Is exogenous tissue plasminogen activator necessary for antithrombotic efficacy of an inhibitor of thrombin activatable fibrinolysis inhibitor (TAFI) in rats?

INTRODUCTION: TAFI indirectly reduces the action of tPA on plasminogen. Whether exogenous tPA is necessary for TAFI inhibitor efficacy is unclear. Potato carboxypeptidase inhibitor (PCI), a TAFI inhibitor, has shown variable tPA dependence in rat models of arteriovenous shunt thrombosis (required) and microthrombosis (not required). This study was designed to further explore the importance of exogenous tPA in revealing PCI activity in rat models of venous and arterial thrombosis and provoked bleeding. METHODS: PCI was given as a bolus (5, 10 mg/kg) +/- infusion (5, 10 mg/kg/h) and with or without low dose tPA (5, 10, 25 microg/kg/min). In each instance tPA was adjusted to produce subthreshold thrombus reduction. Arterial thrombosis was induced by FeCl2; venous thrombosis by tissue factor or FeCl2. Bleeding was induced by kidney incision with PCI given (5 mg + 5 mg/kg/h) in the presence or absence of tPA (10, 150, 200 microg/kg/min). RESULTS: PCI was ineffective without exogenous tPA in all tested thrombosis models. With exogenous tPA, PCI decreased thrombus weight 85% in tissue factor thrombosis, 59% in FeCl2 thrombosis, and 46% in arterial thrombosis. PCI prolonged bleeding only when combined with a relatively high tPA dose (200 microg/kg/min) that increased bleeding alone. CONCLUSIONS: If the current results predict clinical efficacy, the need for exogenous tPA in combination with TAFI inhibition is a potential problem. However, in acute settings where intravenous fibrinolytics are administered, or indications in which tPA production increases, TAFI inhibitors may prove to be safe and moderately effective profibrinolytic agents.

A novel alphaB-crystallin mutation associated with autosomal dominant congenital lamellar cataract.

PURPOSE: To identify the mutation and the underlying mechanism of cataractogenesis in a five-generation autosomal dominant congenital lamellar cataract family. METHODS: Nineteen mutation hot spots associated with autosomal dominant congenital cataract have been screened by PCR-based DNA sequencing. Recombinant wild-type and mutant human alphaB-crystallin were expressed in Escherichia coli and purified to homogeneity. The recombinant proteins were characterized by far UV circular dichroism, intrinsic tryptophan fluorescence, Bis-ANS fluorescence, multiangle light-scattering, and the measurement of chaperone activity. RESULTS: A novel missense mutation in the third exon of the alphaB-crystallin gene (CRYAB) was found to cosegregate with the disease phenotype in a five-generation autosomal dominant congenital lamellar cataract family. The single-base substitution (G-->A) results in the replacement of the aspartic acid residue by asparagine at codon 140. Far UV circular dichroism spectra indicated that the mutation did not significantly alter the secondary structure. However, intrinsic tryptophan fluorescence spectra and Bis-ANS fluorescence spectra indicated that the mutation resulted in alterations in tertiary and/or quaternary structures and surface hydrophobicity of alphaB-crystallin. Multiangle light-scattering measurement showed that the mutant alphaB-crystallin tended to aggregate into a larger complex than did the wild-type. The mutant alphaB-crystallin was more susceptible than wild-type to thermal denaturation. Furthermore, the mutant alphaB-crystallin not only lost its chaperone-like activity, it also behaved as a dominant negative which inhibited the chaperone-like activity of wild-type alphaB-crystallin. CONCLUSIONS: These data indicate that the altered tertiary and/or quaternary structures and the dominant negative effect of D140N mutant alphaB-crystallin underlie the molecular mechanism of cataractogenesis of this pedigree.

[Compare the proteasome activity in the epithelium of human age-related cataract and normal lens].

