[The effect of glutamine on the growth, metabolism and endostatin production of microencapsulated rCHO cells].

Cell transplantation is a promising technology in cancer therapy, however, immunological rejection is the major problem of cell transplantation. Based on the permselective property of microcapsule membrane, encapsulated cells can be immuno-protected. The normal physiological state and function expression of cells can be maintained so as to realize allo- or xenotransplantation. The microencapsulated cells grow in three dimensions, giving a more biologically representative in vivo model, which hints difference in characters of growth and metabolism compared to the monolayer cells. Therefore, characterization of growth and metabolism of microencapsulated recombinant CHO cells is essential for further large-scale culture. In present study, the effect of concentration of glutamine on the growth, metabolism and endostatin production of microencapsulated cells was investigated. In the experimental range of initial glutamine concentrations from 2.69mmol/L to 9.05mmol/L in the culture of microencapsulated recombinant CHO cells, the maximum density of active cells and multiplication ratios almost kept constant. The specific consumption rate of glucose increased with lower initial glutamine concentration (2.69mmol/L). When initial glutamine concentration was much higher (7.91mmol/L to approximately 9.05mmol/L), the specific consumption rates of both glucose and glutamine increased while the efficiencies of glucose and glutamine decreased. The highest efficiencies of glucose and glutamine utilization were observed with initial glutamine concentration of 4.97mmol/L. It was also demonstrated that glutamine had significant effect on the accumulation of endostatin. The accumulative concentration of endostain reached its peak of 546.36 ng/mL with the initial glutamine concentration of 4.97mmol/L.

[PCR-RFLP analysis on Roegneria, Elymus, Hystrix and Kengyilia in Triticeae (Poaceae)].

PCR-RFLP of Plasmon on 23 species of Roegneria, Elymus, Hystrix and Kengyilia and 1 outgroup (Triticum aestivum L.) was carried out. The segments amplified with 3 cpDNA and 3 mtDNA universal primers were digested by 15 restriction endonucleases. Among 329 bands produced, 304 were polymorphic (92.4%). PCR-RFLP results showed that polymorphisms existed between the four genera and between the species in Roegneria, Elymus, Hystrix and Kengyilia, and genetic similarity coefficient was high. Roegneria grandis, R. aristiglumis, R. elytrigioides, R. alashanica and R. magnicaespes were grouped, while R. caucasica, R. ciliaris, R. amurensis and R. japonensis clustered together. Species of Kengyilia clustered independently. Five species of Elymus, 3 of Hystrix and Roegneria kamoji grouped together. The results suggested Kengyilia was a valid genus. As more cytological and molecular data are available, the taxonomic status of Roegneria, Elymus and Hystrix will be recognized.

[Mechanical periodontal treatment combined with tetracycline for aggressive periodontitis].

OBJECTIVE: To evaluate the effect of mechanical periodontal treatment combined with tetracycline on periodontal attachment, distances of cementum-enamel junction (CEJ) to the bone defect bottom and to the alveolar crest, and avidity of serum IgG against Porphyromonas gingivalis (Pg) lipopolysaccharide (LPS) in patients with aggressive periodontitis. METHODS: Twenty-six patients with aggressive periodontitis were divided equally into two groups to receive mechanical periodontal treatment combined with tetracycline or exclusive mechanical periodontal treatment, with another 20 periodontally healthy subjects serving as the control group. In the two patient groups, routine clinical examination of the subjects was performed to record periodontal attachment level and distances of CEJ to the bottom of the bone defects and to the alveolar crest before and 3 and 6 months after the treatment. The avidity of serum IgG against Pg LPS was measured by ELISA with thiocyanate elution before and after periodontal treatment. RESULTS: Compared with patients with exclusive mechanical periodontal treatment, patients with combined treatments showed significant improvement in periodontal attachment level and the distance of CEJ to the bone defect bottom after the treatment (P<0.01 and P<0.05, respectively) with also significantly reduced avidity of serum IgG against Pg LPS (P<0.01). CONCLUSION: Mechanical periodontal treatment combined with tetracycline produces favorable effects in patients with aggressive periodontitis.

