Bi-functional cytokine fusion proteins for gene therapy and antibody-targeted treatment of cancer.

Typically, multiple cytokines act in concert to mediate a desired immunological response, and thus more effective therapeutics may be achieved by combining several cytokines with potentially synergistic activities. We have developed a series of bi-functional cytokine fusion proteins which, when additionally linked to an intact antibody (or the Fc portion of an antibody) in a variety of configurations, can be specifically targeted. We focus here mainly on the synergizing cytokine combination interleukin-2/interleukin-12 (IL-2/IL-12), but also demonstrate the utility of this approach with interleukin-4/granulocyte-macrophage colony-stimulating factor (IL-4/GM-CSF). Cytokine activity was retained in constructs where the cytokines were fused in tandem at the carboxyl terminus of the Fc or antibody heavy (H) chain, as well as in constructs where one cytokine was fused at the carboxyl terminus of the H chain while the second cytokine was fused to the amino terminus of either the H or light (L) chain variable region. Even in such constructs, antigen binding of the antibody-cytokine fusion proteins could be maintained. In the context of bi-functional fusion proteins, hetero-dimeric IL-12 could be expressed either in a single-chain form, or maintained as a heterodimer in which the p40 subunit was fused to IL-2. These IL-12/IL-2 bi-functional fusion proteins were shown to induce extremely high levels of interferon-gamma (IFN-gamma), similar to the synergy normally seen with the combined application of the individual cytokines. In addition, these bifunctional molecules were shown to have striking anti-tumor activity as either gene therapy or as an antibody cytokine(s) fusion protein, and may provide a useful approach to the treatment of cancer.

Chemical transformation is not rate-limiting in the reaction catalyzed by Escherichia coli 6-hydroxymethyl-7,8-dihydropterin pyrophosphokinase.

6-Hydroxymethyl-7,8-dihydropterin pyrophosphokinase (HPPK) catalyzes the transfer of pyrophosphate from ATP to 6-hydroxymethyl-7,8-dihydropterin (HMDP). Because HPPK is essential for microorganisms but is absent from human and animals, the enzyme is an excellent target for developing antimicrobial agent. Thermodynamic analysis shows that Mg(2+) is important not only for the binding of nucleotides but also for the binding of HMDP. Transient kinetic analysis shows that a step or steps after the chemical transformation are rate-limiting in the reaction catalyzed by HPPK. The pre-steady-state kinetics is composed of a burst phase and a steady-state phase. The rate constant for the burst phase is approximately 50 times larger than that for the steady-state phase. The latter is very similar to the k(cat) value measured by steady-state kinetics. A set of rate constants for the individual steps of the HPPK-catalyzed reaction has been determined by a combination of stopped-flow and quench-flow analyses. These results form a thermodynamic and kinetic framework for dissecting the roles of active site residues in the substrate binding and catalysis by HPPK.

Crystal structure of shikimate kinase from Mycobacterium tuberculosis reveals the dynamic role of the LID domain in catalysis.

Shikimate kinase (SK) and other enzymes in the shikimate pathway are potential targets for developing non-toxic antimicrobial agents, herbicides, and anti-parasite drugs, because the pathway is essential in the above species but is absent from mammals. The crystal structure of Mycobacterium tuberculosis SK (MtSK) in complex with MgADP has been determined at 1.8 A resolution, revealing critical information for the structure-based design of novel anti-M. tuberculosis agents. MtSK, with a five-stranded parallel beta-sheet flanked by eight alpha-helices, has three domains: the CORE domain, the shikimate-binding domain (SB), and the LID domain. The ADP molecule is bound with its adenine moiety sandwiched between the side-chains of Arg110 and Pro155, its beta-phosphate group in the P-loop, and the alpha and beta-phosphate groups hydrogen bonded to the guanidinium group of Arg117. Arg117 is located in the LID domain, is strictly conserved in SK sequences, is observed for the first time to interact with any bound nucleotide, and appears to be important in both substrate binding and catalysis. The crystal structure of MtSK (this work) and that of Erwinia chrysanthemi SK suggest a concerted conformational change of the LID and SB domains upon nucleotide binding.

