Affordable oral health care: dental biofilm disruption using chloroplast made enzymes with chewing gum delivery.

Current approaches for oral health care rely on procedures that are unaffordable to impoverished populations, whereas aerosolized droplets in the dental clinic and poor oral hygiene may contribute to spread of several infectious diseases including COVID-19, requiring new solutions for dental biofilm/plaque treatment at home. Plant cells have been used to produce monoclonal antibodies or antimicrobial peptides for topical applications to decrease colonization of pathogenic microbes on dental surface. Therefore, we investigated an affordable method for dental biofilm disruption by expressing lipase, dextranase or mutanase in plant cells via the chloroplast genome. Antibiotic resistance gene used to engineer foreign genes into the chloroplast genome were subsequently removed using direct repeats flanking the aadA gene and enzymes were successfully expressed in marker-free lettuce transplastomic lines. Equivalent enzyme units of plant-derived lipase performed better than purified commercial enzymes against biofilms, specifically targeting fungal hyphae formation. Combination of lipase with dextranase and mutanase suppressed biofilm development by degrading the biofilm matrix, with concomitant reduction of bacterial and fungal accumulation. In chewing gum tablets formulated with freeze-dried plant cells, expressed protein was stable up to 3 years at ambient temperature and was efficiently released in a time-dependent manner using a mechanical chewing simulator device. Development of edible plant cells expressing enzymes eliminates the need for purification and cold-chain transportation, providing a potential translatable therapeutic approach. Biofilm disruption through plant enzymes and chewing gum-based delivery offers an effective and affordable dental biofilm control at home particularly for populations with minimal oral care access.

[Effects of Helicobacter pylori infection on neural expression of stomach and spinal cord: an experiment with mice].

OBJECTIVE: To investigate the effects of Helicobacter pylori (Hp) infection on neural expression in stomach and spinal cord, and to investigate the mechanism of functional dyspepsia after Hp infection. METHODS: Thirty-five female C57BL/6 mice were randomly divided into three groups: Group A (acute infection group, undergoing intragastric gavage of Hp suspension every other day for 3 times and then observed for 2 weeks, 15 mice), Group B (chronic infection group, undergoing intragastric gavage of Hp suspension every other day for 3 times and then observed for 2 weeks, 15 mice) and control group (undergoing intragastric gavage of normal saline every other day for 3 times and then observed for 2 weeks, 5 mice). After the observation the mice were killed and their stomachs were taken out to undergo gastric histology and bacterial colonization by HE staining and Warthin-Starry staining respectively. Their spinal cords of thoracic and lumbar segments were taken out too. Immunohistochemistry was used to detect the expression of Fos, vasoactive intestinal polypeptide (VIP), and calcitonin gene-related peptide (CGRP) in the stomach and spinal cord. RESULTS: Three mice died 12 weeks after Hp infection. The rate of Hp colonization, mainly localized in pyloric gland region, was greater in Group B than in Group A, and was 0 in the control group. The severity of inflammation as shown by mononuclear cell infiltration, and activity of inflammation as shown by polymorphonuclear cell infiltration, in the pyloric gland region, proventriculus-glandular stomach region, and corpus gland region were more pronounced in Groups A and B, especially in Group B, than in the control group. The expression values of Fos, VIP, and CGRP in the stomach of Group A were 3.1 +/- 1.4, 4.5 +/- 1.8, and 2.4 +/- 0.8 respectively, all not significantly different from those of Group B (3.1 +/- 1.3, 3.5 +/- 1.6, and 2.2 +/- 0.8, all P > 0.05). The expression values of Fos, VIP, and CGRP in the spinal cord of Group A were 3.8 +/- 1.2, 3.2 +/- 1.5, and 2.2 +/- 0.6, all not significantly different from those of Group B (3.4 +/- 0.7, 2.6 +/- 1.2, and 2.5 +/- 1.1, all P > 0.05 for all). However, the neural expression values in both acute and chronic infection groups were significantly higher than those in the control group (2.4 +/- 0.9, 1.6 +/- 0.9, and 1.2 +/- 0.8 in stomach; and 2.0 +/- 1.6, 1.2 +/- 1.1, and 1.2 +/- 1.1 in spinal cord, P < 0.05 for all). CONCLUSION: Hp infection, both acute and chronic, induces gastric histological changes such as inflammation and activity, and enhances the Fos, VIP, and CGRP expression in stomach and spinal cord, which can be a basis for symptom generation in dyspeptic patients with Hp infection.

