Positional control of encapsulated atoms inside a fullerene cage by exohedral addition.

The exohedrally functionalized derivative of endohedral metallofullerene, Ce2@C80(Mes2SiCH2SiMes2), was successfully synthesized and fully characterized. X-ray crystallographic and NMR spectroscopic analyses reveal that the free random motion of two metal atoms in Ce2@C80 is controlled inside the cage by exohedral chemical functionalization.

Large-scale separation of metallic and semiconducting single-walled carbon nanotubes.

In the applications of single-walled carbon nanotubes (SWNTs), it is extremely important to separate semiconducting and metallic SWNTs. Although several methods have been reported for the separation, only low yields have been achieved at great expense. We show a separation method involving a dispersion-centrifugation process in a tetrahydrofuran solution of amine, which makes metallic SWNTs highly concentrated to 87% in a simple way.

Chemical reactivity of sc3n @ c80 and la2 @ c80.

Sc3N@C80 has a lower thermal reactivity than La2@C80, although Sc3N@C80 has the same carbon cage (Ih) and oxidation state (C806-) as La2@C80. This result is attributed to the difference in the energy level and distribution of LUMO between Sc3N@C80 and La2@C80.

Novel 4-amino-furo[2,3-d]pyrimidines as Tie-2 and VEGFR2 dual inhibitors.

A novel class of furo[2,3-d]pyrimidines has been discovered as potent dual inhibitors of Tie-2 and VEGFR2 receptor tyrosine kinases (TK) and a diarylurea moiety at 5-position shows remarkably enhanced activity against both enzymes. One of the most active compounds, 4-amino-3-(4-((2-fluoro-5-(trifluoromethyl)phenyl)amino-carbonylamino)phenyl)-2-(4-methoxyphenyl)furo[2,3-d]pyrimidine (7k) is <3 nM on both TK receptors and the activity is rationalized based on the X-ray crystal structure.

Synthesis and characterization of exohedrally silylated M@C82 (M = Y and La).

The silylation of endohedral mono-metallofullerenes (Y@C(82) and La@C(82)) and isolation of the corresponding adducts by HPLC separation have been accomplished. The redox properties of the silylated mono-metallofullerene were first clarified by CV and DPV measurements, indicating that the bis-silylated mono-metallofullerenes have lower oxidation and higher reduction potentials than the parent mono-metallofullerenes. These results reveal that bis-silylation is very effective for producing the electronegatively mono-metallofullerene derivatives as well as empty fullerenes.

Chemical reactivities of the cation and anion of M@C82 (M = Y, La, and Ce).

The chemical reduction and oxidation of M@C82 (M = Y, La, and Ce) afford the corresponding anion and cation, respectively, which show unique and interesting chemical reactivities. It is found that the successful reversible gain or loss of electrons by ionization is useful for controlling the stability and reactivity of M@C82 toward both nucleophiles and electrophiles.

Cell cycle dependence of elicitor-induced signal transduction in tobacco BY-2 cells.

The molecular links between the cell cycle and defense responses in plants are largely unknown. Using synchronized tobacco BY-2 cells, we analyzed the cell cycle dependence of elicitor-induced defense responses. In synchronized cultured apoaequorin-expressing cells, the increase in cytosolic free Ca2+ induced by a proteinaceous elicitor, cryptogein, was greatly suppressed during the G2 and M phases in comparison with G1 or S phases. Treatment with cryptogein during the G1 or S phases also induced biphasic (rapid/transient and slow/prolonged) responses in activation of mitogen-activated protein kinases (MAPKs) and production of reactive oxygen species (ROS). In contrast, elicitor treatment during the G2 or M phases induced only a rapid and transient phase of MAPK activation and ROS production. Their slow and prolonged phases as well as expression of defense-related genes, cell cycle arrest and cell death were induced only after the cell cycle progressed to the G1 phase; removal of the elicitor before the start of the G1 phase inhibited these responses. These results suggest that although cryptogein recognition occurred at all phases of the cell cycle, the recognition during the S or G1 phases, but not at the G2 or M phases, induces the prolonged activation of MAPKs and the prolonged production of ROS, followed by cell cycle arrest, accumulation of defense-related gene transcripts and cell death. Elicitor signal transduction depends on the cell cycle and is regulated differently at each phase.

Simultaneous perturbation learning rule for recurrent neural networks and its FPGA implementation.

Recurrent neural networks have interesting properties and can handle dynamic information processing unlike ordinary feedforward neural networks. However, they are generally difficult to use because there is no convenient learning scheme. In this paper, a recursive learning scheme for recurrent neural networks using the simultaneous perturbation method is described. The detailed procedure of the scheme for recurrent neural networks is explained. Unlike ordinary correlation learning, this method is applicable to analog learning and the learning of oscillatory solutions of recurrent neural networks. Moreover, as a typical example of recurrent neural networks, we consider the hardware implementation of Hopfield neural networks using a field-programmable gate array (FPGA). The details of the implementation are described. Two examples of a Hopfield neural network system for analog and oscillatory targets are shown. These results show that the learning scheme proposed here is feasible.

Effect of catechins on mutagenesis of Salmonella typhimurium TA 102 elicited by tert-butyl hydroperoxide (t-BuOOH).

