Down-regulation of PEDF expression by ribozyme transgene in endothelial and lung cancer cells and its impact on angiogenesis in vitro.

Pigmental epithelial-derived factor (PEDF) is an important anti-angiogenesis factor. It has many different functions in ocular cells. The inhibition on angiogenesis by PEDF is associated with endothelial cell apoptosis. Until now, there have been few reports on the PEDF's role in cancer cell progression. This study examined the effect of a ribozyme transgene, designed to inhibit human PEDF expression, and its impact on in vitro lung cancer, endothelial cell growth and the angiogenesis forming process, in vitro. A transgene encoding ribozymes to specially target human PEDF was constructed using pEF6/v5-his vector. The human lung cancer cell line, A549, and endothelial cells, HECV, were transfected with PEDF ribozyme. MTT assay was used to analyse cell growth alternation after the PEDF gene was knocked out. An in vitro endothelial tubule formation assay was employed to analyse the microtubule forming change after the PEDF gene was knocked out in HECV cells. The PEDF message and protein were successfully removed with the PEDF ribozyme as shown by reverse transcription-PCR and Western blotting, respectively. The growth of both A549 and HECV cells were significantly accelerated when the PEDF gene was knocked out. The HECV cell ability to form microtubules increased following PEDF knockout. Furthermore, HECV significantly enhanced the ability to form microtubules when co-cultured with A549 cells whose PEDF expression was lost by way of the ribozyme transgene. Ribozyme transgenes targeting PEDF in lung cancer and endothelial cells can reduce the growth of these cells. Expression of PEDF in both cancer cells and endothelial cells are correlated with the angiogenic process' paracrine and autocrine pattern.

Characterization of the promoter of 1A6/DRIM, a novel cancer-related gene and identification of its transcriptional activator.

1A6/DRIM (Down-regulated in Metastasis) has been reported to express at a high level in the gastric cancer tissues and the premalignant lesions implicating the involvement of 1A6/DRIM in cell transformation. Although the information regarding the putative functions and distribution of the 1A6/DRIM in different tissues and cell lines has been increasing recently, its promoter and promoter-regulating factors remain unknown. In this study, the transcription initiation site of 1A6/DRIM was confirmed to be located at 147 bp upstream of the ATG codon using the primer extension analysis. The minimal promoter region of the 1A6/DRIM is located between -47 and +42 of the transcription initiation site measured by luciferase reporter assays using a set of deletion constructs. In addition, an E-box is shown to be an essential element for transcriptional regulation of 1A6/DRIM demonstrated by luciferase assay with different deletion and mutation constructs. Finally, a transcription factor, upstream stimulatory factor 2 (USF2) was found to be an activator of the 1A6/DRIM through binding to the E-box demonstrated by luciferase reporter assay, electrophoretic mobility shift assay, and chromatin immunoprecipitation (ChIP) assay. The structural analysis of the 1A6/DRIM promoter and the identification of its potential regulatory effecter may help us to understand its biological functions in regulating cancer development.

[Relationship between polymorphism of IRF-3 gene codon 427 and esophageal cancer in Anyang population of China].

OBJECTIVE: To investigate the association between the single nucleotide polymorphisms (SNPs) of interferon regulatory factor 3 (IRF-3) gene and the susceptibility of esophageal cancer in Anyang area, Henan Province. METHODS: The genomic DNAs of 152 esophageal cancer patients and 191 health controls were extracted from the peripheral blood leukocytes. Three fragments covering codons 96, 194, 377, and 427 of the IRF-3 gene were amplified by polymerase chain reaction (PCR). The SNPs were revealed by DNA sequencing and the genotype of every sample was determined accordingly. The frequency distribution of the SNPs was analyzed by chi(2) test. RESULTS: There were no IRF-3 gene SNPs at codons 96, 194, or 377 in Anyang participants. There was significant difference in the SNPs at codon 427 between the healthy controls and esophageal cancer patients in Anyang area OR=2.38 (95%CI=1.15-4.95,P<0.05). CONCLUSION: The SNPs at codon 427 of the IRF-3 gene may relate to the susceptibility of esophageal cancer. The risk of esophageal cancer in participants with C allele is 2.38-fold compared to those with G allele.

[Preparation of a polyclonal antibody against N-terminal of 1A6/DRIM with MAPs method].

