A highly enantio- and diastereoselective 1,3-dimethylallylation of aldehydes.

A highly enantio- and diastereoselective pentenylation of aldehydes is described. The homoallylic alcohol derived from 1,3-dimethylallylation of (-)-menthone undergoes an efficient allyl-transfer reaction with a wide range of aliphatic aldehydes in the presence of an acid catalyst to give rise to the corresponding 4-methyl-2(E)-penten-4-yl-5-ol products in good yields with high enantio- and 4,5-syn-selectivities.

Tandem cyclization of alkynes via rhodium alkynyl and alkenylidene catalysis.

A rhodium(I)-catalyzed tandem cyclization of alkynes has been developed. The reaction allows for multiple bond formations to occur at both the alpha- and beta-positions of alkynes under mild conditions to yield a variety of fused ring systems as the products. In the presence of triethylamine and the complex derived from [Rh(COD)Cl]2 and P(4-F-C6H4)3, a terminal alkyne is converted to a rhodium alkynyl species which reacts with a tethered alkyl halide at the beta-position to provide a beta,beta-disubstituted alkenylidene complex. The rhodium alkenylidene species then undergoes additional ring closures with a range of pendent functional groups such as alkene, hydroxyl, and phenyl groups through [2 + 2] cycloaddition, nucleophilic addition, and 6pi-electrocyclization processes, respectively.

Total synthesis of kendomycin: a macro-C-glycosidation approach.

Kendomycin, also known as (-)-TAN 2162, is a novel polyketide-derived ansamycin isolated from Streptomyces sp., which exhibits potent antagonist and agonist activities at the endothelin and calcitonin receptors, respectively. This bacterial metabolite also possesses a strong antibiotic activity against a range of gram-positive and -negative bacteria and cytostatic effects on the growth of human cancer cell lines. When a novel macroglycosidation reaction is employed as the key step, the first enantioselective total synthesis of kendomycin has been accomplished. A Friedel-Crafts-type ring closure of the acyclic precursor containing tetrahydropyran and benzofuran moieties produces the macrocycle as a single stereoisomer in good yield, thus establishing the aryl C-glycosidic linkage of the ansa core. This reaction requires a phenolic glycosyl acceptor and appears to proceed through a rapid O-glycosidation followed by a slow rearrangement to an aryl C-glycoside. The requisite secomacrocycle is prepared by the Pd(0)-catalyzed B-alkyl Suzuki-Miyaura cross-coupling of two subunits, which in turn can be expeditiously assembled from readily available building blocks in a modular fashion.

Combinatorial chemistry approach to chiral catalyst engineering and screening: rational design and serendipity.

An efficient asymmetric catalyst relies on the successful combination of a large number of interrelated variables, including rational design, intuition, persistence, and good fortune-not all of which are necessarily well-understood; this renders such practice largely empirical. As a result, the possibility of using combinatorial chemistry methods in asymmetric catalysis research has been widely recognized to be highly desirable. In this account, we attempt to show the principle and application of combinatorial approach in the discovery of chiral catalysts for enantioselective reactions. The concept focuses on the strategy for the creation of a modular chiral catalyst library by two-component ligand modification of metal ions on the basis of molecular recognition and assembly. The self-assembled chiral catalyst with two different ligands indeed exhibited synergistic effects in terms of both enantioselectivity and activity in comparison with its corresponding homocombinations in many reactions. The examples described in this paper demonstrated the powerfulness of combinatorial approach for the discovery of novel chiral catalyst systems, particularly for the development of highly efficient, enantioselective, and practical catalysts for enantioselective reactions. We hope this concept will stimulate further work on the discovery of more highly efficient and enantioselective catalysts, as well as unexpected classes of catalysts or catalytic enantioselective reactions in the future with the help of a combinatorial chemistry approach.

Highly enantioselective Friedel-Crafts reaction of aromatic amines with ethyl glyoxylate catalyzed by chiral titanium(IV) complexes: practical synthesis of aminomandelic acid derivatives.