Chemopreventive effects of rofecoxib and folic acid on gastric carcinogenesis induced by N-methyl-N'-nitro-N-nitrosoguanidine in rats.

OBJECTIVES: Epidemiological and experimental studies indicate that non-steroidal anti-inflammatory drugs (NSAIDs) are chemopreventive agents of gastrointestinal cancers, but few studies on gastric cancer have been carried out. A decrease in folic acid supplement and subsequent DNA hypomethylation are related to gastrointestinal cancers, and it has been shown that high-dose folic acid may interfere with gastric carcinogenesis in dogs. The objective of this study was to investigate the effects of rofecoxib, a selective cyclooxygenase-2 (COX-2) inhibitor, and folic acid on the chemoprevention of gastric cancer induced by N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in Wistar rats, and to evaluate the cell proliferation of gastric mucosa in different experimental groups. METHODS: Eighty male Wistar rats were randomly divided into five groups (16 rats in each group). In the control group, the rats were given pure water and basal diet. In the MNNG group, the rats received MNNG in drinking water (100 mg/L) and basal diet. In the MNNG + low-dose rofecoxib group, the rats were given MNNG and rofecoxib 5 mg/kg per day with basal diet. In the MNNG + high-dose rofecoxib group, the rats were given MNNG and rofecoxib 15 mg/kg per day with basal diet. In the MNNG + folic acid group, the rats were given MNNG and folic acid 5 mg/kg per day with basal diet. The experiment was terminated at 50 weeks, and all rats were killed. Blood samples of 3 mL were obtained for measurement of serum folic acid concentrations in the control group, the MNNG group and the MNNG + folic acid group by using chemiluminescent method. The stomach was removed from all rats for histopathological examination and immunohistochemical study. Proliferating cell nuclear antigen (PCNA) expression in gastric epithelial cells was also determined. RESULTS: In the MNNG group, five of 11 rats (45.5%) developed gastric cancer, while in all other four groups no gastric cancer was found (P < 0.05). The positivity rate of PCNA expression in the cancerous tissues was significantly higher than that in the non-cancerous tissues (80.0%vs 14.1%, P < 0.05). The positivity rate of PCNA expression in the gastric mucosal cells of the MNNG group was significantly higher than that in the other four groups. The mean serum folic acid concentration of rats was significantly higher in the MNNG + folic acid group (193.70 +/- 60.73 ng/mL) than those in the control group (84.21 +/- 25.26 ng/mL) and the MNNG group (72.27 +/- 16.70 ng/mL, P < 0.05). It was shown that both low- and high-dose rofecoxib as well as folic acid interfered with the development of gastric cancer induced by MNNG in Wistar rats. CONCLUSIONS: The results indicate that rofecoxib as well as folic acid interferes with gastric carcinogenesis induced by MNNG in Wistar rats, and the suppression of gastric cell proliferation may play a crucial role in the chemoprevention of gastric cancer by rofecoxib and folic acid. The higher serum folic acid concentration of rats may play an important role in the prevention of gastric cancer.

Study on NO(2) absorption by ascorbic acid and various chemicals.

Study on NO(2) absorption aimed at seeking a better NO(2) absorption chemical at pH 4.5 approximately 7.0 for application to existing wet flue gas desulfurization (FGD). The results from the double-stirred reactor indicated that ascorbic acid has very high absorption rate at this pH range. The rate constant of ascorbic acid reaction with NO(2) (0 approximately 1,000 x 10(-6) mol/mol) is about 3.54 x 10(6) mol/(Ls) at pH 5.4 approximately 6.5 at 55 degrees C.

