[The age-related changes of P53 protein expression in oral mucosa and their effects on oral leukoplakia].

OBJECTIVE: To investigate the age-related changes of P53 protein expression in oral mucous membrane and their effects on pathogenesis of the oral precancerous lesions in elderly people. METHODS: An immunohistochemical technique (SP method) was applied to study the changes of P53 expression in normal oral mucosa and oral leukoplakia(OLK) in different age groups. The staining results were evaluated by semi-quantity method. RESULTS: With aging, an elevating tendency of P53 protein level exhibited in the normal oral mucosa and OLK, respectively. The P53 level in the old with OLK was higher than that in the normal oral mucosa of the old (P < 0.01). CONCLUSION: With aging, P53 protein may undergo dysfunction, lead to imperfect DNA repair and excessive DNA damage accumulation, and ultimately facilitate malignant transformation of the oral mucosa in the old.

[Effects of long-term oral administration of lanthanum nitrate on the liver of rats].

OBJECTIVE: To probe the effects of long-term oral administration of lanthanum nitrate [La(NO(3))(3)] on morphological change in the liver, aftereffect of deposited La in the liver and their mechanism in rats. METHODS: Young Wistar rats were divided into two groups, one fed with 0.1, 0.2, 2.0, 10.0 and 20.0 mg/kg of La(NO(3))(3) for six months and the other for the control. Changes in ratio of liver to body weight were observed after exposure to La(NO(3))(3) at varied doses for six months and one month after six-month exposure, as well as morphology of the liver in the rats with routine histochemistry and transmission electron microscopy (TEM) technique. Content of La in the liver was measured with inductively coupled plasma-mass spectrometry (ICP-MS). RESULTS: Ratio of liver to body weight was significantly higher in the male rats exposed to 20.0 mg/kg of lanthanum for six months than that in the control group. Ratio of liver to body weight restored to normal in the rats exposed to 20.0 mg/kg of La one month after six-month exposure. Infiltration of inflammatory cells in the portal region of the liver, small amount of fat drops in hepatocytic cytoplasm, increased density of mitochondria stroma, lysosome containing highly-electronic-density bodies and dense granules, normal nucleus and slightly deformed nucleus of hepatocytes could be found in the rats exposed to 20.0 mg/kg. Areas of the liver deposited with glycogen after six-month exposure to 20.0 mg/kg of La accounted for (26.1 +/- 1.5)% and (4.1 +/- 1.4)%, respectively for male and female rats, significantly lower than those in the control group [(31.3 +/- 1.4)% and (39.4 +/- 0.9)%, respectively], with a statistical significance and very statistical significance, respectively. There was a little infiltration of inflammatory cells in the portal region of the liver one month after six-month exposure to 20.0 mg/kg of La, and amount of the dense bodies was lower in the rats exposed to La for six months. Liver contents of La in the rats of all experimental groups were lower one month after six-month exposure than those in the rats exposed for six months. CONCLUSIONS: Exposure to a dose of 20.0 mg/kg La(NO(3))(3) for a long term could damage the liver structure to certain extent, but lanthanum deposited in the liver could be eliminated from the body gradually.

Diagnosis, treatment and prognosis of thymoma: an analysis of 116 cases.

OBJECTIVE: To study the diagnosis and treatment of thymoma and to assess its prognostic factors. METHODS: The clinical data of 116 patients with thymoma were collected. A retrospective analysis was performed, by comparing the survival rate calculated by the Kaplan-Meier method with the rate of recurrence or metastasis. RESULTS: The standard posteroanterior and lateral chest radiographs were reliable means of detection of most thymomas. Myasthenia gravis was the most commonly paraneoplastic disease (25.0%, 29/116). The extensive radical resection was beneficial for reducing the rate of recurrence of stage I or stage II thymomas (chi(2) = 4.941, P = 0.0219). The survival time could be prolonged by postoperative radiotherapy and chemotherapy. There was a strong correlation between the clinical stage and the histological classification (according to MH classification), through which the invasive behavior of thymoma could be predicted (chi(2) = 19.76, P = 0.007, RR = 1.47). The 3- 5- and 10-year survival rates were 81.2%, 67.9%, and 40.5%, respectively. Statistical analysis showed a significant negative correlation between the stage and the survival rate (chi(2) = 29.73, P = 0.0000, RR = 0.15). CONCLUSION: The prognosis of thymoma depends mainly on the histological classification, clinical stage and multimodality treatment rather than on the paraneoplastic diseases.

[A study on the variation of goiter rates, urinary iodine and household salt iodine intake among children in West China].