The asymmetric Friedel-Crafts reaction of a variety of N,N-dialkylanilines with ethyl glyoxylate has been achieved by the catalysis of titanium complexes of BINOL derivatives to give corresponding ethyl esters of p-N,N-dialkylaminomandelic acids in high yields (85-99%) and good to excellent enantioselectivities (80-96.6% ee) under the optimized reaction conditions, which has added a new catalyst for this class of reactions. The reaction could also be carried out at a reduced catalyst loading (1 mol %) in gram scale, which provided a practical synthesis of enantiopure aminomandelic acid derivatives.

Situated learning theory: adding rate and complexity effects via Kauffman's NK model.

For many firms, producing information, knowledge, and enhancing learning capability have become the primary basis of competitive advantage. A review of organizational learning theory identifies two approaches: (1) those that treat symbolic information processing as fundamental to learning, and (2) those that view the situated nature of cognition as fundamental. After noting that the former is inadequate because it focuses primarily on behavioral and cognitive aspects of individual learning, this paper argues the importance of studying learning as interactions among people in the context of their environment. It contributes to organizational learning in three ways. First, it argues that situated learning theory is to be preferred over traditional behavioral and cognitive learning theories, because it treats organizations as complex adaptive systems rather than mere information processors. Second, it adds rate and nonlinear learning effects. Third, following model-centered epistemology, it uses an agent-based computational model, in particular a "humanized" version of Kauffman's NK model, to study the situated nature of learning. Using simulation results, we test eight hypotheses extending situated learning theory in new directions. The paper ends with a discussion of possible extensions of the current study to better address key issues in situated learning.

Assembled dendritic titanium catalysts for enantioselective hetero-Diels-Alder reaction of aldehydes with Danishefsky's diene.

A new type of dendritic 2-amino-2'-hydroxy-1,1'-binaphthyl (NOBIN)-derived Schiff-base ligands have been synthesized and applied to the titanium-catalyzed hetero-Diels-Alder reaction of Danishefsky's diene with aldehydes. These reactions afforded the corresponding 2-substituted 2,3-dihydro-4H-pyran-4-ones in quantitative yields and with excellent enantioselectivities (up to 97.2 % ee). The disposition of the dendritic wedges and the dendron size in the ligands were found to have significant impact on the enantioselectivity of the reaction. The recovered dendritic catalyst could be reused without further addition of the Ti source or a carboxylic acid additive for at least three cycles, retaining similar activity and enantioselectivity. The high stability of this type of assembled dendritic titanium catalyst may be attributed to the stabilization effect of large-sized dendron units in the catalyst molecule. The other important phenomenon observed with this catalyst system is that a higher degree of asymmetric amplification has been achieved by attachment of the dendron unit to the chiral ligand, which represents a new advantage of dendrimer catalysts for asymmetric reactions using chiral ligands of lower optical purity.

A novel dual-impedance-analysis EQCM system--investigation of bovine serum albumin adsorption on gold and platinum electrode surfaces.

