[An experimental study on the inhibition effect of corneal allograft rejection in rat by local application of Tetrandrine eye drops].

PURPOSE: To investigate the effect and the best concentration of Tetrandrine (Tet) eye drops in inhibition of the corneal allograft rejection in rat. METHODS: Eighty SD rats were randomly divided for being treated with 0.3%, 0.5%, 1.0% Tetrandrine eye drops, respectively. 0.1% Dexamethasone eye drops-treated and control groups. After the rat model of heterograft-heterotopic graft rejection was made, slit-limp microscope and HE staining, light microscope and microphoto-analyses technique were used to observe the neovascular formation and inflammation at different time points at post-operation. RESULTS: The types of the cells in the surgical area were mainly lymphocytes and mononuclear-macrophage. Corneal neovascularization and inflammatory cells were significantly inhibited in 0.5% Tetrandrine eye drops-treated group (P < 0.05), compared with control group at 7 d, 14 d, 21 d, 28 d post-operation. CONCLUSIONS: The results indicate that 0.5% Tet eye drops significantly inhibit the corneal allograft rejection in rats without obvious side effects.

[Genetic diversity of arbuscular mycorrhizal fungi associated with four Alnus species in Changbai Mountains: a nested PCR-rFLP analysis].

In this paper, the colonization of arbuscular mycorrhizal fungi (AMF) on the root samples of 4 Alnus species in Changbai Mountains was investigated by using nested PCR-RFLP technique, and the PCR results were tested by restriction endonuclease analysis method. The results revealed that the uncultured AMF had a high genetic diversity, and the colonization had a trend from promiscuity to specialization. The AMF from Alnus mandshuica showed the greatest specialization to the host, and Glomus was the dominant colonizer of A. mandshuica. The AMF from A. sibirica var. hirsuta, A. sibirica, and A. tinctoria showed promiscuity, and host had more significant effects on the colonization of AMF than altitude.

[Relationship between GSTT1, GSTM1 and NQO1 gene polymorphism and acute myeloid leukemia and recurrent chromosome translocations].

OBJECTIVE: To investigate the impact of GSTM1, GSTT1 and NQO1 genotypes on susceptibility to acute myeloid leukemia (AML) and recurrent chromosome translocations of AML. METHODS: GSTT1, GSTM1 and NQO1 genotypes were detected in 228 adult patients with de novo AML and 241 controls by PCR or PCR-RFLP. RESULTS: The frequency of GSTM1 null genotype in the AML patients was 62.3%, significantly higher than that in the normal controls (52.7%, P = 0.036), however, no significant difference was found in the incidence of GSTT1 null genotype. The frequencies of NQO1(C609T) C/T and T/T genotypes were 53.1% and 25.0% respectively among the total AML patients (53.1% and 25.0% respectively), 64.3% and 25.0% respectively among the AML patients with t (8; 21) (q22; q22)/AML-ETO fusion gene, and 57.1% and 26.0% respectively among the AML patient with t (15; 17) (q22; q11)/PML-RARalpha fusion gene, all significantly higher than those in the controls (49.4% and 13.7% respectively). The relative risk of t (8; 21) (q22; q22)/AML-ETO (+) AML was 4.487 (95% CI: 1.282-15.705) for the subjects with NQO1(C609T) C/T genotype, and was 6.293 (95% CI: 1.536-25.782) for the subjects with NQO1(C609T) T/T genotype. The relative risk of t (15; 17) (q22; q11)/PML-RARalpha (+) AML was 2.531 (95% CI: 1.286-4.981) for the subjects with NQO1(C609T) C/T genotype, and was 4.149 (95% CI: 1.856-9.275) for the subjects with NQO1(C609T) T/T genotype. CONCLUSION: Determination of the NQO1(C609T) genotypes may be used as a stratification marker to predict high-risk individuals for AML, especially for AML with t (8; 21) (q22; q22)/AML-ETO fusion gene and t (15; 17) (q22; q11)/PML-RARalpha fusion gene.

MEL1S, not MEL1, is overexpressed in myelodysplastic syndromes patients with t(1;3)(p36;q21).

