Global genomic approaches to the iron-regulated proteome.

Iron interacts with cells to regulate the proteome through complex effects on gene expression. In simple organisms such as bacteria and yeast, intra- and extra-cellular iron influences gene expression through defined signal transduction pathways. In higher organisms, effects are probably mediated at the transcriptional level through secondary effects of reactive oxygen species, while post-transcriptional effects operate through well-defined pathways involving iron-regulatory proteins. To investigate the impact of iron levels on gene expression and the proteome, approaches such as differential display and subtractive hybridization have the advantage of surveying the entire genome. However, they are technically demanding and have given way to microarray techniques. To date, numerous microarray experiments with various organisms have not yielded any definitive picture of the role of iron. Common themes throughout such studies are that both iron excess and iron depletion influence expression of proteins related to energy metabolism, cell proliferation, matrix structure, and the metabolism of iron itself. That no consistent set of genes is involved from one study to the next probably results both from the uncertainties inherent in the technique and the biological variability of the systems under study. We briefly describe two types of iron-dependent microarray experiments from our laboratory to examine major cellular targets of iron toxicity. Using Affymetrix oligonucleotide arrays with cardiac cells, we found several hundred genes whose mRNA levels were affected by iron, including an increase in several genes responding to oxidative stress and a decrease in several kinases and phosphatases. In a simpler experiment using a human liver cell line with a limited cDNA array, we targeted 13 genes affected by iron chelation. Metabolic pathway analysis shows links of 5 of these through phorbol ester responsiveness, and additional links through prostaglandin E2. We conclude that definitive understanding of the complex iron-regulated proteome requires global gene approaches and rigorous interlaboratory standardization.

[Therapy of Duchenne muscular dystrophy with umbilical cord blood stem cell transplantation].

OBJECTIVE: To analyze a Duchenne muscular dystrophy(DMD) patient's muscular regeneration, dystrophin expression and locomotive variation before and after he underwent umbilical cord blood stem cell transplantation in order to assess the therapeutic effect. METHODS: A 12-year-old DMD boy who could not walk for 3 years was confirmed by gene analysis and dystrophin protein immune test on his muscle. He had no other chronic disease. By HLA matching, a piece of umbilical cord blood stem cell with 6 HLA sites matching to the boy was found in Guangdong Umbilical Cord Blood Bank. The number of the nucleated cells of the umbilical cord blood stem cell was 24.08x 10(8). After pretreatment for the DMD boy with busulfan, cyclophosphamide and rabbit anti-human thymocyte globulin, the allergenic cord blood stem cells were transplanted into him by intravenous injection. Cyclosporin A, methylprednisolone, MMF, prostaglandin E1 and ganciclovir were given after the transplantation. At the same time, Gran, the granulocytic cell stimulating factor, and gamma globulin were administered. The biochemistry profile including serum creatine kinase (CK), the reconstruction of blood making, the deletion exon of DMD gene, the regenerating muscles, the dystrophin protein expression, and the locomotive function of the DMD boy were tested regularly. RESULTS: (1) The white blood cells (WBC) of peripheral blood decreased gradually to zero after pretreatment. In a period of 15 days after transplantation, the neutrophil increased to 0.5x 10(9)/L; at 25 days, WBC increased to normal level. Blood platelet was more than 20x 10(9)/L at 22 days. The hemoglobin rose to 85-100 g/L. At 140 days, sternal puncture revealed the rapid growth of neutrophil, blood platelet and hemoglobin. (2)At 140 days, the blood type of the DMD boy transformed from type O to type AB (the donor's blood type being AB). There was no grafe versus host reaction. (3) At 18, 30, 43, 55, 74 and 233 days after transplantation, the PCR-short tandem repeat test of the boy's peripheral blood DNA showed that his genotype was completely the same as the donor's. The results of PCR-short tandem repeat tests of the bone marrow cells DNA by sternal puncture at 140, 183 and 235 days were the same as those of the blood DNA. (4) At 60 days, DMD gene analysis by PCR showed that the defected DMD gene (exon 19 deletion) had been corrected by the umbilical cord stem cells transplantation. (5) At 75 days, the biopsy of calf muscle showed there were myoblast cells and muscular tubes growing. The dystrophin expressions were weak, but a few of them were strong. DNA analysis showed that the donor's gene DNA accounted for 1%-13%. At 126 days, obviously increased dystrophin positive muscular fibers of the boy were found. The donor's fibers rose to 2.5%-25%. (6) The serum CK of the boy declined from 5735 U/L to 274 U/L. (7) At 100 days, physical examination revealed improvement in his arms and legs. CONCLUSION: The therapy of Duchenne muscular dystrophy with allogeneic umbilical cord blood hematopoietic stem cell transplantation may reset up the blood-making function, decrease the serum CK level, restore the dystrophin in muscles, and improve the locomotive function of the DMD boy. These data suggest that the allogeneic umbilical cord blood hematopoietic stem cell transplantation may benefit the DMD boys.

