Beneficial effect of tripterine on systemic lupus erythematosus induced by active chromatin in BALB/c mice.

The aim of this study was to determine whether tripterine, isolated from Tripterygium wilfordii Hoog f. in China, had beneficial effects on experimental systemic lupus erythematosus induced by active chromatin in BALB/c mice. BALB/c mice were immunized with active chromatin isolated from concanavalin A-activated syngenetic spleno-lymphocytes on day 0. Tripterine 6 or 12 mg kg(-1) day(-1), or prednisone 5 mg kg(-1) day(-1) was given to BALB/c mice intragastrically from day 35 to day 50. Treatment with tripterine 12 mg kg(-1) day(-1) for 15 days protected renal from glomerular injury with a concomitant reduction of serum autoantibodies and total immunoglobulin G (IgG) also with a improvement of splenocyte proliferation stimulated with concanavalin A and lipopolysaccharide. The effects were associated with reduced interleukin-10 production and serum nitric oxide (NO) level but not interferon-gamma compared with vehicle-treated control group. Tripterine 6 mg kg(-1) day(-1) had no significant protective effect against glomerular injury. It inhibited autoantibodies and interleukin-10 production but had no effect on splenocyte proliferation, serum NO level, and interferon-gamma production. These findings suggested that tripterine had a beneficial effect on systemic lupus erythematosus induced by active chromatin in BALB/c mice.

[Experimental investigation of effects of adenovirus transduced tissue factor pathway inhibitor-2 gene on growth of laryngeal squamous carcinoma].

OBJECTIVE: To investigate the inhibitory effects of adenovirus transduced TFPI-2 gene on the growth of laryngeal squamous carcinoma. METHODS: Recombinant adenoviruses carrying human TFPI-2 gene were amplified and identified. The nude mouse model of laryngeal squamous carcinoma was established by intracutaneous injection of Hep-2 cells. Mice in the treated group were injected with recombinant adenoviruses with Ad-TFPI-2 (adenoviruses-TFPI-2) in peritumor tissue while mice in control group were injected with equivalent null plasmids. After treatment, the tumor weight and volume of tumor in each mouse were measured respectively. The morphological changes of tumor cells were observed using transmission electron microscope and proliferating cell nuclear antigen (PCNA) expression was examined using immunohistochemistry. RESULTS: The Ad-TFPI-2 virus titer was 2.8 x 10(12) PFU/ml after amplification. The average tumor weight and volume in Ad-TFPI-2 treated group were (1.20 +/- 0.34) cm3 and the volume (1.52 +/- 0.39) g, which were significantly lower than the tumor weight (2. 08 +/- 0.52) cm3 and (2.67 +/- 0.47) g in the control group (P < 0. 01). Apoptosis was observed in the tumors of Ad-TFPI-2 treated group. The PCNA index in Ad-TFPI-2 group was (54.9% +/- 12.4%), which was obviously lower than that (75.8% +/- 11.2%)in control group (P < 0.01). CONCLUSION: Peritumor injection of Ad-TFPI-2 can inhibit the growth of laryngeal squamous carcinoma in nude mouse model.

[Expression and distribution of Fas and Fas-L in the nasal polyps].

OBJECTIVES: To confirm the expression and distribution of Fas and Fas-L in the nasal polyps and to illustrate the role of the Fas/Fas-L system in the pathogenesis of human nasal polyps. METHODS: Investigating the transcripts of the Fas/Fas-L gene in 30 human nasal polyp tissues and 30 nasal turbinate mucosa specimens using reverse transcription-polymerase chain reaction. Localization of Fas/Fas-L was performed with immunohistochemistry. RESULTS: The transcripts of the Fas/Fas-L gene were detected at similar levels in both polyps and nasal mucosa. There was a significant overexpression of Fas-L protein on nasal polyps (epithelium: 25 +/- 21, glands: 19 +/- 14) compared to nasal mucosa (epithelium: 14 +/- 13, glands: 12 +/- 10), (t = 1.66, P < 0.01), while Fas was overexpressed on nasal mucosa (epithelium: 17 +/- 11, glands: 17 +/- 13) compared to nasal polyps (epithelium: 13 +/- 10, glands: 11 +/- 9), (t = 1.98, P < 0.01). Fas-L positive cells were localized on the epithelial layers of cystically dilated glands and the down-growing epithelium of nasal polyps. Fas positive cells were localized on the cilia of the epithelial of nasal mucosa and mainly on the infiltrative cells. CONCLUSION: Fas/Fas-L may play an important role on the pathogenesis of human nasal polyps, including cystic degeneration of submucosal glands, apoptosis and conferring of immune privilege to nasal polyp formation.

