[Transcriptional differences between a heterokaryon and its segregants of Fusarium oxysporum f. sp. vasinfectum].

In order to elucidate the mechanism of fungal heterokayosis, a wild type strain of Fusarium oxysporum f. sp. vasinfectum was isolated from the cotton field in Anyang, Henan Province. Through single hyphal-tip isolation, a heterokaryon, Ag149, was obtained, and its two different phenotypic segregants, Ag149-I and Ag149-III, were separated from the mutated sectors on colony of the heterokaryon. They have remarkable differences on color of colony, morphology of hypha and pathogenicity. After analyzing by RAPD on their nuclear DNA with 100 random primers, no polymorphic difference was found among them. On going to find the different expressed gene, the mRNA differential display method was performed. Two kinds of reverse transcriptase AMV and MMLV, and a kit which consists of three kinds of 3' terminal anchor primers and eight kinds of 5' terminal arbitrary primers were used in differential display PCR (DD-PCR). Total RNA as template was reverse transcribed into corresponding cDNA by 3' terminal anchor primers, and the cDNA were amplified by polymerase chain reaction with a set of one same 3' anchor primer and one 5' arbitrary primer. The PCR products were then resolved on denaturing polyacylamimide gel, and the cDNA bands were visualized by silver staining. Among the 144 PCR products, 19 differentially expressed cDNA fragments ranged from 300 bp to 700 bp were purified. All of them were ligated to pGEM-T vector respectively for sequencing and Rev-Northern blotting. Two cDNA fragments (G5 and C6) were observed to be positive after Rev-Northern blotting. The C6 was highly expressed in the heterokaryon Ag149 and its segregant Ag149-I. It is 564 bp in length and can be predicted 77 amino acids from the beginning of the 3rd to 233th base, and then searched from GenBank. The amino acid sequence of C6 shared homologies with the 6th subunit of NADH dehydrogenase found in some bacteria, plants and animals at 30% - 70% level. While the G5 was highly expressed in Ag149 and its segregant Ag149-III. It is 432 bp in length and can be predicted 101 amino acids from the beginning of the 2nd to 304th base, and the amino acid sequence is 35% homologies comparing with the tetracycline efflux protein (OtrB) of Streptomyces rimosus. We also checked these two kinds of the pGEM-T vector harboring cDNA fragments (C6 and G5) by Southern blotting with their nuclear DNA and mitochondrial DNA (mtDNA) separately. The positive identification signals only appeared from nuclear DNA,and it addressed that C6 and G5 locate on their nuclear genome. The result indicated that the difference between the heterokaryon and its segregants is distinct on gene transcriptional level. Thus a molecular evidence for the formation of heterokaryon in filamentous fungi was provided.

[Expression of hormone-sensitive lipase mRNA in diet-induced obesity resistant rats].

OBJECTIVE: In order to explore the expression of hormone-sensitive lipase (HSL) mRNA in white adipose tissue of diet-induced obesity-resistant (DIO-R) rats. METHODS: Fifty male Sprague-dawley (SD) rats were randomly divided into control group and high-fat group and they were fed with basic diet and high-fat diet respectively for 13 weeks. DIO-R and DIO rats were selected according to their body weight and the quantity of energy intake, then observing changing of the body fat content, the level of serum growth hormone was determined by radioimmunoassay and the level of the HSL mRNA in white adipose tissue was determined by RT-PCR. RESULTS: The body fat content of DIO-R rats were lower than those of DIO rats (P < 0.05). The level of serum growth hormone of DIO-R rats were higher than those of DIO rats (P < 0.05). High fat diets increased the level of the HSL mRNA in DIO-R rats. CONCLUSION: Increasing expression of HSL in white adipose tissue may play a part of role in resisting diet-induced obesity of rats.

Efficacy of acupuncture and moxibustion in treating Bell's palsy: a multicenter randomized controlled trial in China.

