Prevention of hepatic oxidative injury by Xiao-Chen-Chi-Tang in mice.

The three purgative Cheng-Chi-Tang decoctions (CCTDs) including Ta-Cheng-Chi-Tang (TCCT), Xiao-Chen-Chi-Tang (XCCT), and Tiao-Wei-Chen-Chi-Tang (TWCCT) are used for treating gastrointestinal disorders, including liver diseases in traditional Chinese medicine. However, the underlying mechanisms as liver disease remedies are far from fully clarified. The objective of the study is to investigate and compare the antioxidant activity of the three purgative CCTDs in order to delineate their hepatic protective potential and mechanism. Antioxidant activity measured with the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging test indicated XCCT as the most potent preparation (IC(50) 8.94 microg/ml). In tert-butylhydroperoxide (TBH, 50mM)-induced lipid peroxidation in ICR mice liver homogenates, XCCT also showed stronger and dose-dependent inhibitory activity against TBH-induced malondialdehyde (MDA, a marker of lipid peroxidation) production (IC(50) 53.66 microg/ml). In addition, XCCT showed dose-dependent protective effect against TBH-induced cytotoxicity in normal human Chung liver cells Furthermore, in carbon tetrachloride (CCl(4))-induced acute liver injury model, mice pretreated with 0.2g/kg and 0.4 g/kg of XCCT extracts showed a decrease of 59.8 and 43.1% in serum glutamic oxaloactetic transaminase (GOT) level, 51.4 and 52% in glutamic pyruvate transaminase (GPT) level, along with a reduction of 31 and 15% in MDA level, respectively, similar to the effects exerted by silymarin. XCCT pretreated mice also showed milder necrotic changes in the microscopic picture of the liver. The results suggest that XCCT has significant antioxidant activity and hepatic protection potential.

MRI of alveolar soft-part sarcoma.

Alveolar soft-part sarcoma (ASPS) is rare. We present a case of a 30-year-old woman with a 6-month history of a palpable mass in her left thigh. MRI showed an 8-cm mass and a satellite nodule at the left gluteus muscle. The main tumor exhibited an isointense signal on T1-weighted images and high-signal-intensity areas with low-signal-intensity scanty solid components on T2-weighted images. MR angiography showed dilated and tortuous veins around the tumor. Histologic findings were compatible with ASPS.

Gastrointestinal stromal tumor of the prostate: a case report and literature review.

We report a case of gastrointestinal stromal tumor (GIST) of the prostate in a 75-year-old man with dysuria and urinary retention. Digital rectal examination revealed a markedly enlarged prostate. The serum level of prostate-specific antigen was 0.2 ng/dL. Imaging studies showed an expanded prostate, measuring 6.7 x 5.6 x 5.5 cm, with heterogeneity in contrast enhancement. No metastatic disease was found. The pathologic diagnosis of prostatic GIST was made based on characteristic morphological features, immunoprofiles, and molecular analysis. The possibility of secondary involvement by a rectal GIST was excluded by radiological and intraoperative findings. To our knowledge, this is the second case of a primary GIST of the prostate reported in the literature.

Protective effect of propolis ethanol extract on ethanol-induced renal toxicity: an in-vivo study.

Acute p.o. administration of absolute ethanol (10 ml/kg) to fasted mice would produce extensive renal failure. Pretreatment with p.o. administration of propolis ethanol extract (PEE) could prevent such renal failure effectively and dose dependently. This renal protective effect of PEE may be contributed, at least in part, to its antioxidative activity. The maximal antioxidative effect against absolute ethanol (AE)-induced renal failure could be observed 1 hour after PEE administration. In order to further investigate the renal protective mechanism of PEE, lipid peroxidation and superoxide scavenging activity were conducted in vivo. PEE exhibited dose-dependent antioxidative effects on lipid peroxidation in mice renal homogenate. Results indicated that mice with acute renal failure have higher malonic dialdehyde (MDA) levels compared with those in PEE administered mice. It was concluded that the renal protective mechanism of PEE could be contributed, at least in part, to its prominent superoxide scavenging effect; hence, it could protect, indirectly, the kidney from superoxide-induced renal damages.

Enhanced expression of transforming growth factor-beta1 in inflammatory cells and secretory granules in Paneth cells in the small intestine of mice infected with Toxocara canis.

