[Effects of water extractives of a qi and blood regulating prescription on early atherosclerosis of apolipoprotein E-deficient mice].

OBJECTIVE: To investigate whether the water extractives of regulating qi and blood prescription (WQBP) had effects on early atherosclerosis of apolipoprotein E-deficient mice (ApoE-mice) at the age of 19 weeks or not, and to explore the possible mechanisms. METHODS: Forty ApoE-mice, six weeks of age, were given high-fat diet and randomly divided into four groups: high-dose WQBP-treated group (360 mg/kg), low-dose WQBP-treated group (72 mg/kg), simvastatin-treated group (25 mg/kg) and untreated group, with ten mice in each group. Meanwhile, ten C57BL/6 mice of same genetic background were allocated to normal control group. Mice in the high- and low-dose WQBP-treated groups and simvastatin-treated group were administered with corresponding drugs from the 15 to 19 weeks. Mice in the untreated and normal control groups were administered with isovolumic water. Sacrificed at 19 weeks, the level of blood-lipid, the plaque construction, plaque integral, and the contents of plaque macrophages and vessel smooth muscle cells of the mice were analyzed by immunohistochemical method and a computer picture processing system. RESULTS: Compared to the untreated group, high-dose WQBP group could obviously decrease the level of low-density lipoprotein cholesterol (LDL-C). Simvastatin group could decrease the levels of LDL-C and total cholesterol (TC) (P<0.01). In high-dose WQBP-treated group and simvastatin-treated group, the thickness of fiber cap and the quantities of vessel smooth muscle cells increased (P<0.05), the quantities of plaque macrophages and the ratio of lipid and plaque reduced (P<0.01). CONCLUSION: WQBP and simvastatin can interfere in early atherosclerosis of ApoE-mice, attenuate and stabilize plaque in some extent. The mechanisms may include adjusting blood lipid, decreasing macrophage number and increasing the quantities of vessel smooth muscle cells.

[Fast and nondestructive discrimination of donkeyhide glue by near-infrared spectroscopy].

Near-infrared (NIR) diffuse reflectance spectroscopy and pattern recognition techniques were applied to develop a fast identification method of a Chinese patent medicine-donkeyhide glue. Samples from different manufactures (eight genuine samples and six counterfeits) were collected, and their NIR spectra were obtained. NIR spectra were pretreated with multiplicative signal correction (MSC) and wavelet transformation to diminish baseline offset. Similarity and Mahalanobis distance methods were separatemy used to qualify donkeyhide glue. For the similarity calculation, spectra of the two real ones were selected as standards, and then others were compared with the standards to obtain the match value. All the samples were rescanned once for the Mahalanobis distance methods, and totally twenty eight spectra were separated into three sets for cross-validation. The two methods can both distinguish the counterfeits of donkeyhide glue successfully. The proposed method is accurate and robust, and could be used in the discrimination of other traditional Chinese medicines.

[Study on real-time control of extraction procedure of Furitus gardenia by near infrared spectroscopy].

A new approach to the real-time control of extraction procedures based on fiber optic near infrared spectroscopy (NIR) was described. Various extraction samples of gardenia were rapidly scanned by NIR, and the SIMCA monitoring model was developed. The model was applied to evaluate the extraction procedures of 26 unknown samples, and all the samples from abnormal procedures were successfully identified. It is suggested that near infrared spectroscopy, combined with SIMCA method, is a rapid, convenient and efficient tool for real-time procedure monitoring, which needs no quantitative determination and can effectively monitor whether the current procedure is in normal or not. The presented approach, therefore, offers a new way for the quality control in real-time of traditional Chinese medicine.

[Determination of salvianolic acid B and paeoniflorin in Shuangdan granules by HPLC].

OBJECTIVE: To develop a method for the determination of salvianolic acid B and paeoniflorin in Shuangdan granules. METHOD: The chromatographic separation was performed on a Zorbax SB C18 column with a linear gradient elution of phosphoric acid-water (0.05:100) and phosphoric acid-acetonitrile-methanol (0.05:4:96) was used. The flow rate was 1 mL x min(-1) and column temperature was set at 25 degrees C. The UV detection wavelength was set at 230 nm. RESULT: The linear ranger was of 0.710-22.7 microg for salvianolic acid B and 0.103-3.30 microg for paeoniflorin. Three batches of Shuangdan granules were analyzed, and the content of salvianolic acid B ranged from 6.46-11.6 mg x g(-1) and paeoniflorin ranged from 1.44-1.78 mg x g(-1). CONCLUSION: The method was accurate, reproducible, reliable, and could be used for quantitative control of Shuangdan granules.