PURPOSE: To compare the chymotrypsin-like and trypsin-like activity of proteasome in the epithelium of human age-related cataract and normal lens, in order to study the mechanism of the proteasome in the formation and development of age-related cataract. METHODS: Cataractous lens were obtained during cataract surgery of Zhongshan Ophthalmic Center, and normal age-matched lens were obtained from Guangdong eye bank. The capsules were frozen and thawed repeatedly to ensure the epithelial cells detached from the capsules, then the capsules were removed. All samples were added with LLVY or VGR, then detected the fluorescent intensity at different time. RESULTS: The proteasome activity of normal lens epithelium was significantly high than cataractous lens epithelium (P < 0.01); the chymotrypsin-like activity of the proteasome of cortical and non-cortical cataract had no difference (P > 0.05); MG-132 can significantly inhibit the proteasome activity of cataractous and normal lens. CONCLUSION: Compared to normal age-matched lens, chymotrypsin-like and trypsin-like activity of proteasome in the epithelium of age-related cataract lens were significantly declined, indicating that the decrease of the proteasome activity may play an important role in the formation and development of age-related cataract.

[Biometry of intraocular lenses using Pentacam Scheimpflug images].

PURPOSE: To compare differences in intraocular lens (IOL) decentration, tilt and anterior chamber depth (ACD) using Pentacam-Scheimpflug System images in eyes having phacoemulsification with implantation of 1 of 3 types of IOLs. METHODS: This study comprised 117 cataractous eyes of 109 patients having phacoemulsification and IOL implantation. The eyes were randomized into 3 groups based on IOL type. The degree of IOL decentration, tilt and ACD was measured using Oculus Pentacam System 1 day and 1, 3, 6 and 12 months after surgery. RESULTS: There was no significant change during the 12 months follow-up period in decentration or tilt in each group (P > 0.05). There was no significant difference among the groups in IOL decentration or tilt at any time point (P > 0.05). The ACD did not change after surgery in the Acrysof SA60AT group (P > 0.05) but showed significant shallowing in the Tecnis Z9001 group and the Sensar AR40e group (P < 0.05). There was significant difference among the groups in ACD only at 1 day after surgery (P < 0.05). CONCLUSIONS: The IOL stability in eyes that received a 1-piece Acrysof SA60AT IOL with flexible haptics was better than the results with normal capsular bag IOL implantation and an intact continuous curvilinear capsulorhexis that received a 3-piece IOL with rigid haptics.

Pigment epithelium-derived factor in cataractous aqueous humor and lens epithelial cells.

PURPOSE: To study the characteristics of PEDF in cataractous aqueous humor and its expression in human lens epithelium. METHODS: The PEDF concentration in the aqueous humor was measured by enzyme-linked immunosorbent assay in senile (130 cases) and congenital (18 cases) cataract patients who underwent cataract phacoemulsification extraction surgery. Anterior lens capsular specimens were obtained from these patients to count lens epithelial cells (LEC) density. The Lens Opacities Classification System III was used to classify the senile cataracts as cortical, nuclear, posterior subcapsular and mixed types of opacity, and quantitative analysis of the nuclear opacities was performed by Pentacam Scheimpflug imaging system. Anterior lens capsular specimens from another senile (10 cases) and congenital (10 cases) cataract were collected for immunofluorescence with polyclonal antibodies specific to human pigment epithelium-derived factor (PEDF). RESULTS: The mean aqueous level of PEDF was (178.9 +/- 87.5) ng/ml, and there was negative linear correlation of PEDF level and age (r = 0.811, P < 0.001) . In senile cases, the aqueous PEDF concentration decreased with increasing nuclear opacities (r = 0.447, P < 0.01), and the mean PEDF level in nuclear cataract was significantly lower than that in posterior subcapsular opacity (P < 0.01). PEDF immunostaining was detected in LEC of all capsular specimens. CONCLUSION: The PEDF level in human aqueous humor is related to age, types of cataracts and lens opacity. PEDF also express in human LEC. The study results suggest PEDF may regulate and/or protect LEC by paracrine and autocrine, and lack of PEDF may play a role in cataractogenesis.

[An experimental wound-temperature study of Bimanual microphacoemulsification using the Whitestar System].

