Translational control by RPL22L1-specific ribosomes enhances DNA repair and chemoresistance.

Ribosome heterogeneity has emerged as a regulatory layer in gene expression, yet its biological roles in cancers remain poorly characterized. Here, we identify RPL22L1, a paralog of the ribosomal protein RPL22, as a key modulator of DNA damage response (DDR) in colorectal cancer cells. DNA damage induces RPL22L1 upregulation and ribosomal incorporation, forming RPL22L1-specific ribosomes. Ribosome profiling reveals that RPL22L1-containing ribosomes preferentially translate mRNAs with highly structured 5' untranslated region (5'UTR). In particular, RPL22L1 enhances the translation of ATRX through a cap-independent mechanism. ATRX subsequently recruits DNA-PKcs to DNA damage sites, thereby enhancing the DNA repair capacity. RPL22L1 loss creates exploitable DDR vulnerabilities, sensitizing cancer cells to cisplatin and PARP inhibitors in vitro and in vivo. Collectively, these findings uncover a specialized ribosome-mediated translational program in DDR and highlight RPL22L1 as a potential therapeutic target in DDR-based cancer therapy.

map3C: a computational tool for processing multiomic single-cell Hi-C data.

SUMMARY: The emergence of multiomic single-cell Hi-C (scHi-C) methods, which simultaneously profile chromatin conformation and other modalities such as gene expression or DNA methylation, creates tremendous opportunities for studying the genome's structure-function relationships. Existing tools for processing multiomic scHi-C datasets lack certain key functions for downstream bioinformatics analysis. We present map3C, a software tool that incorporates additional key functions. Specifically, we demonstrate that map3C facilitates multiomic scHi-C processing, quality control, and identification of structural variant locations in the genome. AVAILABILITY AND IMPLEMENTATION: map3C is available at https://github.com/luogenomics/map3C and is archived at https://doi.org/10.5281/zenodo.20724719.

Dissection of the AtNRT2.1:AtNRT2.2 inducible high-affinity nitrate transporter gene cluster.

Using a new Arabidopsis (Arabidopsis thaliana) mutant (Atnrt2.1-nrt2.2) we confirm that concomitant disruption of NRT2.1 and NRT2.2 reduces inducible high-affinity transport system (IHATS) by up to 80%, whereas the constitutive high-affinity transport system (CHATS) was reduced by 30%. Nitrate influx via the low-affinity transport system (LATS) was unaffected. Shoot-to-root ratios were significantly reduced compared to wild-type plants, the major effect being upon shoot growth. In another mutant uniquely disrupted in NRT2.1 (Atnrt2.1), IHATS was reduced by up to 72%, whereas neither the CHATS nor the LATS fluxes were significantly reduced. Disruption of NRT2.1 in Atnrt2.1 caused a consistent and significant reduction of shoot-to-root ratios. IHATS influx and shoot-to-root ratios were restored to wild-type values when Atnrt2.1-nrt2.2 was transformed with a NRT2.1 cDNA isolated from Arabidopsis. Disruption of NRT2.2 in Atnrt2.2 reduced IHATS by 19% and this reduction was statistically significant only at 6 h after resupply of nitrate to nitrogen-deprived plants. Atnrt2.2 showed no significant reduction of CHATS, LATS, or shoot-to-root ratios. These results define NRT2.1 as the major contributor to IHATS. Nevertheless, when maintained on agar containing 0.25 mm KNO(3) as the sole nitrogen source, Atnrt2.1-nrt2.2 consistently exhibited greater stress and growth reduction than Atnrt2.1. Evidence from real-time PCR revealed that NRT2.2 transcript abundance was increased almost 3-fold in Atnrt2.1. These findings suggest that NRT2.2 normally makes only a small contribution to IHATS, but when NRT2.1 is lost, this contribution increases, resulting in a partial compensation.

[Dynamic observation of brainstem auditory evoked potential and pathological changes of the brain after closed brain injuries in rats].

