[Determination of metal elements in six kinds of desert plants in Inner Mongolia by ICP-AES].

In the present paper, the contents of metal elements from six kinds of desert plants, namely Haloxylon ammodendron, Caragana microphylla, Ammopiptanthus mongolicus, Salix bordensis, Elaeagnus angusti folia, and Salix psammophila in Inner Mongolian were determined by ICP-AES technique. The recovery ratio obtained by standard addition method ranged between 94.98% and 120.25%, and the RSD was lower than 3.4%. The results of determination showed that the major elements Ca, K, Mg, Na and Al, and trace elements essential to plant vital activities such as Fe, Mn, Cu and Zn exhibit different orders in content in the six kinds of desert plants, and the contents of four elements, Fe, Mn, Cu and Zn are lower than the average content of terrestrial plants. The above results provided reliable data and theory bases for improving environment in west China and choosing weather-resistant and sand-fixation tree species.

Association of-238G/A and -857C/T polymorphisms of tumor necrosis factor-alpha gene promoter region with outcomes of hepatitis B virus infection.

OBJECTIVES: To determine whether -238G/A and -857C/T polymorphisms of tumor necrosis factor-alpha (TNF-alpha) gene promoter were associated with outcomes of hepatitis B virus infection. METHODS: A total of 246 HBV self-limited infected subjects and 443 chronic hepatitis B (HB) patients were recruited in this case-control study. TNF-alpha-238G/A and -857C/T gene promoter polymorphisms were examined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP). RESULTS: The frequency of TNF-alpha-238 GG (90.7%) in chronic HB group was significantly lower than that (95.1%) in self-limited group (P = 0.041). The frequency of TNF-alpha-857 CC (79.7%) in chronic HB patients was significantly higher than that (70.9%) in self-limited infected subjects (P = 0.021). Multiple logistic regression analysis revealed that both TNF-alpha-238GA and -857CC were independently associated with chronic HB. CONCLUSIONS: TNF-alpha promoter variants are likely to play a substantial role in influencing the outcomes of HBV infection.

[Study on association between vitamin D receptor gene polymorphisms and the outcomes of HBV infection].

OBJECTIVE: To explore whether the vitamin D receptor gene (VDR) polymorphisms are associated with the outcomes of hepatitis B virus (HBV) infection in Chinese Han population. METHODS: Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to detect the polymorphisms of Fok I locus in exon 2 and Taq I locus in exon 9 of VDR gene. One hundred and eighty-four chronic hepatitis B patients and 205 asymptomatic HBV carriers were recruited to make the comparison of frequencies of genotype and haplotype of the VDR gene between the patients and the carriers. RESULTS: The univariate analysis showed a significant difference in Fok I polymorphism between chronic hepatitis B patients group and asymptomatic HBV carriers group. The FF genotype frequency in chronic hepatitis B patients group was 44.6%,higher than 31.7% in asymptomatic HBV carriers group (P<0.05). After adjusting the confounders by multiple logistic regression analysis, the result still showed a significant difference in Fok I site polymorphism between chronic hepatitis B patients group and asymptomatic HBV carriers group (OR=1.95, P<0.05). The FT haplotype frequency in chronic hepatitis B patients group was higher than that in asymptomatic HBV carriers group (OR=1.45, P<0.05). The fT haplotype frequency in chronic hepatitis B patients group was lower than that in asymptomatic HBV carriers group (OR=0.72, P<0.05). CONCLUSION: VDR gene polymorphism may be an influence factor of genetic susceptibility to HBV infection.

[Acting mechanism of qidan liquid in promoting myocardial angiogenesis in rats after acute myocardial infarction].

OBJECTIVE: To investigate the effect of Qidan Liquid (QL) on myocardial angiogenesis in rats after acute myocardial infarction (AMI) and explore its possible molecular mechanism. METHODS: AMI model was established by ligating the left anterior descending (LAD) coronary artery in Wistar rats, and intervened with QL in high, medium and low doses (QLh, QLm and QL1 group), and isosorbide dinitrate (ID group) respectively. Meanwhile, the model group, the sham group and the normal group were also set up. On the 7th (d7) and 14th day (d14) after modeling, mRNA and protein expressions of vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) were detected with RT-PCR and immunohistochemistry, factor VIII expression in endothelial cells was examined, and microvascular density (MVD) in ischemic myocardium was calculated. RESULTS: MVD in the model rats increased significantly, higher than that in the normal and the sham group (P < 0.01), on d7 and d14, it was lower in the model group than that in the high and medium dose of QL groups and the ID group (P < 0.05). On d7, VEGF protein expression in the model group was higher than that in the normal group (P< 0.05), equivalent to that in the other groups, on d14, it was lower than that in the high and medium dose of QL groups and the ID group (P < 0.05 or P < 0.01). On d7 and d14, the protein expression of bFGF in the model rats increased significantly, higher than that in the normal and the sham group (P < 0.01), it decreased along the time progress between the two time points (P < 0.05), and lower than that in the high dose of QL group and ID group (P < 0.01). The mRNA expressions of VEGF and bFGF were higher in the model group than those in the normal and sham group, but lower than those in all the treated groups on d7 and d14. Comparison of the strips on d7 and d14 among various groups showed that the bFGF mRNA expression in the model group showed a descending trend as the time goes on, but in all the treated groups it was unchanged, while the change of VEGF mRNA expression didn't show any definite tendency. CONCLUSION: QL could up-regulate the protein and gene expressions of VEGF and bFGF persistently and increase MVD in ischemic myocardium to promote the development of collateral circulation in AMI rats.

