pi+2pi+2sigma] reaction of the 1,4 adducts of 1a and 2. The one-pot multistage sequential pericyclic reactions were discussed on the basis of the X-ray crystallographic structures and the MO calculation data.

Assessment of lower-back muscle fatigue using electromyography, mechanomyography, and near-infrared spectroscopy.

We have investigated the etiology of lower-back muscle fatigue using simultaneous recordings of electromyography (EMG), mechanomyography (MMG), and near-infrared spectroscopy (NIRS) in an attempt to shed some light on the electrophysiological, mechanical, and metabolic characteristics, respectively. Eight male subjects performed isometric back extensions at an angle of 15 degrees with reference to the horizontal plane, for a period of 60 s. Surface EMG, MMG and NIRS signals were recorded simultaneously from the center of the erector spinae at the level of L3. NIRS was measured to determine the level of muscle blood volume (BV) and oxygenation (Oxy-Hb). The root mean square amplitude value (RMS) of the EMG signal was significantly increased at the initial phase of contraction and then fell significantly, while mean power frequency (MPF) of the EMG signal decreased significantly and progressively as a function of time. There were also significant initial increases in RMS-MMG that were followed by progressive decreases at the end of fatiguing contractions. MPF-MMG remained unchanged. Muscle BV and Oxy-Hb decreased dramatically at the onset of the contraction and then remained almost constant throughout the rest of the contraction. These results, obtained by simultaneous recordings of EMG, MMG, and NIRS, demonstrate that the restriction of blood flow due to high intramuscular mechanical pressure is one of the most important factors in muscle fatigue in the lower-back muscles. In addition, the simultaneous recording system described here can be used to obtain more reliable information regarding the mechanism(s) of lower-back muscle fatigue.

Comparison of thermogenic sympathetic response to food intake between obese and non-obese young women.

OBJECTIVE: Sympathetic nervous system abnormality in humans is still a matter of debate. The present study was designed to examine diet-induced autonomic nervous system activity and metabolic change in obese and non-obese young women. RESEARCH METHODS AND PROCEDURES: Sixteen age- and height-matched obese and non-obese young women participated in this study. Sympathovagal activities were assessed by means of our newly developed spectral analysis procedure of heart-rate variability during the resting condition and after mixed-food ingestion (480 kcal). Energy expenditure was also measured under these two conditions. RESULTS: There was no significant difference in any of the parameters of the heart-rate variability between the obese group and control group during the resting condition. In the control group, both absolute values (221.5 +/- 54.5 vs. 363.8 +/- 43.7 ms2, p < 0.05) and relative values (0.23 +/- 0.03 to 0.36 +/- 0.02, p < 0.05) of a very-low-frequency component and global sympathetic nervous system index (1.46 +/- 0.19 vs. 3.26 +/- 0.61, p < 0.05) were significantly increased after mixed-food ingestion compared with the values obtained after resting condition. However, no such sympathetic response was found in the obese group. Energy expenditure increased in the two groups after the meal, but the magnitude of the increase above the preprandial resting condition was significantly greater in the control group than in the obese group (11.2 +/- 2.3 vs. 6.7 +/- 0.8%, p < 0.05). DISCUSSION: Our data suggest that despite identical sympathovagal activities at the resting condition, obese young women may possess a reduced sympathetic response to physiological perturbation such as mixed food intake, which might be related to lowered capacity of thermogenesis and the state of obesity.

A theoretical study of neighboring-group participation in thione-to-thiol rearrangement of xanthates. molecular orbital calculation using a conductor-like screening model (COSMO) approach.

The transition structures of the thione-to-thiol rearrangement of O-(2-dimethylaminoethyl and 2-methylthio-) S-methyl xanthates in several solvents were located by semiempirical molecular orbital method (PM3) using the conductor-like screening model (COSMO) approach. Each transition state transforms into the ion-pair intermediate with a three-membered ring structure (aziridinium or thiiranium), indicating that the rearrangement proceeds through an ionic intermediate with the anchimeric assistance of the neighboring group. The intermediary structures in gas phase are also analyzed by ab initio and density functional theory calculations.

Molecular orbital studies on pericyclic reactions of cinnamyl xanthates in beta-cyclodextrin cavities.

