[Dependence of Ca2+ on the acetylcholine-sensitive current in guinea pig type II vestibular hair cells].

OBJECTIVE: To explore the dependence of Ca2+ on the acetylcholine (ACh)-sensitive potassium current in guinea pig type II vestibular hair cells. METHODS: Under the whole-cell patch mode, the current amplitude of the ACh-sensitive potassium current was recorded in response to the concentration change of the extracellular or intracellular Ca2+. RESULTS: Following application of ACh, type II vestibular hair cells displayed the sustained potassium current, which was inhibited by tetraethylammonium chloride (TEA), but not inhibited by 4-aminopyrine (4-AP). The activation of the ACh-sensitive potassium current was strongly affected by the concentration of the extracellular Ca2+. The current amplitude of the ACh-sensitive potassium increased following the increase of Ca2+ concentration from 0 mmol/L to 4 mmol/L At the concentration of 4 mmol/L Ca2+, the current amplitude of the ACh-sensitive potassium current reached the maximal response. Lowering the Ca2 concentration in the external solution from 4 mmol/L to 0. 5 mmol/L, the current amplitude of the ACh-sensitive potassium current was inhibited to (36.5 +/- 6.5)%. However, no difference was found in the presence and in the absence of the intracellular heparin, which was a well-known blocker of the inositol trisphosphate-dependent calcium release channels. In addition, the calcium channel blocker, Cd2+, inhibited the ACh-sensitive potassium current. CONCLUSIONS: The activation of the ACh-sensitive potassium current in guinea pig type II vestibular hair cells was dependent on the extracellular Ca2+ influx through the calcium channel. The application of ACh would stimulate membrane Ca2+ channels; the influx of Ca2+ will then activate the calcium-dependent potassium current in guinea pig type II hair cells to mediate the hyperpolarization effect.

[Progesterone-modulated proteins in human endometrial cancer cell line Ishikawa].

OBJECTIVE: To identify proteins associated with growth inhibitory effects of progesterone in Ishikawa endometrial adenocarcinoma cell line (Ishikawa). METHODS: The total cellular proteins of the control and megestrol acetate (MA)-treated Ishikawa cells were separated by two-dimensional gel electrophoresis. After colloidal Coomassie G-250 staining and scanning, the gel images were analyzed by PDQuest software. Matrix-assisted laser desorption/ionization-time-of-flight mass spectrometry (MALDI-TOF MS) and Mascot software were used to identify the differentially expressed protein. RESULTS AND CONCLUSIONS: Well-resolved and reproducible 2-dimensional patterns of both control and MA-treated Ishikawa cells were obtained. Forty-one proteins displayed differential expression after MA treatment, among which 23 were up-regulated and 18 down-regulated. Peptide mass fingerprints (PMF) by MALDI-TOF-MS analysis and search in NCBInr resulted in the identification of 8 proteins. The up-regulated proteins identified were catechol-O-methyltransferase (COMT), glutathione S-transferase, keratin 8, splicing factor 3a (subunit 3), chaperonin containing TCP1 (subunit 6A isoform a, CCT6A), RAB6A protein and AHA1. The down-regulated protein was peptidyl prolyl isomerase A (isoform 1). These findings can be helpful in further investigation of the molecular mechanism of progesterone.

[Efficient plant regeneration in vitro in Pinus massoniana L].

Pinus massoniana L. is one of the important trees for afforestation in South China. The efficient system of plant regeneration from mature zygotic embryos and seedlings of masson pine was established in this study. The influences of basal media, hormones and methods for buds induction, shoots elongation and rooting were studied. The results indicate that DCR medium with 0.5 mg/L BA and 0.05 mg/L IBA shows the highest differentiation rate of adventitious buds. Induction and multiplication of axillary buds take aseptic seedlings as explants. KT has better effect than BA on the axillary buds induction. The best axillary buds induction medium is DCR medium supplemented with 1 mg/L KT and 0.2 mg/L IBA. After culturing on GD medium with 0.1 mg/L BA and 0.2 mg/L IBA for elongation, the buds were transferred on the 1/2 GD medium with 2 mg/L IBA and 0.05 mg/L BA for adventitious roots induction. Paraffin slice indicates that the adventitious buds developed from the meristematic tissue of cotyledonary epicuticula.