[Screening for endocrine exophthalmos-associated genes/cDNA fragments].

This study was conducted to screen the endocrine exophthalmos-associated genes or cDNA fragments and provide a basis for exploring the pathogenesis. The cDNA from the thyroid tissues of patients with hyperthyroidism and endocrine exophthalmos (HT&EE) was used as tester cDNA, and that from the thyroid tissues of patients with HT but free from EE was used as driver cDNA. The subtractive PCR products between tester and driver cDNA were obtained through two cycles of subtraction hybridization and two cycles of PCR by using suppression subtractive hybridization, and then were inserted into pT-Adv, a cloning vector. The ligated DNAs were transformed into E. coli DH5alpha competent cells and incubated for proper blue/white color development. Forty-eight white colonies were randomly picked and their inserts were colony PCR amplified. The PCR product from one of the colonies contained two inserts; the others contained single insert, having a size of 0.2 kb to 2 kb. The inserts of transformants were arrayed on nylon membrane. After cDNA/Rsa I digestion the thyroid tissues of patients with HT, and of patients with HT&EE, were labeled with digoxigenin; the nylon membranes were then hybridized respectively with the two cDNA probes for a high throughput screening for positive clones. The clones which were hybridized with the cDNA probe of HE&EE patients but not hybridized with the probe of the HT patients or showed only faint signal of hybridization, were chosen as positive clones and their inserts were candidates for the endocrine exophthalmos genes or cDNA fragments. About 50% of the clones were confirmed as positive clones. The cDNA fragments in the positive clones were the endocrine exophthalmos-associated genes or cDNA fragments. Endocrine exophthalmos

[Detection and related analysis to chromosome 22q11 microdeletion in patients with congenital heart diseases].

OBJECTIVE: To ascertain 5 short tandem repeat (STR) markers as qualified tools for detecting chromosome 22q11 deletion and to understand the prevalence and clinical importance of the deletions in patients with congenital heart diseases (CHD) from Chinese Han population. METHODS: The authors selected 5 new tetranucleotide repeat markers, 22D_4_1, 22D_4_2, 22D_4_3, 22D_4_4 and D22S873 located in the proximal region of chromosome 22q11 deletion. One hundred and sixty-three unselected CHD patients and their unaffected parents were analyzed by genotyping of these new tetranucleotide STR markers to detect 22q11 deletion. With fluorescence in situ hybridization (FISH, LSI dual color DNA probe), the deletion status was confirmed in all patients with deletions and some patients without deletions. RESULTS: The heterozygosity of these STR markers in normal population was more than 0.7, except for 22D_4_1 and 22D_4_2 that were 0.65 and 0.52 respectively. Twelve cases of 163 CHD patients (7.36%) had the deletions at chromosome 22q11. The deletions were confirmed in 9 of 12 patients by FISH, except for 2 cases who had unique nested deletion and 1 case who had nested distal deletion. One hundred and ten patients were associated with ventricular septal defect (VSD); and 9 (8.18%) of these cases had microdeletion. Twenty-one patients were associated with tetralogy of Fallot (TOF); and 3 (14.3%) of these cases had microdeletion. CONCLUSION: This study demonstrated that genotyping of 5 STR markers was a useful mean of detecting 22q11 microdeletion in clinical diagnosis owing to its rapid experimental procedure, cost effectiveness and high resolution. 22q11 deletion was common in CHD patients, particularly in VSD and TOF patients, from Chinese Han population.

[Osteopontin as a potential biomarker of metastasis and recurrence for hepatocellular carcinoma].