Balanced chromosomal translocations rarely occur in myelodysplastic syndromes (MDS). We describe here in three further Chinese patients with myelodysplastic syndromes (MDS) whose cytogenetic analysis showed t(1;3)(p36;q21). We detected the expression pattern of MEL1 and MEL1s in BM of two healthy subjects and the three patients, and found that the expression of MEL1s was overexpressed in MDS patients with t(1;3)(p36;q21). Our findings combined previous observations reported in literature support that MDS patients with t(1;3)(p36;q21) should be a new specific subset of MDS. Imbalance of a complete MEL1 message with a PR domain and a short MEL1 message lacking a PR domain (MEL1S) might be involved in the pathogenesis of these patients.

Inhibition of MAPK and NF-kappa B pathways is necessary for rapid apoptosis in macrophages infected with Yersinia.

Macrophages respond to infection with pathogenic Yersinia species by activating MAPK- and NF-kappaB-signaling pathways. To counteract this response, Yersiniae secrete a protease (Yersinia outer protein J (YopJ)) that is delivered into macrophages, deactivates MAPK- and NF-kappaB-signaling pathways, and induces apoptosis. NF-kappaB promotes cell survival by up-regulating expression of several apoptosis inhibitor genes. Previous studies show that deactivation of the NF-kappaB pathway by YopJ is important for Yersinia-induced apoptosis. To determine whether deactivation of the NF-kappaB pathway is sufficient for Yersinia-induced apoptosis, two inhibitors of the NF-kappaB pathway, IkappaBalpha superrepressor or A20, were expressed in macrophages. Macrophages expressing these proteins were infected with Yersinia pseudotuberculosis strains that secrete functionally active or inactive forms of YopJ. Apoptosis levels were substantially higher (5- to 10-fold) when active YopJ was delivered into macrophages expressing IkappaBalpha superrepressor or A20, suggesting that deactivation of the NF-kappaB pathway is not sufficient for rapid Yersinia-induced apoptosis. When macrophages expressing A20 were treated with specific inhibitors of MAPKs, similar levels of apoptosis (within approximately 2-fold) were observed when active or inactive YopJ were delivered during infection. These results suggest that MAPK and NF-kappaB pathways function together to up-regulate apoptosis inhibitor gene expression in macrophages in response to Yersinia infection and that YopJ deactivates both pathways to promote rapid apoptosis. In addition, treating macrophages with a proteasome inhibitor results in higher levels of infection-induced apoptosis than can be achieved by blocking NF-kappaB function alone, suggesting that proapoptotic proteins are stabilized when proteasome function is blocked in macrophages.

Regulation of the polarity protein Par6 by TGFbeta receptors controls epithelial cell plasticity.

The transition of cells from an epithelial to a mesenchymal phenotype is a critical event during morphogenesis in multicellular organisms and underlies the pathology of many diseases, including the invasive phenotype associated with metastatic carcinomas. Transforming growth factor beta (TGFbeta) is a key regulator of epithelial-to-mesenchymal transition (EMT). However, the molecular mechanisms that control the dissolution of tight junctions, an early event in EMT, remain elusive. We demonstrate that Par6, a regulator of epithelial cell polarity and tight-junction assembly, interacts with TGFbeta receptors and is a substrate of the type II receptor, TbetaRII. Phosphorylation of Par6 is required for TGFbeta-dependent EMT in mammary gland epithelial cells and controls the interaction of Par6 with the E3 ubiquitin ligase Smurf1. Smurf1, in turn, targets the guanosine triphosphatase RhoA for degradation, thereby leading to a loss of tight junctions. These studies define how an extracellular cue signals to the polarity machinery to control epithelial cell morphology.

Cloning, expression and characterization of the human NOB1 gene.

The yeast Nob1p (Nin one binding protein) gene is required for proteasome function and RNA metabolism. We report here the cloning and characterization of the human orthologue NOB1 gene and its products. The human NOB1 gene is composed of nine exons and eight introns and is localized on human chromosome 16q22.1. The NOB1 cDNA is 1749 bp long and contains a putative open reading frame of 1239 bp. The predicted NOB1 protein comprises a PIN (PilT amino terminus) domain and a zinc ribbon domain. Western blot analysis showed that the molecular weight of NOB1 is about 50 KDa. RT-PCR analysis of mRNA from human adult tissues showed that NOB1 is expressed mainly in liver, lung and spleen. Expression of NOB1 in mammalian culture cells indicated that the NOB1 protein is mainly localized in the nucleus. Our data provides important information for further study of the function of the NOB1 gene and its products.

Cloning and characterization of the CDZFP gene which encodes a putative zinc finger protein.