[278C > T variant of the alpha-1, 3-galactosyltransferase allele responsible for Bw subgroup].

OBJECTIVE: To investigate the molecular genetic basis of the Bw variant and identify novel alleles at ABO locus in Chinese Han population. METHODS: Serological techniques were performed to characterize erythrocyte phenotype of a proband. Mutations of the ABO gene were screened by polymerase chain reaction, reverse transcription-polymerase chain reaction and DNA sequencing. RESULTS: The proband was identified as Bw phenotype by serological technology and family study. A novel Bw variant allele was identified in the gDNA and cDNA. The novel allele was observed a missense mutation (278 C to T) at the exon 6 which resulted in an amino acid substitution (P93L) compared with B101 allele. The 278 C to T was the first report mutation position in exon 6 among Bw alleles, so the P93L amino acid substitution was different from others Bw variants which had amino acid substitutions in a conserved functional domain reported previously. CONCLUSION: A novel Bw allele (278 C to T) responsible for Bw variant is reported in Chinese population.

[Hepatosplenic gammadelta T cell lymphoma and its relationship with Epstein-Barr virus infection].

To explore the clinical and pathological characteristics of hepatosplenic gammadelta T-cell lymphoma and its relationship with Epstein-Barr virus infection, the clinical features of a 9-year-old girl with hepatosplenic gammadelta T-cell lymphoma were investigated, the smears of bone marrow was stained with Wright' s stain, biopsies of bone marrow and liver specimen were embedded in plastic and sliced about 4 microm in thickness and routinely stained with HE staining, the immunohistochemical staining was used to mark the tumor cells, and EBER probes were used to detect Epstein-Barr virus RNA. The results showed that the girl presented with prolonged fever, anemia, thrombocytopenia, hepatosplenomegaly, chronic active Epstein-Barr virus infection, and elevated levels of serum ferritin and lactate dehydrogenase. Bone marrow aspirate revealed the infiltration of atypical lymphocytes in the bone marrow stroma. The liver biopsy specimen revealed the infiltration of lymphocytes in the sinusoids, which was positive for the T-cell associated marker CD3 and activated cytotoxicity-associated marker granzyme B. In-situ hybridization analysis with EBER probes revealed that the above-mentioned characteristics were negative in neoplastic cells. It is concluded that hepatosplenic gammadelta T-cell lymphoma is a disease with distinctive clinical, histopathologic, and phenotypic characteristics. Hepatic and/or splenic and/or bone marrow biopsy with combined phenotype is beneficial to diagnosis. Epstein-Barr virus infection is late event involving an already transformed gammadelta T-cell clone.

Facile preparation of amperometric laccase biosensor with multifunction based on the matrix of carbon nanotubes-chitosan composite.

The carbon nanotubes-chitosan (CNTs-CS) composite provides a suitable biosensing matrix due to its good conductivity, high stability, and good biocompatibility. Enzymes can be firmly incorporated into the matrix without the aid of other cross-linking reagents. The composite is easy to form insoluble film in solution above pH 6.3. Based on this, a facilely fabricated amperometric biosensor by entrapping laccase into the CNTs-CS composite film has been developed. At pH 6.0, the fungi laccase incorporated into the composite film remains better catalytic activity than that dissolved in solution. The system is in favor of the accessibility of substrate to the active site of laccase, thus the affinity to substrates is improved greatly, such as 2,2'-azino-bis-(3-ethylbenzthiazoline-6-sulfonic acid) diammonium salt (ABTS), catechol, and O2 with Km values of 19.86 microM, 9.43 microM, and 3.22 mM, respectively. The major advantages of the as-prepared biosensor are: detecting different substrates (ABTS, catechol, and O2), possessing high affinity and sensitivity, durable long-term stability, and facile preparation procedure. On the other hand, the system can be applied in fabrication of biofuel cells as the cathodic catalysts based on its good electrocatalysis for oxygen reduction. It can be extended to immobilize other enzymes and biomolecules, which will greatly facilitate the development of biosensors, biofuel cells, and other bioelectrochemical devices.

Effects of maternal immobilization stress on birth weight and glucose homeostasis in the offspring.

Recent epidemiological studies have shown strong associations between low birth weight and the incidence of diabetes in the adult offspring. It has been hypothesized that exposure to maternal glucocorticoids programs cellular changes in the fetus which increases the susceptibility of the offspring to diabetes. Stressors produces large increases in maternal glucocorticoids. The present study determined the effects of immobilization stress during weeks one, two or three of pregnancy on offspring birth weight, glucose homeostasis, and the ability of the offspring to cope with metabolic stress. Immobilization stress produced large increases in maternal levels of ACTH and corticosterone, but did not affect birth weight of the pups. Chronic administration of high fructose diet, a metabolic stressor, to 60 days old control and prenatally stressed offspring produced large increases in plasma levels of triglyceride and insulin. However, there were no differences between the groups either in peak levels, or in the rates of increase and decrease (upon discontinuation of the diet) of plasma triglyceride and insulin concentrations. Basal levels of glucose and insulin, and areas under the glucose and insulin plasma concentration-time curves after an i.p. glucose dose were similar between 120 days old control and prenatally stressed offspring. These results suggest that in young adult rats prenatal immobilization stress did not affect glucose homeostasis or the ability of these rats to cope with chronic metabolic stress.