Characterization of on-road CO, HC and NO emissions for petrol vehicle fleet in China city.

Vehicle emissions are a major source of air pollution in urban areas. The impact on urban air quality could be reduced if the trends of vehicle emissions are well understood. In the present study, the real-world emissions of vehicles were measured using a remote sensing system at five sites in Hangzhou, China from February 2004 to August 2005. More than 48000 valid gasoline powered vehicle emissions of carbon monoxide (CO), hydrocarbons (HC) and nitrogen oxide (NO) were measured. The results show that petrol vehicle fleet in Hangzhou has considerably high CO emissions, with the average emission concentration of 2.71%+/-0.02%, while HC and NO emissions are relatively lower, with the average emission concentration of (153.72+/-1.16)x10(-6) and (233.53+/-1.80)x10(-6), respectively. Quintile analysis of both average emission concentration and total amount emissions by model year suggests that in-use emission differences between well maintained and badly maintained vehicles are larger than the age-dependent deterioration of emissions. In addition, relatively new high polluting vehicles are the greatest contributors to fleet emissions with, for example, 46.55% of carbon monoxide fleet emissions being produced by the top quintile high emitting vehicles from model years 2000-2004. Therefore, fleet emissions could be significantly reduced if new highly polluting vehicles were subject to effective emissions testing followed by appropriate remedial action.

Anti-inflammatory mechanism of oxymatrine in dextran sulfate sodium-induced colitis of rats.

AIM: To investigate the anti-inflammatory mechanism of oxymatrine in dextran sulfate sodium (DSS)-induced colitis of rats. METHODS: Acute colitis was induced by giving 2% DSS orally in drinking water for 8 d. Twenty-six male rats were randomized into oxymatrine-treated group (group A, 10 rats), DSS control (group B, 10 rats) and normal control (group C, 6 rats). The rats in group A were injected muscularly with oxymatrine at the dosage of 63 mg/(kg.d) from d 1 to 11 and drank 2% DSS solution from d 4 to 11. The rats in group B were treated with 0.9% saline in an equal volume as group A and drank 2% DSS solution from d 4 to 11. The rats in group C were treated with 0.9% saline as group B from d 1 to 11 and drank water normally. Diarrhea and bloody stool as well as colonic histology were observed. The levels of serum tumor necrosis factor-alpha (TNF-alpha) and interleukin-6 (IL-6) were determined by ELISA, and nuclear factor-kappaB (NF-kappaB) activity and the expression of inter-cellular adhesion molecule-1 (ICAM-1) in colonic mucosa were detected by immunohistochemistry method. RESULTS: Compared with DSS control group, the inflammatory symptoms and histological damages of colonic mucosa in oxymatrine-treated group were significantly improved, the serum levels of TNF-alpha, IL-6, and the expression of NF-kappaB, ICAM-1 in colonic mucosa were significantly reduced. CONCLUSION: The fact that oxymatrine can reduce the serum levels of TNF-alpha, IL-6, and the expression of NF-kappaB and ICAM-1 in colonic mucosa in DSS-induced colitis of rats indicates that oxymatrine may ameliorate the colonic inflammation and thus alleviate diarrhea and bloody stool.

Biodesulfurization of dibenzothiophene and other organic sulfur compounds by a newly isolated Microbacterium strain ZD-M2.

A new bacterial strain capable of selectively desulfurizing dibenzothiphene (DBT) was isolated from sludge. This organism, designated ZD-M2, clustered most closely with members of the genus Microbacterium, as determined by 16S rRNA gene sequence analysis. Metabolites produced by DBT desulfurization were identified by GC-MS, and two sulfur-free products, 2-methoxybiphenyl (2-MBP) and biphenyl, were detected in addition to 2-hydroxybiphenyl (2-HBP). This strain can also totally degrade 4,6-dimethyl-DBT, thiophene, benzothiophene and 70% diphenylsulfide.