Expression of mRNA for membrane-type 1, 2, and 3 matrix metalloproteinases in human laryngeal cancer.

OBJECTIVE: To investigate correlation of expressions of membrane-type 1, 2, and 3 matrix metalloproteinases (MT1, MT2, and MT3-MMP) to the invasion and metastases in laryngeal cancer. METHODS: Reverse transcription-polymerase chain reaction (RT-PCR) was used to examine the mRNA level of MT1, MT2, and MT3-MMP in 24 patients with laryngeal cancer. The relationships of these three MT-MMP expressions to clinicopathology were analyzed by statistics. RESULTS: The expressions of MT1, MT2, and MT3-MMP were significantly higher in laryngeal cancer tissues than those in para-tumorous tissues (P < 0.01) and had a close relationship with invasive depth (P < 0.05). But no significantly different expressions of these three MT-MMPs were found in different primary location and different histological grade of laryngeal cancer (P > 0.05). The expression of MT1-MMP was obviously higher in patients with metastatic lymph nodes than that in patients without metastatic lymph nodes (P < 0.05). CONCLUSION: MT1, MT2, and MT3-MMP play an important role in the progression of laryngeal cancer, and MT1-MMP may serve as a reliable marker in estimating invasive and metastatic potency of laryngeal cancer. Suppressing expressions of MT1, MT2, and MT3-MMP early may inhibit the invasion and metastases of laryngeal cancer.

[Expression and DNA binding activity of NF-kappaB in mucosal tissue of chronic rhinosinusitis].

OBJECTIVE: To study the expression and activation of nuclear factor-kappa B (NF-kappaB) in mucosa of rhinosinusitis. METHODS: The expressions of NF-kappaB p50 and p65 in the mucosa from 18 patients and 10 norms were detected with the method of immunohistochemistry. The activation of DNA-binding proteins which was labelled with 32P-radiolabeled oligonucleotide probe for NF-kappaB was detected with electrophoretic mobility shift assays (EMSA) in mucosa. RESULTS: Plasma of epithelial and glandular cells displayed a strongly positive staining reaction to p65, and nucleus displayed a strongly positive staining reaction to p50 (7.8%-52.1%). There was significantly difference (chi2 = 22.917, P < 0.01). The DNA-binding proteins activity of 18 samples from patients with chronic rhinosinusitis (28.14 +/- 16.71) was stronger than that (9.28 +/- 2.84) in normal subjects (t = 4.56, P < 0.05). CONCLUSIONS: The expression and DNA-binding proteins activity of NF-kappaB was enhanced. It indicated that NF-kappaB was activated in mucosal inflammation of chronic rhinosinusitis.

[Improved laparoscopic Vechitti procedure for constructing a functioning vagina].

OBJECTIVE: To investigate the clinical effect of improved laparoscopic Vechitti procedure for constructing a functioning vagina. METHODS: By using the method of raising vestibular mucosa, 18 patients with congenital absence of vagina and uterus underwent surgery in our hospital. No cave was made between bladder and rectum. The procedure involved puncturing the vulvar vestibulum pit with an epidural paracentetic needle or specially-made needle into abdominal cavity through rectovesical interspace, two drag-lines was introduced through anterior abdominal wall, using the line to tie a clothes button of 2.0-2.5 cm diameter to the vulva, rasing the lines day by day, the vestibule go upward along with the button, then the vagina was formed. RESULTS: After the procedures, the artificial vagina of all 18 patients could hold a speculum and the mucosa appeared soft and smooth with normal lubrication. The vulvar tissues appeared uninjured and normal in all cases. The married patients were satisfactory to the intercourse. One case of vagino-rectal fistula was observed in a patient after she rode a bicycle with the vaginal mould. CONCLUSION: The improved laparoscopic Vechitti procedure for constructing a functioning vagina has less trauma than conventional operation and is easy to operate. Therefore, the new improved procedure is a preferred way in constructing vagina for treating those patients.