[Surgical pattern of radical hysterectomy and pelvic lymph node dissection for patients with cervical cancer].

OBJECTIVE: To investigate the probability of improving radical surgery to resect uterus thoroughly and to decrease various complications after the surgery. METHODS: We compared the clinical effect of reformed radical hysterectomy for 79 patients with the effect of tradition radical hysterectomy for 60 patients. Reformed surgery had the following features: We firstly resected the uterus and then dissected the pelvic lymph node. Urinary bladder gap and rectum gap were opened. After exposing the route of ureter, we excised the uterus artery at the point between the ureter and the uterus artery. Cardinal ligament and uterosacral ligament were cut off by electric knives. The pelvic lymph node was dissected with a titanium pinch. RESULTS: The average operation time and the time of keeping uterine pipe were shortened. Bleeding during the operation was reduced. No complication was observed. All patients were followed up for 11 to 20 months and no patient died. One patient recurred. CONCLUSION: Reformed surgery can resect the uterus, dissect the pelvic lymph node thoroughly, and reduce various complications. The reformed surgery can not only ensure the curative effect, but also benefit patient's recovery and life quality.

[Effect of erythromycin on apoptosis of epithelial cell in nasal polyps].

OBJECTIVE: To study the effect of erythromycin on apoptosis and expression of Bax and Bcl-2 of epithelial cell in nasal polyps. METHODS: Epithelial cells from thirty nasal polyps and fifteen inferior turbinates were cultured in Dulbecco Eagle and Ham F12 (1:1) and divided into two groups (one group were treated with Erythromycin). Apoptosis was detected by terminal deoxynucleotidyl transferase mediated dUTP nick end labeling and expression of Bcl-2 and Bax detected by immunohistochemistry at 1, 3, 5 days after culture. RESULTS: The apoptosis indexes (AI) of epithelial cell in nasal polyps after cultured for 1, 3, 5 days with erythromycin were (33.23 +/- 6.50)%, (38.21 +/- 7.22)% and (52.63 +/- 7.86)% respectively. The AI of epithelial cell in inferior turbinates were (31.02 +/- 5.60)%, (32.13 +/- 7.15)% and (39.64 +/- 7.48)% respectively. There was significant difference between two groups at 5 day after culture (P < 0.05). Expression of Bax and Bcl-2 of epithelial cell was significantly higher in nasal polyps than that in inferior turbinates (P < 0.01). Expression of Bax of epithelial cell cultured with erythromycin was significantly higher than that in the controls (P < 0.05). Apoptosis was clearly promoted after 5 day culture with Erythromycin (P < 0.05). CONCLUSIONS: Erythromycin promoted expression of Bax and apoptosis of epithelial cell in nasal polyps.

[Preliminary studies on physicochemical properties of Angelica sinensis polysaccharide-iron complex].

OBJECTIVE: To study some physicochemical properties of Angelica sinensis polysaccharide-iron complex (APC). METHOD: Based on the qualitatively identified reactions of iron (III), the qualitatively identified reactions of APC were found out by comparing hydroxide. The content of iron (III) in APC was determined with iodometry. The stability of APC under physiological pH conditions was judged by titrating APC with sodium hydroxide. The deoxidization of APC was tested with colorimetric analysis. RESULT: APC showed the qualitatively identified reactions of iron (III). The content of iron (III) in APC ranged from 10% to 40%, and the water-solubility of APC was related to the content of iron (III). The complex was stable at physiological pH from 3 to 12, without precipitation and dissociation. At 37 degrees C, Fe (III) in the complex was completely reduced to Fe (III) by ascorbic acid in about 6 hours. CONCLUSION: APC can be qualitatively identified by using the qualitatively identified reactions of iron (III). When its iron (III) content is within 20%-25%, APC has a better ability to dissolve in water. And the complex is stable under physiological pH conditions.