PURPOSE: To comparatively study the wound temperature in bimanual microphacoemulsification and conventional phacoemulsification. METHOD: Sixty porcine eyes were equally divided into 4 groups which respectively received bimanual microphacoemulsification and conventional phacoemulsification using Whitestar System with the occlusion mode switched on. The aspiration tube of the phacoemulsification unit was clamped to simulate the state of occluded phaco tip. Wound temperature values at U/S Time 30, 60, 90 and 120 s were measured and recorded using a type K thermocouple digital thermometer. RESULTS: With phaco tips both occluded and un-occluded, the wound temperature measured during bimanual microphacoemulsification was within the range of 21.0-30.7 degrees C, which is lower than corresponding values of the conventional phaco groups. The temperature was statistically different between 2 procedures in the un-occluded state (P < 0.05). And the inter-group difference was statistically significant in the occluded state(P < 0.01). CONCLUSION: Compared to conventional phacoemulsification, bimanual microphacoemulsification using the Whitestar System produced less heat at the surgical incisions.

Refractive error and visual impairment in urban children in southern china.

PURPOSE: To assess the prevalence of refractive error and visual impairment in school-age children in a metropolitan area of southern China. METHODS: Random selection of geographically defined clusters was used to identify children 5 to 15 years of age in Guangzhou. Children in 22 clusters were enumerated through a door-to-door survey and examined in 71 schools and 19 community facilities from October 2002 to January 2003. The examination included visual acuity measurements, ocular motility evaluation, retinoscopy, and autorefraction under cycloplegia and examination of the external eye, anterior segment, media, and fundus. RESULTS: A total of 5053 children living in 4814 households were enumerated, and 4364 (86.4%) were examined. The prevalence of uncorrected, presenting, and best-corrected visual acuity 20/40 or worse in the better eye was 22.3%, 10.3%, and 0.62%, respectively. Refractive error was the cause in 94.9% of the 2335 eyes with reduced vision, amblyopia in 1.9%, other causes in 0.4%, and unexplained causes in the remaining 2.8%. External and anterior segment abnormalities were seen in 1496 (34.3%) children, mainly minor conjunctival abnormalities. Media and fundus abnormalities were observed in 32 (0.73%) children. Myopia (spherical equivalent of at least -0.50 D in either eye) measured with retinoscopy affected 73.1% of children 15 years of age, 78.4% with autorefraction. The prevalence of myopia was 3.3% in 5-year-olds with retinoscopy and 5.7% with autorefraction. Females had a significantly higher risk of myopia. Hyperopia (+2.00 D or more) measured with retinoscopy was present in 16.7% of 5-year-olds, 17.0% with autorefraction. The prevalence of hyperopia was below 1% in 15-year-olds, with both methods. Astigmatism (cylinder of > or = 0.75 D) was present in 33.6% of children with retinoscopy and in 42.7% with autorefraction. CONCLUSIONS: The prevalence of reduced vision because of myopia is high in school-age children living in metropolitan Guangzhou, representing an important public health problem. One third of these children do not have the necessary corrective spectacles. Effective strategies are needed to eliminate this easily treated cause of significant visual impairment.

[Report of gene mutation hot spots analysis in one congenital cataract pedigree].

PURPOSE: To analyse autosomal dominant gene mutation hot spots in a four-generation Chinese congenital cataract pedigree. METHODS: Nineteen family members of the pedigree (including eight affected and eleven unaffected individuals) were enrolled into the study with informed consent. The eight affected individuals underwent a full ophthalmologic and general examination to rude out any concomitant disorders. The eleven unaffected individuals only underwent ophthalmologic examination. Blood samples were taken from the nineteen subjects for genomic DNA preparation. Seventeen distinct loci on ten different genes (including CRYAA, CRYAB, CRYBA1/A3, CRYGD, GJA8, CRYGC, CRYBB2, GJA3, MIP and BFSP2) have been identified as causes of autosomal dominant congenital cataract (ADCC). Polymerase chain reaction (PCR) amplification of ten gene segments encompassing seventeen mutation hot spots were carried out in the nineteen subjects of the pedigree. Sequences and analysis of PCR products were then performed to detect corresponding mutations. RESULT: No mutation was found on the seventeen autosomal dominant mutation hot spots in all nineteen subjects. CONCLUSION: Preliminarily exclude the association of the seventeen published autosomal dominant mutation hot spots with ADCC in this family.

