Aberrant accentuation of neurofibrillary degeneration in the hippocampus of Alzheimer's disease with amyloid precursor protein 717 and presenilin-1 gene mutations.

This study reports correlation of the hippocampal neurofibrillary tangles (NFT) density with beta-amyloid (Abeta) precursor protein (APP) 717 mutation, presenilin (PS)-1 mutation and apolipoprotein E (Apo-E) e4 alleles (E4), being graded as 3 forms (no-E4, one-E4 and two-E4) in autopsied brains from patients with familial and non-familial Alzheimer's disease (AD). We studied the density of NFT-free neurons, intracellular NFT (I-NFT), extracellular NFT (E-NFT) and total NFT (I-NFT plus E-NFT) in the six hippocampal subdivisions: cornu ammonis (CA) 1-CA4, subiculum and entorhinal cortex. The APP mutation cases showed significantly higher total NFT density in the CA1-CA2 region, and the PS-1 mutation cases also showed higher density of total NFT in the CA1-CA3 than non-familial cases. Moreover, high densities of the E-NFT contributed to these high total NFT densities. Non-familial AD cases showed a stereotypical NFT distribution with entorhinal accentuation in the hippocampus irrespective of E4 frequency. Thus, APP and PS-1 mutations predominantly affect the CA regions with profound neurodegeneration, which contributes early and severe clinical features of familial AD.

Blue copper model complexes with distorted tetragonal geometry acting as effective electron-transfer mediators in dye-sensitized solar cells.

The electron self-exchange rate constants of blue copper model complexes, [(-)-sparteine-N,N'](maleonitriledithiolato-S,S')copper ([Cu(SP)(mmt)])(0/)(-), bis(2,9-dimethy-1,10-phenanthroline)copper ([Cu(dmp)(2)](2+/+)), and bis(1,10-phenanthroline)copper ([Cu(phen)(2)](2+/+)) have been determined from the rate constants of electron transfer from a homologous series of ferrocene derivatives to the copper(II) complexes in light of the Marcus theory of electron transfer. The resulting electron self-exchange rate constant increases in the order: [Cu(phen)(2)](2+/+) < [Cu(SP)(mmt)](0/)(-) < [Cu(dmp)(2)](2+/+), in agreement with the order of the smaller structural change between the copper(II) and copper(I) complexes due to the distorted tetragonal geometry. The dye-sensitized solar cells (DSSC) were constructed using the copper complexes as redox couples to compare the photoelectrochemical responses with those using the conventional I(3)(-)/I(-) couple. The light energy conversion efficiency (eta) values under illumination of simulated solar light irradiation (100 mW/cm(2)) of DSSCs using [Cu(phen)(2)](2+/+), [Cu(dmp)(2)](2+/+), and [Cu(SP)(mmt)](0/)(-) were recorded as 0.1%, 1.4%, and 1.3%, respectively. The maximum eta value (2.2%) was obtained for a DSSC using the [Cu(dmp)(2)](2+/+) redox couple under the light irradiation of 20 mW/cm(2) intensity, where a higher open-circuit voltage of the cell was attained as compared to that of the conventional I(3)(-)/I(-) couple.

pH-Dependent reversible transformation of TPPS4 anchored on mesoporous TiO2 film between monomers and J-aggregates.

Protonated TPPS4 monomer and its J-aggregate were formed simultaneously from TPPS4 adsorbed through its sulfonic acid groups on TiO2 porous film by decreasing the pH of the surrounding water and this behavior on TiO2 film can be reversibly repeated depending on pH, indicating a flexible and stable arrangement of TPPS4 through chemical bonds between the sulfonic acid groups and the TiO2 surface.

Individual analysis of EEG band power and clinical drug response in schizophrenia.

The main purpose of this study was to investigate the relationship between short-term clinical outcome and changes in electroencephalogram (EEG) power after drug treatment in patients with schizophrenia, and also to compare two different methods for quantitative EEG analysis. EEG power analysis was performed by both conventional fixed frequency band and adjusted frequency band based on individual alpha frequency (IAF) in 16 drug-naive patients before and after drug administration. In the theta bands determined by both conventional fixed band and IAF methods, the EEG power after treatment was larger than that before treatment. In addition, there was a correlation between EEG power and clinical drug response evaluated by changes in BPRS score. With regard to this correlation, IAF methods showed no apparent advantage over methods using conventional fixed frequency bands. Conventional quantitative EEG analysis can still serve as a useful tool for the assessment of short-term outcome of drug treatment.

