[Cometabolic biodegradation of nitrogenous heterocyclic compounds indole and pyridine under anoxic conditions].

Based on the traditional theory of denitrification under anoxic conditions, and with the addition of nitrate, experiments were conducted to investigate the cometabolic biodegradation of nitrogenous heterocyclic compounds-indole and pyridine. Results show that the optimum ratio of COD to nitrate (C/N) is 8.4 to approximately 8.9. Under the conditions with temperature of 28 degrees C and pH of 7.0 to approximately 7.5, the nitrate reductase activity (NRA) is in the best state. Addition of pyridine can promote NRA and the degradation of indole. When the initial concentration of indole is 150 mg/L, the concentration ratio of indole to pyridine is 1 to approximately 10, and under the optimum C/N conditions, the degradation of indole meet with zero-order kinetics. There was no accumulation of nitrite during the reaction. When the concentration ratio of pyridine to indole is less than 0.25, with the increase of pyridine concentration, there is a faster augment rate for NRA and the degradation of indole than the situation when the concentration ratio of pyridine to indole is more than 0.25.

[Preparation and functional identification of testicular Sertoli cells].

OBJECTIVE: To simplify the method for separation and cultivation of rat testicular Sertoli cells with high viability, quantity and expression efficiency. METHODS: Testicular Sertoli cells from 2 to 3-week-old male Wistar rats were prepared by digestion with collagenase, trypsin and DNase and cultured together with active lymphocytes to observe their killing effect against lymphocytes. After cell culture for 72 h, the Sertoli cells were morphologically observed by different means and identified with transmission electron microscope. Fas ligand and follicle-stimulating hormone receptor (FSHR) were examined immunohistochemically to identify testicular Sertoli cells. SABC method was used for labeling the Fas ligand on the testicular Sertoli cells. RESULTS: The viability of the isolated and cultured Sertoli cells was more than 90%, and in in vitro culture, Sertoli cells, which expressed the Fas ligand, could kill the active lymphocytes. CONCLUSION: This method improves the efficiency in acquisition of rat testicular Sertoli cells expressing Fas ligand, which are believed to be a potential donor for co-transplantation with parathyroid cells to offer immune privilege.

[Experimental research on separate structure jet aerator].

A new type of jet aerator (separate structure jet aerator) was designed and developed, and an idea of separate design was put forth. This type of jet aerator was mainly applied to the treatment of small-scale wastewater and industrial wastewater of high density treatment projects. The experimental set-up was built to investigate the influences of the structural parameters (R1, R2 and R3) on the gas suction flowrate. The experimental results are listed as follows: the gas suction flowrate fluctuates with the changes of R2, and the optimal R2 is relevant to the length of the mixing tube; R3 does not contribute a lot to the suction performance, but choosing a suitable R3 can help to make further use of the gas suction potential of the jet aerator while keeping the other parameters unchanged; R1 has a great impact on the suction performance, and the optimal R1 can be found out in the transitional region. The experiments also prove that it is feasible to apply CFD to designing jet aerator, since its gas suction flowrate is high (reaching 900 approximately 1100 L/min) and it shows prominent performance in agitating liquid.

[Partial dorsal root rhizotomy increases the anterograde transportation of neunotrophic factors in primary sensory neuron].

OBJECTIVE: To investigate whether partial dorsal root rhizotomy promotes the anterograde Five adult cats were transportation of BDNF, NT-3 and GDNF in the primary sensory neuron. METHODS: Subjected to unilateral spared root rhizotomy (the DRGs of L1-L5 and L7-S2 were removed, but L6 DRG was spared) and bilateral dorsal roots of L6 were ligated at the same time. Three days after operation, dorsal roots were taken out and made into frozen sections 20 microm in thickness. The sections were stained using specific BDNF, NT-3, GDNF antibody (1:1500) by ABC method. The immunoreactive density was observed in a site near DRG and a site near spinal cord. RESULTS: In the control group (with spared L6 DRG), there were no marked differences in NT-3 and GDNF immunoreactivity between the site near DRG and the site near spinal cord, while BDNF immunoreactivity was more intense in the site near DRG than that in the site near spinal cord. In the operation group, the immunoreactivity of each neurotrophin in the site near DRG was stronger than that in the site near spinal cord, and the immunoreactivities of BDNF, NT-3, GDNF in the site near DRG of the operation were stronger than those of the control group respectively. CONCLUSION: The increasing of immunoreactivities of neurotrophins near DRG following partial dorsal root rhizotomy suggests that partial dorsal root rhizotomy can promote their anterograde transportation from spared DRG to the spinal cord.