[Association of TNFA polymorphisms with the outcomes of HBV infection].

OBJECTIVE: To explore whether the TNFA promoter single nucleotide polymorphisms (SNPs) are associated with the outcomes of hepatitis B virus(HBV) infection in Chinese Han population. METHODS: One hundred and forty-eight self-limited HBV infection subjects and 207 chronic hepatitis B patients were recruited. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and sequence specific primer-PCR(PCR-SSP) were used to detect the SNPs of five sites in TNFA promoter (-238G/A, -308G/A, -857C/T, -863C/A, -1031T/C). The frequency distributions of genotypes and haplotypes in different groups were analyzed by EPI and EH programs. RESULTS: The frequencies of -238GG genotype in chronic hepatitis B patients were significantly higher than that in self-limited infection subjects (P=0.02). The frequencies of -857TT genotype in chronic hepatitis B patients were clearly lower than that in self-limited infection subjects (P=0.02). Haplotypic frequencies of GGCCT (-238/-308/-857/-863/-1031) in chronic hepatitis B patients was significantly lower than that in self-limited infection subjects (P=0.03), and the frequencies of haplotype GGCAT or GGTAT in chronic hepatitis B patients were clearly higher than those in self-limited infection subjects (P=0.0001; P=0.004). CONCLUSION: TNFA promoter polymorphisms are important host genetic factors affecting the outcomes of HBV infection.

[Proteomic analysis of proliferation and apoptosis in carbon tetrachloride induced rat liver fibrosis].

OBJECTIVE: To study the proliferation and apoptosis in carbon tetrachloride induced rat liver fibrosis. METHODS: Wistar rats were injected subcutaneously with 40% CCl4-olive oil twice weekly for 12 weeks. Liver tissues were obtained at the end of 4, 8, 12 and 16 weeks for histological examination, hydroxyproline (Hyp) assay and proteomic analysis. After two dimension electrophoresis (2-DE), the silver stained gels were analyzed with PDQUEST 2-DE. More than 30 differentially expressed proteins were identified by MALDI-TOF-MS. RESULTS: The degree of collagen deposition and hydroxyproline content of the fibrotic livers increased continuously during the 12 weeks of CCl4 administration, peaked at the end of week 12 (P < 0.05) and declined significantly at week 16 (P < 0.05). Significant differences were observed in two parameters at each time point between the control and the model group. Meanwhile, dramatic change of hepatic proteome in the model group rats was also seen. Differentially expressed proteins identified by MALDI-TOF-MS were categorized as proliferation-related proteins/enzymes (proliferating cell nuclear antigen p120, p40 and cyclin F ubiquitin-conjugating enzyme 7 UBC7), and apoptosis-related proteins, mainly caspase-12 which was absent in the control rats. CONCLUSION: Proliferation and apoptosis related proteins are expressed dynamically in different stages of rat liver fibrosis induced by CCl4.

[Study of small dural window exposure strategy for preventing intraoperative acute brain herniation during removal of huge intracranial meningiomas].

