Mode of action-based local QSAR modeling for the prediction of acute toxicity in the fathead minnow.

The ultimate intention of quantitative structure-activity relationship (QSAR) study in toxicology is to predict the toxic potential of untested compounds with great accuracy. As QSAR has been based on the assumption that compounds from the same chemical domain will behave in similar manner, the QSAR model built upon the analogical chemicals is hypothesized to exhibit better performance than that derived from the miscellaneous data set. In this paper, the acute toxicity, 96 h LC(50) (median lethal concentration) for the fathead minnow from database EPAFHM_v2a_617_1Mar05 served as the interested toxicity endpoint, and the mode of action (MOA) in toxic response was employed as a criterion to compartmentalize the chemical domains. MOA-based local QSAR models were built by partial least squares (PLS) regression for each subset with single mode of action such as Narcosis I, Narcosis II or Reactive, and global model was also developed for the combined data set containing several subsets above. By comparing the performances of these two types of models, the local models were superior to the global model in that the relative standard error (R.S.E.) of the former was much lower for both the training set and the test set of any subset. In addition, the influence of the reliability of MOA determination on the performance of local model was also investigated and the statistical results for subsets with MOAs at A and B confidence level were better than those at C and D confidence level. Therefore, the MOA-based local QSAR models are promising to improve the accuracy of toxicity prediction as long as the assessment of MOA is of high reliability.

Identification and analysis of absorbed and metabolic components in rat plasma after oral administration of 'Shuangdan' granule by HPLC-DAD-ESI-MS/MS.

A valid method using liquid chromatography-diode array detection-electrospray ionization (ESI) ion trap mass spectrometry was established for the study of the absorbed and metabolic components of a Chinese medicine 'Shuangdan' granule in rat plasma after oral administration. The plasma samples were acidified with 1M hydrochloric acid and extracted with ethyl acetate (EtOAc) of two-fold volume for three times. The chromatographic separation was carried out on a Zorbax SB-C18 column with a linear gradient whereas 0.1% acetic acid/water/acetonitrile was used as mobile phase. Mass spectra were acquired in both negative and positive modes. More than 20 components including 16 components from 'Shuangdan' granule and 5 metabolites were simultaneously identified by comparing their mass spectra and retention behavior with reference compounds or literature data. The results proved that the established method could be used to identify the structure of active components responsible for the pharmacological effects of 'Shuangdan' granule.

Analysis of 'SHUANGDAN' granules by high-performance liquid chromatography-diode array detection-electrospray ionization tandem mass spectrometry.

An HPLC-DAD-ESI-MSn method was developed for simultaneous analysis of the chemical constituents in 'SHUANGDAN' granules, a newly developed drug widely used for treating cardiovascular disease. The chromatographic separation were performed on a Zorbax Stable Bond C18 column (4.6 mmx250 mm, 5 microm) with water with 0.5% acetic acid (A) and acetonitrile (B) as mobile phase. According to the characteristic UV absorption profile, the information of molecular weight and structure provided by ESI-MSn, 29 constituents which attributed to Radix Salviae Miltiorrhizae and Cortex Moutan, respectively, were detected and 28 constituents including 14 phenolic acids, 6 diterpenoid quinones, 6 monoterpenoids and 2 other components were identified, while some isomers were distinguished based on the MSn spectra. This method was rapid and reliable for identification of constituents in complex chemical system, 'SHUANGDAN' granules, and the newly fragmentation patterns proposed could be extended to the compounds elucidation with similar framework.

Quality evaluation of cortex moutan by high performance liquid chromatography coupled with diode array detector and electrospary ionization tandem mass spectrometry.

An high performance liquid chromatography (HPLC) coupled with diode array detector (DAD) and electrospray ionization tandem mass spectrometry (ESI/MS(n)) method was developed for quality evaluation of Cortex Moutan through identification of common constituents based on chromatographic fingerprints and determination of key pharmacological compounds. The representative chromatographic fingerprints of Cortex Moutan were obtained by analyzing 10 batches of samples under the optimized HPLC conditions and the results showed that the chromatographic profiles of the analyzed samples were very similar. Total of nineteen common peaks were detected and seventeen of them were identified rapidly by their characteristic UV profile and the information of molecular structure provided by ESI/MS(n) experiments. Simultaneously, five key pharmacological compounds, namely gallic acid, oxypaeoniflorin, paeoniflorin, benzoylpaeoniflorin and paeonol, were determined by the validated HPLC-DAD method. The linear calibration curves were acquired with correlation coefficients higher than 0.999. The precisions of intra-day and inter-day were not exceeding 3.1%, and the recoveries of five analytes were from 92.86 to 99.35%. This developed method that combined the chromatographic fingerprints and quantification assay ensured the phytoequivalence and pharmacological effects of Cortex Moutan and was successfully applied to the quality control of Cortex Moutan.