The solid beta-cyclodextrin (beta-CyD) complex of O-cinnamyl S-methyl dithiocarbonates (xanthate, 1a), upon heating at 45 degrees C, underwent asymmetric [3,31-sigmatropic rearrangement to give the optically S-(1-phenylallyl) S-methyl dithiocarbonate (2a) with 60% ee. Heating the beta-CyD complex of 2a at 120 degrees C caused extrusion of COS to give cinnamyl methyl sulfide (3a) in high yield. The reaction behavior and role of beta-CyD are discussed based on molecular orbital calculation data.

Misexpression of Polycomb-group proteins in Xenopus alters anterior neural development and represses neural target genes.

In Drosophila, the Polycomb-group constitutes a set of structurally diverse proteins that act together to silence target genes. Many mammalian Polycomb-group proteins have also been identified and show functional similarities with their invertebrate counterparts. To begin to analyze the function of Polycomb-group proteins in Xenopus development, we have cloned a Xenopus homolog of Drosophila Polycomblike, XPcl1. XPcl1 mRNA is present both maternally and zygotically, with prominent zygotic expression in the anterior central nervous system. Misexpression of Pcl1 by RNA injection into embryos produces defects in the anterior central nervous system. The forebrain and midbrain contain excess neural tissue at the expense of the ventricle and include greatly thickened floor and roof plates. The eye fields are present but Rx2A, an eye-specific marker, is completely repressed. Overexpression of Pcl1 in Xenopus embryos alters two hindbrain markers, repressing En-2 and shifting it and Krox-20 in a posterior direction. Similar neural phenotypes and effects on the En-2 expression pattern were produced by overexpression of three other structurally unrelated Polycomb-group proteins: M33, XBmi-1, and mPh2. These observations indicate an important role for the Polycomb-group in regulating gene expression in the developing anterior central nervous system.

The muscle sound properties of different muscle fiber types during voluntary and electrically induced contractions.

Soundmyogram (SMG) and electromyogram signals were recorded simultaneously from the relatively fast medial gastrocnemius (MG) and slow soleus (SOL) during voluntary and electrically induced contractions. Using a spike-triggered averaging technique, the averaged elementary sound and corresponding MU spikes were also obtained from about 35 different MUs identified. The rms-SMG of MG increased as a function of force (P < 0.01). On the contrary, these values for SOL increased up to 60% MVC (P < 0.01), but decreased at 80% MVC. The relationship between the peak to peak amplitude of SMG and MU spike indicated significant positive correlations (r = 0.631 to approximately 0.657, P < 0.01). During electrical stimulation at 5 Hz, the SMG power spectral peak frequency (PF) was matched with stimulation frequency in both muscles. At higher stimulation frequencies, e.g., > 15 Hz, only in the MG was SMG-PF synchronized with stimulation frequency; the slow SOL did not show such synchronization. Our data suggest that the SMG frequency components might reflect active motor unit firing rates, and that the SMG amplitude depends upon mechanical properties of contraction, muscle fiber composition, and firing rate during voluntary and electrically induced contractions.

Vascular endothelial growth factor overproduced by tumour cells acts predominantly as a potent angiogenic factor contributing to malignant progression.

To elucidate the role of vascular endothelial growth factor (VEGF), an endothelial cell-specific mitogen, in tumour angiogenesis and malignant progression, an expression vector harboring human VEGF cDNA was stably transfected into three human cancer cell lines with poor VEGF productivity. Though their in vitro growth rate and intrinsic productivity of another angiogenic factor, basic fibroblast growth factor (bFGF), were not changed by transfection, those clones with higher VEGF production were endowed with tumorigenic and angiogenic potentials as follows: firstly, nontumorigenic, lung carcinoma QG90 cells having lower bFGF productivity acquired tumorigenicity as well as significant in vivo angiogenesis-inducing ability, secondly, tumorigenic colorectal carcinoma RPMI4788 cells having higher potency for bFGF production could form more vascularized solid tumour with faster growth rate and thirdly, oestrogen-dependent breast carcinoma MCF-7 cells, which did not produce detectable bFGF, acquired tumorigenicity even in the absence of oestrogen and the solid tumour growth rate was remarkably enhanced, accompanied with increased vascularization, in the presence of oestrogen. These results suggest that tumour progression closely depends on angiogenesis, and VEGF significantly contributes to malignant progression of a variety of tumour cells through its potent angiogenic activity, independent on the bFGF productivity of tumour cells.