Dissociation kinetics of energy-selected Cp(2)Mn(+) ions studied by threshold photoelectron-photoion coincidence spectroscopy.

Threshold photoelectron-photoion coincidence spectroscopy has been used to investigate the dissociation kinetics of the manganocene ion, Cp(2)Mn(+) (Cp = eta(5)-cyclopentadienyl). The Cp loss reaction was found to be extremely slow over a large ion internal energy range. By simulating the measured asymmetric time-of-flight peak shapes and breakdown diagram, the 0 K thermochemical dissociation limit for CpMn(+) production was determined to be 9.55 +/- 0.15 eV. A CpMn(+)-Cp bond energy of 3.43 eV was obtained by combining this CpMn(+) + Cp dissociation limit with the Cp(2)Mn adiabatic ionization energy of 6.12 +/- 0.07 eV. Combining the measured onset with known heats of formation of Cp and Mn(+), the Cp-Mn(+) bond energy was determined to be 3.38 +/- 0.15 eV. These results lead to 298 K heats of formation of Cp(2)Mn(+) and CpMn(+) of 863 +/- 7 and 935 +/- 16 kJ/mol, respectively. Finally, by combining these results with a previous measurement of the CpMn(CO)(3) --> CpMn(+) + 3CO + e(-) dissociation limit, we arrive at a new value for Delta(f)H degrees (298K)(CpMn(CO)(3)) of -424 +/- 17 kJ/mol.

Metal-benzene and metal-CO bond energies in neutral and ionic C6H6Cr(CO)3 studied by threshold photoelectron-photoion coincidence spectroscopy and density functional theory.

photoelectron-photoion coincidence spectroscopy and density functional theory calculations have been used to investigate the dissociation kinetics of the benzene chromium tricarbonyl ion, BzCr(CO)3+ (Bz = C6H6). The dissociation of the BzCr(CO)3+ ion proceeds by the sequential loss of three CO and benzene ligands. The first and third CO and the benzene loss reactions were associated with metastable precursor ions (lifetimes in the microsecond range). By simulating the resulting asymmetric time-of-flight peak shapes and breakdown diagram, the 0 K appearance energies of the four product ions were determined to be 8.33 +/- 0.05, 8.93 +/- 0.05, 9.97 +/- 0.06, and 11.71 +/- 0.06 eV, respectively. Combined with the ionization energy of BzCr(CO)3, 7.30 +/- 0.05 eV, the three successive Cr-CO bond energies in the BzCr(CO)3+ were found to alternate, with values of 1.03 +/- 0.05, 0.60 +/- 0.05, and 1.04 +/- 0.05 eV, respectively, and the Bz-Cr bond energy in BzCr+ is 1.74 +/- 0.05 eV, a trend confirmed by the density functional theory (DFT) calculations. Using the heats of formation of the fully dissociated products, C6H6, Cr+, and CO, the 298 K heats of formation the ionic BzCr(CO)n+ (n = 03) species were determined. By scaling the DFT calculated bond energies for the neutral molecules, the heats of formation of the neutral BzCr(CO)n (n = 03) were also obtained.

[Transesophageal intraluminal ultrasonography in diagnosis and differential diagnosis of esophageal leiomyoma].

OBJECTIVE: To evaluate the diagnostic and differential diagnostic values of transesophageal echo probe (TEEP) or endoscopic ultrasonography (EUS) in the diagnosis and differential diagnosis of esophageal leiomyoma. METHODS: Between September 1996 and August 2000, TEEP or EUS was used among 23 patients with esophageal submucosal tumor and 11 patients with esophagus constricted from outside, among which 32 cases underwent CT too. All the patients except and 3 with constriction by aorta underwent surgical treatment. RESULTS: (1) The accuracy rate of diagnosis of esophageal leiomyoma by TEEP or EUS was 95.7% (22/23) in comparison with that by CT of only 42.9% (9/21, chi(2) = 14.69, P = 0.0196). (2) In the 11 patients with esophagi constricted by masses beyond or around esophagus, TEEP or EUS definitely differentiated the damage beyond or around esophagus from esophageal leiomyoma. CONCLUSION: Transesophageal intraluminal ultrasonography (TEEP and EUS) is a reliable method for the preoperative diagnosis and differential diagnosis of esophageal leiomyoma and is more accurate than CT.