BACKGROUND: Bell's palsy involves acute facial paralysis due to inflammation of the facial nerve. Acupuncture and moxibustion (acu-moxi) is beneficial in treating facial palsy. In order to verify the efficacy of acu-moxi on Bell's palsy, a randomized single-blind, multicenter clinical trial was performed. METHODS: A total of 480 patients from four clinical centers were involved in this trial, of whom 439 completed the trial and 41 did not. All patients were randomly assigned to either the control group or to one of two treatment groups. The control group was treated with prednisone, vitamin B1, vitamin B12, and dibazole; the treatment groups were treated either with acu-moxi alone or in combination with prednisone, Vitamin B1, vitamin B12, and dibazole. Symptoms and signs, the House-Brackmann scale, and facial disability index (FDI) scores were assessed and determined both pre- and post-treatment to evaluate the effectiveness of the treatment methods. RESULTS: The characteristics of the control and two treatment groups were comparable without statistically significant differences before treatment. There were significant differences between the control and treatment groups after treatment (chi(2) = 15.265, P = 0.018). According to evaluations based on the House-Brackmann scale and FDI scores, the effectiveness of treatment in the two treatment groups was better than in the control group and was most effective in patients receiving acu-moxi treatment alone (Z = -2.827, P = 0.005). CONCLUSION: The efficacy of acu-moxi treatment for Bell's palsy is verified scientifically.

[A study on the association between tea consumption and stroke].

OBJECTIVE: The aim of the study was to investigate the association between tea consumption and stroke. METHODS: A cross sectional study was conducted in autumn, 1998. The subjects were from 12 provinces in China. 15 groups of populations were selected by cluster randomized sampling and each group had about 1000 persons, aged from 35 to 60 years old. 14 212 subjects had complete data for analysis. Data regarding tea drinking would include drinking status, dose and type of teas. Logistic regression was used to analyze the association between tea consumption and stroke. RESULTS: There was a strong inverse correlation between tea drinking and stroke after adjusting other risk factors of stroke (P < 0.05). The odds ratio (OR) of stroke was 0.60 [95% confidence interval (CI): 0.42 - 0.85] for subjects who drank tea compared to those who did not. Increased amount of tea consumption per month was associated with decreased stroke prevalence. The association for tea consumption over 150 gram per month and stroke was statistically significant (P < 0.05) with an OR value of 0.56 (95% CI: 0.36 - 0.89). Analytical results indicated that the OR value was 0.24 (95% CI: 0.06 - 1.01) for black tea and other tea (P = 0.05). The OR value was 0.35 (95% CI: 0.18 - 0.72) for green tea (P < 0.01), and 0.75 (95% CI: 0.51 - 1.11) for jasmine tea (P > 0.05). CONCLUSION: Tea drinking was independently associated with prevalence of stroke which might play a role in the prevention of the disease.

[Regularity of flourine release from fluorine-rich coal combustion in the fluorine poisoning area].

Laboratory experiments and in situ measurements inside farmer's houses indicate that the fluorine concentration of indoor air was positively related with the fluorine content in the consumed coal (R = 0.612, P<0.01), but fluorine content in the coal had no effect on the release rate of coal fluorine under the usual combustion temperature of about 1000 degrees C. The release rate of coal fluorine varied versus combustion temperature ranging from 200 to 1200 degrees C, following a "S" curve pattern, increasing slowly under 300 to 700 degrees C, but quickly under 700 to 1000 degrees C, and then slowly again before finally reaching 100% under 1100 to 1200 degrees C, this curve can be fit by Logistic equation. The release rate of coal fluorine usually depends upon the existence status of fluorine and the chemical and mineral components of the coal. The fluorine in soft coal or anthracite from Guizhou is released more quickly than in bone coal from Southern Shanxi, with the former almost completely released under 1100 degrees C but the later almost completely released under 1200 degrees C. The combustion temperature of civil stoves is at most 1000 degrees C, under which averagely 86.9% of the fluorine in coal from Guizhou and 80.6% of that in bone coal from Southern Shanxi is released.