Changes of blood aqueous barrier after phacoemulsification in rabbits.

PURPOSE: To evaluate the function and the morphological changes of the blood aqueous barrier (BAB) after phacoemulsification and to provide the physiological mechanism of BAB dysfunction. METHODS: Phacoemulsification was performed on 12 rabbit eyes. Aqueous humor was abstracted and the concentrations of aqueous humor protein were examined preoperatively and on postoperative days 1, 7, 14, 21, 30 with Lowry. After the iris and the ciliary body were removed from the rabbit eyes, we selected lanthanum nitrate as a tracer, then ultrastructural changes of BAB were investigated with transmission electron microscopy. RESULTS: Highly statistically significant differences (P < 0.05) were found between preoperative aqueous humor protein concentration and those measured on days 1, 7, 14, 21 and 30 after surgery. Under transmission electron microscopy, a part of the endothelial cells and BAB damaged as shown by the leakage of lanthanum tracing into intravessels through the broad space were observed. CONCLUSION: The results suggest that there is a blood aqueous barrier breakdown after phacoemulsification. The morphological basis of BAB damage is the reversible opening of tight junctions between endothelial cells.

[Phacoemulsification in eyes with posterior polar cataract and foldable intraocular lens implantation].

PURPOSE: To study the surgical technique of phacoemulsification with foldable intraocular lens(IOL) implantation in eyes with posterior polar cataract and the managements of intraoperative or postoperative complications. METHODS: Phacoemulsification with removal of posterior plaque, or continuous curvilinear posterior capsulorhexis and foldable IOL implantation were carried out after topical anesthesia in 46 cases (61 eyes) with age ranging from 38 to 78 years old (average 57). RESULTS: All eyes were implanted foldable IOL. Fifty-one eyes revealed intact posterior capsule, six of which revealed posterior capsule opacification. Ten eyes developed posterior capsule rupture. However, four eyes need to perform anterior vitrectomy. Pars plana vitrectomy was performed in one eye with uveitis and secondary glaucoma induced by soft shell of nucleus sank into the vitreous cavity. Fifty-eight in 61 IOL were implanted in the capsular bags, while 3 IOLs were fixed in the ciliary sulcus. CONCLUSION: Phacoemulsification with foldable IOL implantation is an effective method to treat posterior polar cataract. Dealing with the posterior plaque during phacoemulsification carefully may decrease intraoperative and postoperative complications and remarkably improve postoperative visual acuity.

[Role of p38MAPKs pathway in the growth inhibition of rabbit lens epithelial cells induced by EGCG].

PURPOSE: Study the role of p38MAPKs pathway in the growth inhibition of rabbit lens epithelial cells(LECs) induced by green tea extract (-)-Epigallocatechin-3-gallate (EGCG). METHODS: (1) MTT colormetric assay was used to study the effect of p38 specific inhibitors SB203580 on LECs growth. (2) MTT colormetric was used to study the effect of SB203580 on LECs growth inhibition induced by EGCG. (3) Western blotting methods was used to study the effect of EGCG on the kinase phosphorylation- and nonphosphorylation-level of p38. RESULTS: (1) When LECs were preincubated with 25 mumol/L, 50 mumol/L SB203580 for 1 h, 100 mumol/L EGCG had little influence on LECs proliferation. It showed great protects effect significantly when increased to 200 mumol/L. (2) Basic phosphorylation-level of p38 were very weak in LECs, but it increased with EGCG concentration increased and reached maximum at 200 mumol/L. 15 min after 200 mumol/L was added, the phosphorylation-level was the highest and began to fall, but even at the end of our test, it still stayed higher than that of basic level. The nonphosphorylation-level of p38 remained stable in all test. CONCLUSIONS: The LECs proliferation inhibition induced by EGCG is through p38 pathway at lease partially.

