Decoding age-stratified clinical and molecular heterogeneity in male breast cancer through multiomic profiling.

OBJECTIVE: Age-associated molecular heterogeneity is well described in female breast cancer but remains insufficiently characterized in male breast cancer (MBC). We profiled age-stratified clinical and molecular differences between younger (&#x2264;55 years) male breast cancer (YMBC) and older (>55 years) male breast cancer (OMBC). METHODS: We retrospectively analyzed 347 patients with MBC diagnosed at Fudan University Shanghai Cancer Center by integrating clinicopathological data, RNA sequencing, and whole-exome sequencing (WES). Survival, differential expression, and mutational signature analyses were performed. Tumor microenvironment features were inferred using xCell and ESTIMATE, and weighted gene co-expression network analysis (WGCNA) was conducted to identify age-associated co-expression modules. Candidate therapeutics were prioritized using the Genomics of Drug Sensitivity in Cancer (GDSC) resource and evaluated using patient-derived organoids (PDOs). RESULTS: Compared with OMBC, YMBC more frequently had human epidermal growth factor receptor 2 (HER2)-positive status (14.91% vs. 4.02%) and triple-negative tumors (4.92% vs. 1.78%), and had worse 5-year recurrence-free survival (hazard ratio=2.19, P=0.018). Transcriptomic analyses indicated enrichment of neural-related programs and reduced immune-related signaling in YMBC, and xCell/ESTIMATE supported lower immune infiltration. Consistently, WGCNA identified age-associated modules linking neural-related programs with reduced immune infiltration. Immunohistochemistry supported increased perineural invasion and lower CD8+ T cell infiltration in YMBC. GDSC-guided prioritization with PDO testing nominated sepantronium bromide (YM155) as a candidate vulnerability in YMBC. WES showed a higher NBPF10 mutation frequency in YMBC (54.5% vs. 14.3%, P<0.05). CONCLUSIONS: Integrated multi-omics profiling revealed age-stratified clinical and molecular heterogeneity in MBC. YMBC patients demonstrated inferior recurrence-free survival, neural signaling enrichment, an immune-cold microenvironment, and enriched NBPF10 mutations. These findings support age as a meaningful stratification variable in MBC risk assessment and treatment planning, and highlight the need for caution when considering treatment de-escalation in younger patients, while nominating YM155 as a candidate agent for prospective evaluation.

[Study on the extracting of ursolic acid by uniform design].

Uniform design method was adopted to research the extraction of ursolic acid. The ethanol concentration, the extraction time, the liquid to solid ratio and the extraction times were researched by U7 (7(4)). The optimal conditions were achieved by optimization treatment and confirmed by experiment:the ethanol concentration being 75%, liquid to solid ratio being 7, extraction time 60 minutes, extracted two times.

[Evapotranspiration of natural Quercus litaotungensis and Tilia paucicostata secondary stands in Liupan Mountains of Ningxia].

With heat dissipation probe technique and combined with microlysimeter and hydrological methods, this paper studied the evapotranspiration of secondary Quercus liaotungensis and Tilia paucicostata stands, and its relationship to forest structure from August to September 2004. The results indicated that the stem sap flux density (SFD) of Quercus liaotungensis and Tilia paucicostata changed regularly from day to night in later growth season. In relatively still period (nighttime), the SFD kept low values continuously, usually below 0.05 microl x cm(-2). min(-1), while in active period (daytime), it increased quickly, usually below 0.25 microl x cm(-2) x min(-1). The daily transpiration of whole-tree was estimated based on the calculation of daily cumulative SFD, which showed that there existed a significant difference between two tree species. The daily transpiration of Quercus liaotungensis reached 5.31 and 2.48 L x d(-1) in sunny- and cloudy-days, 2.35 and 3.75 folds as that of Tilia paucicostata, respectively. There was no significant difference in daily leaf transpiration rate between these two shrub species. During measurement periods, the average daily stand evapotranspiration was 1.45 mm x d(-1), including transpiration (0.72 mm x d(-1)), soil evaporation (0.19 mm x d(-1)), and canopy interception (0.54 mm x d(-1)), which accounted for 49.6%, 13.3% and 37.1% of the total evapotranspiration, respectively. The results showed that the difference of whole-tree transpiration between tree species contributed significantly to the difference of tree canopy transpiration, while the difference of shrub canopy transpiration was on account of the leaf amount in the canopy. The effects of vertical layers in the stand on total stand evapotranspiration also differed, with a percentage of 65.8%, 20.9% and 13.3% for tree canopy, shrub canopy and floor, respectively, which meant that the transpiration and interception of tree canopy contributed most to the total evpotranspiration, followed by shrub transpiration, soil evaporation, and grass transpiration.

[Rainffall interception model of forest canopy: a preliminary study].

Canopy interception is an important hydrological process in forest ecosystem, and its modelling is of significance to understand and estimate the rainfall interception by the canopy. In this paper, a canopy rainfall interception model was established by dividing a rain incident into a set of short period, calculating the rainfall distribution intercepted by the canopy, and educing the process of the rain incident. This model considered the effects of the dryness of canopy and trunk on the evaporation from wet canopy and trunk during one rain incident, and introduced two factors, leaf area index (LAI) and surface area of trunk per unit area of ground (SAI), when computing the evaporation. The application of the model to simulate the rainfall interception process in a Larix principis-rupprechtii plantation in Guyuan, south Ningxia Hui autonomous region of China showed that the simulated and measured throughfall were identical, and the absolute deviation between simulated and measured results was within +/- 1 mm. But, when the precipitation was smaller than 6 mm, the simulated throughfall was lower than the measured one. The values of simulated stemflow were lower than the measured values, and the relative deviation between simulated and measured values was smaller when the precipitation was larger. The process of throughfall in the forest was also simulated, with the results fitted well to the measured one.

[Clonal growth of stoloniferous herb Potentilla anserina on degraded and non-degraded alpine meadow soil].

The clonal growth of Potentilla anserina on degraded and non-degraded Kobresia humilis meadow soil was studied by a transplanting experiment in the field. No significant differences in numbers of stolons, height, and leave size per mother ramet were observed between the two soils, but the numbers of leaves per mother ramet, length and width of stolon, and spacer length were significantly different. There were more leaves per mother ramet, longer stolon and spacer, and wider stolon on degraded soil, where available nutrient was poor than in non-degraded soil. Under degraded alpine meadow soil condition, the clonal plant species might produce much more photosynthetic product to support stolon growth by the increase of leave numbers per mother ramet, and the longer and wider stolon could intensify the foraging ability of the mother ramet that would benefit to daughter ramet. Both mother and daughter ramet of Potentilla anserina invested much more biomass to their underground part (root system) in non-degraded soil to increase the survival rate of daughter ramet.
