[A colorimetric method of HEC toxin hemolysis test for diagnosis of paroxysmal nocturnal hemoglobinuria].

The purpose of the study is to establish a colorimetric method of HEC toxin hemolysis test for diagnosis of paroxysmal nocturnal hemoglobinuria (PNH). RBCs from normal persons and patients with PNH and non-PNH anemia were treated with HEC toxin secreted by Aeromonas hydrophila J-1 strain and the absorbance at 630 nm was measured to quantitate the extent of hemolysis. The results demonstrated that the RBCs from PNH patients showed resistance to the toxin hemolysis, which was in accord with the percentages of CD59(-) cells, while the RBCs from normal persons and non-PNH anemic patients were nearly totally lysed. It is concluded that the method can be considered as a simple, specific and reliable method for the diagnosis of PNH.

[KCNQ4 gene mutations affected a pedigree with autosomal dominant hereditary hearing loss].

OBJECTIVE: To investigate if the KCNQ4 gene contributes to a Chinese non-syndromic hearing loss pedigree and to detect the gene mutations in the pedigree using candidate approach. METHODS: PCR-SSCP and clone sequencing were performed to identify the mutations and polymorphism in PCR products of KCNQ4 coding sequence in the six-generations pedigree of autosomal dominant hereditary hearing loss. RESULTS: Mutations and polymorphism detection were performed on the KCNQ4 coding sequence in 36 family members of the pedigree. A molecular polymorphism marker located in the exon2 and exon3 intron sequence, which resulted from a copy variation of 47 base pairs insertion or deletion, was found in KCNQ4 sequence. CONCLUSION: A new molecular polymorphism marker with different genotypes was proved to locate at the intron sequence between at exon2 and exon3. The correlation between genotype and phenotype was analyzed. Deaf individuals were accompanied by the increase of the intron copies in the family. These findings suggest that the changes of the copies of intron between exon2 and exon3 of KCNQ4 might be a specific marker for the hearing loss of the pedigree.

Liver transplantation in patients with fulminant hepatitis B: experience in Canton, China.

OBJECTIVE: To investigate the effect of orthotopic liver transplantation on fulminant hepatitis B and the preventive efficiency of lamivudine on recurrence of hepatitis B in China. PATIENTS AND METHODS: Ten patients with fulminant hepatitis B received orthotopic liver transplantation under veno-venous bypass. All patients had preoperatively serious jaundice, ascites and coagulopathy, and of whom 7 with encephalopathy, 2 with acute renal failure, and 1 with gastro-hemorrhage. RUSULT: Seven of the 10 patients have survived for 3 approximately 18 months, but 3 died of multi-organ failure or recurrence of fulminant hepatitis B. Seven survivors took lamivudine and 6 of them have survived for 3 approximately 18 months without the signs of recurrence of hepatitis B. CONCLUSION: Orthotopic liver transplantation is an effective therapy for fulminant hepatitis B, and lamivudine may prevent recurrence of hepatitis B after transplantation.

[GVHD Following Autologous Peripheral Blood Stem Cell Trasplantation Reduced Malignancy Relapse]

Administration of the immunosuppressive drug cyclosporine (CSA) after autologous peripheral blood stem cell transplantation (APBSCT) induces a systemic auto-immune syndrome resembling graft-versus-host disease (GVHD), this syndrome termed autologous GVHD has significant antitumor activity, it can reduce the incidence of tumor relapse after APBSCT. The antitumor effect of this auto-aggression syndrome can be enhanced by the administration of gamma-interferon (gamma-IFN). Five consecutive patients who received APBSCT received therapy inducing autologous GVHD. Intravenous administration of CSA [1 mg/(kg.d) for 28 days] was begin on the day of transplantation. gamma-interferon (0.025 mg/m(2) qod) was administered sub-cutaneously from days 7 throngh 28 after transplatation. Results showed that four of five occured autologous GVHD-skin demage, five in control didn't occur autologous GVHD. The relapse rate of the treated cases was 20% (1/5) versus 60% (3/5) of the control, and the median survival time of the treated cases was 20 (4 - 30) months versus 10 (2 - 20) months of the control. The data indicates that autologous GVHD results in low relapse rate of the patients rececving APBSCT.

A Promoter Overlapping with that of nodF is Responsible for a New RNA Molecule Transcription in Rhizobium leguminosarum.

The expression of nod genes of rhizobia is controlled at the transcriptional level in a complicated way. In this work, a new promoter, which is responsible for an RNA molecule transcription in the opposite direction to that of nodF, was identified in Rhizobium leguminosarum. This new promoter was characterized to overlap with that of nodF, and the size of px(2), the RNA molecule initiated from this promoter was determined to be of approximately 0.72 kb in length. The study on px2 showed that its transcription depended on the Escherichia coli sigma(70)- like factor of Rhizobium leguminosarum, and that the in vitro transcription of px(2) was inhibited by Px, a newly identified regulatory protein in our lab. The involvement of px(2) in the regulation of nod gene expression is suggested.

How big are educational and racial fertility differentials in the U.S.?

Using pooled data from the 1980, 1985, 1990 and 1995 CPS and 1988 and 1995 NSFG surveys, we show that shifts in fertility timing have occurred disproportionately for the more educated and for whites (compared to the less educated and to African Americans). Such timing shifts imply that the underlying period quantum of fertility is considerably higher for college-educated women and for whites than suggested by the standard total fertility rate. Applying the Bongaarts-Feeney model (1998), we decompose observed racial and educational differences in age-order-specific fertility rates and TFR into tempo and quantum components. We find that a modest part of educational differences and a substantial part of racial difference in period fertility can be attributed to differential changes in tempo. Analysis by race and education shows a clear interaction: higher fertility among African Americans is confined to the less educated.

Centrosome impairments and consequent cytokinesis defects are possible mechanisms of taxane drugs.

Taxol and taxotere are two of the most promising anticancer drugs. To determine the mechanisms responsible for cell death after exposure to low doses of taxane, PC3 cells were treated with taxol and taxotere, and observed with immunofluoroscence microscopy. Pericentriolar material dissociation and blockage of normal centrosome separation were found to result in two different abnormal spindle types; multipolar and monopolar spindles, respectively. The majority of abnormal spindles induced by taxol were monopolar spindles, whereas taxotere mostly induced abnormal multipolar spindles. Consequently, monopolar spindle mitosis resulted in cleavage failure, while multipolar spindle mitosis led to the formation of both cleavage failure and multipolar cell division. Multinucleation characterized interphase cells which had undergone cytokinesis defects. These cells subsequently became giant multinucleated cells after several rounds of cell cycle with sustained cleavage failure, and were gradually eliminated through cell death.