OBJECTIVE: To investigate and assess the value of small dural window exposure strategy on preventing brain herniation during the operation for huge intracranial meningiomas. METHODS: Different classical operative approaches were used for the removal of intracranial meningiomas situated in different locations. When the dura was found to be under high tension, it was imperative that measures should be taken to prevent damage to the brain tissue in the important cortical areas as a result of acute intraoperative brain swelling and herniation. The strategy to be taken was to make a small dural window for exposure, thus the dura was used as a natural barrier to prevent acute brain swelling and herniation. The concept was to tailor the location and size of the dural opening to the location and size of the tumor, so that through the window only a part of the tumor was exposed, without exposing any important structures, such as bridging veins to the sagittal sinus and middle cerebral artery in the Sylvian fissure or any part of brain tissue initially. When necessary only a part of non-important brain tissue adjacent to the tumor could be exposed. Through this small dural window a huge tumor could be removed piecemeal till the high pressure under the dura was relieved. Then the dura could be opened more to remove the whole tumor. RESULTS: In 6 consecutive cases with extremely high tension under the dura, total tumor removal was successfully performed with complete avoidance of the risk of acute intraoperative brain swelling and herniation, and all the patients had uneventful recovery, and they were able to resume normal life. The locations of the said tumors were: in parasagittal falx region in 2 cases, and 1 case each in sphenoid, cerebellopontine angle, middle cranial fossa and temporal convexity region. CONCLUSION: The small dural window exposure strategy can guarantee safe total removal of huge intracranial meningiomas avoiding the risk of acute intraoperative brain swelling and herniation. Peritumorous brain tissue can be kept intact. Preoperative spinal drainage of cerebrospinal fluid for lowering intracranial pressure is unnecessary in the majority of such cases. Some new knowledge for our understanding the etiological mechanisms and the mechanisms of the surgical strategy to avoid acute intraoperative brain swelling and herniation in removal of huge intracranial meningiomas can be learned.

[Apoptosis induced by cantharidin in human pulmonary carcinoma cells A549 and its molecular mechanisms].

OBJECTIVE: To investigate the apoptosis-inducing effect of cantharidin in human lung cancer cells A549 and its molecular mechanisms. METHODS: MTT assay was used to determine A549 cells proliferation. Light and electron microscopy, FACScan, Annexin V-FITC staining and DNA gel electrophoresis were used to detect apoptosis. The expression of bcl-2, Bax and survivin were examined by Western blot. RESULTS: Cantharidin inhibited the proliferation of A549 cells. The cells treated with cantharidin showed a typical apoptotic morphology and hypodiploid peak before G(1) phase. Flow cytometry analysis with annexin quantitatively further confirmed the increase of cell apoptosis. DNA of treated A549 cells depicted a ladder pattern characteristic of apoptosis, indicating the presence of DNA fragmentation. Western blot assay showed that cantharidin increased the level of Bax expression and inhibited the level of bcl-2 and survivin expression. CONCLUSION: Cantharidin can induce A549 cells apoptosis mainly via regulation of Bax, bcl-2 and survivin expression.

[Computational study on the electronic absorption transitions for 2,3-diaryl maleic anhydrides].

The electronic absorption transitions for 2, 3-bis (2, 4-dimethylthiophen-3-yl) maleic anhydride were calculated by time-dependent method with 6-31G(d) basis at structures optimized at B3LYP/6-31G(d) level. The S1, S2, and S3 transitions calculated for the open and the closed forms of the compound are 390, 360.5, 339.4; 584.6, 395.8, 370.2 nm, respectively. The results are comparable to the electronic absorption spectrum of 2,3-bis(2,4,5-trimethylthiophen-3-yl) maleic anhydride which bears extra methyl groups that should not have much effects on the spectrum. The oscillator strengths are well correlated to the spectrum peak heights.

[Protective effects of sini decoction on cardiomyocytes in oxidative stress damage induced by peroxide].