OBJECTIVE: To obtain the antibody against N-terminal of 1A6/DRIM, and thereafter get the profile of 1A6/DRIM expression in different cell lines. METHODS: The N-terminal of 1A6/DRIM (aa 577-714) was cloned into pGEX-4T-3. Multiple antigenic peptides MAPs (aa638-661) was synthesized as the antigen with Fmoc/PyBOP method. Rabbits were immunized by injecting the MAPs and the immunized sera were analyzed with ELISA and Western Blot. The Western Blot and immunofluorescence were performed to analyze the expressing profiles of the 1A6/DRIM in different tumor cell lines. RESULTS: The antibody specifically recognized the full length of 1A6/DRIM as a 310 kDa band, which was also recognized by C-terminal monoclonal antibody shown by Western Blot. 1A6/DRIM is expressed in multiple tumor cell lines and mainly located in the nuclei. CONCLUSION: Preparation of the antibody with MAPs is a useful technique when the fusion proteins can not be induced in E. coli. The antibody we got via MAPs has supplied a good tool for further studies on the functions of the novel gene 1A6/DRIM.

p53 polymorphism and p21WAF1/CIP1 haplotype in the intestinal gastric cancer and the precancerous lesions.

The development of intestinal gastric carcinoma involves several precancerous stages. The environmental factor plays an important role in gastric carcinogenesis, while the host's genetic makeup may influence the susceptibility to cancer. In this study we investigated correlations of the p53 variations at codon 72 and p21(WAF1/CIP1) haplotype with the risk of intestinal gastric carcinoma. Forty-eight intestinal gastric carcinoma cases (GC), 96 chronic atrophic gastritis (CAG), 96 intestinal metaplasia (IM) and 96 dysplasia (DYS) controls were enrolled in this study. The p53 codon 72 proline allele carriers were found to be more susceptible to progress to GC than to IM (OR = 2.22, 95%CI = 1.05-4.70, P = 0.038). Patients carrying homozygous p21(WAF1/CIP1) haplotype A, which contains the serine at codon 31, the cytidine at the 16th base of the second intron, and the cytidine at the 70th base of the exon 3 were more prone to develop GC than to reach the IM or DYS stage (IM versus GC, OR = 3.35, 95%CI = 1.11-10.15; DYS versus GC, OR = 3.27, 95%CI = 1.09-9.80, P = 0.035). The combination of p53 codon 72 variation with the p21(WAF1/CIP1) haplotype further distinguished the risk of GC from IM precancerous lesion (OR = 9.31, 95% CI = 1.77-48.85, P = 0.08). These results suggest that p53 and/or p21(WAF1/CIP1) genotype may influence the progression during gastric tumorigenesis.

Human papillomavirus 16 E6 oncoprotein interferences with insulin signaling pathway by binding to tuberin.

Tuberous sclerosis complex (TSC) is a genetic disorder caused by mutations in either TSC1 or TSC2 tumor suppressor gene. TSC1 and TSC2 products, Harmatin and Tuberin, form the functional complex to serve as the negative regulator for insulin-induced phosphorylation of S6 kinase and elF4E-binding protein 1. High-risk human papillomavirus (HPV) infection is the necessary cause for cervical cancer. E6 oncoprotein encoded by HPV plays a pivotal role in carcinogenesis by interference with the host intracellular protein functions. In this study, we show that HPV16 E6 interacts with tumor suppressor gene TSC2 product, Tuberin, and results in the phosphorylation of S6 kinase and S6 even in the absence of insulin. The overexpression of Tuberin overcomes the effect of E6 on S6 kinase phosphorylation. Binding with HPV16 E6 causes the proteasome-mediated degradation of Tuberin. A DILG motif and an ELVG motif located in the carboxyl-terminal of Tuberin are required for E6 binding. In addition, the Tuberin interaction region in E6 has been mapped in the amino-terminal portion of HPV16 E6, which is different from the binding domain with p53. These results provide a possible link between E6-induced oncogenesis and the insulin-stimulated cell proliferation signaling pathway.

An assessment of World Health Organization criteria for severe acute respiratory syndrome in patients with cancer.