We report here the cloning and characterization of a novel human cytoplasm-distribution zinc finger protein (CDZFP) gene, isolated from human ovary cDNA library, and mapped to 4p12 by searching the UCSC genomic database. The CDZFP cDNA is 1793 base pairs in length and contains an open reading frame (ORF) encoding 236 amino acids. The CDZFP gene consists of 7 exons and encodes a putative zinc finger protein with a transmembrane region and two zinc finger motifs. Subcellular localization demonstrated that CDZFP protein was located in the cytoplasm when overexpressed in Hela cells and northern blot analysis revealed that CDZFP was ubiquitously expressed in 16 human tissues.

A leucine-rich motif targets Pseudomonas aeruginosa ExoS within mammalian cells.

Type III cytotoxins contribute to the ability of bacterial pathogens to subvert the host innate immune system. ExoS (453 amino acids) is a bifunctional type III cytotoxin produced by Pseudomonas aeruginosa. Residues 96 to 232 comprise a Rho GTPase activating protein domain, while residues 233 to 453 comprise a 14-3-3-dependent ADP-ribosyltransferase domain. An N-terminal domain (termed the membrane localization domain [MLD]) targets ExoS to the Golgi-endoplasmic reticulum (Golgi-ER) of mammalian cells. This study identifies an amino acid motif that is responsible for the membrane binding properties of the MLD. Deletion mapping showed that the MLD included a symmetrical leucine-rich motif within residues 51 to 77 of ExoS. The terminal dileucines and internal leucines and an isoleucine within the MLD, but not charged or other hydrophobic residues, targeted a reporter protein to the Golgi-ER region of HeLa cells. Mutations of the leucines within the MLD did not affect type III secretion or translocation into HeLa cells but limited the ability of ExoS to ADP-ribosylate Ras GTPases. Mutations of charged residues within the MLD did not affect type III secretion, delivery into HeLa cells, or the ability of ExoS to ADP-ribosylate Ras GTPases. The organization of the leucines within the MLD of ExoS is different from that of previously described leucine-rich motifs but is present in several other bacterial proteins. This implies a role for intracellular targeting in the efficient targeting of mammalian cells by type III cytotoxins.

[Autologous peripheral blood stem cells transplantation in treatment of 62 cases of lower extremity ischemic disorder].

OBJECTIVE: To observe the clinical efficacy of autologous peripheral blood stem cells (PBSC) transplantation in 62 cases with ischemic lower extremity disorder. METHODS: Totally 62 patients with 34 cases of diabetic foot and 28 cases of various lower extremity ischemic disorders received recombinant human granulocyte-macrophage colony-stimulating factor (rhGM-CSF) 450 - 600 microg/d by hypodermic injection for 5 days to mobilize stem cells. On the sixth day, PBSC were collected by COBE 6.1 Spectra Version with an amount of 82 - 148 ml; the number of mononuclear cells (MNC) is (718.2 - 224.6) x 10(9)/L. CD34+ cells were tested. The PBSC were injected into the ischemic lower extremity and foot intramuscularly at 3 cm x 3 cm distance. The clinical and laboratory findings were monitored from first day to 24th week. RESULTS: In 62 patients with PBSC transplantation, free of severe pain was found in 54 cases (87.1%) from 7 to 30 days, improvement of foot cool feeling in 56 patients (90.3%) from 7 to 30 days, improvement of foot ulcer in 16 cases (40.0%) from 4 to 16 weeks. Ankle/brachial index (ABI) increased in 12 cases (34.3%), TcPO2 improved in 26 cases (42.3%). Digital subtraction angiographic scores were performed in 5 patients after 8 - 12 weeks, there was formation of new collateral vessels. No related complication or adverse effect was observed except in 2 patients with diabetic foot and cerebral infarction exacerbation of symptoms during the process of stem cells mobilization in all process. CONCLUSION: Autologous PBSC transplantation might be a safe and effective method for lower extremity ischemic disorder. It could improve the quality of life of many patients as amputation of lower extremity or foot might be avoided.

[Research on noninvasive blood glucose measurement with simulate sample by NIR spectroscopy].

The article puts milk forward as simulate sample for blood glucose measurement. With the new simulate sample, the authors make deep researches on measuring method and wavelength choice in the field of noninvasive blood glucose measurement by near-infrared diffuse reflectance spectroscopy. Depending on the absorption coefficient of lactose, the authors choose the two wavebands from 1560 to 1750 nm and form 2090 to 2190 nm. The model was built by partial least-squares (PLS), several pretreatment methods (including multiplicative scatter correction, first derivative and vector normalization) and different spectral regions were compared. The best correlation values for lactose were 0.99 and the best RMSEP values for lactose were 0.045. The results provide us with valuable theory reference and practical experiences and lead us to next research on noninvasive blood glucose measurement.