The influences of ambiguity phase aberration profiles on focusing quality in the very near field--part I: single range focusing on transmission.

Most phase aberration measurement algorithms have an ambiguity for constant and tilted phase aberration profiles. Based on the Fresnel (near field) approximation with single range focusing and the Fraunhofer (far field) approximation, constant and tilted phase aberration profiles change the position of the focal point only and do not influence the image focusing quality. Therefore, ambiguity phase aberration profiles are generally considered to be harmless and ignored in those algorithms and related theoretical analyses. However, Fresnel and Fraunhofer approximations may become invalid under many medical ultrasound imaging situations, e.g., when the imaging field is in the very near field (f-number approximately 1). In the very near field, although it is known that constant and tilted phase aberration profiles may degrade the focusing quality, it seems that there is a lack of quantitative analysis results in the literature about their influences, and this is the purpose of the current paper. In this paper, a quantitative analysis with a very near field approximation is performed for single range focusing on transmission, which is a commonly used transmission focusing method in medical ultrasound imaging. The tolerable levels of constant and tilted phase aberration profiles are derived as a function of the imaging system's f-number and wavelength. Because some phase aberration measurement algorithms may also have an ambiguity for quadratic phase aberration profiles, they are also included in the analysis. The theoretical results are compared with numerical and simulation results. These results have shown that the influences of tilted and quadratic phase-aberration profiles can be ignored only under certain conditions in the very near field.

The influences of ambiguity phase aberration profiles on focusing quality in the very near field part II: dynamic range focusing on reception.

In Part I of this work, the influences of ambiguity phase aberration profiles, including constant, tilted, and quadratic profiles, on focusing quality have been quantitatively analyzed with the very near field approximation for single range focusing on transmission. In this paper, their influences are analyzed in a very different situation: dynamic range focusing on reception, which is commonly used in medical ultrasound imaging for beam formation on reception. It is shown that the results for dynamic range focusing on reception are dramatically different from those for single range focusing on transmission. For example, constant phase aberration profiles are harmless to focusing quality for single range focusing on transmission but become harmful for dynamic range focusing on reception. The analysis also shows that, compared with single range focusing on transmission, dynamic range focusing on reception is much more sensitive to ambiguity phase aberration profiles, which have adverse effects on focusing quality even in the near field and far field. These significant differences are caused by the fundamental differences between single range focusing and dynamic range focusing as well as between transmission and reception. Numerical and simulation results are also derived to test the correctness and accuracy of the theoretical results.

Intestinal damage mediated by Kupffer cells in rats with endotoxemia.

AIM: To determine the in vivo effects of phagocytic blockade of Kupffer cell (KC) on the release of proinflammatory cytokines in small intestinal lesion and on the integrity of intestinal tract by using gadolinium chloride (GdCl(3)) during early endotoxemia. METHODS: Wistar rats were divided into three groups: Group A, rats were injected with endotoxin (E. coli O111:B(4), a dose of 12 mg x kg(-1)) only; Group B, rats were pretreated intravenously with 25 mg of GdCl(3) per kg 24 h are given endotoxin; and Group C, sham operation only. All animals were sacrificed 4 h after endotoxin injection. In portion of the rats of three groups, bile duct was cannulated, which the bile was collected externally. Morphological changes of ileum were observed under light microscopy and electronic microscopy. The KC were isolated from rats by collagenase perfusion and in KC, expression of TNF-alpha and IL-6 mRNA were determined by RT-PCR analysis. Plasma and bile TNF-alpha and IL-6 Levels were determined by enzyme-linked immunosorbent assay (ELISA). RESULTS: In group A, there were neutrophil infiltration and superficial epithelial necrosis of the ileal villi, sloughing of mucosal epithelium, and disappearance of some villi. In group B, the ileal mucosal damage was much reduced. which in group C, no significant morphological changes were seen. GdCl(3) pretreatment decreased significantly the expression of TNF-alpha and IL-6 mRNA in group B (4.32+/-0.47 and 4.05+/-0.43) when compared to group A (9.46+/-1.21 and 9.04+/-1.09) (P<0.05). There was no significant expression of TNF-alpha and IL-6 mRNA in group C (1.03+/-0.14 and 10.4+/-0.13). In rats of group A, the levels of TNF-alpha and IL-6 in bile and plasma were 207+/-29 ng x L(-1), 1032+/-107 ng x L(-1), 213+/-33 ng x L(-1), and 1185+/-127 ng x L(-1), respectively. In group B, they were 113+/-18 ng x L(-1), 521+/-76 ng x L(-1), 147+/-22 ng x L(-1), and 572+/-54 ng x L(-1), respectively. In group C, they were 67+/-10 ng x L(-1), 72+/-13 ng x L(-1), 109+/-18 ng x L(-1), and 118+/-22 ng x L(-1) respectively. There were significant difference between the three group (P<0.05). CONCLUSION: KC release cytokines TNF-alpha and IL-6 causing damage to the integrity of intestinal epithelium and play a crucial role in the initiation and progression of intestinal mucosal damage during early endotoxemia.