Both quartz crystal micro-balance (QCM) impedance and electrochemical impedance spectroscopy (EIS) methods are widely used in interface studies. This paper presents details about a new strategy for simultaneous, mutual-interference-free and accurate measurements of QCM impedance and EI, through connecting a suitable capacitance in series with the piezoelectric quartz crystal (PQC) between QCM impedance and EIS measurement instruments. Combined and individual measurements of QCM impedance and EIS during silver deposition gave results comparable with each other, demonstrating the reliability of the proposed method. Bovine serum albumin (BSA) adsorption on gold and platinum electrodes in Britton-Robinson (B-R) buffers was investigated, and the Fe(CN)6(3-)/Fe(CN)6(4-) couple was used as an electrochemical probe to characterize BSA adsorption. While the reversibility of Fe(CN)6(3-)/Fe(CN)6(4-) couple on bare Au and Pt electrodes changed very slightly with decreasing solution pH from pH approximately 7 to pH approximately 2, the standard rate constant (ks) of this couple increased abruptly with solution pH below pH approximately 4.5 at a BSA-modified Au electrode, but decreased with solution pH at a BSA-modified Pt electrode. By analyzing the QCM impedance data with a modified BVD equivalent circuit and the EI data with a modified Randle's equivalent circuit, inflexion changes at pH approximately 4.5 were all found at pH-dependent responses of the resonant frequency, the double-layer capacitance, the capacitance of the adsorbed BSA layer, the peak-absorbance values of BSA solutions at 277.5 and 224.5 nm, and so on. It was also found that a BSA adsorption layer can effectively inhibit gold corrosion during ferrocyanide oxidation in a ferrocyanide-containing BR solution. Some preliminary explanations of these findings have been given. The proposed method is highly recommended for wider applications in surface science.

To probe the origin of activation effect of carboxylic acid and (+)-NLE in tridentated titanium catalyst systems.

The activation effect of carboxylic acid and positive nonlinear effect ((+)-NLE) in tridentated titanium catalyst systems for hetero Diels-Alder reaction and aldol-type reactions has been first elucidated on the basis of X-ray crystal structural analysis of homochiral (pseudo-octahedral coordination geometry with C2 symmetry) and heterochiral (pseudo-octahedral coordination geometry with C1 symmetry) titanium complexes and the observation of their different reactivities with carboxylic acid additive.

Dramatically synergetic effect of carboxylic acid additive on tridentate titanium catalyzed enantioselective hetero-diels-alder reaction: additive acceleration and nonlinear effect.

This paper describes the successful development of a group of highly efficient chiral tridentate titanium catalysts for hetero-Diels-Alder reaction of Danishefsky's diene and a variety of aldehydes through ligand and additive diversity. Dramatically synergetic effect of carboxylic acid additives and influence of substituent in chiral Schiff base ligands on the enantioselectivities of the reaction are reported. It was found that a chiral drug Naproxen (A21) was a highly efficient additive for Ti-catalyzed HDA reaction, affording 2-substituted 2,3-dihydro-4H-pyran-4-one in up to 97 % ee and >99 % yields. Quantitative study of the effect of chiral carboxylic acid A21 revealed that the reaction could be accelerated by one order of magnitude. Another interesting feature of present catalytic system is the existence of significant positive nonlinear effect, which indicates that the heterochiral Schiff base-titanium complexes may have higher stability than their homochiral counterparts. As a result, the homochiral Schiff base-titanium complexes with higher ee than that of starting ligand will react with carboxylic acid additive to form the more active species and predominate the catalytic process. The isolation and characterization of stable heterochiral ((+/-)-L1)2Ti complex has also been successful, which strongly supported the explanation for positive nonlinear effect observed in the catalytic system.

Acid-free aza Diels-Alder reaction of Danishefsky's diene with imines.

[reaction: see text] A highly efficient aza Diels-Alder reaction of Danishefsky's diene with imines was found to occur in methanol in the absence of any acids at room temperature to give corresponding 2-substituted dihydro-4-pyridone derivatives in high yields. This reaction can be also carried out in a three-component one-pot reaction manner. The reaction was found to proceed through a Mannich-type condensation mechanism.

Assay and analysis for anti- and pro-oxidative effects of ascorbic acid on DNA with the bulk acoustic wave impedance technique.