OBJECTIVE: To investigate protective effects of Sini decoction on cardiomyocytes from oxidative stress damage induced by peroxide (H2O2). METHODS: Ventricular cardiomyocytes were prepared from 1-to-2-day-old Sprague-Dawley rats and cultured with DMEM culture media containing 15% neonatal bovine serum. 4 days later, culture media were replaced with neonatal bovine serum-free DMEM with 15%, 1.5% and 0.15% SD rats serum containing Sini decoction, respectively. Viablity percentage of rat cardiomyocytes was detected by flow cytometry using Annexin V-FITC to screen the appropriate SD rat serum concentration containing Sini decoction in DMEM culture media. Cardiomyocytes were exposed to 1 mmol/L H2O2 for 0.5 h, 1 h, 2 h, 4 h and 8 h respectively. MDA and SOD in cardiomyocytes were measured. Apoptosis and necrosis percentage of cardiomyocytes were measured by flow cytometry using Annexin V-FITC as well. Bcl-xL and Bcl-xS mRNA in cardiomyocytes were detected by polymerase chain reaction. RESULT: 15% SD rat serum with Sini decoction in DMEM culture midia was the appropriate concentrate for cell growth. MDA and SOD in cardiomyocytes were changed with time elapse of cardiomyocytes exposure to H2O2. Sini decoction could inhibit rat cardiomyocytes from oxidative stress damage induced by H2O2. CONCLUSION: To some extend, Sini decoction inhibit rat cardiomyocytes from oxidative stress damage induced by peroxide through promoting expression of Bcl-xL mRNA and decreasing expression of Bcl-xS mRNA in cardiomyocytes.

[Protective effects of Sini decoction on adriamycin-induced heart failure and its mechanism: role of superoxide dismutase].

OBJECTIVE: To investigate the role of superoxide dismutase (SOD) in Adriamycin (ADR)-induced heart failure and the protective effects of Sini decoction (SND). METHOD: SD rats were randomly divided into three groups, control group, heart failure group and SND group. ADR was injected in the rats of heart failure group and SND group by caudal vein. After injection, the rats in SND group were given SND (3.75 g x kg(-1) x d(-1), p.o.). Three weeks later, cardiac function, content of malondialdehyde (MDA) of both myocardium and mitochondria and activity of Cu-Zn SOD and Mn SOD were measured. The mRNA expression of Cu-Zn SOD and Mn SOD were also detected by RT-PCR. RESULT: Compared with control group, LVSP and +/- dp/dt max were obviously decreased, while LVEDP was markedly increased in the heart failure group. The mRNA expression and the activity of Cu-Zn SOD and Mn SOD in heart failure group were obviously lower than that in the controls'. In addition, the MDA content of both myocardium and mitochondria were clearly increased in heart failure rats. In SND-treated rats, the cardiac function, the activity and the mRNA expression of Cu-Zn SOD and Mn SOD were significantly elevated and the content of MDA was reduced, which had no statistic difference with the rats in control group. CONCLUSION: The data suggest that oxidative stress is present in the mitochondria of myocardium in ADR-induced heart failure rats and it can be eased by SND. The mechanism may be closely related to SOD.

[Evaluation on the case management program of hypertension in Zhongguancun community, Beijing].

OBJECTIVE: To explore a suitable model regarding community-based case management of hypertension in the urban areas. METHODS: Based on the guidelines set by the Chinese Hypertension League (CHL), the goal of case management of hypertension was set as: to maintain blood pressure at the normal range. Family doctors were guided to take care of case management. Hypertension patients who had been contracted to the family doctors were followed by a team of family doctors for half a year. RESULTS: After the 6-month intervention, the rate of hypertension under control had increased from 50.44% to 69.84% (P < 0.0001) while the means of systolic blood pressure decreased by 3.72 mm Hg and diastolic blood pressure decreased by 2.67 mm Hg (P < 0.0001) respectively. The means of SBP decreased by 8.59 mm Hg and diastolic blood pressure decreased by 5.26 mm Hg in patients whose baseline blood pressure were not under control. The rates of smoking, higher salt intake and no physical exercise had significantly decreased (P < 0.05). The mean number of hospital visits in the six-month follow-up period was 7.69 +/- 2.37. The "rate of control" among those who had followed schedule was higher than that of those who were not on schedule (P < 0.0001). The rate among those who had followed the schedule tended to have increased with aging and the level of education (P < 0.001). CONCLUSION: The community-based case management program on hypertension played an important and effective role in the control of hypertension in urban areas. Patients received great benefit when following the guidance from their doctors.