Vascular endothelial growth factor is increased in aqueous humor of glaucomatous eyes.

PURPOSE: To assess the concentrations of vascular endothelial growth factor (VEGF) in aqueous humor in eyes with and without glaucoma. METHODS: Concentrations of VEGF were measured using a sandwich ELISA kit in aqueous humor aspirates taken during anterior segment surgery from 87 patients, of whom 54 had glaucoma (27 primary open-angle glaucoma, 8 angle-closure glaucoma, 16 exfoliative glaucoma) and 33 had cataract only. RESULTS: Vascular endothelial growth factor was detected in all samples. The concentration in eyes with cataract only without glaucoma was 102.4 +/- 29.7 pg/mL (mean +/- SD), which was significantly lower than that from eyes with glaucoma (146.7 +/- 51.8 pg/mL). There were no significant differences between primary open-angle glaucoma (140.4 +/- 51.0 pg/mL), angle-closure glaucoma (142.8 +/- 40.2 pg/mL), and exfoliative glaucoma (158.6 +/- 58.9 pg/mL). An unusually high VEGF concentration was detected in one eye with neovascular glaucoma (759 pg/mL) and two eyes with uveitic glaucoma (322 pg/mL). No effect of age, gender, or previous history of medical, laser, or surgical treatment of the aqueous humor VEGF concentration could be detected ( > 0.05). Aqueous humor and plasma VEGF concentrations were measured and compared in 46 patients. The aqueous humor VEGF concentration (144.2 +/- 107.9 pg/mL) was significantly higher ( < 0.01) than the plasma concentration (79.2 +/- 46.1 pg/mL). No significant correlation was found between aqueous humor and plasma VEGF concentrations. CONCLUSION: Aqueous VEGF concentration is increased in eyes with glaucoma.

[Study on apoptosis in myelodysplastic syndromes by DNA in situ end labelling combined with APAAP].

OBJECTIVE: To investigate the total in situ apoptotic cell number and the apoptotic situation in erythroid cell and megakaryocytes in patients with myelodysplastic syndromes (MDS). METHODS: Apoptosis cell number and the apoptotic situation of erythroid cell and megakaryocytes were analysed on cold embedded bone marrow sections from 25 MDS patients by DNA in situ end labelling (ISEL)/alkaline phosphatase anti-alkaline phosphatase (APAAP) double stained techniques. Fourteen cases of iron deficiency anemia (IDA) were taken as control. RESULTS: Mean apoptotic cell numbers in MDS and control group were (39.44 +/- 29.34)/mm(2) and (13.43 +/- 8.39)/mm(2) respectively (P < 0.01). RA/RAS subtypes had a higher apoptosis ratio (47.56 +/- 32.86/mm(2)) than that in RAEB/RAEB-t subtypes (21.87 +/- 13.65/mm(2)) (P < 0.05). Double staining showed similar apoptosis percentage in erythroid cell and megakaryocytes in MDS patients comparing with that of controls (P > 0.05). Some apoptotic cells showing erythroid or megakaryocytic morphologic characteristics expressed no cluster differentiation antigen. CONCLUSION: Overapoptosis existed in MDS, RA/RAS group had a higher apoptosis ratio than RAEB/RAEB-t group. No obvious increased apoptosis in erythroid cell and megakaryocytes was observed in MDS perhaps due to the loss of surface antigens in later stages of apoptotic cells.

[Clinical use of capsular tension ring during phacoemulsification in subluxated lens].