[Physiologic response to proportional assist ventilation in patients with acute exacerbation of chronic obstructive pulmonary disease].

OBJECTIVE: To investigate the effect of different assist levels during proportional assist ventilation (PAV) on patients with acute exacerbation of chronic obstructive pulmonary disease (COPD). METHODS: Nine patients with acute exacerbation of COPD received different levels of assist ventilation during PAV. The change of inspiratory muscle effort and breathing pattern of the patients were observed. RESULTS: (1) The average tidal volume (V(T)), minute ventilation (V.(E)) and respiratory frequency (RR) were slightly higher during PAV, as compared to spontaneous breathing (SB). The change of breathing pattern of the patients was not significant at different assist levels during PAV. (2) The transdiaphragmatic pressure (Pdi), the pressure-time product (PTP) and work of breathing (Wi) reduced significantly during PAV as compared with SB (differently reduced 8.36 cm H2O, 11.49 cm H2O.s(-1).L(-1) and 0.53 J/L). The change in Pdi, PTP and Wi was small among different levels during PAV. (3) Dyspnea was improved during PAV. CONCLUSIONS: The present study confirms the feasibility of noninvasive ventilation PAV in treating patients with acute exacerbation of COPD. Based on the patient's assessment, the ventilatory support level which the patients could comfortably tolerate was (57 +/- 11)%. noninvasive ventilation PAV set at a level of comfort determined by the patient can unload inspiratory muscles; at the comfortable level of ventilatory support, Wi, Pdi and PTP decreased by 57%, 72% and 65% respectively. It also improved breathing pattern and dypnea.

[Simultaneous determination of the residues of pyrethriods, organochlorines and polychlorinated biphenyl congeners in unpolished rice by gas chromatography with gel permeation chromatography purification].

The separation behaviors of organochlorine pesticides, pyrethriod pesticides, polychlorinated biphenyl congeners (PCBs) and unpolished rice matrixes were systematically investigated with gel permeation chromatography (GPC) because of its outstanding capability in removing lipid and pigments from sample solutions. A separation method was proposed for the quantitative determination of 35 pesticides and PCBs in unpolished rice by GPC and gas chromatography simultaneously. Samples were extracted with ethyl acetate, purified by GPC, and then determined by gas chromatography and electron capture detection (GC-ECD). With external standard method, the recoveries of 3 standard addition levels of low, medium, high concentrations were 70.1% - 107.4%, 70.3% - 109.9%, 70.8% - 109.5%, respectively with relative standard deviations from 2.3% to 13.3%. The detection limits were 0.07 microg/kg for organochlorines and PCBs, and 0.44 microg/kg for pyrethriods.

[Fluorescence study by simulating the metabolizability of carotenoid and porphyrin during cancer development].

The purpose of the present work was to investigate the effects of carotenoid on the spectroscopic examination of human blood serum by simulating the metabolizability of the carotenoid and porphyrin during the development of cancer and by studying the fluorescence of them. Blood serum was prepared with blood sample from elbow vein and the luminescence center of blood serum was studied using photo induced-luminescence techniques. The major factors of blood serum fluorescence were electron transitions in serological protein, carotenoid and porphyrin molecules with conjugated groups. The intensity of the fluorescence spectrum at 530 nm was enhanced while the minimum intensity at 614 nm was also enhanced with the content of carotenoid increasing, thus the value of diagnostic symbol I decreased, namely the veracity of cancer examination decreased.

[Determination of mercury in battery by microwave digestion cold atomic absorption spectrometry].