OBJECTIVE: To investigate the dynamic experiences of pathological changes in brain and the injured role of brainstem auditory evoked potential (BAEP) after closed Mild Traumatic Brain Injury (MTBI) in rat. METHODS: A closed diffuse brain injury model was successfully produced in rat by hit the parietal bone with a spring-droved steel stick. The auditory brainstem response evoked by click at 50Hz stimulating frequency were recorded at pre-trauma, 15 min and 1, 3, 6, 12 h and 1, 2, 4, 7, 10, 14, 21 d after brain injury. The pathological changes of brain were observed under light microscope and the brain edema was detected by wet weight/dry weight rations. RESULTS: The I-V, III-V of BAEP wave inter peak latency (IPL) in MTBI group increased immediately in 15 minutes after injury. The III, V wave peak latency (PL) and I-V, III-V wave IPL became longer than the pre-trauma level in 6, 12 h post-injury (P<0.05). The III, V wave PL and I - III, I - V, III - V wave IPL became longer than the pre-trauma level in 1, 2 d after injury (P < 0.001), the BAEP retuned to normal level in 14 d after brain injury. At 15 minutes after injury, the water content in brain increased and met its peak in 1 d, decreased gradually in 4 d, then decreased slowly and returned to normal level in 10 d after injury. CONCLUSION: The regular changes of BAEP could be an objective tool for evaluating hearing dysfunction after closed mild diffuse traumatic brain injury.

Prebiotic effects of neoagaro-oligosaccharides prepared by enzymatic hydrolysis of agarose.

To investigate the prebiotic properties of neoagaro-oligosaccharides (NAOS), obtained from enzymatic hydrolysis of agarose, the in vitro and in vivo effects of NAOS on bacterial growth were studied. In vitro NAOS were found to be highly resistant to enzymes of the upper gastrointestinal tract, which remained intact after 24h incubation with different amylolytic enzymes. NAOS significantly stimulated the growth of bifidobacteria and lactobacilli in Man-Rogosa-Sharp (MRS) medium, anaerobically. Compared with fructo-oligosaccharides (FOS), 1% (w/v) NAOS promoted the specific growth rate of beneficial bacteria by about 100%. The decreases of media pH with NAOS were almost the same as that with FOS. In vivo, NAOS significantly increased the numbers of lactobacilli and bifidobacteria (P<0.05) in fresh feces or cecal content while reducing putrefactive microorganisms. Mice fed with 2.5% (w/v) NAOS for 7 days had larger increases in colonic beneficial bacteria population than those fed with even 5% (w/v) FOS for 14 days. No side effects, such as eructation and bloating, were found. Interestingly, NAOS with higher degrees of polymerization (DP) showed better prebiotic activity. These results indicated that NAOS had great prebiotic effect, which could be beneficial to the host.

Potential bridges for HIV infection to men who have sex with men in Guangzhou, China.

To assess the potential for HIV acquisition among men who have sex with men (MSM) in Guangzhou, China, we conducted a cross-sectional, anonymous, face-to-face survey of MSM in the metropolitan area of Guangzhou, China. As a pilot recruitment for a cohort study, participants were recruited by convenience sampling through newspaper and television advertising, website information, and respondent referral. Blood samples were tested for HIV, hepatitis B (HBV), hepatitis C (HCV), and syphilis. Client-centered HIV and STD counseling was provided.A total of 201 MSM were interviewed and 200 blood samples were tested. The prevalence of HIV antibody was 0% (97.5% CI 0-1.8%); 17.5% of MSM were HBV surface antigen positive; 1.0% had HCV antibodies; 10.5% had antibodies to syphilis. Syphilis seropositivity was associated with sex with a foreign MSM in the last six months and 10.4% reported sex with a foreign MSM overall. The majority (54.7%) reported unprotected anal sex with other men. Nearly one-third (31.8%) had regular female partners; 25.9% were currently married to a woman; 6% had casual female partners; 4.5% had sex with a female sex worker; 4.5% had sex with a male sex worker; and 12.9% had unprotected vaginal sex and unprotected anal sex with a man in the past six months. Only one MSM reported injection drug use (0.5%). The currently low prevalence of HIV but high level of unprotected anal sex, high prevalence of syphilis infection, and sexual networks that include foreign MSM point to a transient window for HIV prevention among MSM in Guangzhou. We recognize challenges to recruiting a representative sample of MSM and retaining them in longitudinal cohort studies.

