Mode of action-based local QSAR modeling for the prediction of acute toxicity in the fathead minnow.

The ultimate intention of quantitative structure-activity relationship (QSAR) study in toxicology is to predict the toxic potential of untested compounds with great accuracy. As QSAR has been based on the assumption that compounds from the same chemical domain will behave in similar manner, the QSAR model built upon the analogical chemicals is hypothesized to exhibit better performance than that derived from the miscellaneous data set. In this paper, the acute toxicity, 96 h LC(50) (median lethal concentration) for the fathead minnow from database EPAFHM_v2a_617_1Mar05 served as the interested toxicity endpoint, and the mode of action (MOA) in toxic response was employed as a criterion to compartmentalize the chemical domains. MOA-based local QSAR models were built by partial least squares (PLS) regression for each subset with single mode of action such as Narcosis I, Narcosis II or Reactive, and global model was also developed for the combined data set containing several subsets above. By comparing the performances of these two types of models, the local models were superior to the global model in that the relative standard error (R.S.E.) of the former was much lower for both the training set and the test set of any subset. In addition, the influence of the reliability of MOA determination on the performance of local model was also investigated and the statistical results for subsets with MOAs at A and B confidence level were better than those at C and D confidence level. Therefore, the MOA-based local QSAR models are promising to improve the accuracy of toxicity prediction as long as the assessment of MOA is of high reliability.

A comparative study on the individual and combined effects of baicalin and jasminoidin on focal cerebral ischemia-reperfusion injury.

To compare the individual effects of baicalin and jasminoidin with the combined effect of them on cerebral ischemia-reperfusion injury, and test whether the combined administration of baicalin and jasminoidin can improve the therapeutic effect. Male Sprague-Dawley rats underwent focal cerebral ischemia for 1.5 h and reperfusion for 24 h. Just before reperfusion, tested drugs (baicalin, jasminoidin, a drug combination consisting of baicalin and jasminoidin, or nimodipine) were intravenously treated. Diffusion weighted imaging (DWI) of magnetic resonance imaging (MRI), behavior examination, 2,3,5-triphenyltetrazolium chloride (TTC) staining, histological examination, and real-time PCR for BDNF and caspase-3 were performed. All of the drug treatments could significantly ameliorate the results of TTC and histological examination, and the baicalin/jasminoidin combination did so most prominently. This combination could also significantly ameliorate DWI of MRI and behavior examination results, and promote the expression of BDNF and inhibit the expression of caspase-3. On the whole, both baicalin and jasminoidin have a preventive effect against ischemic stroke, although their effects are not very strong. However, the combination of baicalin and jasminoidin can significantly improve their effectiveness. This may be related to its better regulation on the BDNF and caspase-3.

[Effects of active components extracted from Qixue Bingzhi Recipe on proliferation of vascular smooth muscle cells and expressions of platelet-derived growth factor and its receptor genes].

OBJECTIVE: To observe the effects of paeoniflorin and total flavones extracted from Qixue Bingzhi Recipe on proliferation of vascular smooth muscle cells (VSMCs) cultured in endothelial cell conditioned medium (EC-CM) induced by oxidized low-density lipoprotein (ox-LDL) and the expressions of platelet-derived growth factor and its receptor genes. METHODS: The VSMCs were cultured in normal culture medium, EC-CM, simvastatin-medicated EC-CM, paeoniflorin and total flavones (low, medium and high dose) -medicated EC-CM respectively. The growth activity of VSMCs was assessed by thiazolyl blue tetrazolium bromide (MTT) assay. The expressions of platelet-derived growth factor (PDGF)-BB and platelet-derived growth factor receptor-alpha (PDGFR-alpha) mRNAs were detected by reverse transcription polymerase chain reaction (RT-PCR). RESULTS: The growth activity of VSMCs cultured in the EC-CM induced by ox-LDL was significantly higher than that in the normal culture medium (P<0.01), and the expressions of PDGF-BB and PDGFR-alpha mRNAs were obviously increased as compared with those in the normal culture medium. The growth activity of VSMCs cultured in each paeoniflorin and total flavones-medicated EC-CM was significantly lower than that in the EC-CM (P<0.01), and the expressions of PDGF-BB and PDGFR-alpha mRNAs were obviously decreased as compared with those in the EC-CM. CONCLUSION: The paeoniflorin and total flavones extracted from Qixue Bingzhi Recipe may be beneficial to the prevention and treatment of arteriosclerosis, and this efficacy may be correlated with down-regulating the expressions of PDGF-BB and PDGFR-alpha mRNAs which are related to the proliferation of VSMCs cultured in EC-CM induced by ox-LDL.

[Origination and development of syndrome concept in traditional Chinese medicine].

In the opinion of dialectical materialism, concept is developing with the deepening and broadening of human being's mind. The developing process of syndrome concept in traditional Chinese medicine also takes the same way. In the Sui and Tang Dynasties, the connotation of syndrome concept was defined and the embryonic form of syndrome differentiation of zang-fu viscera was formed. In the Song, Jin and Yuan Dynasties, the development of syndrome concept embodied in optimizing the theoretical research based on clinical practice and experiences according to many medical masters in different denominations. The syndrome differentiation of eight principles was established to standardize and perfect the principles of syndrome differentiation in the Qing Dynasty. Modern research of syndrome concept is manifested in objectifying the process of research of syndrome and diseases in different levels and aspects by use of advanced scientific technology.