BACKGROUND: The differential diagnosis of severe acute respiratory syndrome (SARS) in patients with cancer can be challenging. Although diagnostic criteria for SARS have been issued by the World Health Organization (WHO), simple adoption of the established criteria may lead to overdiagnosis in patients with cancer or to an increase in the risk of spreading SARS within cancer hospitals. METHODS: The authors report their experience with the exclusion and quarantine of patients with cancer during the peak of the SARS epidemic in Beijing, China. The patients included 4 males and 7 females with a median age of 66 years (range, 39-73 years). RESULTS: All 11 patients met the WHO diagnostic criteria for probable SARS. Among those 11 patients with probable SARS, only 1 had confirmed SARS; for the other 10 patients, the possibility of SARS infection was ruled out. CONCLUSIONS: Special attention must be paid to patients with cancer who have symptoms similar to those seen in SARS. Although the WHO diagnostic criteria for SARS should be widely accepted, they are not definite or practical in certain populations.

5-aza-2'-deoxycytidine activates the p53/p21Waf1/Cip1 pathway to inhibit cell proliferation.

In addition to its demethylating function, 5-aza-2'-deoxycytidine (5-aza-CdR) also plays an important role in inducing cell cycle arrest, differentiation, and cell death. However, the mechanism by which 5-aza-CdR induces antineoplastic activity is not clear. In this study, we found that 5-aza-CdR at limited concentrations (0.01-5 microm) induces inhibition of cell proliferation as well as increased p53/p21(Waf1/Cip1) expression in A549 cells (wild-type p53) but not in H1299 (p53-null) and H719 cells (p53 mutant). The p53-dependent p21(Waf1/Cip1) expression induced by 5-aza-CdR was not seen in A549 cells transfected with the wild-type human papilloma virus type-16 E6 gene that induces p53 degradation. Furthermore, deletion analysis and site-directed mutagenesis of the p21 promoter reveals that 5-aza-CdR induces p21(Waf1/Cip1) expression through two p53 binding sites in the p21 promoter. Finally, 5-aza-CdR-induced p21(Waf1/Cip1) expression was dependent on DNA damage but not on DNA demethylation as demonstrated by comet assay and bisulfite sequencing, respectively. Our data provide useful clues for judging the therapeutic efficacy of 5-aza-CdR in the treatment of human cancer cells.

Aberration of X chromosome in liver neoplasm detected by fluorescence in situ hybridization.

BACKGROUND: A diverse range of cytogenetic alterations of autosomal chromosomes has been reported to date. However, few studies have addressed the abnormalities of X chromosome in hepatocellular carcinoma (HCC) except sporadic reports on the deletion of band F1 in X chromosome, and the clonal analysis of methylation pattern of the X chromosome-linked human androgen receptor gene. Identification of specific X chromosome alterations during the course of neoplastic development would be essential to defining the genetic basis of HCC. Therefore, we studied the regularity of aberration of X chromosome in liver cancer. METHODS: Hepatocarcinoma cellular lines and tumor tissues were detected respectively through DNA probes of X chromosome after fluorescence in situ hybridization (FISH). RESULTS: Increased copies of X chromosome were observed in all samples, and four signals of hybridization were of the major type. CONCLUSIONS: Increased copy number of X chromosome frequently occur in liver cancer. The relationship between copy number of X chromosome and liver cancer genesis needs further investigation. This study is the first of its kind determining the copy number of X chromosome in liver cancer by using FISH.

[Studies of the relationship of HLA polymorphisms and the infection of H. pylori in the population of Linqu in Shandong Province].

In order to analyze the relationship of HLA polymorphisms and the infection of H. pylori in the population of Linqu County in Shandong Province, polymerase chain reaction with sequence-specific primers(PCR-SSP) was used to determine the alleles of HLA type I and II in 90 Hp-positive persons and 49 Hp-negative controls. The results showed that among the 68 alleles of HLA type I ,4 alleles were found significantly different between Hp-positive and Hp-negative population, while no significant difference was found among the 22 alleles of HLA type UI. Hp-positive persons had a lower allele frequency of A* 02 (OR=0.56, 95% CI=0.33 approximately 0. 94; P=0.029), B* 48 (OR=0.15, 95% CI=0.03 approximately 0.72; P=0. 007), CW* 08 (OR=0. 32, 95% CI=0.15 approximately 0.69; P=0. 003) and a higher allele frequency of CW* 15 (OR=5.11, 95% CI=0.63 approximately 40.90; P=0. 024) compared with Hp-negative controls. Our results indicated that the polymorphisms of HLA type I is involved in the genetic susceptibility of Hp infection in Linqu County, while the polymorphisms of HLA type I may have no relationship with the genetic susceptibility of Hp infection. It was shown that among the alleles of HLA type I , CW* 15 might be a susceptible gene of Hp infection while A * 02 ,B * 48 and CW* 08 might be protective genes.