[A chromatographic fingerprinting-based approach to evaluating the stability of production process of traditional Chinese medicine].

OBJECTIVE: To study a new method for the evaluation of the stability of production process of Traditional Chinese Medicines (TCM) based on chromatographic fingerprinting. METHOD: Taking the extraction process of Red Ginseng for example, the fingerprints of extracts produced with fluctuating processing factors, i.e. ratio of solvent to liquid, solvent concentration, extraction durations and extraction times, were acquired. Meanwhile, the similarity of fingerprinting based on Euclidean distance was calculated and each fingerprint was fitwith cubic spline. RESULT: Similarities of fingerprints of the extracts responded to the fluctuation of processing factors, and it showed that taking similarity as index, quantitative relation between chromatographic fingerprints and processing factors could be established. CONCLUSION: The method proposed in the paper was able to describe the quality of products resulting from fluctuating conditions in extracting process, and therefore, can be developed as a practicable strategy to evaluate the stability Traditional Chinese Medicines in production process.

[Transport mechanism of danshensu across Caco-2 monolayer model].

OBJECTIVE: To study the absorption mechanism of Danshensu by using Caco-2 monolayer model . METHOD: Caco-2 cell monolayer model was used to study the bi-direction transport of Danshensu. An LC-MS method was developed to measure the concentration of Danshensu in cell culture medium and calculate the apparent permeability coefficients (P(app)). The effects of time, drug concentration and inhibitor on the absorption of Danshensu were studied. RESULT: Transport of Danshensu was time and concentration dependent, and it was also effected by P-glycoprotein inhibitor. P(app) increased with time increase and tended to become saturated at some point. It, however, decreased while concentration of Danshensu increased. P(ratio) is larger than 1.5 . Verapamil can cause significantly effect on transport of Danshensu: P(app,A-B) increased and P(app,B-A) decreased . CONCLUSION: The absorption of Danshensu in Caco-2 cell model may be an active transportation mediated by P-glycoprotein transporter.

An easy, simple and rapid micellar electrokinetic capillary chromatography (MEKC) method was developed for the separation of two active components paeonol (PN) and paeoniflorin (PF) within 7 min. Capillary electrophoresis (CE) was performed using a 50.0 cm (42.0 cm to the detector window) x 75 microm i.d. fused-silica capillary. The optimal running buffer containing 10mM borate and 25 mM SDS at pH 9.54 was employed. The applied voltage 15 kV and the temperature 25 degrees C was used in CE separation. The linearities between peak areas and the concentrations of the analytes were investigated, and they exhibited excellent linear behavior over the investigated concentration ranges (R(2): 0.9945 for PN and 0.9992 for PF). The method was successfully applied to the determination of these two components contained in Cortex Moutan and 'Shuangdan' granule. The average recoveries ranged between 97.6 and 105.3% for PN and 95.3 and 106.1% for PF, respectively.

An approach to develop two-dimensional fingerprint for the quality control of Qingkailing injection by high-performance liquid chromatography with diode array detection.

An approach was proposed to develop two-dimensional fingerprint (2D fingerprint) by means of principal component analysis (PCA) of high-performance liquid chromatography with diode array detection (HPLC/DAD) data. The approach was applied to establish 2D fingerprints of various Qingkailing injections which were produced by different manufacturers and procedures. In comparison with common one-dimensional fingerprint (ID fingerprint) at fixed wavelength, 2D fingerprint compiled additional spectral data and was hence more informative. Principal component analysis of the 2D fingerprint data was performed in this study, and it led to an accurate classification of various samples on their manufacturers and procedures. The quality of Qingkailing samples was further evaluated by similarity measures and the same results were achieved. For comparison, four conventional ID fingerprints were also applied to the quality assessment for the same samples. Finally, we demonstrated that 2D fingerprint was a more powerful tool to characterize the quality of samples, and could be used to comprehensively conduct the quality control of traditional Chinese medicines.

Quantitative study of electrostatic and steric effects on physicochemical property and biological activity.