[Exploration on thinking approach and methodology of syndrome standardization].

The researching thinking approach of "descending the dimension with the lowering rank, ascending the rank with the raising of nsion of dimension", which could exclude the intervention of non-crucial fact ors and reveal the complexity of syndrome as well, provides a feasible approach for syndrome standardization. A study was conducted by analyzing some syndromes and their correlated factors selected from the "Ancient Good-sized Medical Records Database" through unconditional logistic multivariate stepwise regression by SAS 6.12 universal statistical software to sieve the variable, and the mathematical mode of regression equation was obtained eventually, expressed as logit (p) = alpha + beta1X1 + beta2X2 + ... + betamXm. With that, the research of syndrome standardization was proceeded on the syndromes with diagnostic significance to provide references for standardization of syndrome differentiation. I

[Research of kingsbrain on improving the learning and memory ability of MCAO rats].

OBJECTIVE: To explore the influence of Kingsbrain (GETO) on the learning memory impairment of rats with cerebral ischemia. METHOD: Rats with cerebral ischemia were administered GETO orally once a day for one month. The ability of spatial-learning memory of rats was evaluated by Morris Water Maze (MWM). Duxil was used as a positive control. RESULT: the results of place navigation of MWM showed that at the 3rd time of swimming training, the escape latency of rats of the GETO group, Duxil group and Sham group were shorter than that of model group. The escape latency were (54.1 +/- 43.94), (55.9 +/- 43.49), (50.4 +/- 34.99) and (85.4 +/- 42.8) s, respectively; but there was no significantly difference. After the 6th time of swimming training, the escape latency of rats of the GETO group (37.8 +/- 38.69) s, the Duxil group (37.4 +/- 38.03) s and the sham group (26.9 +/- 21.63) s were significantly shorter than that of model rats (77.5 +/- 47.59) s, P < 0.05, respectively. Comparison of the swimming distance among groups were similar to the escape latency among groups. In the test of spatial probe, results of the ratio of the swimming time of platform quadrant (tP) vs the total swimming time (tT) and the ratio of the swimming distance of platform quadrant (dP) vs the total swimming distance (dT) indicated that the ratios of the GETO group (0.347 +/- 0.0662, 0.344 +/-0.055 1), the Duxil group (0.345 +/- 0.0984, 0.34 +/- 0.0934) and the sham group (0.35 +/- 0.0662, 0.349 +/- 0.0589) were significantly higher than those of the Model group (0.261 +/- 0.0689, 0.274 +/- 0.0544), P < 0.05, respectively. CONCLUSION: GETO can significantly improve the spatial learning and memory ability of rats with cerebral ischemia, which provides the pharmacodynamics evidence for its clinical application of improveing the learning and memory ability in poststroke patients.

[Methodological thoughts on clinical efficacy evaluation with multiple clinical trials].

Some clinical trials showed that the efficacy of traditional Chinese medicine was not good as the efficacy of western medicine even thoush clinical doctors have been using traditional medicine effectively. Such a result originated from improper application of western medical thoughts in routine clinical efficacy evaluation. To find differences from effective cases with non-effective cases and put the differences into the indication of next clinical trial would conform to the differentiation theory in traditional Chinese medicine. The differences, whether positively or negatively related to the efficacy, could be obtained from all information including symptom evaluation, tongue appearance, pulse feeling and biological parameters with non-linear and other statistical methods. More specific indication finding would be the key task for clinical efficacy evaluation via multiple clinical trials.

Suppressive effects of a Chinese herbal medicine qing-luo-yin extract on the angiogenesis of collagen-induced arthritis in rats.

Qing-Luo-Yin (QLY), a traditional Chinese herbal medicine for the treatment of rheumatoid arthritis, is a combination of the extracts of Sophora flavescens Ait., Phellodendron amurense Rupr., Sinomenium acutum Rehd. et Wils. and Dioscorea hypoglauca Palib. The suppressive effect of QLY on the development of angiogenesis was investigated in a rat model of collagen-induced arthritis (CIA). QLY (0.3 g/kg) was orally administered daily for 27 days. Neo-angiogenesis, pannus and cartilage damage, the expression of metalloproteinases (MMP)-3 messenger RNA (mRNA) and the level of the tissue inhibitor of matrix metalloproteinase (TIMP)-1 in the synovium were examined by histology, in situ hybridization and immunohistochemiscal assays, respectively. It was observed that the articular morphological alterations, the over-expression of MMP-3 mRNA and the reduced production of TIMP-1 in CIA rats were significantly ameliorated by QLY. QLY performed about as effectively as tripterygium glycosidorum tablets (0.1 g/kg) extracted from Tripterygium wilfordii Hook. f.. These results indicate that QLY exerts a suppressive effect on the angiogenesis of CIA rats, and suggest that the therapeutic effect of QLY could be due to restoring the balance of MMP-3 and TIMP-1 in rheumatoid synovium.
