Investigating the Role of MicroRNA396 (miR396) Gene in Regulating Wheat Yield and Grain Nitrogen Concentration.

Nitrogen (N) is essential for crop growth, yet excessive fertilization causes environmental issues, highlighting the need to sustain yield and grain N concentration under reduced N input. miR396s are known to regulate plant development and stress responses. Here, we examined whether and how miR396 affects wheat yield and N status under high and low N conditions. TaMIM396 (transforming with the target mimicry construct of miR396) overexpression significantly increased plant height, spike length, grain yield, and grain N concentration under both N treatments. Physiological data showed TaMIM396 enhanced dry matter (DM) and N accumulation at anthesis and maturity, as well as improved post-anthesis remobilization of DM and N to grains. RNA-seq analysis revealed that, under low N, TaMIM396 specifically upregulated key photosynthetic antenna genes, including Lhca3 and Lhcb1/2/3/5, which are critical for light harvesting, suggesting improved photosynthetic efficiency that promotes DM accumulation under N limitation. Collectively, our results demonstrate that TaMIM396 acts as a broad-spectrum N-efficiency gene, coordinating carbon and N remobilization while boosting photosynthetic capacity, thereby supporting stable yield and grain N concentration across N supply levels. Therefore, TaMIM396 is a promising candidate for breeding N-efficient wheat cultivars compatible with sustainable high-yield agriculture.

Complexation of fatty acids and fatty acid-CoAs with synthetic O-methylated polysaccharides.

An experimentally simple, but highly reproducible and reliable method has been developed to follow the complexation event of fatty acid (FA) and FA-CoA with polysaccharides. This method was based on the recent discovery of the unique blue-shifted UV absorption associated with the aggregation of tetraenoic fatty acids (TE-FAs) in aqueous solution. Complexation was monitored by recording the intensity of UV absorption at 250 nm ([free TE-FA]) and 303 nm ([complexed TE-FA]), and the K aggregate of C(20) t,t,t,t-TE-FA exhibited the ideal property for this purpose. Synthetic 3-O-methyl-D-mannose- and 6-O-methyl-D-glucose-containing lipopolysaccharides were found to exhibit a broad range of the binding affinities with C(20) t,t,t,t-TE-FAs as well as saturated FAs/FA-CoAs.

Approaches to the study of N-linked glycoproteins in human plasma using lectin affinity chromatography and nano-HPLC coupled to electrospray linear ion trap--Fourier transform mass spectrometry.

In this publication, we will describe the combination of lectin affinity chromatography with nano high performance liquid chromatography (HPLC) coupled to a linear ion trap Fourier transform mass spectrometer (capillary LC-LTQ/FTMS) to characterize N-linked glycosylation structures in human plasma proteins. We used a well-characterized glycoprotein, tissue plasminogen activator (rt-PA), which is present at low levels in blood, as a standard to determine the dynamic range of this approach. N-linked glycopeptides derived from rt-PA could be characterized at a ratio of 1:200 in human plasma (rtPA: total plasma protein, w/w) by accurate mass measurement in the FTMS and fragmentation (MS(n)) in the linear ion trap. We demonstrated that this platform has the potential to characterize the general N-linked glycosylation structures of abundant glycoproteins present in human plasma without the requirement for antibody-based purification, or additional carbohydrate analytical protocols. This conclusion was supported by the determination of carbohydrate structures for three glycoproteins, IgG, haptoglobin, and alpha-1-acid glycoprotein, at their natural levels in a human plasma sample, but only after the lectin enrichment step.

Motion compensation via redundant-wavelet multihypothesis.

Multihypothesis motion compensation has been widely used in video coding with previous attention focused on techniques employing predictions that are diverse spatially or temporally. In this paper, the multihypothesis concept is extended into the transform domain by using a redundant wavelet transform to produce multiple predictions that are diverse in transform phase. The corresponding multiple-phase inverse transform implicitly combines the phase-diverse predictions into a single spatial-domain prediction for motion compensation. The performance advantage of this redundant-wavelet-multihypothesis approach is investigated analytically, invoking the fact that the multiple-phase inverse involves a projection that significantly reduces the power of a dense-motion residual modeled as additive noise. The analysis shows that redundant-wavelet multihypothesis is capable of up to a 7-dB reduction in prediction-residual variance over an equivalent single-phase, single-hypothesis approach. Experimental results substantiate the performance advantage for a block-based implementation.