The physicochemical properties and biological activities are highly dependent on the electrostatic and steric effects of compounds. The characterization of these structural features will help to elucidate the quantitative structure-property/activity relationship (QSPR/QSAR). In this paper, two novel structural descriptors, lone-pair electrons index (LEI) and molecular volume index (MVI) were developed to quantify the molecular electrostatic and steric effects, respectively. One of the most attractive traits of these two descriptors is that the calculation is very easy, especially for LEI, which only takes the heteroatoms into account. Four data sets on diverse physicochemical properties and biological activities were selected as examples to evaluate the utility of these descriptors. One or two-variable multiple linear regression (MLR) models against the four data sets obtained such correlation results as follows: correlation coefficient R = 0.9998 and standard deviation s = 0.095 for the partition coefficient of halide benzenes; R = 0.9689 and s = 0.248 for the toxicity of heterocyclic nitrogen-containing compounds to miracidium; R = 0.9645 and s = 0.209 for the antifungal activity of benzyl alcohols to aspergillus niger; R = 0.9530 and s = 0.206 for the toxicity of substituted phenols against tetrahymena pyriformis. Leave-one-out cross validation was also carried out to evaluate the stability of each model and good results were obtained.

A sensitive and specific HPLC-MS method for the determination of sophoridine, sophocarpine and matrine in rabbit plasma.

A sensitive and specific method was developed for the determination of sophoridine (SRI), sophocarpine (SC) and matrine (MT) in rabbit plasma by HPLC-MS. After an administration of Kuhuang by injection, blood samples were collected and extracted with methanol. The extract solutions were analysed by HPLC-MS method. The separation was performed on a ZORBAX Extend-C18 column using methanol/water/diethylamine (50:50:0.07, v/v/v) as mobile phase. The quinolizidine alkaloids were detected by using mass spectrometry in the SIM mode. There was a good linear relationship between peak area and concentration of analytes over the concentration range of 13.2-995.0 ng mL(-1) for SRI, 7.0-530.0 ng mL(-1) for SC and 8.8-655.0 ng mL(-1) for MT, respectively. The absolute recovery of this method was more than 57% for SRI, 87% for SC and 91% for MT. The accuracy of assay was more than 90%. The limits of detection (LODs) were 6.8 ng mL(-1) for SRI, 3.5 ng mL(-1) for SC and 4.2 ng mL(-1) for MT, respectively. The limits of quantitation (LOQs) were 13.2 ng mL(-1) for SRI, 7.0 ng mL(-1) for SC and 8.8 ng mL(-1) for MT, respectively. The intra-day and inter-day coefficients of variation (RSDs) were less than 10.1, 6.3 and 5.8% for SRI, SC and MT, respectively. The developed method was applied to determine the concentration-time profiles of SRI, SC and MT in rabbit plasma after injection of Kuhuang.

Phthalide Lactones from Ligusticum chuanxiong inhibit lipopolysaccharide-induced TNF-alpha production and TNF-alpha-mediated NF-kappaB Activation.

The dried rhizome of Ligusticum chuanxiong Hort. (Umbelliferae) is a traditional Chinese medicine (TCM) herb for the prevention and treatment of inflammatory and cardiovascular diseases. However, the role of phthalide lactones from Ligusticum chuanxiong in the therapeutic actions is not yet fully understood. In the present study, two phthalide lactones from the herb, Z-ligustilide and senkyunolide A, were identified and characterized as inhibitors of lipopolysaccharide (LPS)-induced TNF-alpha production in monocytes. The results of gene expression studies showed that the observed TNF-alpha suppression was related to their inhibitory activity on TNF-alpha mRNA transcription. Furthermore, the two phthalides exhibited significant suppressive effects on TNF-alpha-mediated nuclear factor-kappaB (NF-kappaB) activation in reporter gene assays. Taken together, the results suggest that Z-ligustilide and senkyunolide A may have potential applications in the treatment of inflammation and related diseases based on their inhibitory activity on TNF-alpha production and TNF-alpha bioactivity. New insights into the therapeutic basis of the TCM herb, Ligusticum chuanxiong, are presented.

Simultaneous determination of major bioactive components in Qingkailing injection by high-performance liquid chromatography with evaporative light scattering detection.

High-performance liquid chromatography with evaporative light scattering detection (HPLC/ELSD) was established for simultaneous determination of seven major bioactive components of Qingkailing injection including adenosine, geniposide, chlorogenic acid, baicalin, ursodeoxycholic acid, cholic acid, and hyodeoxycholic acid. The proposed method was applied to analyze ten various Qingkailing injections and produced data with acceptable linearity, repeatability, precision and accuracy having a limit of detection (LOD) of 10-50 ng. In comparison with UV detection, HPLC/ELSD permits the determination of non-chromophoric compounds without prior derivatization, and shows good compatibility to the multi-components of complex analytes. The proposed method is a useful alternative for routine analysis in the quality control of traditional Chinese medicine.