[Detection of human papillomavirus type 16 E6 mRNA in carcinomas of upper digestive tract].

OBJECTIVE: To investigate the relationship between human papillomavirus (HPV) infection and the development of upper digestive tract carcinomas. METHODS: In situ hybridization (ISH) technique was used to detect the expression of HPV16 E6 mRNA in 40 specimens of esophageal squamous cell carcinoma, 74 specimens of gastric cardiac adenocarcinoma, 40 specimens of adenocarcinoma of gastric corpus, 62 specimens of adenocarcinoma of gastric antrum, all resected during operation, and 58 specimens of normal tissues of gastric mucosa, obtained by endoscopy. RESULTS: The positive rate of HPV16 E6 mRNA was 70% (28/40) in esophageal squamous cell carcinomas, 67.6% (50/74) in gastric cardiac adenocarcinomas, 47.5% (19/40) in adenocarcinoma of gastric corpus, 35.5% (22/62) in adenocarcinoma of gastric antrum, and 20% (10/50) in normal tissues of gastric mucosa respectively, without a significant difference between esophageal squamous cell carcinoma and gastric cardiac adenocarcinoma (P = 0.955). The positive rate in gastric adenocarcinoma was significantly lower than that in gastric cardiac adenocarcinoma (P < 0.05) or in esophageal squamous cell carcinomas (P < 0.01). The positive rate in normal tissue of gastric mucosa was significantly lower than those in gastric adenocarcinoma (P < 0.05) in gastric cardiac adenocarcinoma (P < 0.01), and in esophageal squamous cell carcinoma (P < 0.01). The positive rate in gastric cardiac carcinomas was negatively correlated with TNM stage (P < 0.05). However, there was no correlation between HPV16 and gender, age, tumor size, depth of penetration, differentiation, lymph node metastasis, vascular invasion and prognosis of these patients with gastric cardiac carcinoma (all P > 0.05). CONCLUSION: HPV infects not only squamous epithelium, but also glandular epithelium. HPV16 may be an etiologic factor in the development of upper digestive tract carcinomas, especially esophageal and cardiac carcinomas.

Detection of human papillomavirus in Chinese esophageal squamous cell carcinoma and its adjacent normal epithelium.

AIM: To investigate the putative role of human papillomavirus (HPV) infection in the carcinogenesis of esophageal squamous cell carcinoma in China. METHODS: Twenty-three esophageal squamous cell carcinoma samples and the distal normal epithelium from Shanxi Province, and 25 more esophageal squamous cell carcinoma samples from Anyang city, two areas with a high incidence of esophageal cancer in China, were detected for the existence of HPV-16 DNA by PCR, mRNA in situ hybridization (ISH) and immunohistochemistry (IHC) targeting HPV-16 E6 gene. RESULTS: There were approximately 64 % (31/48) patients having HPV-16 DNA in tumor samples, among them nearly two-thirds (19/31) samples were detected with mRNA expression of HPV-16 E6. However, in the normal esophageal epithelium from cancer patients, the DNA and mRNA of HPV-16 were found with much less rate: 34.7 % (8/23) and 26.1 % (6/23) respectively. In addition, at protein level detected by IHC assay, 27.1 % (13/48) tumor samples had virus oncoprotein E6 expression, while only one case of normal epithelium was found positive. CONCLUSION: HPV infection, especially type 16, should be considered as a risk factor for esophageal malignancies in China.

[Analysis of the mutations of BRCA1 in 9 familiar breast cancer patients].