OBJECTIVE: To study the factors leading to the variation of children's total goiter rates, urine iodine and salt iodine in West China. METHODS: Design effect (DEFF) was used to evaluate the variation of indicators. RESULTS: DEFF of children's total goiter rate was about 3.0 in 9 provinces, and the proportion was 75.0%. The DEFF of urine iodine was 1.0 - 3.5 in 6 provinces, and the proportion was 50.0%. The DEFF of intake rate of qualified iodized salt was over 3.0 in 11 provinces. The DEFF of covering rate of iodized salt was about 3.0 in 4 provinces. CONCLUSION: In order to provide scientific basis for IDD surveillance in China, other than bias due to methods of no-sampling, sample size should be further calculated in West China. Thus the main influencing factors of indicator variation could be measured with scientific and reasonable basis.

[Effects of soybean phospholipid on the content of malondialdehyde and lipofuscin in heart, liver and spleen in mice].

In order to observe the effects of soybean phospholipids on the SOD activity in liver and the content of malondialdehyde and lipofuscin in heart, liver and spleen in mice, forty male mice were divided into 4 groups. Experimental mice were fed with different doses of soybean phospholipids (2.5, 5 and 10 g/kg BW) and the control mice were fed with normal saline by tube feeding for 18 days. After that, the SOD activity in liver and the content of malondialdehyde and lipofuscin in heart, liver and spleen were measured. The results showed that the three doses of soybean phospholipid could significantly increased the SOD activity of liver (P < 0.05), and also could remarkably decrease the malondialdehyde content of in heart(P < 0.01) and in liver (P < 0.05). The soybean phospholipid at dose of 2.5 g/kg BW could significantly decrease the malondialdehyde content of in spleen(P < 0.05). But the three doses of soybean phospholipid did not obviously affect the content of lipofusin in liver, heart and spleen. The study indicated that the soybean phospholipid was effective on the inhibition of lipid peroxide.

[Study on the treatment of high dose mifepristone and progesterone in endometrial carcinoma].

OBJECTIVE: To investigate the effect of high dose mifepristone and high dose progesterone in the treatment of patients with endometrial carcinoma and to explore the possible mechanisms associating with them. METHODS: Thirty untreated patients diagnosed as endometrial carcinoma through dilation and curettage of the uteri were divided into 3 groups at random. Each group was given medroxyprogesterone acetate (MPA), (500 mg/day) or mifepristone (MIF), (100 mg/day) or MIF (100 mg/day) + MPA (500 mg/day) for 5 days respectively. On the sixth day, hysterectomy was performed on these patients. The endometrial cancer specimen of post-hysterectomy was compared with the one of pre-administrating. The morphologic changes of the endometrial cancer cells were observed through light microscope. Immunohistochemistry assay (SP method) was applied to determine the localization and immunoreactive intensity of proliferating cell nuclear antigen (PCNA), estrogen receptor (ER), progesterone receptor (PR), B-cell leukemia lymphoma-2 (bcl-2), bcl-2 associated X protein (bax) and CD(44v6). RESULTS: Better differentiation degree and active excretion were observed in all of the post-hysterectomy endometrial specimen. In the same time, apoptosis of carcinoma cells was observed. The most significant changes were seen in the MIF + MPA group. In the MPA group, the pre-treatment and post-treatment expression of PR (2.9 +/- 1.1, 1.6 +/- 0.8), ER (2.8 +/- 0.9, 1.4 +/- 0.9), PCNA (0.84 +/- 0.10, 0.60 +/- 0.12), bcl-2 (0.236 +/- 0.089, 0.157 +/- 0.981) and CD(44v6) (4.6 +/- 1.8, 2.5 +/- 1.9) were all decreased (all P < 0.01); the expression of bax (0.20 +/- 0.10, 0.42 +/- 0.07) was increased (P < 0.01). In the MIF group, the expression of PR (3.4 +/- 1.0, 1.9 +/- 0.8), ER (2.7 +/- 0.9, 1.2 +/- 0.7), PCNA (0.80 +/- 0.15, 0.65 +/- 0.10), bcl-2 (0.214 +/- 0.097, 0.121 +/- 0.073) were all decreased (all P < 0.01); the expression of bax (0.21 +/- 0.05, 0.44 +/- 0.09) was increased (P < 0.01); no significant change in the expression of CD(44v6) (4.2 +/- 2.0, 4.3 +/- 1.7) was seen (P > 0.05). In the MIF + MPA group, the expression of PR (3.2 +/- 1.0, 0.8 +/- 0.8), ER (2.7 +/- 0.9, 0.7 +/- 0.9), PCNA (0.81 +/- 0.09, 0.25 +/- 0.09), bcl-2 (0.225 +/- 0.091, 0.066 +/- 0.009) and CD(44v6) (4.5 +/- 1.9, 2.7 +/- 1.6) were all decreased (all P < 0.01); the expression of bax (0.22 +/- 0.06, 0.59 +/- 0.09) was increased (P < 0.01); there were significant different expression of PCNA, ER, PR, bax and bcl-2 as compared with the MIF group and the MPA group, respectively (all P < 0.01). The expression of CD(44v6) was significantly different (P < 0.01) between the MIF + MPA group, and the MIF group, but not significantly different between the MIF + MPA group and the MPA group. CONCLUSIONS: The study indicates that high dose progesterone could inhibit the growth, promote apoptosis and inhibit metastasis of endometrial carcinoma, MIF could inhibit the growth and promote apoptosis, MIF + MPA could more strongly inhibit the growth, promote apoptosis and inhibit metastasis of endometrial carcinoma than MIF or MPA, and synergistic effect was observed on the expression of PCNA, ER, PR, bax and bcl-2.