[Sodium-enhanced limestone wet FGD in rotating-stream tray scrubber].

Adding sodium sulfate to limestone slurry can increase SO2 removal efficiency. In this paper, sodium-enhanced limestone flue gas desulfurization(FGD) tests were conducted in rotating-stream tray scrubber. Changes of SO2 removal efficiency and pH value with time were experimentally studied and, at different pH range, the dissolution rates of limestone etc. were analyzed. The effects of plate number on SO2 removal efficiency and pressure drop were investigated at temperature approximated to industrial operating value. The average plate efficiencies were calculated. According to the experimental results, increasing plate number could increase SO2 removal efficiency, but the average plate efficiency decreased. Under the experimental conditions, when the plate number was increased from 1 to 4, the removal efficiency was increased from 25.5% to 48.6% at the liquid-to-gas ratio of 4 L/m3, but the average plate efficiency was decreased from 25.5% to 15.3%. The equation of the relation between the average plate efficiency and plate number was obtained.

Suicide and the unique prevalence pattern of schizophrenia in mainland China: a retrospective observational study.

BACKGROUND: Unlike almost every other country in the world, the prevalence of both schizophrenia and suicide in China is higher in women than in men. Schizophrenia and suicide are important public-health problems for China that might be related to each other. We present prevalence data for schizophrenia and estimate relative and attributable risk of suicide in people with schizophrenia in mainland China. METHODS: We used data from the national psychiatric epidemiology study, the Ministry of Health's mortality registry, the census, and the national psychological autopsy study to estimate frequencies and rates of schizophrenia, suicide, and suicide in people with schizophrenia aged 15 years and older in mainland China during 1995-99. FINDINGS: We estimated 4.25 million people with schizophrenia in China, and 284614 suicides and 28737 suicides in people with schizophrenia yearly. Prevalence of schizophrenia was higher in women than men (relative risk 1.77 [95% CI 1.15-2.72]), and greater in urban than rural areas (1.62 [1.10-2.40]). Risk of suicide was greater in women than men (1.22 [1.20-1.23]) and in rural than urban areas (3.61 [3.56-3.66]). Relative risk of suicide in individuals with schizophrenia compared with those without was 23.8 (18.8-30.2); the proportion of all suicides attributable to schizophrenia was 9.7% (7.7-12.1). Relative risk of suicide in rural residents with schizophrenia versus those without was higher in men than in women (ratio of two relative risks 2.02 [1.13-3.63]), but in urban residents with schizophrenia the ratio was lower in men than women (0.56 [0.21-.49]). INTERPRETATION: Risk factors for suicide vary in people with different mental disorders, so identification of illness-specific risk profiles would improve prediction of suicide and help tailor prevention efforts. The difference in suicide rates by sex and residential location in individuals with schizophrenia might be one of several contributing factors to the unique epidemiological pattern of schizophrenia in China.