BACKGROUND & OBJECTIVE: Osteopontin (OPN), a secretory phosphorylated glycoprotein and integrin-binding protein, has been involved in tumorigenesis, progression, invasion, and metastasis. OPN is over-expressed in a variety of human tumors. This study was to evaluate the value of OPN detection in forecasting the postoperative metastasis and recurrence of hepatocellular carcinoma (HCC). METHODS: Plasma OPN level of 56 patients with HCC was detected before operation by enzyme-linked immunosorbent assay (ELISA), and plasma samples of 23 healthy volunteers were also analyzed as control group. The correlation of plasma OPN level to metastasis and recurrence of HCC was analyzed. RESULTS: Plasma OPN level was significantly higher in HCC group than in control group [(864.4+/-391.9) microg/L vs. (382.5+/-169.4) microg/L, P<0.001]. During the follow-up, 25 patients had tumor recurred. Plasma OPN level was significantly higher in the patients with early HCC recurrence than in those without recurrence [(961.0+/-411.6) microg/L vs. (744.6+/-323.9) microg/L, P=0.039]. Cox multivariate analysis showed that plasma OPN level, cirrhosis, and tumor size were independent predictors for HCC recurrence and metastasis. CONCLUSION: Plasma OPN level may be a potential diagnostic marker for predicting metastasis and recurrence of HCC.

[In situ identification of ANAMMOX bacteria in freshwater sediments].

In situ identification of ANAMMOX bacteria was conducted using 16S rRNA approach for sediment samples from the Xinyi River in Jiangsu Province, China. 16S rRNA clone library including 6 clone sequences was constructed. The alignment of these sequences and treeing were conducted using ARB package. Results show that the sediment samples contained 16S rRNA genes closely related to the known ANAMMOX bacterium Candidatus "Brocadia anammoxidans" (similarity of 91%). They also contained 16S rRNA gene sequences from a new branch of Planctomycetes distantly related to the ANAMMOX sequence cluster. However, the microbiological characteristics of these Planctomycetes are to be studied in the future. The detection of ANAMMOX bacteria will lead to further research on ANAMMOX process in the remediation and restoration of freshwater aquatic ecosystems and new understanding of the nitrogen cycle.

Cardiovascular disease risk factor awareness in American Indian communities: the strong heart study.

OBJECTIVE: To use data from the longitudinal Strong Heart Study (SHS) to determine the level of awareness about risk factors for heart disease among 13 populations of American Indians in Arizona, Oklahoma, and South/ North Dakota. The aim of this study is to assess awareness of nine major risk factors for heart disease among participants in SHS. METHODS: During July 1993 to December 1995 (phase II of SHS), 3638 participants ages 46 to 80 years (mean age 60) were asked if nine known risk factors for cardiovascular disease affect a person's chances of getting heart disease; 3226 (89%) participants completed the study and met the method reliability criteria for inclusion. RESULTS: Among each of the nine risk factors, the percentage of correct answers provided by study participants ranged from 70% (family history of heart disease) to 90% (being very overweight). Participants with hypertension (90% vs 86%, P<.05) and diabetes mellitus (81% vs 71%, P<.05) were more likely than those without these disorders to know they were heart disease risk factors. For all nine risk factors, the percentage of correct answers was lower (P<.05) among smokers than among nonsmokers. In multivariate logistic regression analyses, female sex, advanced education, and being from Oklahoma were significantly associated with heart disease awareness. CONCLUSION: Although overall risk factor awareness for heart disease was high, subgroups were identified who could benefit from culturally appropriate health education and other interventions to motivate health prevention actions, especially for smoking.

[In vitro study on proliferation and multi-lineage differentiation potential of adipose-derived cells].

Adipose were obtained from patients underwent liposuction treatment (total 19 female donors, 31.5 +/- 5.8 years old). Liposuction tissues were digested with type I collagenase, cells were isolated and cultured up to passage 10. To evaluate the proliferation potential of ADCs, growth curve and cumulative population doubling were achieved by cell counting. CD29,CD105, CD106, CD166, CD49d, CD34, CD31, 3G5 were analyzed by flow cytometry and immunocytochemistry to characterize the cell population. The multi-lineage potential of ADCs was testified by differentiating cells with osteogenic, chondrogenic and adipogenic inducer. A total of 5x10(7) nucleared cells could be obtained from 300ml liposuction tissues. After in vitro cultivation, cumulative population doubling number reached 15.53 at passage 10 (average 1.59 +/- 0.224 /passage). Flow cytometry and immunocytochemistry showed that ADCs expressed high level (>60%) of stem cell-related antigen (CD29, CD105, CD106, CD166), while cells expressed hematopoiesis-related antigen CD34 and CD31 around 7.3% and 29.2% respectively. Collagen II (both in mRNA and protein level) was detected in chondrogenic differentiation. The calcified nodules were observed by von Kossa and Alizarn Red staining and the expressions of AKP and Osteonectin were detected by RT-PCR in osteogenic differentiation. PPARr2, GLU-4, and Leptin genes were detected in adipogenic differentiation and intracellular lipid droplets could be observed by Oil Red staining. ADCs can be abundantly harvested and have high proliferative and multi-lineage differentiation potential.