[A comparative study of unrelated donor bone marrow transplantation and peripheral blood stem cell transplantation for their therapeutic effects on leukemia].

OBJECTIVE: To compare the effect of unrelated donor bone marrow (BM) transplantation and peripheral blood stem cell (PBSC) transplantation in light of hemopoietic reconstitution, immune reconstitution, infection, incidence of graft-versus-host disease (GVHD) and other complications in patients with leukemia. METHODS: The clinical outcomes of 16 patients receiving unrelated PBSC graft mobilized by granulocyte colony-stimulating factor (G-CSF) were compared with 30 patients receiving unrelated BM transplantation. RESULTS: Engraftment was achieved in 97.83% of the total patients. Compared with BM transplantation group, PBSC graft contained significantly more nucleated cells (P=0.000), resulting in a significantly shorter time-to-neutrophil (16.21-/+3.09 vs 12.81-/+4.15 days, P=0.003) and platelet engraftment (20.31-/+7.19 vs 15.50-/+6.91 days, P=0.035). T cell reconstitution differed little between the two groups at different time points after transplantation. The incidences of early-stage infection were 37.50% and 50.00% (P=0.644) in the PBSC and BM groups, respectively. In PBSC and BM groups, the incidences of grades I to IV acute GVHD (aGVHD) were 56.25% and 70.00% (P=0.456), 18.75% and 13.79% (P=0.661) for grades III to IV aGVHD, and 30.77% and 36.36% (P=0.413) for chronic GVHD (cGVHD), respectively. The nonrelapse transplant-related mortality (TRM) rates were 18.75% in PBSC group and 33.33% in BM group (P=0.295). The relapse occurred in 18.75% and 6.90% (P=0.226) of the patients in the two groups, respectively, and the 2-year disease-free survival (DFS) rates were 62.19% and 56.23% (P=0.615), respectively. CONCLUSION: G-CSF-mobilized PBSCs allow more rapid engraftment in unrelated donor recipients in comparison with conventional BM, but T cell reconstitution and the incidence of infection between the two groups differ little, nor are there significant differences in the incidence or severity of aGVHD and cGVHD, nonrelapse TRM or 2-year DFS rates between the two groups.

Is the hypoxia-inducible factor-1 alpha mRNA expression activated by ethanol-induced injury, the mechanism underlying alcoholic liver disease?

BACKGROUND: Excessive alcohol consumption can result in multiple organ injury, of which alcoholic liver disease (ALD) is the most common. With economic development and improvement of living standards, the incidence of diseases caused by alcohol abuse has been increasing in China, although its pathogenesis remains obscure. The aim of this study was to investigate the role of hypoxia in chronic ALD. METHODS: Twenty-eight male Sprague-Dawley rats were randomized into a control group (n=12) with a normal history and an experimental group (n=16) fed with 10 ml/kg of 56% (vol/vol) ethanol once per day by gastric lavage for 24 weeks. At 24 weeks, blood samples were collected and then the rats were killed. Liver samples were frozen at -80 degrees C and used for RT-PCR; other liver samples were obtained for immunohistochemical staining. RESULTS: When the period of alcohol consumption increased, the positive rate of expression of hypoxia-inducible factor-1 alpha (HIF-1alpha) mRNA was more significantly elevated in the liver of the alcohol group than in the control group (P < or = 0.05). The HIF-1alpha protein located in the cytoplasm was seldom expressed in the control group, but significantly in the alcohol group (P < or = 0.01). CONCLUSION: HIF-1alpha mRNA expression was activated by ethanol-induced injury in this study, suggesting that hypoxia is involved in the underlying mechanism of ALD.

[Effects of hydraulic retention time (HRT) on nitrification performance and microbial community of conventional activated sludge (CAS)].