The small intestine is the initial organ which Toxocara canis larvae invade. Information on intestinal pathological changes associated with transforming growth factor-beta1 (TGF-beta1) and secretory granules (SG) in Paneth cells (PCs) caused by T. canis is unclear. Mice orally inoculated with 250 T. canis infective eggs were evaluated by pathological and immunohistochemical assessments with a 294-day investigation. Pathologically, the inflammatory reactions with or without trapped larvae in the submucosa were observed only within the first 28 days post-infection (DPI), with inflammatory injury ranging from severe during 2 DPI to mild between 7 and 28 DPI. The crypts of Leiberkuhn were major larval penetration sites. Enhanced expression of SG in PCs appeared earlier than those of TGF-beta1 in infiltrating cells. The significance of both effectors might be related to the host's defense against larval invasion in the intestinal phase of toxocaral infection.

Antioxidative natural product protect against econazole-induced liver injuries.

The study objective of this research is in order to investigate the hepatoprotective and therapeutic effects of propolis ethanol extract (PEE) on acute econazole-induced liver injury. Positive control of various concentrations of PEE on liver function and the dose-response relationship of liver injury induced by various doses of econazole were firstly observed from biochemical assay of serum level of aspartate transaminase (SGOT) and serum alanine transaminase (SGPT) and histopathological microscopic examination. The hepatoprotective effects of various concentration of PEE on liver damage induced by hepatotoxic dose (300 mg/kg) of econazole were observed by the obvious decrement of SGOT and SGPT level and further confirmed by hepatohistological microscopic examination. The inhibitory effects of PEE on FeCl(2)-induced (in vitro) or econazole-induced (in vivo) lipid peroxidation were investigated from the measurement of the formed malonic dialdehyde (MDA) level in the rat liver homogenate. The IC(50) (microM) of various concentrations of PEE in the superoxide scavenging activity in econazole (300 mg/kg)-damaged rat liver homogenate were assessed by cytochrome c reduction method and compared with that of (+)-alpha-tocopherol. It could be postulated that the hepatoprotective effect of PEE may be, at least in part, due to their inhibitory ability on membrane lipid peroxidation and free radical formation or due to their free radical scavenging ability.

Milium-like syringoma: a case study on histogenesis.

BACKGROUND: Milium-like syringoma is a variant of syringoma first described in 1987. Only few cases have been reported in the literature. It may be misleading clinically, and its histogenesis has not been clarified. CASE REPORT: We present a case of periorbital milium-like syringoma, with studies on the histopathologic, histochemical, and immunohistochemical features. RESULTS: Histology showed a large keratin-filled cyst in the upper portion of the lesion approximating the epidermis. Serial sections revealed that the cyst connected with the underlying syringomatous epithelial strands. Melanin was absent in the wall of the cyst, as demonstrated by Fontana-Masson stain. Cytokeratin 7 was expressed neither in the milia nor in the solid epithelial parts. Carcinoembryonic antigen (CEA) reactivity was seen in the luminal cells of the keratinous cysts. However, in the largest keratin-filled cyst clinically suggesting a milium, only the lower half of the cyst was positive for CEA. These results proved that the milia were part of syringoma with eccrine duct differentiation. Fusion of the upper half of the largest cyst with the epidermis may explain the absence of CEA positivity in this part, analogous with eccrine duct milia. Review of the literature indicated that this variant of syringoma occurs more often in Asians. CONCLUSION: We present evidence, and propose the histogenesis of milium-like syringoma, that it is a variant of syringoma with a prominent cystic component showing features of eccrine duct milia. Recognition of its nature is of therapeutic significance. Further studies are required to verify its clinical characteristics as compared with ordinary syringomas.

Infectivity and pathogenicity of 14-month-cultured embryonated eggs of Toxocara canis in mice.