Background correction in near-infrared spectra of plant extracts by orthogonal signal correction.

In near-infrared (NIR) analysis of plant extracts, excessive background often exists in near-infrared spectra. The detection of active constituents is difficult because of excessive background, and correction of this problem remains difficult. In this work, the orthogonal signal correction (OSC) method was used to correct excessive background. The method was also compared with several classical background correction methods, such as offset correction, multiplicative scatter correction (MSC), standard normal variate (SNV) transformation, de-trending (DT), first derivative, second derivative and wavelet methods. A simulated dataset and a real NIR spectral dataset were used to test the efficiency of different background correction methods. The results showed that OSC is the only effective method for correcting excessive background.

[The effect of preparation method on the quality of shen-mai injection].

OBJECTIVE: To investigate the effect of different preparation method on the quality of Shen-mai injection. METHOD: The Shen-mai injection samples were prepared using three different methods. Fingerprints of Shen-mai extracts red ginseng, and its intermediates were obtained using an HPLC analytical procedure. The contents of ginsenoside Rg1, Rc and Rb1, and the gross saponins of Shen-mai extract were quantitatively mensured with HPLC procedures. RESULT: There was significant difference in fingerprints and chemical contents of the injections prepared by the three different methods. CONCLUSION: The quality of Shen-mai injection was greatly influenced by the preparation method. HPLC fingerprinting method can be applied for the determination of the Shen-mai preparations.

[Simultaneous determination of 4 anthraquinones in Kuhuang injection by RP-HPLC].

OBJECTIVE: To develop a new high-performance liquid chromatographic method for simultaneous quantitative determination of 4 anthraquinones in Kuhuang injection. METHOD: The chromatographic separation was performed on a Lichrospher C18 column (4.6 mm x 250 mm, 5 microm,), A linear gradient elution of A (CH3COOH: H2O = 1:100) and B (CH3COOH: H2O: CH3CN = 1:20:80) was used. The flow rate was 0.8 mL x min(-1) and column temperature was set at 35 degrees C. The UV detection wavelength was set at 254 nm. RESULT: The recoveries of rhein, emodin, chrysophanol and physcion were 98.9%, 100.5%, 102.5% and 99.0%, respectively. Their linear ranges were 0.0875-1.75 microg for rhein, 0.0825-1.65 microg for emodin, 0.159-3.17 microg for chrysophanol and 0.0525-1.05 microg for physcion, respectively. CONCLUSION: The method was accurate, repeatable and suitable to determine the contents of 4 anthraquinones in Kuhuang injection.

[A method for predicting activity of traditional Chinese medicine based on quantitative composition-activity relationship of neural network model].

OBJECTIVE: To study a method for evaluating the quality of traditional Chinese medicine (TCM) according as their activity. METHOD: Combined with partial least squares (PLS), BP and RBF neural networks were selected to establish the model of quantitative composition-activity relationship (QCAR) due to their strong approximation capabilities for nonlinear function respectively. The activity of TCM was predicted with the QCAR model, and the quality of TCM was evaluated according to the predicted activity. RESULT & CONCLUSION: The proposed method was applied to evaluate the quality of Chuanxiong. The results indicated that, in the indexes including training error, prediction error and correlation coefficient, the established model is better than the model established by principal component regression or PIS regression. The new model can accurately represent the complicated nonlinear relationship between the components and the bioactivity of Chuanxiong. Consequently, this method has potential to evaluate the quality of TCM according to bioactivity.

[Applications of chitosan and its derivatives in pharmaceutical industry of Chinese medicine].

Recently, chitosan has been widely used as a clarifier in clarification procedure for many kinds of TCM, which is better than alcohol in retaining active substances, such as polysaccharids, as well as the removal of heavy metals. Because it is a biocompatible polymer with low toxicity and can be biodegradable in vivo, chitosan with its derivatives is one of the ideal materials for drug controlled release systems to enhance the efficacy and reliability of TCM drug therapy. On the other hand, chitosan, the only natural alkalic polysaccharid, also has several therapeutic effects such as anti-bacteria, antipyrotic and ulcer healing. Chitosan and its derivatives wide application prosperity in pharmaceutical industry of Chinese medicine.