Volatile solvent-free solid-state polymer-sensitized TiO2 solar cells with poly(3,4-ethylenedioxythiophene) as a hole-transporting medium.

Novel, volatile solvent free, solid-state solar cells were fabricated with mesoporous TiO2 electrodes sensitized using thiophene derivatives containing carboxyl groups and in situ electropolymerized poly(3,4-ethylenedioxythiophene) as a hole-transporting material together with the ionic liquid 1-ethyl-3-methylimidazolium bis(trifluoromethylsulfonyl)amide and lithium bis(trifluoromethanesulfone)imide as additives for charge transport promotion.

Roles of electrolytes on charge recombination in dye-sensitized TiO(2) solar cells (2): the case of solar cells using cobalt complex redox couples.

Dye-sensitized solar cells (DSC) were prepared from nanoporous TiO(2) electrodes with two different cobalt complex redox couples, propylene-1,2-bis(o-iminobenzylideneaminato)cobalt(II) {Co(II)(abpn)} and tris(4,4'-di-tert-buthyl-2,2'-bipyridine)cobalt(II) diperchlorate {Co(II)(dtb-bpy)(3)(ClO(4))(2)}. The performances of the DSCs were examined with varying the concentrations of the redox couples and Li cations in methoxyacetonitrile. Under 1 sun conditions, short-circuit currents (J(sc)) increased with the increase of the redox couple concentration, and the maximum J(sc) was found at the Li(+) concentration of 100 mM. To rationalize the observed trends of J(sc), electron diffusion coefficients and lifetimes in the DSCs were measured. Electron diffusion coefficients in the DSCs using cobalt complexes were comparable to the previously reported values of nanoporous TiO(2). Electron lifetime was independent of the concentration of the redox couples when the concentration ratio of Co(II)(L) and Co(III)(L) was fixed. With the increase of Li(+) concentration, the electron lifetime increased. These results were interpreted as due to their slow charge-transfer kinetics and the cationic nature of Co complex redox couples, in contrast to the anionic redox couple of I(-)/I(3)(-). The increase of the lifetimes with Li(+) was interpreted with the decrease of the local concentration of Co(III) near the surface of TiO(2). The addition of 4-tert-butylpyridine (tBP) with the presence of Li(+) increased J(sc) significantly. The observed increase of the electron lifetime by tBP could not explain the large increase of J(sc), implying that tBP facilitates the charge transfer from Co(II)(L) to dye cation, with the association of the change of the reorganization energy between Co(II) and Co(III).

Role of electrolytes on charge recombination in dye-sensitized TiO(2) solar cell (1): the case of solar cells using the I(-)/I(3)(-) redox couple.

Performance of dye-sensitized solar cells (DSCs) was investigated depending on the compositions of the electrolyte, i.e., the electrolyte with a different cation such as Li(+), tetra-n-butylammonium (TBA(+)), or 1,2-dimethyl-3-propylimidazolium (DMPIm(+)) in various concentrations, with and without 4-tert-butylpyridine (tBP), and with various concentrations of the I(-)/I(3)(-) redox couple. Current-voltage characteristics, electron lifetime, and electron diffusion coefficient were measured to clarify the effects of the constituents in the electrolyte on the charge recombination kinetics in the DSCs. Shorter lifetimes were found for the DSCs employing adsorptive cations of Li(+) and DMPIm(+) than for a less-adsorptive cation of TBA(+). On the other hand, the lifetimes were not influenced by the concentrations of the cations in the solutions. Under light irradiation, open-circuit voltages of DSCs decreased in the order of TBA(+)> DMPIm(+) > Li(+), and also decreased with the increase of [Li(+)]. The decreases of open-circuit voltage (V(oc)) were attributed to the positive shift of the TiO(2) conduction band potential (CBP) by the surface adsorption of DMPIm(+) and Li(+). These results suggest that the difference of the free energies between that of the electrons in the TiO(2) and of I(3)(-) has little influence on the electron lifetimes in the DSCs. The shorter lifetime with the adsorptive cations was interpreted with the thickness of the electrical double layer formed by the cations, and the concentration of I(3)(-) in the layer, i.e., TBA(+) formed thicker double layer resulting in lower concentration of I(3)(-) on the surface of the TiO(2). The addition of 4-tert-butylpyridine (tBP) in the presence of Li(+) or TBA(+) showed no significant influence on the lifetime. The increase of V(oc) by the addition of tBP into the electrolyte containing Li(+) and the I(-)/I(3)(-) redox couple was mainly attributed to the shift of the CBP back to the negative potential by reducing the amount of adsorbed Li cations.