A conventional activated sludge system was used to treat the ammonium-bearing inorganic wastewater for 260 days under decreased HRTs. When treating 500 mg x L(-1) NH4+ -N wastewater, over 98% ammonia removal could be obtained at HRT > or =20 h. Sharp washout of biomass occurred when HRT was further decreased, resulting in the accumulation of ammonium and nitrite. Changes in the community structure were monitored by microbial quinone and conventional analytical methods. Quinone profiles indicated that ammonia-oxidizing bacteria (AOB) in beta-Protevbacteria and Nitrobacteria in alpha-Proteobacteria were the predominant species to oxidize ammonia and nitrite, respectively. The number of AOB decreased with the reduction of biomass in bioreactor, while that of nitrite-oxidizing bacteria (NOB) changed little and fluctuated at 10(5) CFU x L(-1)

[Expression and clinical significance of nm23-H1 and MUC-1 in peripheral T-cell lymphoma].

BACKGROUND & OBJECTIVE: Peripheral T-cell lymphoma (PTCL) is a group of heterogeneous malignancy with poor prognosis. The role of international prognostic index (IPI) in PTCL remains to be determined. It is necessary to find new molecular markers for PTCL. This study was to evaluate the clinical significance of nm23-H1 and MUC-1 in predicting the prognosis of PTCL. METHODS: The expression of nm23-H1 and MUC-1 proteins in 96 specimens of PTCL was detected by SP immunohistochemistry. The correlations of nm23-H1 and MUC-1 expression to clinical features, objective response, and overall survival of PTCL patients were analyzed. RESULTS: Of the 96 patients, 78 (81.2%) were nm23-H1-positive, 56 (58.3%) were MUC-1-positive. Neither of the expression of nm23-H1 and MUC-1 was correlated to the pathologic subtype of PTCL (P>0.05). The high expression of nm23-H1 was associated with some poor prognostic factors such as stage III-IV, performance status (PS)> or =2, extranodal involvement, and more than one site of extranodal involvement (P<0.05). The high expression of MUC-1 was only associated with stage III-IV and more than one site of extranodal involvement (P<0.05). Of the 89 patients with evaluable disease, the overall response rate was 87.8% with a complete remission (CR) rate of 56.7%. The CR rate was significantly higher in nm23-H1-negative patients than in nm23-H1-positive patients (66.7% vs. 55.4%, P<0.05), and significantly higher in the patients with low nm23-H1 expression than in those with high nm23-H1 expression (79.9% vs. 44.0%, P<0.05); the CR rate was higher in MUC-1-negative patients than in MUC-1-positive patients, and higher in the patients with low MUC-1 expression than in those with high MUC-1 expression, but the differences were not significant. The median follow-up of the whole group was 30 months (range, 2-98 months), and the median survival time was 32 months [95% confidence interval (CI)= 26-34 months]. The overall 5-year survival rate of the whole group was 35.1%. The overall 5-year survival rate was significantly higher in nm23-H1-negative patients than in nm23-H1-positive patients (86.7% vs. 24.9%, P=0.001), and significantly higher in the patients with low nm23-H1 expression than in those with high nm23-H1 expression (52.3% vs. 21.7%, P<0.001). The overall 5-year survival rate was slightly higher in MUC-1-negative patients than in MUC-1-positive patients (47.9% vs. 28.5%, P>0.05), and slightly higher in the patients with low MUC-1 expression than in those with high MUC-1 expression (46.2% vs. 22.2%, P>0.05). Multivariant analysis showed that IPI score and nm23-H1 expression were independent prognostic factors of PTCL. CONCLUSIONS: Overexpression of nm23-H1 is related to poor prognosis of PTCL; it may be a potential prognostic index of PTCL. Overexpression of MUC-1 is not related to.

[Comparison of clinical outcomes between unrelated donor peripheral blood stem cell transplantation and bone marrow transplantation for leukemia].