[A new approach to the fast measurement of content of amino acids in Cordyceps sinensis by ANN-NIR].

A new method for fast determining the content of amino acid in Cordyceps sinensis by means of near infrared (NIR) spectroscopy was developed. Colorimetry was first employed to measure the actual content of amino acid in Cordyceps sinensis. BP neural network was introduced to model the quantitative correlations between the NIR spectra and the contents of glycine, arginine and total amino acid. By comparing several preprocessing procedures and wavelength ranges, the optimal models could be obtained in the range of 7501.7-6097.8 cm(-1) and 5453.7-4246.5 cm(-1) with first derivative NIR spectra. Standard errors of prediction set (RMSEP) for glycine, arginine and total amino acid were 0.08, 0.07 and 0.36, respectively. The ultimate experimental results indicated that the proposed artificial neural network model was far superior to those of partial least square regression (PLS) and principal component regression (PCR). As an effective nonlinear multivariate calibration strategy, this paper could offer a new approach to the fast measurement of content of chemical components in traditional Chinese medicine by NIR spectroscopy.

[Identification of the habitat of Ligusticum chuangxiong with chromatographic fingerprinting].

OBJECTIVE: To establish the chromatographic fingerprinting for identifying the habitat of Ligusticum chuangxiong. METHOD: HPLC system was applied to obtain the chromatograms of L. chuangxiong samples from different areas, and 15 peaks were measured from the chromatograms. Then some computer-based methods including principle component analysis, clustering analysis, similarity calculation and fisher factor analysis were applied for data analysis. RESULT: There was obvious difference among chromatographic fingerprints of L. chuangxiong samples from different areas. The 15 measured peaks could be used as the fingerprint features. CONCLUSION: Chromatographic fingerprinting can be used for identifying the habitat of L. chuangxiong.

[Study on the solubility changes of Rhizoma Coptidis' main chemical constituents influenced by the proportion of Rhizoma Coptidis and Fructus Evodiae].

OBJECTIVE: To investigate the relationship between the solubility of Rhizoma Coptidis' main chemical constituents and the proportion of Rhizoma Coptidis and Fructus Evodiae. METHOD: 9 samples were selected in which the proportion of Rhizoma Coptidis and Fructus Evodiae were 1:0, 6:1, 3:1, 2:1, 1:1, 1:2, 1:3, 1:6 and 0:1, respectively. The samples' water-extracts were analyzed by RP-HPLC. Then solubility changes of eight chemical constituents were calculated, and the regression equations were established. RESULT: Among the eight chemical constituents, one was independent on the change of proportion of Rhizoma Coptidis and Fructus Evodiae, six having linear relationship. A new constituent was detected, and there was nonlinear relationship between its quantity and the proportion of the couple. CONCLUSION: The solubility of Rhizoma Coptidis' main chemical constituents declines almost in the same scale when Rhizoma Coptidis and Fructus Evodiae are used as a couple, and a new constituent has been detected which can't be found in the mono-extraction.

[Some engineering problems on developing production industry of modern traditional Chinese medicine].

Based on the review of some engineering problems on developing modern production industry of Traditional Chinese Medicine (TCM), the differences of TCM production industry between China and abroad were pointed out. Accelerating the application and extension of high-tech and computer integrated manufacturing system (CIMS) were suggested to promote the technology advancement of TCM industry.

[A novel fingerprint method for quality evaluation of Chinese medicinal plants based on analytical data visualization].

OBJECTIVE: To investigate new method for evaluating the quality of Chinese Medicinal Plants (CMP). METHOD: A visualization technique for representing instrumental analytical data was developed by applying the fundamental of Data Visualization, with Principal Component Analysis (PCA) and spatial projection transformation, original IR spectral data were projected into a low-dimensional subspace so that the dimensionality of original data space was decreased and tiny fingerprint features were extracted. The data set in the subspace was visualized by means of two-dimensional grayscale images. Consequently, the characteristic fingerprint for appraising the quality of CMP was obtained. RESULT: 42 mulberry root-bark samples from three different quality classes were identified with the proposed method, which showed that the fingerprint images had satisfactory resolution and classification accuracy as high as 90.5%. CONCLUSION: The proposed method is a useful technique for appraising the quality of CMP.