[A cross-sectional study on the usage condition of hypertension related examinations and antihypertensive drugs among 40 district and community based hospitals in Beijing].

OBJECTIVE: To study the use of hypertension related physical examinations, laboratory tests and anti-hypertension drugs among district and community based hospitals in Beijing and to analyze the possible hardware (examination, test, drug) problems under the application of the "China Hypertension Guide" in those hospitals. METHODS: The survey was carried out in 40 hospitals in Chaoyang and Haidian districts, including 9 district level hospitals, and the rest at community level. Questionnaire survey and in-depth interview were used to collect information from the leaders of related departments in those hospitals. RESULTS: (1) The hardware condition of district hospital satisfied the implicit requirements of hypertension evaluation and treatment in "China hypertension Guide". (2) 64.5% of the community level hospitals had the basic equipments and routine laboratory tests for diagnosis and treatment on hypertension, but 35.5% of them lack of tests on blood chemistry (potassium, sodium, creatinine) and urine protein. (3) 71.0% of community level hospitals could not judge the patient's "target organ damage" independently. (4) Both district and community level hospitals had all major types of anti-hypertension drugs in there pharmacy except ARB. CONCLUSION: When necessary laboratory tests for both district and community level hospitals were provided, they could accomplish the tasks of hypertension treatment and management. However, the community level hospitals should cooperate with hospitals at higher level to have a comprehensive clinical understanding of patients with high blood pressure.

[Study of cerebral protective effects of naosaitong in animals].

OBJECTIVE: To investigate the influence of Naosaitong (NST) on the cerebral blood flow (CBF), the infarct areas and blood rheology in animals. METHOD: NST's cerebral protective effects were investigated by using middle cerebral artery occlusion (MCAO), bilateral common carotid artery ligation, and carrogeenin-induced thrombus model rats, being administrated with medicine for seven days. RESULTS: Three dosage groups of NST increased CBF in anesthetized rabbits, reduced the infarct areas in MCAO rats, decreased the physical sign indexes, and water quantities. They increased the activities of Glutathione peroxidase (GPX) and Catalase (CAT), decreased the contractions of Lipid peroxidase (LPO) and Lactate (LD) in the cerebral ischemia-reperfusion rats; shortened the length of thrombus and improved the blood rheology in the carrogeenin-induced thrombus model rats, and prolonged hypoxia-resisting time in mice. CONCLUSION: NST can evidently increase CBF in rabbits, improve the cerebral edema brain tissues' injure and nervous physical sign indexes in the cerebral ischemia-reperfusion rats, reduce the infarct areas in MCAO rats, postpone thrombosis course and have antioxidation effects, which show that NST can obviously protect the brain tissues in the experimental cerebral infarct model rats.

[Variations of water-soluble carbohydrate and nitrogen contents in Leymus chinensis and Phragmites communis under different stocking rates].

The variations of the contents of water-soluble carbohydrate and nitrogen in L. chinensis and Ph. communis under different stocking rates were studied by fencing grazing experiment on L. chinensis grassland. The results indicated that the water-soluble carbohydrate content in the base stem of L. chinensis and Ph. communis was the lowest (7.12% and 3.95%, respectively) at the beginning of growth period, and then increased gradually with seasons. There was no significant variation in the content of leaf water-soluble carbohydrate in the grasses. L. chinensis germinated earlier than Ph. communis, resulted in the difference between the water-soluble carbohydrate content in the base stem and the leaf of the grasses at the beginning of the experiment in May. Proper stocking rates were favorable for the increase of water-soluble carbohydrate content, and could accelerate the re-growth capability of the herbage. The nitrogen content was the highest at the beginning of growth period, and then decreased gradually with seasons, which was accorded with their phenology. Proper stocking rates could stimulate the assimilation of nitrogen in soil, and bring it to the ground, which improved the nutrition value of the herbage.