Helicobacter pylori "test-and-treat" strategy is not suitable for the management of patients with uninvestigated dyspepsia in Shanghai.

OBJECTIVE: The safety of Helicobacter pylori "test-and-treat" and "test-and-endoscopy" strategies for the management of young patients with uninvestigated dyspepsia has not been evaluated in Shanghai. MATERIAL AND METHODS: A total of 14,101 consecutive patients with dyspepsia receiving endoscopy in our hospital from October 2002 to December 2003 were retrospectively studied. The detection rates of esophageal or gastroduodenal malignancies and alarm symptoms were investigated, and H. pylori status was assessed. RESULTS: A total of 202 (1.4%) gastrointestinal (GI) malignancies were found, including 162 cases (1.15%) of gastric cancer, 4 of gastric lymphoma, 35 (0.25%) of esophageal cancer and 1 case of duodenal cancer. Among those patients with GI malignancies, 99 (49.0%) were infected with H. pylori and 108 (53.5%) presented with alarm symptoms. Eighteen patients (0.46%, 18/3952) under 45 years of age were diagnosed as having gastric cancer. Of these patients, 5 (27.8%) presented with alarm symptoms and 13 (72.2%) were infected with H. pylori. If the H. pylori "test-and-treat" strategy were used in dyspeptic patients under the age of 45 years without alarm symptoms in the Shanghai region, then 13 cases (72.2%) of gastric cancer would be missed. If the H. pylori "test-and-endoscopy" strategy were applied, then 3 cases (16.7%) of gastric cancer would be missed. CONCLUSIONS: H. pylori "test-and-treat" and "test-and-endoscopy" strategies are both not suitable for the management of patients with uninvestigated dyspepsia in Shanghai. For most Shanghai dyspeptic patients, prompt endoscopy should be recommended as the first-line initial management option.

Experimental study on the inhibition of biological reduction of Fe(III)EDTA in NO(x) absorption solution.

Scrubbing of NO(x) from the gas phase with Fe(II)EDTA has been shown to be highly effective. A new biological method can be used to convert NO to N(2) and regenerate the chelating agent Fe(II)EDTA for continuous NO absorption. The core of this biological regeneration is how to effectively simultaneous reduce Fe(III)EDTA and Fe(II)EDTA-NO, two mainly products in the ferrous chelate absorption solution. The biological reduction rate of Fe(III)EDTA plays a main role for the NO(x) removal efficiency. In this paper, a bacterial strain identified as Klebsiella Trevisan sp. was used to demonstrate an inhibition of Fe(III)EDTA reduction in the presence of Fe(II)EDTA-NO. The competitive inhibition experiments indicted that Fe(II)EDTA-NO inhibited not only the growth rate of the iron-reduction bacterial strain but also the Fe(III)EDTA reduction rate. Cell growth rate and Fe(III)EDTA reduction rate decreased with increasing Fe(II)EDTA-NO concentration in the solution.

[Microbial reduction of Fe(III) (EDTA) in the system of nitric oxide removal by metal chelate absorption].

Reduction of Fe(III) (EDTA) with cultivated microorganisms in the system of nitric oxide removal by metal chelate absorption was investigated. Supplemental glucose stimulates the formation of Fe(II) (EDTA) more than ethanol and methanol as the carbon sources. Ammonium salt was used to be as the nitric source instead of nitrate, which inhibits the reduction of Fe(III) (EDTA) due to the competition between the two electron acceptors. The optimal pH value was from 6 to 7. The reduction percentage of Fe(III) (EDTA) varied little with the range of 30 degrees C - 40 degrees C and decreased quickly with a temperature higher than 40 degrees C. The bio-reduction could be achieved efficiently with enough carbon source and cell inoculation. The reduction rate did not increase with adding more amount of carbon source or cell inoculation. The bio-reduction rate could be described by Michaelis-Menten equation and fitted to the first order reaction kinetics. The maximum reaction rates gamma max and the Michaelis constant k(m) were 1.3 mmol x (L x h)(-1) and 53.5 mmol x L(-1)respectively.