Infectivity and pathogenicity to mice of embryonated eggs of Toxocara canis, that had been maintained in 2% formalin for 14 months at 4 degrees C, were evaluated by immunological and pathological assessment at 1, 4, 8, 12, 16, 20, 24, 28, 42 and 67 weeks post-infection (WPI). On each date, three infected mice and two age-matched uninfected mice were sacrificed for serum collection and histological processing of the liver, lungs, musculature, and brain. Infectivity assessment by enzyme-linked immunosorbent assay (ELISA) revealed that the overall immunological pattern of infected mice tended to be towards the Th2 type response. Serum IgG1 antibody titers in infected mice were significantly higher than that of the uninfected control mice throughout the trial (P<0.05). On the other hand, no significant difference in titers of IgG3 antibody, an indicator for the Th1 type response, was observed between the infected and control mice, except at eight WPI (P<0.05). Pathogenicity was assessed semiquantitatively by comparing the mean number or diameter of inflammatory foci as well as histopathological changes in the liver, musculature, brain, or lungs of the infected mice and the control mice. Each hematoxylin and eosin (H&E) stained tissue section slide was examined under 100x magnification and 15 random fields were counted. Degree of inflammatory injury among the four organs was scored and categorized into four levels: normal (0), mild (1+), moderate (2+), and severe (3+). An index of inflammatory injury (mean score of experimental group/mean score of 10 control groups of 20 uninfected mice) of 2-3 is considered as moderate to severe, 1-2 as mild to moderate, and 0-1 as normal to mild. Histopathological changes were moderate to severe in the liver and lungs, mild to moderate in the musculature, and only normal to mild in the brain throughout the trial. It is noteworthy that apocrine-like change in epithelial cells of the bile duct was observed in most of the infected mice from eight WPI onward. Furthermore, larvae trapped by organized granulomas were found in soft tissue near the musculature at 12, 20, and 28 WPI. Altogether, not only were the infectivity and pathogenicity of the 14-month-cultured T. canis embryonated eggs retained, the hatched larvae were also capable of eliciting some special pathological changes in the murine host.

F-18 fluorodeoxyglucose positron emission tomography in pulmonary cryptococcoma.

Cryptococcosis is not uncommon. Potential interpretation pitfalls should be kept in mind when fluorodeoxyglucose (FDG) positron emission tomography (PET) is used for differentiating pulmonary nodules and for discriminating infection from malignancy, especially in areas where the prevalence of granulomatous infection is high and in immunocompromised patients. In this case, a nodular mass was shown on chest radiography and computed tomographic (CT) scanning. A consolidated infection or a bronchioloalveolar carcinoma was suspected because the CT scan showed air bronchograms within the mass and another perihilar infiltration. The FDG PET scan clearly delineated the lesion and had intermediately high glucose uptake (standard uptake value, 3.8-4.0), which led to the exclusion of the possibility of bronchioloalveolar carcinoma because most of these tumors had normal or mildly increased FDG accumulation. Cryptococcoma was finally diagnosed. Hence, the CT scan and FDG PET played complementary roles in the differential diagnosis of this nodular mass.

Hepatoprotective effects of Arctium lappa Linne on liver injuries induced by chronic ethanol consumption and potentiated by carbon tetrachloride.

Arctium lappa Linne (burdock) is a perennial herb which is popularly cultivated as a vegetable. In order to evaluate its hepatoprotective effects, a group of rats (n = 10) was fed a liquid ethanol diet (4 g of absolute ethanol/ 80 ml of liquid basal diet) for 28 days and another group (n = 10) received a single intraperitoneal injection of 0.5 ml/kg carbon tetrachloride (CCl(4)) in order to potentiate the liver damage on the 21st day (1 day before the beginning of A. lappa treatment). Control group rats were given a liquid basal diet which did not contain absolute ethanol. When 300 mg/kg A. lappa was administered orally 3 times per day in both the 1-day and 7-day treatment groups, some biochemical and histopathological parameters were significantly altered, both in the ethanol group and the groups receiving ethanol supplemented with CCl(4). A. lappa significantly improved various pathological and biochemical parameters which were worsened by ethanol plus CCl(4)-induced liver damage, such as the ethanol plus CCl(4)-induced decreases in total cytochrome P-450 content and NADPH-cytochrome c reductase activity, increases in serum triglyceride levels and lipid peroxidation (the deleterious peroxidative and toxic malondialdehyde metabolite may be produced in quantity) and elevation of serum transaminase levels. It could even restore the glutathione content and affect the histopathological lesions. These results tended to imply that the hepatotoxicity induced by ethanol and potentiated by CCl(4) could be alleviated with 1 and 7 days of A. lappa treatment. The hepatoprotective mechanism of A. lappa could be attributed, at least in part, to its antioxidative activity, which decreases the oxidative stress of hepatocytes, or to other unknown protective mechanism(s).