In this paper, a method of microwave digestion followed by cold atomic absorption spectrometry (CAAS) for the determination of mercury in battery was developed. Microwave digestion conditions and operational conditions of CAAS were optimized. The digestive solution was a mixture of hydrochloric acid-sulfuric acid (4:1, volume ratio) and the digestive time was 3 min at a microwave power of 1.0 MPa. The reaction medium was 0.5 mol x L(-1) sulfuric acid at room temperature. The characteristic concentration was 0.25 microg x L(-1). Five battery samples were determined, the recoveries were from 99.9% to 113%, and the relative standard deviations were from 4.85% to 11.9% (n = 6). The analytical results were satisfactory. The method is fast, simple, accurate and solvent-saving.

[Alcohol consumption and incidence of ischemic stroke in male Chinese].

OBJECTIVE: To assess the relationship between incidence of ischemic stroke and alcohol consumption. METHODS: The information on alcohol consumption and other cardiovascular risk factors such as SBP, DBP, TC was collected among 10 populations in northern and southern parts of China according to the international standardized methods. A total number of 30 560 men and women aged 35 - 59 were enrolled and followed up for an average of 15.2 years. Complete data on 12,352 men was analyzed. RESULTS: The risk on ischemic stroke incidence showed a graded association. Compared with nondrinkers, the relative risks of incidence of ischemic stroke for those who drank alcohol less then 15 gram per day, 15-30 gram per day, 30-60 gram per day, above 60 gram per day were 0.86 (95% confidence interval 0.57-1.27), 1.20 (0.87-1.65), 1.26 (0.86-1.84) and 1.96 (1.30-2.93), respectively. CONCLUSION: Our study revealed that in male Chinese, heavy alcohol intake significantly increased the risk while mild alcohol drinking did not increase the risk of ischemic stroke.

Hypothalamus-brain stem circuitry responsible for vagal efferent signaling to the pancreas evoked by hypoglycemia in rat.

Circulating glucose levels significantly affect vagal neural activity, which is important in the regulation of pancreatic functions. Little is known about the mechanisms involved. This study investigates the neural pathways responsible for hypoglycemia-induced vagal efferent signaling to the pancreas and identifies the neurotransmitters involved. Vagal pancreatic efferent nerve activities were recorded in anesthetized rats. Insulin-induced hypoglycemia, a decrease of blood glucose levels from 114 +/- 5 to 74 +/- 6 mg dl(-1), stimulated an increase in pancreatic efferent nerve firing from a basal rate of 1.1 +/- 0.3 to 19 +/- 3 impulses 30 s(-1). In contrast, vagal primary afferent neuronal discharges recorded in the nodose ganglia were unaltered by systemic hypoglycemia. Vagal afferent rootlet section plus splanchnicotomy had no effect on hypoglycemia-induced vagal efferent firing, suggesting a central site of action. Decerebration reduced the increase in nerve firing stimulated by hypoglycemia from 21 +/- 4 to 9.6 +/- 2 impulses 30 s(-1). Chemical ablation of the lateral hypothalamic area, but not the arcuate nucleus, inhibited pancreatic nerve firing evoked by hypoglycemia. Microinjection of the orexin-A receptor antagonist SB-334867 into the dorsal motor nucleus of the vagus (DMV) inhibited pancreatic nerve firing evoked by insulin-induced hypoglycemia by 56%. In contrast, injection of orexin-A (20 pmol) into the DMV elicited a 30-fold increase in pancreatic nerve firing. We concluded that systemic hypoglycemia stimulates pancreatic efferent nerve firing through a central mechanism. Full expression of pancreatic nerve activities during hypoglycemia requires both the forebrain and the brain stem. In addition to activating neurons in the brain stem, central neuroglucopenia activates subpopulations of neurons in the lateral hypothalamic area that contain orexin. The released orexin acts on DMV neurons to stimulate pancreatic efferent nerve activities and thus regulate pancreatic functions.

A deletion mutation in the betaA1/A3 crystallin gene ( CRYBA1/A3) is associated with autosomal dominant congenital nuclear cataract in a Chinese family.