OBJECTIVE: To investigate the safety of capsular tension ring implantation during phacoemul- sification in the subluxated lens. METHODS: Capsular tension ring was implanted in 13 cases (14 eyes) of subluxated lens after continuous curvilinear capsulorrhexis during phacoemulsification. The luxated area was ranged from one third to three quarters of a circle. The capsular tension ring was necessary to be fixated on sclera if the luxated area was larger than half-quarter of a circle. After the ring was implanted, phacoemulsification was performed and foldable intraocular lens (IOL) was implanted. RESULTS: The capsular tension ring was successfully implanted in 14 eyes, including 11 IOLs fixated in the bag and 3 in ciliary sulcus. IOL positions in 12 cases were normal, and in 2 eyes, had slight tilt. The most common intraoperative complication was the rupture of capsular bag, and secondly, the loss of vitreous body. CONCLUSION: Capsular tension ring implantation is safe in the patient of lens subluxation during phacoemulsification. It can prevent the IOL decentration and decrease the vitreoretinal complication.

[Macular image changes of optical coherence tomography after phacoemulsification].

OBJECTIVE: To investigate the effects of phacoemulsification on the macula following uncomplicated phacoemulsification by optical coherence tomography (OCT). METHODS: Eighty eyes of the senile cataract were chosen randomly. The uncomplicated phacoemulsification was performed. OCT was examined preoperatively and 1 week after the surgery. Preoperative visual acuity, the retinal thickness and phaco power were compared with those after surgery. RESULTS: In 80 eyes, the preoperative mean foveal thickness was (142.9 +/- 16.7) micrometer and the postoperative (157.9 +/- 36.7) micrometer, the difference being not significant (P > 0.05). Three eyes had macular edema 1 week after surgery. In 11 eyes with Tyndall sign (+ +), the mean postoperative foveal thickness was thicker than the mean preoperative value (P < 0.05). In lower phaco power group, the mean postoperative foveal thickness was (156.2 +/- 18.3) micrometer and the higher phaco power group was (172.6 +/- 32.9) microm (P < 0.05). The best corrected visual acuity after surgery had a negative correlation with the retinal thickness. CONCLUSIONS: The retinal thickening and macular edema can be found after uncomplicated phacoemulsification. The higher phaco power results in significant inflammation and thicker retina. The visual consequences were proportional to the degrees of macular thickening.

[Tear film changes after phacoemulsification].

OBJECTIVE: To evaluate tear film changes after phacoemulsification. METHODS: This prospective randomized study involved 68 consecutive patients with age-related cataract (79 eyes) who underwent phacoemulsification. Tear break-up time, Schirmer I test (S I t) value, corneal fluorescein staining, tear film pattern by DR-1 tear interfermetry and the height of tear meniscus were measured preoperatively and 1 day and 2, 7, 14, 30, 180 days postoperatively. RESULTS: At 1 day and 2 days postoperatively, the mean tear break-up time reduced greatly, and the number of patients who showed III to V tear film pattern, the mean S I t value, the height of tear meniscus and fluorescein staining scores increased significantly (P < 0.001 for day 1, P < 0.005 for day 2). S I t value at 7 days postoperatively (P = 0.831) and the height of tear meniscus at 14 days returned to their preoperative values (P = 1.000). The tear break-up time, corneal fluorescein staining scores and the grades of tear film pattern all recovered at 30 days postoperatively (P > 0.05). At 30 days postoperatively, 19.3% of patients showed shorter tear break-up time and less S I t value than that of preoperative day. In addition, 1/9 of the patients with normal preoperative tear film experienced dry eye at 30 days postoperatively. The cases with tear break-up time < 10 seconds compared to patients with tear break-up time >/= 10 seconds were more easily to show tear instability postoperatively (relative risk 13.5, 8.25, 20.0, 39.0, 8.6, 3.9 at 1 day and 2, 7, 14, 30, 180 postoperative days, respectively). CONCLUSION: Phacoemulsification significantly alters the tear break-up time, S I t value, corneal fluorescein staining, tear film pattern and the height of tear meniscus. Some patients with normal tear film may experience dry eye after surgery. Patients with preexisting tear break-up time < 10 seconds are easily at risk of experiencing the tear film instability postoperatively.

[Detection of cell cycle related gene expression in cultured immortalized human lens epithelial cell].