Decomposition of simulated odors in municipal wastewater treatment plants by a wire-plate pulse corona reactor.

Decomposition of simulated odors in municipal wastewater treatment plants was investigated experimentally by a wire-plate pulse corona reactor. A new type of high pulse voltage source with a thyratron switch and a Blumlein pulse forming network (BPFN) was adopted in our experiments, and the testing malodorants were ammonia, ethanethiol and tri-methyl amine, respectively. The maximum output power of the pulse voltage source and the maximum peak voltage were 1 kW and 100 kV. The experiments were conducted at the gas-flow rate of 4.0-23.0 m3 h(-1). Important parameters, including peak voltage, pulse frequency, capacitance (inductance) of the BPFN, gas-flow rate, initial concentration, which influenced on the removal efficiency, were investigated. The results show that the odors can be treated effectively. Almost 100% removal efficiency was obtained for 32 mg m(-3) ammonia at the gas-flow rate of 4.0 m(3) h(-1). The maximum removal efficiencies of 85 mg m(-3) ethanethiol and 750 mg m(-3) tri-methyl amine at 10.0 m(3) h(-1) were 98% and 91%, respectively. The energy yield of 110 mg m(-3) ammonia was 2.99 g kWh(-1) when specific energy density was 106 Jl(-1). In the cases of ammonia, ethanethiol and tri-methyl amine removal, ozone and nitrogen oxides were observed in the exit gas. The carbon and sulfur elements of ethanethiol and tri-methyl amine were mainly converted to carbon dioxide, carbon monoxide and sulfur dioxide. Moreover, the ammonium nitrates and sulfur were discovered in the reactor.

Characterization of sorption mechanisms of VOCs with organobentonites using a LSER approach.

To fully utilize the sorption traits of organobentonites to control volatile organic compounds (VOCs) pollution, the sorption mechanisms of VOCs with organobentonites need to be understood adequately. The sorption of VOCs as vapors to a typical organobentonite, modified with cetyltrimethylammonium bromide (CTMAB-bentonite), was characterized using a linear solvation energy relationship (LSER) of the type log Kc = c + rR2 + s pi2H + a sigma(alpha2)H + b sigma(beta2)H + l log L16. The fitted LSER equation, log Kc = 0.434 + 0.968R2 - 0.0886pi2H + 2.170sigma(alpha2)H + 1.611sigma(beta2)H + 0.417 log L16, was obtained by a multiple regression of the partition coefficients of 22 probe solutes against the solvation parameters of the solutes. The coefficients of the LSER equation show that CTMAB-bentonite is a sorbent with nonsignificant dipolarity/polarizability, interacts with solutes partly through pi-/n-electron pairs, behaves both as hydrogen-bond donor and hydrogen-bond acceptor, and can interact with solutes by cavity/dispersion interactions. The related terms in LSER suggest that the potential factors governing the sorption of VOCs onto CTMAB-bentonite are dispersion interactions, hydrogen-bond acidity interactions, hydrogen-bond basicity interactions, and pi-/n-electron interactions. The dispersion interaction is recognized to be the predominant parameter for most solutes, whereas the contributions of the other parameters depend on specific solutes. The derived LSER equation successfully predicted the VOC partition coefficients and the selectivity of CTMAB-bentonite for the VOCs. The relationship between LSER and adsorption/partition model was compared. The classification of sorption mechanisms by LSER goes on the molecular interaction types between sorbate and sorbent, and classification by adsorption/partition model goes on the property difference among various components of sorbent. The LSER approach coupled with inverse gas chromatography (IGC) is a comparatively simple and reliable tool to rapidly characterize the sorption mechanism of VOCs with solid sorbents such as CTMAB-bentonite, and may potentially be applied to the design of an organoclay sorbent for control of VOCs.

Development of gastric adenocarcinoma in Mongolian gerbils after long-term infection with Helicobacter pylori.