Y55 and D78 are crucial amino acid residues of a new IgE epitope on trichosanthin.

Trichosanthin (TCS) possesses many biological and pharmaceutical activities, but its strong immunogenicity limits its clinical application. To reduce the immunogenicity of TCS, we modified the reported method for the prediction of antigenic site and identified two crucial amino acid residues (Y55 and D78) for a new epitope. We mutated these two residues into glycine and serine, respectively, and obtained three mutants, Y55G, D78S, and Y55G/D78S. These mutants induced less amount of Ig and IgG antibodies in C57BL/6J mice than wild-type TCS (wTCS) (p<0.01) and almost lost the ability to induce IgE antibody production. The mutants stimulated fewer TCS-specific B cells in C57BL/6J mice than wTCS (p<0.01). Compared with wTCS, Y55G, D78S, and Y55G/D78S lost 26.9%, 17.9%, and 98.7% specific binding ability to anti-TCS monoclonal antibody TCS4E9, respectively. These mutants still retained RNA N-glycosidase activity. In conclusion, Y55 and D78 are two crucial amino acid residues of a new IgE epitope on TCS, and their mutation reduces the immunogenicity of TCS, but still retained the enzymatic activity.

Implications of utilizing a phonics-based reading curriculum with children who are deaf or hard of hearing.

Extensive literature has reiterated the reading difficulties of students who are deaf or hard of hearing. Building and expanding upon the work of B. J. Trezek and K. W. Malmgren, this study demonstrated that given 1 year of instruction from a phonics-based reading curriculum supplemented by Visual Phonics, kindergarten and first-grade students who were deaf or hard of hearing could demonstrate improvements in beginning reading skills as measured by standardized assessments of (a) word reading, (b) pseudoword decoding, and (c) reading comprehension. Furthermore, the acquisition of beginning reading skills did not appear to be related to degree of hearing loss. In this study, students with various degrees of hearing loss benefited equally well from this phonics-based reading curriculum supplemented by Visual Phonics.

High-affinity nitrate transport in roots of Arabidopsis depends on expression of the NAR2-like gene AtNRT3.1.

The NAR2 protein of Chlamydomonas reinhardtii has no known transport activity yet it is required for high-affinity nitrate uptake. Arabidopsis (Arabidopsis thaliana) possesses two genes, AtNRT3.1 and AtNRT3.2, that are similar to the C. reinhardtii NAR2 gene. AtNRT3.1 accounts for greater than 99% of NRT3 mRNA and is induced 6-fold by nitrate. AtNRT3.2 was expressed constitutively at a very low level and did not compensate for the loss of AtNRT3.1 in two Atnrt3.1 mutants. Nitrate uptake by roots and nitrate induction of gene expression were analyzed in two T-DNA mutants, Atnrt3.1-1 and Atnrt3.1-2, disrupted in the AtNRT3.1 promoter and coding regions, respectively, in 5-week-old plants. Nitrate induction of the nitrate transporter genes AtNRT1.1 and AtNRT2.1 was reduced in Atnrt3.1 mutant plants, and this reduced expression was correlated with reduced nitrate concentrations in the tissues. Constitutive high-affinity influx was reduced by 34% and 89%, respectively, in Atnrt3.1-1 and Atnrt3.1-2 mutant plants, while high-affinity nitrate-inducible influx was reduced by 92% and 96%, respectively, following induction with 1 mm KNO(3) after 7 d of nitrogen deprivation. By contrast, low-affinity influx appeared to be unaffected. Thus, the constitutive high-affinity influx and nitrate-inducible high-affinity influx (but not the low-affinity influx) of higher plant roots require a functional AtNRT3 (NAR2) gene.

Biodegradable and complexed microspheres used for sustained delivery and activity protection of SOD.