Activation/driving force relationships for cyclopropylcarbinyl --> homoallyl-type rearrangements of radical anions.

By using direct and indirect electrochemical methods, rate constants (ko) for cyclopropane ring opening of radical anions derived from the one-electron reduction of trans-1-benzoyl-2-phenylcyclopropane, trans-1-benzoyl-2-vinylcyclopropane, 2-methylenecyclopropyl phenyl ketone, spiro[anthracene-9,1'-cyclopropan-10-one], 3-cyclopropylcyclohex-2-en-1-one, and 3-(1-methylcyclopropyl)cyclohex-2-en-1-one were determined. Qualitatively, rate constants for ring opening of these (and other cyclopropyl- and cyclobutyl-containing radical anions) can be rationalized on the basis of the thermodynamic stability of the radical anion, the ability of substituents on the cyclopropyl group to stabilize the radical portion of the distonic radical anion, and the stability of the enolate portion of the distonic radical anion. On the basis of this notion, a thermochemical cycle for estimating deltaG(o) for ring opening was presented. For simple cyclopropyl-containing ketyl anions, a reasonable correlation between log(ko) and deltaG(o) was found, and stepwise dissociative electron transfer theory was applied to rationalize the results. Activation energies calculated with density functional theory (UB3LYP/6-31+G*) correlate reasonably well with measured log(ko). The derived log(ko) and deltaG(o) and log(ko) vs E(a) plots provide the basis for a "calibration curve" to predict rate constants for ring opening of radical anions derived from carbonyl compounds, in general.

[Study on the effect of leptin on fibroblast proliferation and collagen synthesis in vitro in rats].

OBJECTIVE: To investigate the effect of leptin on fibroblast proliferation and collagen synthesis as to elucidate that fibroblasts play a role in leptin's effect on wound healing. METHODS: Purified dermal fibroblasts were derived from sucking wistar rat skin and exposed to leptin at concentration of 0, 10, 50, 100, 200, and 400 ng/ml. The survived fibroblasts were assessed by the colorimetric thiazolyl blue (MTT) assay. Replication of fibroblast was quantified by the incorporation of 3H-thymidine. Collagen synthesis of fibroblast cell was measured by the incorporation of 3H-proline into collagenase-sensitive protein. RESULTS: The absorption of fibroblast exposed to leptin at concentration of 200 and 400 ng/ml 0.082 +/- 0.013, 0.091 +/- 0.018 was higher than that of control group 0.063 +/- 0.010, P < 0.05. The incorporations of 3H-thymidine of fibroblast exposed to leptin at concentration of 200 and 400 ng/ml 379 +/- 101 cpm, 326 +/- 33 cpm were significantly higher than those of control group 219 +/- 56 cpm, P < 0.05. The incorporations of 3H-proline of fibroblast exposed to leptin at concentration of 200 and 400 ng/ml 911 +/- 55 cpm, 1 072 +/- 259 cpm were significantly higher than that of control group 679 +/- 176 cpm, P < 0.05. CONCLUSION: Leptin can promote rat cutaneous fibroblast proliferation and collagen synthesis in vitro. This suggests that cutaneous fibroblast plays a role in leptin's promoting skin wound healing and it may be one of the main mechanisms by which leptin enhances skin wound healing.

Capillary isoelectric focusing and affinity capillary electrophoresis approaches for the determination of binding constants for antibodies to the prion protein.

For the development of specific immunological assays, the binding of a specific antibody (Ab) to the target antigen (Ag) has to be relatively strong. In this study, we have utilized affinity capillary electrophoresis (ACE), a form of capillary zone electrophoresis (CZE) to determine the binding constant (Kb) of specific Abs against bovine serum albumin (BSA) and the healthy prion protein (PrPc), in buffer solutions at fixed pHs, approximating in vivo conditions. We have also utilized capillary isoelectric focusing (cIEF) to determine the complexity and recognition of the various isoforms of PrPc Abs towards their Ag, PrPc. Only ACE and CZE have been used to derive Kb values. The selected Abs for the prion protein can recognize both healthy and diseased states of the protein and are commercially available. The Kb values of PrPc Abs appear to be as strong as the anti-BSA (Ab to BSA) and other reported Kb values for proteins of similar size to PrPc. This appears to be one of the few reports on Kb values for any PrPc Abs, and their applications for in vitro immunoassays (e.g., enzyme-linked immunosorbent assays (ELISAs)). Such assays are being used to detect the infectious agent, PrPres, in brain and related matter/tissues.