Green tea, one of the most popular beverages consumed in Asian countries, has been reported to possess anticarcinogenic and antimutagenic properties. The aim in this study is to test the radical scavenging effect of catechins and caffeine, which were major components of green tea, and if they really prevent oxygen radical-induced mutagenesis. We used TA102 strain of Salmonella typhimurium which is sensitive to hydroxyl radical in the Ames mutation assay. We found that caffeine did not show any effects on mutagenesis in this system, but catechin significantly reduced mutagenesis or genotoxicity caused by hydroxyl radical. This radical-scavenging action of catechins may indeed contribute to the anticarcinogenic activity of green tea as has been proposed.

Prewashing with acidified sodium chlorite reduces pathogenic bacteria in lightly fermented Chinese cabbage.

Efficacy of prewashing with acidified sodium chlorite (ASC) for the sanitation of lightly fermented Chinese cabbage was evaluated. The population of the natural microflora on the cabbage leaves was reduced about 2.0 log CFU/g just after washing with ASC, a significant reduction compared with the control distilled water wash (P < or = 0.05). In the control experiment, viable aerobic bacteria increased gradually when incubated at 10 degrees C; however, ASC-washed cabbage maintained a lower microbial concentration. The treatment of Chinese cabbage with ASC reduced the population of artificially inoculated Escherichia coli O157:H7, Salmonella Enteritidis, Staphylococcus aureus, and Listeria monocytogenes by 2.4 log CFU/g. The sanitation efficacy of ASC was 1.6 log CFU/g higher than that of distilled water washing. The viable cell counts of all pathogenic bacteria tested remained constant during 8 days of storage at 10 degrees C for both washing treatments, with the exception of L. monocytogenes, whose viable cell counts increased gradually with time for both treatments. No significant differences in color, odor, taste, and texture in raw leaves were observed after the ASC wash compared with after the distilled water wash. These results indicate that prewashing with ASC could control bacterial growth in lightly fermented Chinese cabbage without changing the product quality.

Effect of components of green tea extracts, caffeine and catechins on hepatic drug metabolizing enzyme activities and mutagenic transformation of carcinogens.

Green tea contains catechins and caffeine as major constituents. Treatment of rats with green tea (2.5% w/v) significantly increased 7-ethoxycoumarin O-deethylase (7-ECOD), caffeine N-1 demethylase (CN1D) and UDP-glucuronyltransferase (UGT) activities. Treatment with caffeine similarly activated CYP1A2 and related monooxygenases as well as UGT, while treatment with catechins induced UGT activity but not 7-ECOD or CN1D activity. Numbers of benzo[a]pyrene (BP)-induced revertant colonies in an Ames test (mutation assay) with S. typhimurium TA98 as the test strain were markedly larger when BP was preincubated with the liver S-9 (9000 x g supernatant of liver homogenate) from green tea-treated rats than when preincubated with that from control rats. In a modified Ames assay system in which UGT was activated by the addition of UDP-glucuronic acid to the preincubation mixture, the numbers of revertant colonies in the assay using liver S-9 from green tea-treated rats decreased to a similar level to that in the assay using S-9 from controls. The acceleration of two enzymatic reactions may contribute to the rapid elimination of BP; the first step, the formation of a metabolic intermediate (which is mutagenic) by CYP1A2 and the second, the conjugation of active metabolic intermediates by UGT. We speculated that green tea can reduce the amount of time carcinogens reside in the body and the chance that body tissues will be exposed to active metabolites of carcinogens thorough rapid elimination due to the simultaneous induction of CYP1A2 and UGT activities.

Isolation, characterization, and theoretical study of La2@C78.

A new metallofullerene, La2@C78, has been synthesized by DC arc discharge method, isolated by high-performance liquid chromatography, and characterized by laser desorption time-of-flight mass spectrometry, UV-vis-NIR absorption, differential pulse voltammetry, 13C NMR spectroscopy, and theoretical calculations. The La2@C78/CS2 solution is dark violet and presents several characteristic absorption features at 647, 561, 533, and 386 nm, with an onset around 1000 nm. With respect to empty D3-C78, the capability of La2@C78 as an electron acceptor or donor is stronger. Addition of 1,1,2,2-tetrakis(2,4,6-trimethylphenyl)-1,2-disirane to La2@C78 photochemically, as well as thermally, affords bis- and mono-adducts. Theoretical studies and 13C NMR spectroscopic analysis of La2@C78 indicate that it possesses a D3h-C78 cage (78:5).

4-Acylamino-6-arylfuro[2,3-d]pyrimidines: potent and selective glycogen synthase kinase-3 inhibitors.

Modeling studies of a furo[2,3-d]pyrimidine GSK-3 hit compound 1 superimposed onto the X-ray crystal structure of a legacy pyrazolo[3,4-c]pyridazine GSK-3 inhibitor 2 led to the identification of 4-acylamino-6-arylfuro[2,3-d]pyrimidine template 3. Synthesis of analogues based on template 3 has resulted in a number of potent and selective GSK-3beta inhibitors. The most potent and selective compound was the m-pyridyl analogue 24.
