[Neuroprotection of chloride channel blockers against NMDA-induced apoptosis of cultured rat hippocampal neurons].

Activation of N-methyl-d-aspartic acid (NMDA) receptor plays an important role in neuronal apoptosis induced by cerebral ischemia but the underlying mechanisms are still unclear. The present study examined the neuroprotection of three chloride blockers in an in vitro cell model of cerebral ischemia established by treatment of cultured rat hippocampal neurons with NMDA. Hoechst 33258 staining and MTT assay were used to detect neuronal apoptosis and cell viability, respectively. The neuroprotective effects of chloride channel blockers on the cell viability and neuronal apoptosis were only observed when the blockers were applied before NMDA exposure. In comparison with DIDS, SITS showed more potent protective effect in a dose-dependent manner, whereas NPPB showed no significant neuroprotective effect. The results demonstrate that pretreatment with both SITS and DIDS have protective effect against neuronal apoptosis, which is achieved by blocking both NMDA receptor and chloride channel.

[The effects of tumor antigen gene OVA66 on biological characteristics of tumor cells].

AIM: To explore the effects of tumor antigen gene OVA66 on the biological characteristics of tumor cells. METHODS: The eukaryotic expression vector pcDNA3.1-mock and recombined vector pcDNA3.1-OVA66 were transfected into hepatoma cell line SMMC-7721 by LipofectAMINETM 2000. After stable selection and clone proliferation of cells, the expression of OVA66 gene and protein was detected by RT-PCR, Western blot and immunocytochemical (ICC) staining. The cell growth cycle was analyzed by FACS. The ultrastructure was observed under electron microscope. The experiments in cell migration and invasion were performed in vitro. The OVA66-associated genes were analyzed by genechip technique after gene transfection. RESULTS: OVA66 was highly expressed in 7721 cells transfected with pcDNA3.1-OVA66 at mRNA level. Western blot result demonstrated that OVA66 protein content was notably increased compared with that in control cells. Electron microscope and FACS results indicated that OVA66 gene promoted the growth and proliferation of tumor cells. In vitro, the experiments in cell migration and invasion showed that this gene enhanced migration and invasion of tumor cells. The result of genechip demonstrated that the over- expression of OVA66 gene increased the expression of plasminogen activator inhibitor-1 gene (PAI-1), suggesting that it had some effects on tumor invasion and metastasis. CONCLUSION: Tumor antigen gene OVA66 and it's protein demonstrate a series of biological characteristics of tumor cells, which can promote the proliferation, migration and invasion of tumor cells.

[DC conductivity of DNA solution].

DNA with different length was solved into ultra-pure water, the conducing behavior of the suspension was investigated at varied temperature and DNA concentration. The electrical conductivity of the samples increased with the increment of temperature and concentration of the solution. In terms of the length, the short-strand DNA (less than 50bp) showed a better conducting capability than the long-strand DNA(more than 1000bp). The electrical conductivity of short-strand DNA arrives at 1.08 x 10(-3) S/cm at room temperature. The conductance of the two DNA samples became similar at high temperature as well as their uptrend when DNA samples were heated.

[Protective effects of allopurinol on white matter damage in premature rats].

OBJECTIVE: To investigate the protective effects of allopurinol (ALLO) on white matter damage in premature rats. METHODS: An animal model for white matter damage was established by bilateral carotid artery occulation (BCAO). Eighty-four newborn SD rats (1 day old) were used in this study and were divided randomly into three groups [sham surgery (Sham); BCAO group (BCAO); allopurinol-treated group (ALLO)]. Pathological changes were studied 7 days and 14 days after BCAO, respectively. Myelin basic protein (MBP) was detected by immunohistochemistry 7 days and 14 days after BCAO, respectively. MBP-mRNA expression was determined 7 days and 14 days after BCAO respectively by reverse transcription-polymerase chain reaction (RT-PCR) with fluorescent quantitative method. RESULTS: In BCAO group, mild or severe rarefaction was found in 10 cases in the corpus callosum area, especially at the cingulum. Pathological changes of white matter were found in 4 cases in internal capsule. Subcortex white matter rarefaction was found in 8 cases. The extent of white matter rarefaction in ALLO group was reduced significantly. Enlargement of bilateral ventricles was found in 6 of 8 cases in BCAO group. The average ventricle size in ALLO group (2.44 +/- 0.71)% was reduced significantly as compared with that in BCAO group (3.27 +/- 0.73)% (P < 0.05). Strong MBP positive staining was found in sub-cortex, corpus callosum, hippocampus gyrus, and internal capsule of P14 sham surgery group. In BCAO group the MBP staining extent was reduced. The extent of MBP staining of ALLO group was between the other two groups. The optical density (OD) of MBP positive staining in BCAO group (6.60 +/- 0.68) was found higher than that in sham surgery group (9.40 +/- 0.53), the difference was statistically significant (P < 0.05). Compared with BCAO group, OD value in ALLO group (7.10 +/- 0.18) increased significantly (P < 0.05). RT-PCR data showed that MBP-mRNA copies (log10) in P7 and P14 rats of both BCAO and ALLO groups were lower than that in sham surgery group (P < 0.01); However, MBP-mRNA copies in ALLO group were higher than that in BCAO group (P < 0.05). CONCLUSIONS: BCAO could be used in newborn rats (1 day old) to establish a premature white matter damage (WMD) animal model. Allopurinol may have a potential protective effect on premature SD rat with ischemic WMD.