[Preliminary screening of nocturnal sleep breathing disturbance in patients with ischemic cerebral stroke].

OBJECTIVE: To investigate nocturnal sleep breathing disturbance in patients with ischemic cerebral stroke. METHODS: Forty-one patients with cerebral infarction undertook all-night screening of sleep breathing. The clinical features between cerebral infarction concomitant with mild and severe sleep breathing disturbance were compared, and the effects of different infarcted sites and sizes, the infarction history and symptoms on the breathing disorders were analyzed. RESULTS: The history of cerebral infarction was significantly shorter in patients with AHI greater than 20 (P < 0.01) whose breathing frequencies were also significantly lower (P = 0.043). Breathing disturbance was remarkably severe in patients with brainstem involvement (P = 0.045), with larger infarctions and with abnormalities of ingestion, pharyngeal reflex, speech, tongue thrust, consciousness, and motion coordination. CONCLUSIONS: Newer cerebral infarctions are commonly accompanied with relatively severe breathing disturbance. The occurrence of hypopnea would increase when brainstem is involved, and more severe breathing disturbance is associated with larger infarctions. The functioning abnormalities of tissues surrounding the upper airways resulted from infarctions are considered to be involved in the exacerbation of sleep disturbance.

[Identification of drought tolerant germplasm and inheritance and QTL mapping of related root traits in soybean (Glycine max (L.) Merr.)].

Fifty nine accessions of soybean (Glycine max (L.) Merr.) selected from 301 ones in Huang-Huai-Hai and Middle-Lower Changjiang Valleys were tested in two years for their tolerance to drought by using the mean membership index value averaged over those of plant height, leave number, dry root weight and dry stem and leaf weight. Four most tolerant accessions (Rank 1) and two most sensitive ones (Rank 5) were identified. There existed very significant correlations between drought tolerance and relative values of dry root weight, total root length, and root volume (per plant dry weight basis), respectively, which could be used as root indicators of drought tolerance. The RIL population derived from Kefeng 1 x Nannong 1138-2 was used to analyze the inheritance of the three related root traits by using the segregation analysis of quantitative traits under the major gene plus polygene mixed inheritance model. The results showed that between the two parents (Rank 1 x Rank 4), the relative values of dry root weight, total root length and root volume were respectively controlled by two major genes (linked together for the latter two traits, recombination value being 4.30% and 1.93%, respectively) plus polygenes with their major gene heritability values of 62.26%-91.81% and polygene heritability values 2.99%-24.75%, indicating that the major genes, especially the one with larger effect, accounted for a major part of the genetic variation between the two parents. It was identified that five, three, and five QTLs located on N6-C2, N8-D1b + W, N11-E, and N18-K linkage groups for relative dry root weight, total root length and root volume, respectively. Each of the traits appeared to have one locus (Dw1, R/1, and Rv1) with relatively large effect in comparison with their other loci, and those major ones were located near the same site of the same linkage group N6-C2. The results of segregation analysis and QTL mapping appeared pretty consistent with each other, which could be used as a demonstration of each other.

[A 73-year-old woman with depression, dementia, and parkinsonism].

We report a 73-year-old woman who had depression, dementia, and parkinsonism. She had postural tremor since her fortics. She was losing her weight since age 66 years. She noted difficulty in walk at age 72 (2001). She could not stand without assistance on July 2001, and she became hypobulic. On admission to our hospital on November 2001, she had dementia and revised Hasegawa dementia scale (HDS-R) was 8/30. She had mild limitation of the upward gaze, and rigidity in the neck, but not in the limbs. Postural tremor was seen. No muscle weakness was noted and tendon reflexes were normal. She was treated with levodopa/carvidopa, but she did not improve. She did not eat much. She was transferred to another hospital and she suddenly died on January 2002. The patient was discussed in a neurological CPC, and a chief discussant arrived at a conclusion that the patient had Parkinson disease with dementia. Some participants thought the diagnosis was progressive supranuclear palsy or diffuse Lewy body disease. The examination at autopsy revealed mild neuronal loss and Lewy bodies in the substantia nigra. Many Lewy bodies were observed in the cerebral cortex which corresponded to the neocortical type of DLB, and Lewy neurites were seen in the CA2 of the hippocampus by immunohistochemistry for alpha-synuclein. Spongy change was seen in the parahippocampus. Pathological diagnosis was diffuse Lewy body disease.