BACKGROUND AND AIM: The experimental evidence that long-term colonization of Helicobacter pylori results in the development of gastric cancer in Mongolian gerbils has been reported only by two Japanese groups to date. This study aimed to investigate the carcinogenicity of H. pylori infection in a Mongolian gerbil model. METHODS: Thirty-six Mongolian gerbils (inner Mongolian origin) were divided into two groups (male to female ratio, 1:1) and orally inoculated with a standard H. pylori strain (ATCC43504) or H. pylori161 (isolated from a Chinese patient with gastric adenocarcinoma), respectively, once a week for 5 weeks. Another 10 control gerbils were given phosphate-buffered saline. The animals were killed 8, 20, 28 and 84 weeks after inoculation for bacterial and histological examination. RESULTS: Seven inoculated gerbils died at the week 42. Overall, H. pylori colonization was detected in 24 (83%) of the 29 available inoculated gerbils. The gastric lesions were aggravated gradually over time. At week 84, moderate to severe gastritis, characterized by diffuse infiltration of mononuclear cells and formation of multiple lymphoid follicles in mucosa and submucosa, and even the lymphoepithelial lesions, were observed. Epithelial hyperplasia were dominant in almost all gerbils. Four (24%) of the 17 animals had hyperplastic polyps. Intestinal metaplasia were rarely seen (in three gerbils). Well-differentiated gastric adenocarcinomas developed in three (18%) of the 17 gerbils after 84 weeks. Of the three gerbils, one female gerbil was infected with H. pylori161 and the others (one male and one female) were infected with ATCC43504. CONCLUSIONS: The present study reconfirms that H. pylori infection alone can induce gastric adenocarcinoma in Mongolian gerbils and suggests that different species of gerbil and both standard and clinically isolated H. pylori strains can be used for investigating the carcinogenesis of H. pylori. This is the first report of the development of gastric cancer in female gerbils, which highlights the importance of using both sexes to investigate the pathogenesis of H. pylori and whether host susceptibility is influenced by sex.

Regeneration of nitric oxide chelate absorption solution by two heterotrophic bacterial strains.

Ferrous chelate absorption is deemed a promising method for NO removal from flue gas, but the key problem is the difficulty to regenerate the absorption solution, i.e. the complexes of Fe(II)(EDTA)NO and Fe(III)(EDTA) in the solution. Two bacterial strains isolated recently from the sludge of the denitrification step of a municipal wastewater treatment plant could be applied effectively to regenerate the absorbent were Pseudomonas sp. and klebsiella trevisan sp. Pseudomonas sp. exhibited high reduction ability on Fe(II)(EDTA)NO and the klebsiella trevisan sp. was more suitable for Fe(III)(EDTA) re-duction.

Dissimilatory reduction of FeIII (EDTA) with microorganisms in the system of nitric oxide removal from the flue gas by metal chelate absorption.

In the system of nitric oxide removal from the flue gas by metal chelate absorption, it is an obstacle that ferrous absorbents are easily oxidized by oxygen in the flue gas to ferric counterparts, which are not capable of binding NO. By adding iron metal or electrochemical method, FeIII(EDTA) can be reduced to FeII(EDTA). However, there are various drawbacks associated with these techniques. The dissimilatory reduction of FeIII(EDTA) with microorganisms in the system of nitric oxide removal by metal chelate absorption was investigated. Ammonium salt instead of nitrate was used as the nitrogen source, as nitrates inhibited the reduction of FeIII due to the competition between the two electron acceptors. Supplemental glucose and lactate stimulated the formation of FeII more than ethanol as the carbon sources. The microorganisms cultured at 50 degrees C were not very sensitive to the other experimental temperature, the reduction percentage of FeIII varied little with the temperature range of 30-50 degrees C. Concentrated Na2CO3 solution was added to adjust the solution pH to an optimal pH range of 6-7. The overall results revealed that the dissimilatory ferric reducing microorganisms present in the mix-culture are probably neutrophilic, moderately thermophilic FeIII reducers.