[Cost-effectiveness analysis of emergency endoscopic variceal ligation plus octreotide in the treatment of acute esophageal variceal bleeding in cirrhotic patients].

OBJECTIVE: To compare and evaluate the cost and effectiveness of endoscopic variceal ligation (EVL) at emergency plus octreotide versus octreotide alone in the treatment of acute esophageal variceal bleeding in cirrhotic patients. METHODS: Seventy-eight patients with active variceal bleeding under emergency endoscope, were assigned to two groups receiving either combined therapy of EVL at emergency and octreotide ('EVL' group) or a continuous infusion of octreotide alone ('octreotide' group). Both efficacy and cost-effectiveness were observed. RESULTS: There were no significant differences between the two groups in patients' characteristics, supporting treatment or general treatment. In group EVL, there appeared a significantly higher rate in controlling bleeding and lower complication rate than that of octreotide group(94.4% vs.78.6%, P = 0.045 and 19.4% vs. 42.9%, P = 0.027, respectively). Early rebleeding and mortality rate were also lower in group EVL, but with no significant differences between them (2.9% vs. 7.7%, P = 0.358 and 5.6% vs. 14.3%, P = 0.205, respectively). The combined therapy had a significantly shorter time of hemostasis, less administration of octreoid, fewer units of blood transfusion and shorter hospital stay (P < 0.001). The median costs of the combined therapy and octreotide alone were RMB 9046.5 Yuan and 13 743.6 Yuan, respectively (P = 0.045). The cost-effective ratio of group EVL seemed superior to that of octreoid group. CONCLUSION: The therapeutic scheme of emergency EVL plus octreotide was a more cost-effective one for controlling acute esophageal variceal bleeding.

[Effects of shenmai injection on the clinical efficacy of noninvasive ventilation in patients with severe respiratory failure caused by chronic obstructive pulmonary disease].

OBJECTIVE: To study the effect of Shenmai Injection (SMI) on the clinical efficacy of noninvasive positive-pressure ventilation (NPPV) in treating patients with severe respiratory failure (RF) caused by chronic obstructive pulmonary disease (COPD). METHODS: Patients in the control group (n=23) were treated with conventional therapy plus NPPV, and those in the treatment group (n=25) received the same treatment but with intravenous infusion of SMI 30 ml once a day additionally. The treatment lasted for 10 days. Arterial oxygen saturation (SaO2) was continuously monitored in all patients, arterial blood gas values was determined before treatment and at the 6th hour, the 24th hour, the 3rd day and the 7th day after treatment, and consciousness, breath, heart rate and severity of dyspnea were observed during the treating period. RESULTS: All patients in the treatment group were markedly improved and discharged from hospital, while in the control group 20 were improved and discharged from hospital, but the other 3 failed to be remitted and received the conventional invasive mechanical ventilation. The duration of hospitalization and ventilation were 11.3 +/- 2.5 d and 5.8 +/- 2.8 d in the treatment group, and 13.2 +/- 2.7 d and 7.5 +/- 3.0 d in the control group, showing significant difference in the two indexes between the two groups (P < 0.05). The dyspnea score and the arterial blood gas values determined on the 3rd and 7th day were significantly improved in the treatment group than those in the control group (P < 0.05 or P < 0.01). The incidence of endotracheal intubation caused adverse reaction, such as facial-skin crush wound, abdominal distention and contaminated infection in the treatment group was obviously lessened as compared with the control group. CONCLUSION: SMI could shorten the duration of hospitalization and mechanical ventilation, reduce trachea cannulation, and decrease the incidence of complications and adverse reactions in the patients with severe RF due to COPD treated with NPPV.
