Transforming growth factor-beta stimulates parathyroid hormone-related protein and osteolytic metastases via Smad and mitogen-activated protein kinase signaling pathways.

Transforming growth factor (TGF)-beta promotes breast cancer metastasis to bone. To determine whether the osteolytic factor parathyroid hormone-related protein (PTHrP) is the primary mediator of the tumor response to TGF-beta, mice were inoculated with MDA-MB-231 breast cancer cells expressing a constitutively active TGF-beta type I receptor. Treatment of the mice with a PTHrP-neutralizing antibody greatly decreased osteolytic bone metastases. There were fewer osteoclasts and significantly decreased tumor area in the antibody-treated mice. TGF-beta can signal through both Smad and mitogen-activated protein (MAP) kinase pathways. Stable transfection of wild-type Smad2, Smad3, or Smad4 increased TGF-beta-stimulated PTHrP secretion, whereas dominant-negative Smad2, Smad3, or Smad4 only partially reduced TGF-beta-stimulated PTHrP secretion. When the cells were treated with a variety of protein kinases inhibitors, only specific inhibitors of the p38 MAP kinase pathway significantly reduced both basal and TGF-beta-stimulated PTHrP production. The combination of Smad dominant-negative blockade and p38 MAP kinase inhibition resulted in complete inhibition of TGF-beta-stimulated PTHrP production. Furthermore, TGF-beta treatment of MDA-MB-231 cells resulted in a rapid phosphorylation of p38 MAP kinase. Thus, the p38 MAP kinase pathway appears to be a major component of Smad-independent signaling by TGF-beta and may provide a new molecular target for anti-osteolytic therapy.

New rigid angular dicarboxylic acid for the construction of nanoscopic supramolecules: from a molecular rectangle to a 1-D coordination polymer.

A new rigid angular bridging ligand, 7-oxa-dibenzofluorene-3,11-dicarboxylic acid (H(2)L), was synthesized by cyanation of known rac-6,6'-dibromo-1,1'-bi-2-naphthol followed by ring closure and hydrolysis with concentrated sulfuric acid and used for the self-assembly of nanoscopic molecular rectangle [Cu(4)(L)(4)(Py)(8)].2DMF.10H(2)O, 1, and 1-D coordination polymer [Co(2)(L)(2)(Py)(4)].2DMF.2H(2)O, 2. Both 1 and 2 contain open channels occupied by DMF and water guest molecules. Effects of extracorporeal shock wave lithotripsy on plasma levels of nitric oxide and cyclic nucleotides in human subjects.

PURPOSE: The role of extracorporeal shock wave lithotripsy (ESWL, Dornier Medizintechnik GmbH, Wessling Munchen, Germany) on plasma nitric oxide (NO) and cyclic nucleotides was investigated. It is known that ESWL increased plasma levels of vasoactive substances. MATERIALS AND METHODS: A total of 28 patients undergoing ESWL for renal (14) and lower ureteral (14) stones participated in a prospective study. Peripheral blood samples were analyzed for plasma nitrite, NO metabolite, cyclic 3', 5'-guanosine monophosphate (cGMP), cyclic 3',5'-adenosine monophosphate and plasma renin activity before, immediately after, 30 and 60 minutes after ESWL. Urine samples were also analyzed for nitrite before, immediately after, 30 and 60 minutes after ESWL. Blood pressure and pulse rate were measured before and after ESWL. RESULTS: ESWL increased mean plasma nitrite levels. Before, immediately after, 30 and 60 minutes after ESWL mean plasma nitrate plus or minus standard error in patients with renal versus lower ureteral stones was 43.6 +/- 8.5, 52.8 +/- 7.4, 54.0 +/- 8.0 and 52.2 +/- 9.1 versus 27 +/- 5.8, 24.6 +/- 4.7, 23.6 +/- 4.8 and 28.0 +/- 7.1 nM., respectively. Before, immediately after, 30 and 60 minutes after ESWL mean plasma cGMP in patients with renal versus lower ureteral stones was 1.21 +/- 0.1, 1.64 +/- 0.16, 1.52 +/- 0.12 and 1.58 +/- 0.14 versus 1.45 +/- 0.18, 1.52 +/- 0.13, 1.52 +/- 0.15 and 1.51 +/- 0.11 fmol., respectively. Cyclic 3',5'-adenosine monophosphate was not increased by ESWL in patients with renal stones. Urine nitrite increased slightly but not significantly after ESWL. Plasma cyclic nucleotides and nitrite, and urine nitrite were not changed by ESWL in patients with lower ureteral stones. Plasma renin activity decreased significantly in patients with renal stones 60 minutes after ESWL. Blood pressure and pulse rates increased mildly without statistical significance after ESWL for renal stones. CONCLUSIONS: ESWL increases plasma nitrite and cGMP in patients with renal stones, suggesting that shock waves stimulate the NO-cGMP signaling pathway.

The epidemiology of childhood alopecia areata in China: a study of 226 patients.

To study the clinical and epidemiologic profile of childhood alopecia areata, we performed a survey in which a total of 226 childhood patients less than 16 years old were enrolled. Statistical analysis and heritability were performed using EPI INFO 6.0, SPSS10.0, and the Falconer method. The median age of onset was 10 years. The majority of patients (84.96%) presented with limited alopecia. The male : female ratio was 1.4:1. Boys appeared to have more severe involvement. The earlier the age of onset, the greater the severity of the disease. Sixty-seven patients (29.65%) had previous episodes of alopecia areata. Greater severity and longer duration were seen in the relapsing patients than in the primary patients. Six patients (2.65%) had an associated disease. A positive family history was reported in 25 patients (11.06%). The prevalence figures for alopecia areata in first-, second-, and third-degree relatives of the probands were 2.87%, 0.40%, and 0.13%, respectively. The heritabilities of AA in first-, second-, and third-degree relatives were 51.20%, 46.25%, and 25.65%, respectively. It can be speculated that the effect of genetic factors is important in the occurrence of this disease.