OBJECTIVE: To compare the hemopoietic reconstitution, immune reconstitution, infection, incidence of graft-versus-host disease (GVHD) and clinical outcome between unrelated donor peripheral blood stem cell (PBSC) transplantation and bone marrow (BM) transplantation for leukemias. METHODS: The clinical results of 21 leukemia patients receiving G-CSF mobilized PBSC graft from unrelated donors were compared with that of 32 patients receiving unrelated BM transplants. RESULTS: Compared with BM grafts, the PBSC graft contained significantly more nucleated cells (P = 0.000), and resulted in a significantly shorter time-to-neutrophil (12.43 +/- 3.67 vs 16.16 + 2.99 days) and platelet engraftment (14.67 +/- 6.19 vs 21.23 +/- 8.25 days), (P = 0.000 and 0.003, respectively). T cell reconstitution between the two groups differed little after transplantation. The incidences of early-stage infection (42.86% vs 53.13%), the probabilities of acute graft-versus-host disease (aGVHD) (61.90% vs 71.88%), the grades III to IV aGVHD (23.81% vs 15.63%), the chronic GVHD (47.06% vs 43.48%) and the probabilities of relapse (6.90% vs 12.50%) between PBSC and BM groups all has no statistical significance (NS). The 2-year disease free survival (DFS) rates of the two groups were (50.14 +/- 12.00) % and (59.81 +/- 8.99)%, respectively also have no NS. CONCLUSION: G-CSF-mobilized unrelated donor PBSCs engraft more rapidly in the recipients as compared with conventional BM grafts. The T cell reconstitution, the incidence of infection, the incidence and severity of aGVHD and cGVHD, and the 2-year DFS rates between the two groups all have no significant differences.

Phonatory impairment in Parkinson's disease: evidence from nonlinear dynamic analysis and perturbation analysis.

Many persons with Parkinson's disease (PD) will eventually experience vocal impairment as their condition advances. Using standard perturbation analyses (parameters like jitter and shimmer) to measure fluctuations in phonatory signal may inhibit researchers from recognizing severely disordered patterns that seem to be present in the voices of some PD patients. Nonlinear dynamic analysis can quantify these aperiodic patterns, which indicate severe pathology that is usually characterized perceptually by hoarseness. Here, sustained vowel phonations of a heterogeneous group of PD subjects (20 women and 21 men) were compared with those of a control group (22 women and 18 men) based on results of nonlinear dynamic analyses (D(2)) and perturbation analyses. Results showed PD subjects as a whole to have significantly higher D(2) values than control subjects (P = 0.016), which indicates increased signal complexity in PD vocal pathology. Differences in the comparison of these two groups were significant in jitter (P = 0.014) but nonsignificant in shimmer (P = 0.695). Furthermore, the performance on these three measures was affected by subject sex. Nonlinear dynamic analysis showed significantly higher D(2) in the female PD group than in the female control group (P = 0.001), but jitter and shimmer did not show such a difference. The male PD group had statistically higher jitter than the male control group (P = 0.036), but these groups did not differ in D(2) or shimmer. Overall, nonlinear dynamic analysis may be a valuable method for the diagnosis of Parkinsonian laryngeal pathology.

Theoretical studies on vibrational spectra of some halides of group IVB elements.

The vibrational spectra of group IVB elements halides MX4 (M=Ti(IV), Zr(IV), Hf(II); X=F, Cl, Br and I), have been investigated by ab initio RHF, MP2 and density functional theory B3LYP method with LanL2DZ basis sets. The optimized geometries, calculated vibrational frequencies and Far-IR intensities of MX4 are evaluated via comparison with experimental data. The vibrational frequencies, calculated by these methods, are compared to each other. The results indicate that B3LYP method is more reliable than RHF and MP2 methods for the frequencies calculations for these compounds. With this method, some vibrational frequencies of M2X6(2+)(M=Ti(IV), Zr(IV) and Hf(II); X=F, Cl, Br and I) are also predicted.

Effects of synergistic reinforcement and absorbable fiber strength on hydroxyapatite bone cement.