A bulk acoustic wave (BAW) impedance sensor has been applied for in situ monitoring of the whole process of DNA oxidative damage induced by the vitamin C (Vc)-Fe (III) system, based on its real-time responses to the density-viscosity change of the tested solution due to the damages occurring on the DNA molecules. The results showed that Vc exhibited two conflicting effects, i.e., pro-oxidation and anti-oxidation on the DNA at different Vc concentrations in the damage system, and the "threshold" concentration of Vc for these two effects was estimated to be about 100 micromol/L. The end-point frequency change of the sensor (Deltaf(m)) was found to be linearly related to the initial concentration of the soybean DNA (C(DNA)) in the range of 40-1000 microg/mL, and the exponential relationship between the frequency change (Deltaf(0)) vs damaging time suggested that the Fe (III)-mediated DNA damage by Vc could be described as a first-order kinetics reaction. The effects of variations in concentrations of Vc and Fe3+ on the DNA oxidative damage were discussed, and based on investigations for the enhancing influence of H2O2 and inhibiting influence of HO* scavengers on the DNA damage, the nature and physiological toxicity of the damage in biological system were also examined. In addition, UV-vis spectra and electrophoresis analysis were also used, and the experimental observations were in good agreement with the above results.

Piezoelectric quartz crystal impedance study of the Pb2+-induced precipitation of bovine serum albumin and its dissolution with EDTA in an aqueous solution.

The piezoelectric quartz crystal impedance technique (QCI) was employed to monitor in situ the Pb2+-induced precipitation of BSA onto a gold electrode and the precipitate dissolution with EDTA in an aqueous solution. The critical precipitation concentration of Pb2+, at which the resonant frequency decreased significantly, was estimated to be 4.78 x 10(-4) mol/L. The saturated adherence of the precipitate on the electrode was observed when the concentration of Pb2+ was greater than 7.53 x 10(-2) mol/L. The frequency response was mainly caused by the mass effect of the precipitate adherence to the electrode, rather than the changes in the physico-chemical properties of the contacting liquid. An excess addition of Na2EDTA after the Pb2+-BSA dissolution led to new precipitation, probably due to the formation of an EDTA precipitate in this medium (pH approximately 3). The pH effect on the response of the resonant frequency was analyzed by using the sum of two exponential functions. A larger frequency response occurred at a pH greater than pI. These findings have been reasonably explained. Also, a decrease in the concentration of the background electrolyte increased the frequency response.

Functional Difference between N Domain and C Domain of hEGF and hTGF-alpha.

By exchanging the N domain and C domain of hEGF and hTGF-alpha genes by PCR, two chimeras E-TGF(EGF(1-32)-TGF-alpha(34-50))and T-EGF(TGF-alpha(1-33)-EGF(33-53))were constructed. The wild and chimeric molecules were expressed in E.coli under phoA system. The expressed hEGF, hTGF-alpha and two chimeras were purified. The EGF receptor competitive binding affinity of the four molecules was hEGF > hTGF-alpha and E-TGF > T-EGF and the cell proliferation stimulating activity of them was hTGF-alpha and E-TGF > T-EGF > hEGF. The result suggests that the N domain of hEGF and hTGF-alpha may play a major role in receptor binding activity and C domain of them may be responsible for stimulating cell proliferation.

The Effect of Partially Conserved Residues of hTGF-alpha C Domain in Its Structure and Function.

There is a highly homologous region in the C domain of the EGF family, some of its residues are semi-conserved. We constructed three hTGF-alpha mutants, hTGF-alphaV35, hTGF-alphaQ44, hTGF-alphaY45R46, by site-directed mutagenesis to replace the semi-conserved residues in the C domain of hTGF-alpha with the corresponding residues of hEGF. We observed that although the binding affinity of hEGF to hEGF receptor was about two fold that of hTGF-alpha, but the receptor binding affinity of the three mutants was respectively decreased to about 22 %, 13.4 % and 25 % compared of that of hTGF-alpha. On the other hand, the stimulating action of hEGF on NRK-49F cell proliferation was only 10 % that of hTGF-alpha, but those of the threemutants was about 4 fold, 10 fold and 5 fold more active than hTGF-alpha. Thus, the three mutants did not become more similar to hEGF in function. The functional difference between hEGF and hTGF-alpha was not simply determined by any single semi-conserved residue, but substitution at those sites in the C domain have altered the characters of hTGF-alpha sharply.