To develop a new protein delivery system for superoxide dismutase (SOD), biodegradable materials like poly(DL-lactide-co-glycolide) (PLGA), alginate, and chitosan were used for preparing PLGA microspheres and alginate-chitosan microspheres, which were used for encapsulating protein. Alginate-chitosan microspheres showed much higher entrapment efficiency (91.08% +/- 1.28%) than that of PLGA microspheres (36.42% +/- 1.81%). In vitro release study showed that SOD presented a sustained release character in the preparation of these biodegradable materials. After 15 days, 43.72% +/- 0.43% of protein was released from alginate-chitosan microspheres, while there was 62.96% +/- 3.95% of protein release from PLGA microspheres. However, alginate-chitosan demonstrated that it was a better material to control the burst release of protein from microspheres. Furthermore, SOD activity in microspheres was evaluated, and the results showed that microspheres protected the activity of protein to some extent. Finally, PLGA-alginate-chitosan complex microspheres were constructed and the release character in vitro demonstrated that this preparation could not only prolong the release of drug but also decrease the burst release.

A case of overlapping Bickerstaff's brainstem encephalitis and Guillain-Barré syndrome.

OBJECTIVE: There is no report on Bickerstaff's brainstem encephalitis (BBE) patients in China. We here report the first case of BBE in China. METHODS: Clinical features, results of electromyography, electroencephalography (EEG), magnetic resonance imaging (MRI) and cerebrospinal fluid (CSF) examination were studied to clarify the characteristics of this syndrome. RESULTS: A 44-year-old man presented himself at our inpatient department with somnolence and dizziness as his initial symptoms. He developed multiple cranial nerves paralysis especially internal and external ophthalmoplegia, ataxia and tetraparesis within 1 week. His condition rapidly deteriorated, and he experienced coma. Electromyography showed indications of peripheral nerve dysfunction, electroencephalography revealed loss of basic rhythm, MRI demonstrated high-intensity abnormalities on T(2)-weighted images of medulla oblongata, and CSF albuminocytological dissociation was defined abnormally as high protein. Ten months later, he almost completely recovered. CONCLUSION: BBE, fisher syndrome (FS) and Guillain-Barré syndrome (GBS) are similar clinically; BBE and FS were proposed to be the variant of GBS.

[A study on expression of caspase-8 in organs of rats after electrical injury at antemortem or postmortem].

OBJECTIVE: This study was conducted to detect the expression of caspase-8 in organs of rats after The electrical injury so as to elucidate whether caspase-8 is useful in identifying electrical lesion. METHODS: experiment included two parts. In the first part (the antemortem electrical injury part), thirty-five healthy male SD rats were randomly divided into seven groups (n = 5 per group), i. e. the group of rats subjected to instantaneous electrothanasia; the groups of rats subjected to cervical dislocation at 1 h, 2 h, 4 h, 8 h after electrical injury; the sham group and the normal control group. In the second part (the postmortem electrical injury part), twenty-five healthy male SD rats wererandomly divided into five groups (n=5 per group), i. e. the groups of rats electrically injured just after death, and at 15 min, 30 min, 60 min after death; and the postmortem sham group. All experimental rats were given respectively an electric shock with two metal clamps that were connected with two poles of 220 V alternating current by clamping the rats' left hind limbs and right forelimbs. The rats of sham group after death were clamped but not electrified. The brain, lung, heart, liver, spleen, kidney, the muscle of electrified limb, the cutis of electrified limb of all experimental rats and those organs of control groups were dissected to detect the expression of caspase-8 by immunohistochemistry staining, and the staining intensities were assessed by image analysis system. RESULTS: In the antemortem electrical injury groups, the expression of caspase-8 was positive in brain, heart, liver and kidney; the strongest staining intensity appeard at 4 h after electrical injury and decreased at 8 h after electrical injury. In the group of rats electrically injuryed just after death, the expression of caspase-8 was faint, and the expression of caspase-8 in spleen, lung, muscle and cutis was negative in the other groups. The expression of caspase-8 in all detected organs was negative in the other rats that were electrified after death. CONCLUSION: Caspase-8 can be regarded as an index in identifying electrical injury and distinguishing between antemortem and postmortem electrical injuryies.
