Mutant presenilin 2 causes abnormality in the brain lipid profile in the development of Alzheimer's disease.

Mutation in the presenilin 2 (PS2mt) is known to be one of factors involved in the development of Alzheimer's disease (AD). It was recently revealed that an abnormality of lipid metabolism is a phenomenon occurring in AD. Therefore, the aim of this study was to investigate the potential relationship between the mutation of PS2 and alterations of the lipid profile within the brain. The results showed there increases in the levels of cholesterol, low density lipoprotein and triglyceride, but a decrease in the level of high density lipoprotein in brain tissues expressing mutant PS2. These findings indicated that PS2mt is involved in the abnormalities of the lipid profile, which could cause or result in the development of AD.

Water surface contacting cover system--the basic study for improving the oxygen transfer coefficient and the BOD removal capacity.

In order to construct an environmental aeration basin in a sewage treatment plant, the suggested novel aeration basin was examined for the oxygen transfer coefficient by using tap water on a lab-scale and pilot-scale. We called it water surface contacting cover system (WSCCS) that was installed with a cover plate on the wafer surface and a slant plate in aeration basin, compared to a standard aeration basin (SRAS, spiral roll aeration system). Finally, the novel basin could be compared by adopting the slant and cover plates into the actual sewage treatment plant (W x L x D (m)=5.0 x 2.65 x 4.25, treatment capacity: 600 m3/d). As a result, only by adding a slant plate, the oxygen transfer coefficient (K(L)a) increased approximately by 15% more than that with a SRAS basin. And the slope of slant plate was optimized at 10 degrees . Additionally, in the WSCCS, the increasing efficiency of the oxygen transfer coefficient, affected only by the cover, was proved at about 25% on the lab-scale aeration basin and about 20% on the pilot plant. After all, the increasing effect of the oxygen transfer coefficient on WSCCS that was installed with the water contacting cover and the slant plate on standard aeration basin was proved to be above 35%. The distance between the air diffuser and the slant plate was 20-30 cm in the pilot-scale plant. As an effective factor for increasing the oxygen transfer rate, the width/depth ratio of aeration basin was proved to be 2. Through the short-term results of actual sewage treatment plant, it was known that BOD removal efficiency could be improved by about 3% by adopting tile WSCCS not but SRAS.

Mutant presenilin 2 increases acetylcholinesterase activity in neuronal cells.

A presenilin 2 mutation is believed to be involved in the development of Alzheimer's disease. In addition, transgenic mice with a presenilin 2 mutation have been reported to have learning and memory impairments. In this study, exposing PC12 cells expressing mutant presenilin 2 to 50 microM AP25-35, 30 mM L-glutamate and 50 microM H2O2 caused a significant increase in acetylcholine esterase activity. An in vivo study revealed high levels of this enzyme activity in the mutant presenilin 2 transgenic brains compared with the wild type presenilin 2 transgenic and nontransgenic samples. These results suggest that a mutant presenilin 2-induced neurodegeneration in Alzheimer's disease might be involved in the increase in acetylcholinesterase activity. These findings might help in the development of an appropriate therapeutic intervention targeting mutant presenilin 2-induced Alzheimer's disease.

Scapulothoracic arthrodesis: indications, technique, and results.

UNLABELLED: Twenty-two patients (24 shoulders) had a scapulothoracic arthrodesis for various clinical disorders including facioscapulohumeral muscular dystrophy, scapular winging from serratus anterior palsy, painful scapular crepitation, and cleidocranial dysostosis. All patients were extremely disabled with pain and loss of function because of their symptomatic scapular winging, and many of the patients had multiple previous procedures on their shoulders before the scapulothoracic arthrodesis. The surgical indication was stabilization of painful scapulothoracic articulation to provide pain relief and allow functional use of the involved arm for activities of daily living. The surgical technique involved use of a semitubular plate and wire construct along the medial border of the scapula with the use of autograft (iliac crest) or allograft bone or both between the scapula and the rib cage. Patients were immobilized postoperatively for 12 weeks. Complications occurred in more than (1/2) of the patients and included pulmonary complications, hardware failure, pseudarthrosis, and persistent pain. Postoperatively, 20 of 22 (91%) patients thought that the pain in their shoulder complex was improved and were satisfied with their functional outcome. Scapulothoracic arthrodesis can improve function and reduce pain in the shoulder complex in patients with debilitating complex scapulothoracic dysfunction. Regulation of intracellular pH in rat renal inner medullary thin limbs of Henle's loop.

Regulation of intracellular pH (pHi) was studied in isolated rat renal inner medullary thin limbs of Henle's loop in bicarbonate/phosphate-buffered medium with high pCO2, high osmolality ( congruent with670 mosmol/kg H2O; 270 mM urea; 180 mM NaCl), organic osmolytes, and a pH of 6.8 to approximate the physiological in vivo environment. The pH-sensitive fluorescent dye 2',7'-bis(2-carboxyethyl)-5,6-carboxyfluorescein (BCECF) was used to measure pHi. Resting pHi was always acid and significantly more acid in descending thin limb (DTL) cells than in ascending thin limb (ATL) cells from pure or mixed-type thin limbs. Resting pHi was slightly but significantly higher in both DTLs and ATLs in high osmolality ( approximately 670 mosmol/kg H2O) than in low osmolality ( approximately 290 mosmol/kg H2O) medium but not when sucrose replaced urea. In both DTLs and ATLs the rate of recovery of pHi following additional acidification with an NH4Cl pulse was reduced by Na+ removal from the medium and by the addition of 60 microM HOE642 (an inhibitor of the Na+/H+ exchanger, NHE1), 55 microM S1611 (inhibitor of Na+/H+ exchanger, NHE3), 1 microM bafilomycin A1 (an inhibitor of vacuolar H+ -ATPase), or 20 microM Schering 28080 (an inhibitor of H+ -K+ -ATPase) to the medium. Resting pHi was also reduced by 60 microM HOE642, 55 microM S1611, and 20 microM Schering 28080. In both DTLs and ATLs, RT-PCR revealed message for NHE1, NHE3, and vacuolar H+ -ATPase; immunocytochemistry demonstrated the expression of the protein for NHE1 (basolateral membrane), NHE3 (luminal membrane), and H+ -K+ -ATPase (luminal membrane). These data suggest that pHi in rat inner medullary thin limbs is regulated by urea and by basolateral and luminal H+ extrusion via NHE1, NHE3, vacuolar H+ -ATPase, and H+ -K+ -ATPase.

[Taxonomical study on the Fascioloidae in Korea]

In order to study species of the Fascioloidae in Korea, a series of morphological and taxonomical studies were carried out and the following results were obtained. 1)Among the species of the Fascioloidae in Pusan area, intermediate form (F. indica?) were 70 %, F. gigantica 25 % and F. hepatica, 5 %. 2)None experimental, natural infective rate of the Fasciolosis was 3.3~3.4 % among the cattles (Bos taurus var. domesticus) in Pusan area.