[Comparative study of pneumatic lithotripsy and holmium laser lithotripsy for ureteral stones].

OBJECTIVE: To compare the efficacy and safety of endoscopic laser lithotripsy (LL) and endoscopic pneumatic lithotripsy (PL) for ureteral stones. METHODS: We retrospectively analyzed the clinical data of 415 patients with ureteral calculi treated with endoscopic laser lithotripsy (n = 214 ) and pnumatic lithotripsy (n = 201 ). RESULTS: The overall successful fragmentation rate of all ureteral stones in a single session of the LL group was higher than that of the PL group (95% vs. 69%, P < 0.01). The average stonefree time of the LL group was shorter than that of the PL group (18 days vs. 31 days, P < 0.01). No complications such as perforation during the operation were observed in the LL group whereas 3 perforations occurred in the PL group. CONCLUSION: LL has its advantage over PL in its better clinical effect for the stone fragmentation and low complication rate and is an effective and safe treatment for ureteral stones.

[Construction of an engineered Acidithiobacillus caldus with high-efficiency arsenic resistance].

Using the recombinant technique in vitro, a new arsenic resistance plasmid pSDRA4 was constructed by subcloning the arsenic resistance genes from plasmid pUM3 into the wide-host-range IncQ plasmid pMMB24 with the hybrid trp-lac ( tac ) promoter, and followed by deleting the regulative gene of the promoter, the lacIQ gene. Then plasmid pSDRA4 was introduced from E. coli into extremely acidophilic obligately chemolithotrophic Acidithiobacillus caldus by conjugative transfer with a frequency of( 1.444 +/- 0.797) x 10(-4), and the engineered strain of Acidithiobacillus caldus (pSDRA4) for biomining was constructed. The successful transfer demonstrates the development of a conjugational system between strains of E. coli and A. caldus. The recombinant plasmid pSDRA4 is stable in A. caldus. Compared with wild type A. caldus, the level of the arsenic resistance of A. caldus (pSDRA4) is greatly raised from 10mmol/L to 45mmol/L.

[Effects of soil temperature and humidity on soil respiration rate under Pinus sylvestriformis forest].

Employing root-wrenching method and LI-6400-09 soil respiration chamber, this paper measured the diurnal changes of soil respiration rate with and without roots in situ on June 17, August 5, and October 10, 2003. The seasonal changes of soil respiration were also measured from May to September, 2004. The results showed that both the total and the root-wrenched soil respiration appeared single diurnal pattern, with the peaks presented during 12:00-14:00. The diurnal fluctuation of soil respiration on August 5 was smaller than that on June 17 and October 10. There were also obvious seasonal changes in total and root-wrenched soil respiration, as well as in root respiration, which were higher from June to August but lower in May and September. The average total soil respiration, root-wrenched soil respiration, and root respiration were 3.12, 1.94 and 1.18 micromol CO2 x m(-2) s(-1), respectively, and the contribution of roots to total soil respiration ranged from 26.5% to 52.6% from May to September, 2004. There were exponential correlations between respiration rate and soil temperature, and linear correlations between respiration rate and soil humidity. The Q10 values were 2.44, 2.55 and 2.27 for total soil respiration, root-wrenched soil respiration, and root respiration, respectively. The effect of soil temperature on root-wrenched soil respiration was lager than that on total soil respiration and root respiration. Soil humidity had a larger effect on total soil respiration than on root respiration and root-wrenched soil respiration.