Approximately a million bone grafts are performed each year in the United States, and this number is expected to increase rapidly as the population ages. Calcium phosphate cement (CPC) can intimately adapt to the bone cavity and harden to form resorbable hydroxyapatite with excellent osteoconductivity and bone-replacement capability. The objective of this study was to develop a strong CPC using synergistic reinforcement via suture fibers and chitosan, and to determine the fiber strength-CPC composite strength relationship. Biopolymer chitosan and cut suture filaments were randomly mixed into CPC. Both suture filaments and composite were immersed in a physiological solution. After 1-day immersion, cement flexural strengths (mean +/- SD; n = 6) were: (2.7 +/- 0.8) MPa for CPC control; (11.2 +/- 1.0) MPa for CPC-chitosan; (17.7 +/- 4.4) MPa for CPC-fiber composite; and (40.5 +/- 5.8) MPa for CPC-chitosan-fiber composite. They are significantly different from each other (Tukey's at 0.95). The strength increase from chitosan and fiber together in CPC was much more than that from either fiber or chitosan alone. The composite strength became (9.8 +/- 0.6) MPa at 35-day immersion and (4.2 +/- 0.7) MPa at 119 days, comparable to reported strengths for sintered porous hydroxyapatite implants and cancellous bone. After suture fiber dissolution, long macropore channels were formed in CPC suitable for cell migration and tissue ingrowth. A semiempirical relationship between suture fiber strength S(F) and composite strength S(C) were obtained: S(C) = 14.1 + 0.047 S(F), with R = 0.92. In summary, this study achieved substantial synergistic effects by combining random suture filaments and chitosan in CPC. This may help extend the use of the moldable, in situ hardening hydroxyapatite to moderate stress-bearing orthopedic applications. The long macropore channels in CPC should be advantageous for cell infiltration and bone ingrowth than conventional random pores and spherical pores.

Dysregulation of receptor interacting protein-2 and caspase recruitment domain only protein mediates aberrant caspase-1 activation in Huntington's disease.

Caspase-1 plays a role in the pathogenesis of a variety of neurological diseases. Caspase-1 activation is an early event in models of Huntington's disease (HD). However, mechanisms regulating the activation of this apical caspase in cell death are not known. Receptor interacting protein-2 (Rip2) and caspase recruitment domain (CARD) only protein (Cop) are two CARD proteins with significant homology to the caspase-1 CARD and modulate caspase-1 activation in inflammation. Rip2 is a caspase-1 activator, and Cop is a caspase-1 inhibitor. We demonstrate in models of HD that caspase-1 activation results from dysregulation of caspase-1 activation pathways. Associated with disease progression, we detect elevation of the caspase-1 activator Rip2 and reduction of the caspase-1 inhibitor Cop. Knocking down endogenous Rip2/Cop respectively results in reduced/increased sensitivity to neurotoxic stimuli. Our data provide evidence that caspase-1-mediated cell death is regulated, at least in part, by the balance of Rip2 and Cop, and alterations of this balance may contribute to aberrant caspase-1-mediated pathogenesis in Huntington's disease.

Increased staining for phospho-Akt, p65/RELA and cIAP-2 in pre-neoplastic human bronchial biopsies.

BACKGROUND: The development of non-small cell lung carcinoma proceeds through a series of well-defined pathological steps before the appearance of invasive lung carcinoma. The molecular changes that correspond with pathology changes are not well defined and identification of the molecular events may provide clues on the progression of intraepithelial neoplasia in the lung, as well as suggest potential targets for chemoprevention. The acquisition of anti-apoptotic signals is critical for the survival of cancer cells but the pathways involved are incompletely characterized in developing intra-epithelial neoplasia (IEN). METHODS: We used immunohistochemistry to determine the presence, relative levels, and localization of proteins that mediate anti-apoptotic pathways in developing human bronchial neoplasia. RESULTS: Bronchial epithelial protein levels of the phosphorylated (active) form of AKT kinase and the caspase inhibitor cIAP-2 were increased in more advanced grades of bronchial IEN lesions than in normal bronchial epithelium. Additionally, the percentage of biopsies with nuclear localization of p65/RELA in epithelial cells increased with advancing pathology grade, suggesting that NF-kappaB transcriptional activity was induced more frequently in advanced IEN lesions. CONCLUSION: Our results indicate that anti-apoptotic pathways are elevated in bronchial IEN lesions prior to the onset of invasive carcinoma and that targeting these pathways therapeutically may offer promise in prevention of non-small cell lung carcinoma.