Ionic liquid crystal as a hole transport layer of dye-sensitized solar cells.

Use of a new ionic liquid crystal, 1-dodecyl-3-methylimidazolium iodide, and iodine as an electrolyte of dye-sensitized solar cells leads to a high short circuit photocurrent density and a high light-to-electricity conversion efficiency, due to a self-assembled structure of the imidazolium cations, resulting in high conductivity of the electrolyte.

Changes in EEG and autonomic nervous activity during meditation and their association with personality traits.

Meditation is the attainment of a restful yet fully alert physical and mental state practiced by many as a self-regulatory approach to emotion management, but the psychophysiological properties and personality traits that characterize this meditative state have not been adequately studied. We quantitatively analyzed changes in psychophysiological parameters during Zen meditation in 20 normal adults, and evaluated the results in association with personality traits assessed by Cloninger's Temperament and Character Inventory (TCI). During meditation, increases were observed in fast theta power and slow alpha power on EEG predominantly in the frontal area, whereas an increase in the normalized unit of high-frequency (nuHF) power (as a parasympathetic index) and decreases in the normalized unit of low-frequency (nuLF) power and LF/HF (as sympathetic indices) were observed through analyses of heart rate variability. We analyzed the possible correlations among these changes in terms of the percent change during meditation using the control condition as the baseline. The percent change in slow alpha EEG power in the frontal area, reflecting enhanced internalized attention, was negatively correlated with that in nuLF as well as in LF/HF and was positively correlated with the novelty seeking score (which has been suggested to be associated with dopaminergic activity). The percent change in fast theta power in the frontal area, reflecting enhanced mindfulness, was positively correlated with that in nuHF and also with the harm avoidance score (which has been suggested to be associated with serotonergic activity). These results suggest that internalized attention and mindfulness as two major core factors of behaviors of mind during meditation are characterized by different combinations of psychophysiological properties and personality traits.

Quantitative evaluation of age-related white matter microstructural changes on MRI by multifractal analysis.

Multifractal analysis has been applied to evaluate biological tissues, which are composed of complex structures. We carried out multifractal analyses in a group of healthy young and elderly subjects to examine age-related white matter microstructural changes on T2-weighted MR images without any visible abnormal intensity, and to correlate such changes with age-related cognitive decline. Comparison between the two age groups showed that Deltaalpha (established as the most suitable index of heterogeneity in our previous report) in the frontal region was significantly higher in the elderly group, but no significant group difference was found in Deltaalpha in the parieto-occipital region. The Trail-Making Test score (a measure of executive dysfunction) was significantly higher in the elderly group. In the elderly group, the Trail-Making Test score was positively correlated with Deltaalpha in the frontal region, but not in the parieto-occipital region. These results suggest that microstructural changes in the white matter preferentially occur in the frontal region with normal aging, and these changes are associated with executive cognitive decline reflective of frontal-subcortical dysfunction.

[Effect of gamma-aminobutyric acid-rich germinated brown rice on indexes of life-style related diseases].

We examined the effects of gamma-aminobutyric acid-rich germinated brown rice (germinated brown rice) on principal indexes of life-style related diseases in 67 volunteers aged 71 +/- 8. They were divided into two groups; germinated brown rice group, which had an equal amount of the germinated brown rice to polished rice for 11 to 13 months, and control group, which had polished rice alone for the same period. Differences of indexes before and after the examination between the two groups were compared. Significant increases in body fat ratio, hemoglobin A1c and mean red cell volume and a significant decrease in mean red cell hemoglobin concentration were observed in the germinated brown rice group. However, there was no difference of changes in body mass index, blood pressure, serum lipid, hepatic and renal functions, bone metabolic markers, bone density, depression score, red blood cell counts, hemoglobin, hematocrit, and homeostasis model assessment of insulin resistance between the two groups. These findings suggested that germinated brown rice might not improve glucose metabolism.

Different mechanisms of hypoxic injury on white matter and gray matter as revealed by dynamic changes in glucose metabolism in rats.