[The uptake of fluoroquinolone agent drugs by cultured human conjunctival epithelial cells and cultured SIRC rabbit corneal cells].

PURPOSE: To investigate the uptake of drugs by cultured human conjunctival epithelial cells (HCEC) and cultured rabbit corneal cell lines (SIRC). METHOD: The drugs examined were ofloxacin (OFLX), lomefloxacin (LFLX), and norfloxacin (NFLX). The amount of drug uptake in the cultured cells that were exposed to the drugs was measured by high-performance liquid chromatography (HPLC). RESULT: The amount of LFLX, OFLX, and NFLX uptake in HCEC was 1.38 +/- 0.21 nmol/10(6) (mean +/- standard deviation) cells (5 min), 0.79 +/- 0.05 nmol/10(6) cells (5 min), and 0.56 +/- 0.03 nmol/10(6) cells (5 min), respectively. The amount of LFLX, OFLX, and NFLX uptake in SIRC was 1.08 +/- 0.18 nmol/10(6) cells (5 min), 0.58 +/- 0.17 nmol/10(6) cells (5 min), and 0.41 +/- 0.35 nmol/10(6) cells (5 min), respectively. The uptake levels of LFLX by both cultured cells were higher than those of OFLX and NFLX. CONCLUSION: This method may be useful as a screening method for intraocular drug dynamics studies.

[Peak oxygen uptake, ventilatory threshold and leg extension power in apparently healthy Japanese].

Peak oxygen uptake, ventilatory threshold (VT) and leg extension power were measured in 832 apparently healthy subjects (421 males, 411 females) aged 30-60 decades who performed medical examination at 4 health check up facilities. 1) Peak oxygen uptake was greater in male than female, and decreased with age in both sexes. Single regression formula on age were Y (ml/kg/min) = 46.6 - 0.36 x AGE, (r = 0.447, p < 0.001) in male and Y (ml/kg/min) = 35.3 - 0.23 x AGE, (r = 0.407, p < 0.001) in female. 2) VT was greater in male than female, and less decreased with age compared to maximal aerobic power. Single regression formula on age were Y (ml/kg/min) = 22.6 - 0.13 x AGE, (r = 0.371, p < 0.001) in male, Y (ml/kg/min) = 20.0 - 0.10 x AGE, (r = 0.301, p < 0.001) in female. 3) Leg extension power was greater in male than female, and decreased with age in both sexes. Single regression formula on age were Y (W/kg) = 31.9 - 0.27 x AGE, (r = 0.539, p < 0.001) in male and Y (W/kg) = 20.9 - 0.18 x AGE, (r = 0.510, p < 0.001) in female. Subjects in the present study showed height, weight and walk steps per day similar to those in National Nutrition Survey. Comparison of Peak oxygen uptake, ventilatory threshold and leg extensor power to previous studies revealed that these parameters were quite similar to the counterparts in the previous studies with less selection bias. It was suggested that subjects in this study were quite similar to normal Japanese population, and that Peak oxygen uptake, VT and leg extension power in the present study were of value for the evaluation of health related fitness in community and workplace.

1998 ISEK Congress Keynote Lecture: The use of electromyography in applied physiology. International Society of Electrophysiology and Kinesiology.

Electromyogram (EMG) analyses (surface, intramuscular and evoked potentials) in studies of muscle function have attracted increasing attention during recent years and have been applied to assess muscle endurance capacity, anaerobic and lactate thresholds, muscle biomechanics, motor learning, neuromuscular relaxation, optimal walking and pedalling speeds, muscle soreness, neuromuscular diseases, motor unit (MU) activities (MU recruitment and rate coding), and skeletal muscle fatigue. This paper deals with the use of EMG analyses employed in the area of applied physiology and is divided into three sections: surface EMG analyses; intramuscular EMG analyses; and evoked potential analyses.