Free flow electrophoresis coupled with liquid chromatography-mass spectrometry for a proteomic study of the human cell line (K562/CR3).

The requirement for prefractionation in proteomic analysis is linked to the challenge of performing such an analysis on complex biological samples and identifying low level components in the presence of numerous abundant housekeeping and structural proteins. The employment of a preliminary fractionation step results in a reduction of complexity in an individual fraction and permits more complete liquid chromatography/mass spectrometry (LC/MS) analysis. Free flow electrophoresis (FFE), a solution-based preparative isoelectric focusing technique, fractionates and enriches protein fractions according to their charge differences and is orthogonal in selectivity to the popular reversed phase high performance liquid chromatography (HPLC) fractionation step. In this paper, we explored the advantages of a combination of FFE and liquid chromatography/mass spectrometry to extend the dynamic range of a proteomic analysis of a complex cell lysate. In this study, the whole cell lysate of a chronic myelogeneous leukemia cell line, K562/CR3, was prefractionated by FFE into 96 fractions spanning pH 3-12. Of these, 35 fractions were digested with trypsin and then analyzed by LC/MS. Depending on the algorithm used for peptide assignment from MS/MS data, at least 319 proteins were identified through database searches. The results also suggested that pI could serve as an additional criterion besides peptide fragmentation pattern for protein identification, although in some cases, a pI shift might indicate post-translational modification. In summary, this study demonstrated that free flow electrophoresis provided a useful prefractionation step for proteomic analysis and when combined with LC/MS allowed the identification of significant number of low level proteins in complex samples.

A simple route towards CuO nanowires and nanorods.

CuO nanowires and nanorods were synthesized through a novel controllable solution-phase hydrothermal method using a nonionic surfactant polyethylene glycol (PEG) as the structure-directing template. The lengths of obtained 1D CuO nanostructures could be successfully controlled through choosing different molecular weights of PEG. The phase structures and morphologies were investigated by XRD, TEM, HRTEM and SAED. The formation mechanisms of the nanorods and nanowires were investigated and discussed on the basis of the experimental results.

A new type of single-helix coordination polymer with mixed ligands [M2(phen)2(e,a-cis-1,4-chdc)2(H2O)2]n (M=Co and Ni; phen=1,10-phenanthroline; chdc=cyclohexanedicarboxylate).

A new family of single-stranded helices coordination polymers with mixed ligands, [M2(phen)2(e,a-cis-1,4-chdc)2(H2O)2]n (1, M=Co; 2, M=Ni; chdc=cyclohexanedicarboxylic acid; phen=1,10-phenanthroline), were prepared under hydrothermal conditions and characterized by elemental analyses, IR spectra, TG analysis, and single-crystal X-ray diffraction analysis. X-ray crystal structural analyses reveal that 1 and 2 are isomorphic and belong to the monoclinic system. C40H36Co2N4O10, P2(1)/c, a=10.0566(5) A, b=8.8843(5) A, c=20.2912(14) A, beta=100.052(3) degrees, Z=2 for 1; and C40H36Ni2N4O10, P2(1)/c, a=9.8921(6) A, b=9.0151(4) A, c=20.1628(17) A, beta=100.31(2) degrees, Z=2 for 2. In the structures of 1 and 2, the 1,4-chdc ligand possesses only one kind of e,a-cis-conformation although there are both cis- and trans-conformations in the raw material. Two oxygen atoms of one carboxyl in 1,4-chdc ligand and another oxygen atom of contraposition carboxyl link adjacent Co or Ni atoms into an infinite 1-D zigzag chain. The most attractive structural feature of 1 and 2 is that they both exhibit an infinite chiral chainlike structure with 2(1) helices along the b axis. In addition, the right-handed and the left-handed chains are alternate. Meanwhile, the adjacent chains of 1 and 2 are linked via hydrogen bonds into 2-D network structures, which further form 3-D frameworks via pi-pi interactions of 1,10-phen.

Derivatization of phospholipids.

Phospholipids are major components of biological membranes. Without chemical derivatization, it is difficult to identify and quantitate phospholipids in biological samples. Chemical derivatization can improve both the selectivity and sensitivity of the analytes. This paper gives a full review, through March, 2002, of derivatization methods used for phospholipids in HPLC, CE and GC as well as the spray reagent used for TLC in the early days.

Self-assembled multilayer films based on a Keggin-type polyoxometalate and polyaniline.