[The effect of surgical treatment on thyroid-associated ophthalmopathy].

OBJECTIVE: To evaluate the effect of muscle surgery on strabismus caused by extraocular muscle for thyroid-associated ophthalmopathy (TAO), and assess the change of vision acuity, intraocular pressure, visual field and blood flow velocity in central retinal artery that made by surgery treatment. METHODS: Recession and release of the extraocular muscles were used in 17 TAO patients (20 eyes) with strabismus caused by the change of extraocular muscles. In those patients one of them was undergone superior rectus tendon resection and other one was undergone superior rectus suspension. Another patient right eye was undergone inferior rectus recession and the left one with superior rectus recession. For rest of 14 patients with 17 eyes inferior rectus recession was performed. The vision acuity, intraocular pressure, visual field, and blood flow velocity in central retinal artery were measured pre- and post-operatively. RESULTS: In 17 patients (20 eyes), 5 eyes vision acuity improved two or more lines and 15 eyes was unchanged postoperatively. Intraocular pressure and mean defect of visual field showed statistically significance (P < 0.01). The peak systolic velocity (PSV) and the end diastolic velocity (EDV) were significantly increased, and the resistance index (IR) was significantly reduced (P < 0.05). CONCLUSION: Muscle surgery of strabismus caused by extraocular muscle for TAO in the quiescent period can eliminate strabismus, decrease intraocular pressure, improve visual field and blood flow velocity in central retinal artery.

[Investigation of GSTT1, GSTM1 and NQO1 genes polymorphisms in myelodysplastic syndromes].

OBJECTIVE: To investigate the relationship between GSTM1, GSTT1 and NQO1(C609T) genotypes and myelodysplastic syndromes (MDS) susceptibility and chromosome abnormalities. METHODS: GSTT1, GSTM1 and NQO1(C609T) genotypes were detected in 52 MDS patients and 241 unrelated controls by PCR or PCR-RFLP. RESULTS: The incidence of GSTT1 and GSTM1 null genotype was significantly increased in MDS patients as compared with controls (P = 0.001 and P < 0.001, respectively). In individuals with GSTT1 and GSTM1 null genotype, the odds ratios for MDS risk were elevated to 2.873 (95% CI: 1.491-5.537) and 3.591 (95% CI: 1.717-7.508), respectively. A significantly increased frequency of GSTT(1) null genotype among MDS patients with normal karyotype and increased frequency of GSTM1 null genotype among MDS patients with chromosome abnormalities were found as compared to controls (OR = 5.336, P = 0.005 and P = 0.003, OR = 3.740, respectively). There was no difference in the incidence of NQO1(C609T) genotypes between MDS patients and controls. CONCLUSION: Determination of the GSTM1 and GSTT1 genotypes may be used as a stratification marker to predicate high-risk individuals for MDS.

Direct ab initio dynamics studies of the hydrogen abstraction reactions of hydrogen atom with n-propyl radical and isopropyl radical.

The kinetics of the hydrogen abstraction reactions of hydrogen atom with n-propyl radical and isopropyl radical were studied using the direct ab initio dynamics approach. BHandHLYP/cc-pVDZ method was employed to optimize the geometries of stationary points as well as the points on the minimum energy path (MEP). The energies of all the points for the two reactions were further refined at the QCISD(T)/cc-pVTZ level of theory. No barrier was found at the QCISD(T)/cc-pVTZ//BHandHLYP/cc-pVDZ level of theory for both reactions. The forward and reverse rate constants were evaluated with both canonical variational transition state theory (CVT) and microcanonical variational transition state theory (mu VT) in the temperature range of 300-2,500 K. The fitted three-parameter Arrhenius expression of the calculated CVT rate constants at the QCISD(T)/cc-pVTZ//BHandHLYP/cc-pVDZ level of theory are k(CVT) (n-C3H7) = 1.68 x 10(-14) T(0.84) e((319.5/T)) cm3 molecule(-1) s(-1) and k(CVT) (iso-C3H7)=4.99 x 10(-14) T(0.90) e((159.5/T)) cm3 molecule(-1) s(-1) for reactions of n-C3H7 + H and iso-C3H7 + H, respectively, which are in good agreement with available literature data. The variational effects were analysed.
