A novel real-time quantitative PCR method using attached universal template probe.

A novel real-time quantitative polymerase chain reaction (PCR) method using an attached universal template (UT) probe is described. The UT is an approximately 20 base attachment to the 5' end of a PCR primer, and it can hybridize with a complementary TaqMan probe. One of the advantages of this method is that different target DNA sequences can be detected employing the same UT probe, which substantially reduces the cost of real-time PCR set-up. In addition, this method could be used for simultaneous detection using a 6-carboxy-fluorescein-labeled UT probe for the target gene and a 5-hexachloro-fluorescein-labeled UT probe for the reference gene in a multiplex reaction. Moreover, the requirement of target DNA length for UT-PCR analysis is relatively flexible, and it could be as short as 56 bp in this report, suggesting the possibility of detecting target DNA from partially degraded samples. The UT-PCR system with degenerate primers could also be designed to screen homologous genes. Taken together, our results suggest that the UT-PCR technique is efficient, reliable, inexpensive and less labor-intensive for quantitative PCR analysis.

Influence of heat treatment of rayon-based activated carbon fibers on the adsorption of formaldehyde.

The influence of heat treatment of rayon-based activated carbon fibers on the adsorption behavior of formaldehyde was studied. Heat treatment in an inert atmosphere of nitrogen for rayon-based activated carbon fibers (ACFs) resulted in a significant increase in the adsorption capacities and prolongation of breakthrough time on removing of formaldehyde. The effect of different heat-treatment conditions on the adsorption characteristics was investigated. The porous structure parameters of the samples under study were investigated using nitrogen adsorption at the low temperature 77.4 K. The pore size distributions of the samples under study were calculated by density functional theory. With the aid of these analyses, the relationship between structure and adsorption properties of rayon-based ACFs for removing formaldehyde was revealed. Improvement of their performance in terms of adsorption selectivity and adsorption rate for formaldehyde were achieved by heat post-treatment in an inert atmosphere of nitrogen.

[The effect of triptolide on the proliferation of airway smooth muscle and the expression of c-fos and c-jun in asthmatic rats].

OBJECTIVE: To study the effect of triptolide on airway smooth muscle (ASM) proliferation and expressions of c-fos and c-jun in asthmatic rats. METHODS: 70 healthy male sprague-dawley rats were randomly divided into seven groups of 10 each. The asthmatic model was established by ovalbumin inhalation and injection. The mRNA expression of c-fos and c-jun were examined by RT-PCR and their protein expression by immunohistochemical method. The airway wall thickness, the ASM thickness, the number of ASM nucleus and pulmonary tissue changes were observed with microscope. RESULTS: c-fos and c-jun mRNA and protein expressions of asthma groups A and five therapy groups (C, D, E, F and G) were significantly higher than those in the control group B (P < 0.01). The expressions were lower in group C, D, E, F and G than those in group A (P < 0.01). There was no significant difference in the expressions among group C, D, E, F and G (P > 0.05). Triptolide decreased ASM proliferation pathologically in asthmatic rats. CONCLUSIONS: ASM proliferation is a feature of asthma. c-fos and c-jun may promote ASM proliferation in asthma. The remarkable inhibition of ASM proliferation by triptolide was probably due to its inhibitory effect on the expression of c-fos and c-jun.