Multilayer ultrathin films were fabricated from partially doped polyaniline (PAN) and a Keggin-type polyoxometalate [alpha-SiW(12)O(40)](4-) (alpha-SiW(12)) in aqueous solution via the layer-by-layer self-assembly technique and characterized by UV-vis, FTIR, and X-ray photoelectron spectra (XPS), ellipsometry, scanning electron microscopy (SEM), and atomic force microscopy (AFM). UV-vis spectroscopy shows that the absorbance values at characteristic wavelengths of the multilayer films increase almost linearly with the number of PAN/alpha-SiW(12) bilayers, suggesting that the deposition process is regular and highly reproducible from layer to layer. FTIR and XPS spectra confirm the incorporation of alpha-SiW(12) and PAN into the films. Ellipsometric measurements show that the PAN/alpha-SiW(12) bilayer thickness increases with the increasing PAN solution concentration. SEM and AFM images indicate that the film surface is relatively uniform and smooth. In addition, the electrical conductivities of the multilayer films doped with hydrochloric acid were also measured.

Novel hydrogen-bonded three-dimensional networks encapsulating one-dimensional covalent chains: [M(4,4'-bipy)(H2O)(4)](4-abs)(2).nH2O (4,4'-bipy = 4,4'-bipyridine; 4-abs = 4-aminobenzenesulfonate) (M = Co, n = 1; M = Mn, n = 2).

Two novel compounds, [Co(4,4'-bipy)(H(2)O)(4)](4-abs)(2).H(2)O (1) and [Mn(4,4'-bipy)(H(2)O)(4)](4-abs)(2).2H(2)O (2) (4,4'-bipy = 4,4'-bipyridine; 4-abs = 4-aminobenzenesulfonate), have been synthesized in aqueous solution and characterized by single-crystal X-ray diffraction, elemental analyses, UV-vis and IR spectra, and TG analysis. X-ray structural analysis revealed that 1 and 2 both possess unusual hydrogen-bonded three-dimensional (3-D) networks encapsulating one-dimensional (1-D) covalently bonded infinite [M(4,4'-bipy)(H(2)O)(4)](2+) (M = Co, Mn) chains. The 4-abs anions in 1 form 1-D zigzag chains through hydrogen bonds. These chains are further extended through crystallization water molecules into 3-D hydrogen-bonded networks with 1-D channels, in which the [Co(4,4'-bipy)(H(2)O)(4)](2+) linear covalently bonded chains are located. Crystal data for 1: C(22)H(30)CoN(4)O(11)S(2), monoclinic P2(1), a = 11.380(2) A, b = 8.0274(16) A, c = 15.670(3) A, alpha = gamma = 90 degrees, beta = 92.82(3) degrees, Z = 2. Compound 2 contains interesting two-dimensional (2-D) honeycomb-like networks formed by 4-abs anions and lattice water molecules via hydrogen bonding, which are extended through other crystallization water molecules into three dimensions with 1-D hexagonal channels. The [Mn(4,4'-bipy)(H(2)O)(4)](2+) linear covalent chains exist in these channels. Layer-by-layer self-assembled ultrathin multilayer films of lanthanide polyoxometalates and poly(allylamine hydrochloride) and their photoluminescent properties.

Ultrathin multilayer films of two lanthanide polyoxometalates (LPOMs), K(17)[Eu(P(2)W(17)O(61))(2)] (EPW) and K(13)[Eu(SiW(11)O(39))(2)] (ESW), and poly(allylamine hydrochloride) (PAH) have been prepared by layer-by-layer self-assembly from dilute aqueous solutions. UV-vis spectroscopy and ellipsometry respectively show that the absorbance values at characteristic wavelengths and the thicknesses of the multilayer films increase linearly with the number of LPOM/PAH bilayers, suggesting that the deposition process is linear and highly reproducible from layer to layer. Average thicknesses of ca. 3.4 and 2.4 nm were determined for the EPW/PAH and ESW/PAH bilayers by ellipsometry, respectively. In addition, scanning electron microscopy (SEM) and atomic force microscopy (AFM) images provide the surface morphology of the LPOM/PAH films, indicating that the film surface is relatively uniform and smooth. The photoluminescent properties of these films have also been investigated by fluorescence spectroscopy. The LPOM/PAH multilayer film has a good thermal stability as shown by UV-vis, X-ray photoelectron, and fluorescence spectra.