Expressions of cyclin E2 and survivin in acute leukemia and their correlation.

Cyclin E2 is present in solid tumors, while its expression and clinical value in acute leukemia is unknown. This study was aimed to investigate the expression of cyclin E2 and survivin gene in bone marrow cells from patients with acute leukemia and their relationship. Reverse transcription polymerase chain reaction was used for detection of the expression of cyclin E2 and survivin mRNA in 84 adult patients with acute leukemia which included 16 cases of relapse, 60 cases of de novo acute leukemia, 8 cases of continuously complete remission, and 20 normal persons as controls. The results showed that (1) positive expression of cyclin E2 (70.24%) in acute leukemia patients was significantly higher than that (0%) in controls, positive expression of survivin (72.62%) in acute leukemia patients was higher than that (30%) in control. (2) the expression of cyclin E2 positively correlated with that of survivin in acute leukemia patients. (3) remission rate in cyclin E2-positive patients (55.81%) was lower than that (88.24%) in cyclin E2-negative patients, the rate of cyclin E2 expression in relapse group was the highest among the three groups; while that in continuously complete remission group was the lowest among the three groups. (4) positive rate of cyclin E2 expression (59.32%) in patients with acute myelocytic leukemia was lower than that (96%) in patients with acute lymphocytic leukemia, no correlation between cyclin E2 expression and white blood cell counts of patients was found. It is concluded that the overexpression of cyclin E2 has been confirmed for the first time to positively correlate with the expression of the survivin in acute leukemia patients, and implicate the poor prognosis. Cyclin E2 may be used as a marker for examination of minimal residual disease.

[Clinical investigation of chronic blepharitis and evaporative loss dry eye].

OBJECTIVE: To investigate the relationship between chronic blepharitis and evaporative loss dry eye. METHODS: Forty-one patients (82 eyes) were diagnosed as chronic blepharitis from November 2002 to April 2003 in Beijing Tongren Eye Center, of which 22 cases were male and 19 female. All patients' ocular surface was judged by Schirmer I test, tear film break up time (BUT) and Tearscope. Meibomian gland secretion was used for bacteria culture and antibiotic sensitivity test. Thirty patients (60 eyes) of glaucoma, cataract and LASIK before surgery were used as control, which included 16 male and 14 female. Microbiological results of meibomian gland secretion were compared and the statistical difference was evaluated by chi(2) test. RESULTS: Of the blepharitis group, ST value was less than 10 mm in 30 eyes (36.6%), BUT less than 10 s in 56 eyes (68.3%) and tear film tearscope evaluation more than 3 grade in 46 eyes (56.1%). The bacteria spectrum of meibomian secretion in both groups was similar, which included staphylococcus epidermidis, staphylococcus aureus and corynebacterium. However, the bacterium culture positive rate and antibiotic sensitivity showed significant difference (P < 0.05). CONCLUSION: The chronic blepharitis is one of important cause in evaporative loss dry eyes.

[Expression of anaplastic lymphoma kinase and survivin proteins in anaplastic large cell lymphoma and its significance].

OBJECTIVE: To study the expression of anaplastic lymphoma kinase (ALK) and survivin proteins in anaplastic large cell lymphoma (ALCL) and there clinical significance. METHODS: The morphologic characteristics were studied by routine light microscopy. Immunohistochemical staining for ALK and survivin proteins was performed using LSAB method. RESULTS: ALK protein was positive in 51 cases (63%) and negative in 30 cases (37%) of the 81 cases of ALCL studied. The prognosis of patients with ALK protein expression was better than those without ALK expression (P < 0.05). As for survivin protein, there were various degrees of expression in all the 77 ALCL cases studied. High level of survivin protein expression was observed in 33 cases (42.9%), while low level of expression was seen in 44 cases (57.1%). The expression of survivin protein did not correlate with that of ALK protein (P > 0.05). The survival rate was significantly lower in patients with high survivin protein expression (P < 0.05). In cases with ALK protein expression, the prognosis was less favorable if there was also high co-expression of survivin protein (P < 0.05). In ALK protein negative cases, prognosis did not significantly correlate with the expression of survivin protein (P > 0.05). In addition, multivariate analysis confirmed the prognosis value of ALK protein expression, survivin protein expression and constitutional symptoms. CONCLUSION: Survivin protein expression can serve as an independent prognostic predictor of unfavorable clinical outcome in patients with ALCL, especially when ALK protein is positive.

[Elevation of IFN-gamma transcription level in peripheral blood mononuclear cells of EIAV vaccinated horses].