Congenital cataracts are an important cause of blindness worldwide. In a family of Chinese descent, a dominant congenital nuclear cataract locus was mapped to chromosome 17q11.1-12. The maximum LOD score, 2.49, at recombination fraction 0, was obtained for marker D17S1294. The results of both linkage and haplotype analyses defined a disease-gene to an 11.78-cM region harboring the gene coding for betaA1/A3 crystallin ( CRYBA1/A3). Mutation analysis of the CRYBA1/A3 gene identified a 3-bp deletion in exon 4, which cosegregated with the disease risk in this family and was not observed in 100 normal chromosomes. This mutation resulted in the deletion of a highly conserved glycine at codon 91 (DeltaG91) and could be associated with an incorrect folding of betaA1/A3 crystallin. It highlights the physiological importance of crystallin and supports the role of CRYBA1/A3 in human cataracts formation.

Homogeneous assays for adenosine 5'-monophosphate-activated protein kinase.

Allosteric activation of 5(')-AMP-activated protein kinase (AMPK) is currently of interest as an approach for the treatment of metabolic disorders because AMPK plays multiple roles in glucose and lipid metabolism. The availability of ultrafast, ultrasensitive, and robust assays suited to high-throughput screening (HTS) is key to obtaining small-molecule AMPK activators. In the absence of high-affinity and selective antiphospho Ser/Thr antibodies for AMPK substrates, we have developed two homogeneous AMPK assays with the commercially available antibody Anti-pS(133)-CREB and an engineered peptide ACC-CREBp. Anti-pS(133)-CREB antibody was raised against the phospho-CREB peptide derived from cAMP response element binding protein (CREB). ACC-CREBp was a variant (Arg to Pro) of ACC-CREB, a hybrid peptide consisting of a 9-amino-acid peptide from rat acetyl-CoA carboxylase (ACC), CREB peptide, and the addition of two hydrophobic Leu residues. ACC-CREBp showed increased suitability as a substrate for AMPK, eliminated phosphorylation by PKA, and preserved antibody binding. The homogeneous time-resolved fluorescence and AlphaScreen AMPK assays were developed using both Anti-pS(133)-CREB antibody and ACC-CREBp that are either labeled with a fluorescent probe or linked to a photoactivated bead, respectively. Thus, ACC-CREBp phosphorylation can be measured as a signal change resulting from the formation of antibody-antigen complex. This approach of adapting known antibody and antigenic peptide pairs to monitor alternate Ser/Thr kinases may be of general use.

Hypothalamic regulation of pancreatic secretion is mediated by central cholinergic pathways in the rat.

The vago-vagal reflex plays an important role in mediating pancreatic secretion evoked by cholecystokinin and non-cholecystokinin-dependent luminal factors. We hypothesize that the vago-vagal reflex mediating pancreatic secretion in the rat is under central control and regulated by cholinergic pathways in the hypothalamus. To test this hypothesis, we demonstrated that chronic decerebration decreased basal pancreatic enzyme secretion from 318 +/- 12 to 233 +/- 9 mg h-1 and reduced the net increase in pancreatic secretion stimulated by intraduodenal infusion of 5 % peptone and hypertonic NaCl by 54 % and 45 %, respectively. Intracerebroventricular administration of methscopolamine (MSCP, 50 nmol (5 mul)-1), a blood-brain barrier-impermeant cholinergic muscarinic receptor antagonist, evoked results similar to those achieved by chronic decerebration. To localize the sites of action, we demonstrated that microinjection of MSCP (20 nmol) into the lateral hypothalamic nucleus or the paraventricular nucleus resulted in inhibition of both basal pancreatic protein secretion and luminally stimulated pancreatic secretion by 48 % and 52 %, respectively. Intracerebroventricular injection of hemicholinium-3 at doses known to deplete the endogenous ACh store produced similar inhibitory results. In addition, microinjection of ACh (5 pmol) or the muscarinic M1 receptor agonist McN-A-343 (30 ng) into the lateral hypothalamic nucleus increased pancreatic secretion over basal levels by 46 % and 40 %, respectively. Selective lesions of lateral septal cholinergic neurons decreased basal pancreatic secretion and inhibited peptone-induced pancreatic secretion by 30 %. Destruction of the lateral parabrachial nucleus produced a 44 % inhibition of peptone-induced pancreatic section. Finally, microinjection of glutamate into the lateral septum or the lateral parabrachial nucleus stimulated vagal pancreatic efferent nerve firings from a basal level of 0 +/- 0.5 impulses (30 s)-1 to 4.5 +/- 0.5 and 14 +/- 2 impulses (30 s)-1, respectively, and pancreatic protein output increased 50 % and 84 % over basal levels. Administration of MSCP to the paraventricular nucleus eliminated these effects. These observations suggest that cholinergic neurons of the lateral septum and lateral parabrachial nucleus regulate pancreatic secretion. Further, cholinergic input from the lateral parabrachial nucleus to the hypothalamus plays a major role in the modulation of vagal pancreatic efferent nerve activity and pancreatic secretion evoked by the vago-vagal reflex.