OBJECTIVE: To investigate the gene expression of cyclin E, cyclin-dependent-kinase 2 (CDK(2)) and CDK inhibitor (CKI) p21 during cell cycle of the human lens epithelial cell (HLEC) for elucidating the molecular mechanism of proliferation of HLEC and the formation of posterior capsular opacification (PCO). METHODS: HLE-B3 cell line was cultured in vitro under MEM medium with 15% fetal bovine serum. The cell cycle was analyzed by flow cytometry (FCM) system. The cell cycle was synchronized by double thymidine block method. The mRNA was detected by reverse transcription polymerase chain reaction (RT-PCR). The total RNA was separated from cells at different phases. Protein expression was detected with immunohistochemistry or western blotting method. The primary antibodies were rabbit anti-human cyclin E polyclonal antibody, mouse anti-human p21 and CDK(2) monoclonal antibody, respectively. RESULTS: FCM showed that G(1) phase was about 33.5%, S phase cells about 46.1%, and the G(2) phase about 20.4%, total cells in G(2) and S phase were over 60%. It was suggested that HLE- B3 cells have relatively more capacity of mitosis than normal LEC. The mRNA expression of CDK(2) was found at all phases of cell cycle during synchronization, but cyclin E mRNA only expressed during synchronized cells, and not expressed in non-synchronized cells. Only Sphase cells could express very low p21 mRNA. There was no any positive signal of cyclin E by immunohistochemistry or western blot. CDK(2) protein detected by Western blot expressed at all phases. The p21 protein was expressed in a few cells by immunohistochemistry, but p21 could not detected by western blotting. These results showed cyclin E and CDK(2) were higher expressed than p21 in transcriptional level and translational level. CONCLUSIONS: There is mRNA expression of cyclin E, CDK(2) and p21 during the cell cycle in HLE-B3 cell line, but it means the imbalance of the association of CDK and CKI. These results suggest that the imbalance expression of cyclin, CDK and p21 be the molecular mechanism of PCO.

[To establish and observe the experimental lens regeneration model in rabbits].

PURPOSE: To establish the lens regeneration model in New Zealand rabbits, observe the lens regeneration process to investigate the mechanism of lens epithelial cell growth and lens regeneration. METHODS: Lens extraction by endocapsular phacoemulsification was performed on 8 experimental eyes in New Zealand albino rabbits weighing approximately 1.5 - 2.5 kg. To restore a relative intact capsule bag, a little needle tip was inserted through the limbal wound to perform a 2 mm-lined anterior capsulotomy, which just permitted the insertion of a 21G phacoemulsification tip. Phacoemulsification extraction was applied to remove all lens cortical material and then Healon was injected intracapsularly to distend the capsule bag. With slitlamp, the procedure of le ns regeneration were observed and taken photographs. A histologic study on regenerated lens were also performed. RESULTS: The relative 3 different results on experimental eyes according to the size of anterior capsulotomy in surgical procedure. In 4 eyes, when the size of anterior capsulotomy is approximately 2 mm and the capsule bag is relatively intact, lens regeneration occured and it has a relatively normal translucent cortex and central nuclear opacity. Among other 4 surgical eyes, the lower half part of lens regeneration was still observed in one eye though the upper half of anterior capsule was damaged, whereas in other 3, owing to the whole anterior capsule lack nearly, lens regrowth was not noted in areas of pupil under the slitlamp biomicroscopy observation and the posterior capsule appeared as gray-white membrane opacity. Histologic study showed that lens epithelial cells in equatorial zone proliferate and differentiate into lens fibers and continues to refill the whole capsule bag. Lens fiber alignment was regular in the periphery of regenerated lens, whereas in the center opacity nuclear, the bulk of the regenerated lens material consisted of an eosinophilic disorganized lens fibers. In addition, there were much of irregular cellular proliferation in capsule wrinkling. CONCLUSION: Capsule is the support material of lens epithelial cells regrowth. Lens regeneration occurred in rabbits after restoring lens capsular integrity following endocapsule phacoemulsification lens extraction, which was the result of the lens epithelial cells proliferation and differentiation. The cellular proliferation and differentiation was associated with the of capsule morphosis.