AIM: To evaluate the relationship between the transcriptional level of IFN-gamma mRNA in peripheral blood mononuclear cells (PBMC) and immune protective response driven by inoculated horses with donkey leukocyte attenuated vaccine of EIAV(DLV), and to elucidate the immune mechanism of DLV. METHODS: A real-time PCR method was established for quantitative detection of IFN-gamma mRNA level from horse PBMCs. Twelve horses were divided into vaccination group, healthy control group, challenging control group and EIAV natural infection group. The transcriptional level and distribution of IFN-gamma mRNA in PBMCs were analyzed. The temperature and other parameters of the inoculated horses were monitored or a daily basis. Horses inoculated with DLV vaccines were challenged post-inoculated 8 months, and the change of transcriptional level of IFN-gamma mRNA during pre/post-challenge was evaluated. RESULTS: Compared with the healthy control and natural infection group. The transcriptional level of IFN-gamma mRNA was much higher in vaccination group (P<0.01), and kept increasing even after challenging with virulent EIAV strains. In the challenging control group, the transcriptional level of IFN-gamma mRNA fluctuated with the development of disease and reduced markedly during febrific episodes. CONCLUSION: This study for the first time reveals that EIAV-attenuated vaccine could induce high level transcription of IFN-gamma mRNA, with is associated closely with immune protective response induced by DLV. This finding provides some clues to elucidate the mechanism of immunity protection of DLV at the molecular level.

[Fluid management in orthotopic liver transplantation].

OBJECTIVE: To investigate the influential factors of fluid management during orthotopic liver transplantation surgery. METHODS: Ninety-six patients scheduled for orthotopic liver transplantation were divided into two groups according to the liver function: the decompensatory group (Child-Pugh score C, n=50) and the compensatory group (Child-Pugh score A or B, n=46). According to the intraoperative bleeding amount, the compensatory group was further divided into subgroup A (< 2,000 ml) and subgroup B (> or = 2,000 ml). Plasma albumin concentration and the parameters of blood coagulation were measured before and after the surgery. The intraoperative bleeding amount and the amount of the blood products infused were recorded. RESULTS: Before the surgery, prothrombin time (PT) and activated partial thromboplastin time (APTT) were significantly longer in decompensatory group than in compensatory group (both P<0.05); blood platelets (PLT) and fibrinogen were significantly lower in decompensatory group than in compensatory group (both P<0.05). After the surgery, there were no significant differences in PT, APTT, PLT, and fibrinogen between the two groups (all P>0.05). The amounts of the blood products infused intraoperatively in subgroup A were significantly lower than those in the decompensatory group (all P<0.05). There was no significant difference in the amount of the infused blood products between the subgroup B and decompensatory group (all P>0.05). CONCLUSION: During orthotopic liver transplantation surgery, patients with decompensatory liver function, or those who suffered large amounts of intraoperative bleeding, need infusion of larger amount of blood products.

[Changes of orthodontic tooth in different healing extraction sites].

OBJECTIVE: To compare the canines root resorption and alveolar bone height when they were moved into different healing extraction sites. METHODS: Twenty patients with overjet were treated by extracting the maxillary first premolars. By random determination, distalization of different lateral upper canines started after 1 week and 12 weeks. Every 4 weeks use a force of approximately 100 g to distalize the canines. The experiment was ended when bilateral canines have contacted with the second maxillary bicuspids completely. Periapical radiography of the canine in the end of the distalization was made. RESULTS: External apical root resorption and alveolar bone height of the canines have no significant difference between the different healing extraction sites. CONCLUSION: Tooth extraction site healing has not much effect on the canine external apical root resorption and alveolar bone height, when it was distalization. In practice, tooth movement into extraction sites can start early.

[Allelic loss of 6q16.3 microsatellite DNA in childhood acute lymphoblastic leukemia and bioinformatics analysis].

OBJECTIVE: To locate the cluster region of loss of heterozygosity (LOH) in children with acute lymphoblastic leukemia (ALL), and explore the new tumor suppressor gene. METHODS: Allelic loss was analyzed by PCR with 15 microsatellite markers mapping on 6q16.3. The LOH was analyzed by bioinformatics. The relationship between LOH and clinical factors was further analyzed. RESULTS: The frequency of LOH at least at one loci on 6q16.3 was 32.7%. The LOH in relapsed patients was higher than those in not relapsed. The higher frequency of LOH was observed in two regions of D6S1709-D6S1028 and D6S2160-D6S1580 at 6q16.3. GRIK2 may be a candidate of tumor suppressor gene. There are 12 ESTs may carry out new anti-oncogene. Patients with 6q LOH had higher WBC counts (P < 0.01), blast cells percentage (P < 0.01), relapse rate (P < 0.05) and chromosomal aberration (P < 0.05). CONCLUSION: D6S1709-D6S1028 and D6S2160-D6S1580 are two regions of minimus deletion on 6q16.3 in which tumor suppressor gene may exist. The LOH on 6q16.3 may be a prognostic index of children with ALL.