OBJECTIVE: To analyze the mutations of BRCA1 in 9 Chinese familiar breast cancer patients. METHODS: Peripheral blood samples were obtained from 9 patients enrolled from 9 breast cancer families, one normal control, 32 sporadic breast cancer patients and 33 normal donors. DNA extracted from lymphocytes was amplified by polymerase chain reaction (PCR). The 22 exons and partial introns of BRCA1 were screened by PCR-denaturing high performance liquid chromatography (PCR-DHPLC) and confirmed by direct sequencing. RESULTS: Among these 9 familiar breast cancer patients, a deleterious mutation was detected in one case in exon 11 (3870delTGTC) which was a 4-base deletion and caused a frameshift in turn. One novel and unique amino acid substitution (E867R) was detected in one case. Eight patients were detected to have a known variation in intron 18 (IVS18 + 65G-->A), and the ratio of this variation detected was 88.9% (8/9). The ratio of this variation was 37.5% (12/32) in sporadic breast cancer patients or 33.3% (11/33) in normal control. This variation was found to be accompanied all the time with a known missense variation in exon 11 (P871L) and a polymorphism in intron 9 (IVS8-57delT). Those three variants were also detected in homozygous in one case, which implies the linkage of the 3 sites. The linkage had not been reported. Two patients had been found with a known polymorphism in exon 13 (S1436S). Another known polymorphism was found in one case (L771L). In addition, intronic variants (IVS2 + 48C-->T, IVS2 + 133C-->T, IVS12 + 112C-->A) were detected. CONCLUSION: The mutations of BRCA1 in Chinese familiar breast cancer patients are different from the hot spots reported in Caucasian and Jewish. It is important that further study be conducted to seek for specific mutations of this gene or other possible relevant genes in Chinese familiar breast cancer patients.

[Expression of Fas, FasL and IFN-gamma in gastric cancer].

OBJECTIVE: To study the expression of Fas, Fas ligand (FasL) and IFN-gamma in gastric cancer and its possible significance. METHODS: Fifty-eight gastric paraffin wax embedded cancer tissues and fifty-three normal tissues adjacent gastric cancer were tested for the expression of Fas and FasL protein by immunohistochemistry and IFN-gamma mRNA by in situ hubridisation respectively. RESULTS: The positive rate of Fas in cancer cells of gastric cancer tissues was significantly lower than that in gastric epithelial cells of the tissues adjacent cancer(19.0% and 64.2%, respectively; chi 2 = 23.46, P = 0.00). The positive rate of FasL in cancer cells of gastric cancer tissues was significantly higher than that in gastric epithelial cells of the tissues adjacent cancer(63.8% and 45.3%, respectively; chi 2 = 3.83, P = 0.05). The positive rate of IFN-gamma in cancer cells of gastric cancer tissues was significantly lower than that in gastric epithelial cells of the tissues adjacent cancer(0.0% and 49.1%, respectively; chi 2 = 37.16, P = 0.00). CONCLUSION: The Fas-FasL system was unbalanced, and the expression of IFN-gamma was low in gastric cancer cells in this study. These may be related to the carcinogenesis of gastric epithelial cells and might be responsible for the immune escape of these cells.

[Analysis of Lewis gene polymorphism in high and low incidence area of gastric cancer in Shandong province].

To explore the cause leading to the difference in incidence of gastric cancer between Linqu and Cangshan populations, Shandong Province, and to provide evidence for the possible mechanism of high incidence of gastric cancer in Linqu County, the distribution of T59G mutation in Lewis gene was screened between Linqu and Cangshan populations by PCR-sequencing.The frequency of individuals with T59G mutation was 34.5% in Linqu population and 31.6% in Cangshan population, respectively, with no significant difference, P>0.05, and OR is 1.14 (95% CI,0.59-2.19). This suggests that Linqu and Cangshan populations may share the same genetic background.T59G mutation of Lewis gene could not be used as a genetic marker for Linqu and Cangshan populations and is not relevant to the difference in incidence of gastric cancer between them.

[Analysis of polymorphisms on Lewis blood group antigen related SE gene in the populations in Shandong with high-risk of gastric cancer and Beijing].

To study the relation between host genetic background and the susceptibility to H.pylori associated gastric cancer, PCR-sequencing and PCR-RFLP were used to screen SECRETOR gene polymorphisms in 142 subjects including 69 cancer patients and 73 non-cancer individuals from high-risk area of gastric cancer in Shandong and 93 control individuals from Beijing. Results showed that the difference in se(w)/se(w) distribution between non-cancer individuals and Beijing population was significant (P<0.01,OR is 3.06,95% CI,1.28-7.30), but that between cancer patients and non-cancer individuals was not with significance. SE gene polymorphism was not relevant to H.pylori infection. We concluded that Shandong population from high-risk area of gastric cancer shared a high distribution of se(w)/se(w) genotype, which could be considered as one of the genetic markers.
