Tissue tropism of the TTV in experimentally infected rhesus monkeys.

OBJECTIVE: To determine whether the transfusion-transmitted virus (TTV) is hepatotropic. METHODS: Total DNA was extracted from various tissues of 5 experimentally infected Rhesus monkeys during the viremic period. A dot hybridization was done with viral double stranded DNA probes or single antisense probes. RESULTS: The double-stranded probe was hybridized with DNA from the liver, bone marrow, spleen,stomach, small intestine and colon. The single-stranded antisense probe was hybridized with DNA from the liver, small intestine and bone marrow of all 5 monkeys, but not with that from other tissues. CONCLUSIONS: As the viral genome was of negative polarity, the plus-stranded fragment identified in our study might be a replicative intermediate, and was only demonstrated in the liver, small intestine, and bone marrow by dot blot hybridization with single-stranded antisense probes. It is suggested that the TTV replicates in the liver, bone marrow and small intestine, and TTV might be hepatotropic.

[Effect of B7-2 expression plasmid on the specific immune response induced by HBV DNA vaccine].

OBJECTIVE: To determine whether B7-2 could improve hepatitis B virus (HBV)-specific immunity induced by inoculation of a plasmid encoding HBV preS2+S HBV DNA vaccine. METHODS: B7-2 expression plasmids were inoculated with the DNA vaccine in the gastrocemius muscles. Then the cytolytic T Lymphocyte activity (CTL), the delayed-type hypersensitivity (DTH) response and the titers of anti-HBs were analysed. RESULTS: The DTH response and CTL activity were significantly enhanced when B7-2 expression plasmids were coinoculated with the DNA vaccine (P<0.01), but the titers of anti-HBs were unaffected (P>0.05). CONCLUSIONS: The results suggest that B7-2 expression plasmid could improve HBV specific cell-mediated immunity induced by HBV DNA vaccine, and the gene-based coinoculation strategy using HBV viral antigen and B7-2 costimmulating could be a powerful means of combating HBV infection.