Fresh rat brain slices were incubated with [18F]2-fluoro-2-deoxy-D-glucose ([18F]FDG) in oxygenated Krebs-Ringer solution at 36 degrees C, and the fractional rate constant (=k3*) of [18F]FDG proportional to the cerebral glucose metabolic rate in white matter and gray matter was investigated with positron autoradiography. In both white matter and gray matter, the k3* value with > or = 20 min hypoxia was markedly lower than the unloaded control value, indicating irreversible hypoxic injury. Next, the neuroprotective effect against hypoxia induced by the addition of an N-methyl-D-aspartate receptor antagonist or a free radical scavenger was assessed by determining whether a decrease in the k3* value after hypoxia loading was prevented. In gray matter, both agents exhibited a neuroprotective effect against 20 min hypoxia. In white matter, however, only the free radical scavenger was effective. These results suggest a similarity in the degree of vulnerability to hypoxia between white matter and gray matter as well as a difference in the developmental mechanism of hypoxic injury, i.e. the involvement of both glutamate and free radicals in gray matter, and the more selective involvement of free radicals in white matter.

Audio-visual integration in temporal perception.

In situations of audio-visual interaction, research has generally found that audition prevails over vision in temporal perception, while vision is dominant over audition for spatial perception. Modality appropriateness to a given task generally determines the direction of this inter-modality effect. However, we found a reverse effect in some situations where a change in the frequency of visual stimuli was associated with a perceived change in the frequency of auditory stimuli. In our experiment, 12 participants were asked to judge the change in the frequency of visual and auditory stimuli using a visual flicker and auditory flutter stimuli. In some conditions either the auditory or the visual information was ambiguous. In addition to confirming the expected finding that a change in the frequency of the auditory stimuli induced a perceived change in the frequency of the visual stimuli, we found a new phenomenon. When ambiguous auditory temporal cues were presented, the change in the frequency of the visual stimuli was associated with a perceived change in the frequency of the auditory stimuli. This suggests that cross-modal asymmetry effects are influenced by the reliability of visual and auditory information as well as modality appropriateness.

Glyceraldehyde 3-phosphate dehydrogenase and endothelin-1 immunoreactivity is associated with cerebral white matter damage in dentatorubral-pallidoluysian atrophy.

DRPLA is a rare neurodegenerative disorder caused by CAG triplet elongation on chromosome 12p. In addition to neurodegeneration of both the dentatorubral and pallidoluysian systems, there is cerebral white matter damage, especially in older cases. Intracellular accumulation of DRPLA protein is widespread in the central nervous system, and DRPLA protein has been shown to immobilize glyceraldehyde 3-phosphate dehydrogenase (GAPDH), which regulates glycolysis and controls mRNA of tissue-type plasminogen activator (tPA) in tissue restoration. However, little is known about the pathogenesis regarding the formation of cerebral white matter damage in DRPLA. Therefore, the pathology of this damage was investigated by examining markers of glycolysis and related processes. Nine clinically and pathologically confirmed DRPLA cases were used in the present study. CAG triplet elongation on chromosome 12p was confirmed in all cases where tissue was available for genotyping (seven cases). PAS and immunohistochemistry with antibodies to GFAP, GAPDH and endothelin-1 were used to demonstrate astrocytosis. The polysaccharides storage state with PAS-positive astrocytes was detected in seven cases. GAPDH- and endothelin-1-positive endothelium and astrocytes were observed in two cases with GFAP-positivity. Based on the biochemical process together with the present results, GAPDH and endothelin-1 immunoreactivity is associated with this damage and the mismetabolism of polysaccharides caused by CAG triplet elongation on chromosome 12p may contribute to the formation of the cerebral white matter damage in DRPLA.

Sequential EEG analysis during intermittent photic stimulation in never-medicated patients with schizophrenia.

To investigate sequential changes in electroencephalograms (EEGs) during intermittent photic stimulation (IPS), quantitative EEG analysis for the alpha band (related to stimulus frequency) was performed in 18 drug-naïve schizophrenic patients and 18 sex- and age-matched control subjects. IPS consisted of white flickers at 10 flashes/sec, lasting 10 sec and repeated six times at 10 sec intervals. The differences between the two groups were assessed for two conditions (i.e., during photic stimulation (PS) and inter-PS). During stimulus, the absolute 9-11 Hz band power of the patient group was higher at the posterior than that at the anterior sites throughout the 10 sec periods. However, this difference between sites was not seen during the first 1 sec in the control group. During the nonstimulus period, posterior dominance of 9-11 Hz band power was prominent in the patient group throughout. In the control group, however, this difference was minor, especially during the latter half of the period. These results suggest that schizophrenic patients show fewer changes in posterior alpha activity during both stimulus and nonstimulus, and this continuity of posterior dominance may reflect hyperarousal, which counteracts any decrease in vigilance throughout the IPS.