Functional repair of the corticospinal tract by delayed transplantation of olfactory ensheathing cells in adult rats.

Adult rats were trained to use their forepaws to retrieve a piece of food. Destruction of the dorsal corticospinal tract on one side at the level of the first cervical segment abolished the use of the ipsilateral forepaw for retrieval for at least 6 months after operation. Where a variable amount of the corticospinal tract was spared, there was a proportionate persistence of retrieval. In lesioned rats that had shown no retrieval for 8 weeks after operation, a suspension of olfactory ensheathing cells was injected into the lesion site. Starting between 1 and 3 weeks after transplantation, all rats with transplants bridging the lesion site resumed retrieval by the ipsilateral forepaw. Biotin dextran anterograde tracing shows regenerating corticospinal axons crossing the bridge, traveling caudally for approximately 10 mm in the distal part of the corticospinal tract and forming terminal arborizations in the spinal gray matter. Functional recovery can occur when only approximately 1% of the corticospinal tract axons are present.

Artemisinin derivatives bearing Mannich base group: synthesis and antimalarial activity.

Novel artemisinin derivatives bearing Mannich base group were prepared and tested for their antimalarial activity. These water-soluble artemisinin derivatives were more stable than sodium artesunate and few compounds were found to be more active against Plasmodium berghei in mice than artesunic acid by oral administration. Two most potent derivatives 17b and 17d were examined for their antimalarial activity against Plasmodium knowlesi in rhesus monkeys.

Transplanted olfactory ensheathing cells promote regeneration of cut adult rat optic nerve axons.

Transplantation of olfactory ensheathing cells into spinal cord lesions promotes regeneration of cut axons into terminal fields and functional recovery. This repair involves the formation of a peripheral nerve-like bridge in which perineurial-like fibroblasts are organized into a longitudinal stack of parallel tubular channels, some of which contain regenerating axons enwrapped by Schwann-like olfactory ensheathing cells. The present study examines whether cut retinal ganglion cell axons will also respond to these cells, and if so, whether they form the same type of arrangement. In adult rats, the optic nerve was completely severed behind the optic disc, and a matrix containing cultured olfactory ensheathing cells was inserted between the proximal and distal stumps. After 6 months, the transplanted cells had migrated for up to 10 mm into the distal stump. Anterograde labeling with cholera toxin B showed that cut retinal ganglion cell axons had regenerated through the transplants, entered the distal stump, and elongated for 10 mm together with the transplanted cells. Electron microscopy showed that a peripheral nerve-like tissue had been formed, similar to that seen in the spinal cord transplants. However, in contrast to the spinal cord, the axons did not reach the terminal fields, but terminated in large vesicle-filled expansions beyond which the distal optic nerve stump was reduced to a densely interwoven mass of astrocytic processes.
