[Treatment of ischemia in lower extremities by primary arterization in situ of Vana saphena magna].

In order to summarize the experience in the treatment of ischemic necrosis of lower extremities resulted from thrombotic occluded angittis, 15 cases were reported, which were treated by primary arterization in situ of V. Saphena magna. With a period of follow-up, 4-26 months on the average, it was found that symptoms in 14 cases were much allayed obviously, except 1 case with little relief. It suggested that primary arterization in situ of V. saphena magna could improve the circulation of the ischemic extremity rapidly without any influence of venous reflux.

Phosphatidylethanolamine N-methyltransferase from liver.

Phosphatidylethanolamine N-methyltransferase (PEMT) converts phosphatidylethanolamine to phosphatidylcholine. Most PEMT activity (PEMT1) is associated with endoplasmic reticulum. A second form of the enzyme (PEMT2) has been localized to the mitochondria-associated membrane. PEMT2 is a 22.5-kDa protein that has been purified from rat liver. The rat liver PEMT2 cDNA and the murine PEMT gene have been cloned and characterized. The PEMT gene encodes both forms of the enzyme. Deletion of the PEMT gene eliminates all activity in liver that converts phosphatidylethanolamine to phosphatidylcholine. The activity of PEMT is regulated by supply of the substrates, phosphatidylethanolamine and S-adenosylmethionine, and by the product S-adenosylhomocysteine. The expression of the gene is regulated during development and by the supply of choline in the diet. There is reciprocal regulation of the Kennedy pathway for phosphatidylcholine biosynthesis (via CDP-choline) and phosphatidylethanolamine N-methyltransferase. Several experimental approaches suggest that this enzyme might play a role in regulation of hepatocyte growth and cell division.

Induction of hepatocyte proliferation after partial hepatectomy is accompanied by a markedly reduced expression of phosphatidylethanolamine N-methyltransferase-2.

Expression of phosphatidylethanolamine N-methyltransferase (PEMT)-2 in rat hepatoma cells caused an increase in the time for cell division from 18 to 50 h [Cui et al. (1994) J. Biol. Chem. 269, 24531-24533]. We investigated whether or not a similar inverse relationship might exist for liver proliferation in vivo. Thus, partial hepatectomized rats were used to investigate the expression of PEMT2 during liver regeneration. Enhanced biosynthesis of phosphatidylcholine after partial hepatectomy was due to increased activity and amount of CTP:phosphocholine cytidylyltransferase. On the other hand the total activity of PEMT was markedly decreased during the first days of rat liver regeneration. Maximal decrease of total PEMT activity (45%) and loss of PEMT2 protein (90%) coincided with maximal DNA synthesis and CTP:phosphocholine cytidylyltransferase activity 24 h after partial hepatectomy in both male and female rats. Supplementing dietary choline in the diets of female rats shifted this pattern from 24 h to 36 h after partial hepatectomy, whereas the pattern in male rats was not affected. Northern blot studies showed that the amount of PEMT2 mRNA was decreased accordingly, suggesting regulation of the amount and activity of PEMT2 at a pre-translational level. Thus, our data show a reciprocal regulation of CTP:phosphocholine cytidylyltransferase and PEMT2 at the level of gene expression in regenerating rat liver. These results implicate PEMT2 in the regulation of hepatocyte cell growth in a physiologically relevant model.

Inverse correlation between expression of phosphatidylethanolamine N-methyltransferase-2 and growth rate of perinatal rat livers.

Our previous studies have implicated the liver-specific phosphatidylethanolamine N-methyltransferase-2 (PEMT2) in suppression of hepatocarcinoma proliferation (Cui et al. (1994) J. Biol. Chem. 269, 24531-24533). It was not known if this phenomenon in cell culture had relevance to liver growth and PEMT2 expression in an intact animal. Hence, we investigated the relationship between normal proliferation of liver and the expression of PEMT2 during the perinatal period of developing rats. PEMT2 protein was completely absent, and PEMT activity was very low, in prenatal livers in which liver growth is rapid. At birth, a decrease of liver growth coincided with the rapid appearance in liver of a high level of PEMT2 protein that was sustained throughout adult life. Northern blots revealed that the postnatal expression of PEMT2 correlated with the level of its mRNA. Immunohistochemical staining of liver sections showed a distinctive pattern of PEMT2 expression at birth. A high level of PEMT2 was expressed in defined extranuclear regions of hepatocytes from newborn rats whereas the protein was dispersed in the extranuclear areas in adult hepatocytes. The inverse correlation between the rate of liver growth and PEMT2 expression together with other results suggest that this enzyme, or its product, is involved in control of normal liver proliferation.

Transient inactivation of phosphatidylethanolamine N-methyltransferase-2 and activation of cytidine triphosphate: phosphocholine cytidylyltransferase during non-neoplastic liver growth.

Phosphatidylethanolamine N-methyltransferase-2 (PEMT2) may contribute to the control of hepatocyte cell division, since its inactivation is associated with several types of liver proliferation including tumorigenesis [Cui, Houweling and Vance (1994) J. Biol. Chem. 269, 24531-24533]. To determine if the inactivation of PEMT2 was involved in non-neoplastic proliferation of hepatocytes, we studied the expression of this enzyme in a model of lead nitrate-induced liver proliferation in vivo in rats. A maximal decrease in PEMT activity (60%) and loss of PEMT2 protein (95%) coincided with maximal DNA synthesis and maximal cytidine triphosphate:phosphocholine cytidylyltransferase activity 36 h and 48 h after lead nitrate stimulation in male and female livers respectively. The decrease in expression of PEMT2 corresponded to a decrease in its mRNA. Compared with males, female rats exhibited a 12 h delay in the peak of DNA synthesis, in cytidylyltransferase activity and in the minimum of PEMT2 expression. Supplementation of the rats with dietary choline shifted the female pattern of PEMT2 inactivation, DNA synthesis and activation of cytidylyltransferase to 12 h earlier so that it was similar to the time frame of the expression of these activities in males. These results are consistent with the proposal that the inactivation of PEMT2 may have a role in the regulation of non-neoplastic growth of liver.

Estradiol stimulates growth hormone production in female goldfish.

The effects of estradiol (E2) on growth hormone (GH) production was investigated in gonad-intact female goldfish. It was first necessary to generate a specific antibody for use in immunocytochemistry, Western, and dot-blot analyses of GH production. To accomplish this, grass carp GH (gcGH) cDNA was cloned by the reverse transcription polymerase chain reaction (RT-PCR) and expressed in Echerichia coli and a specific polyclonal antibody to recombinant gcGH was generated in the rabbit. In Western blot, the anti-gcGH antibody specifically immunoreacted with recombinant gcGH, purified natural common carp GH, and with a single 21.5-kDa GH form from pituitary extracts of grass carp, common carp, goldfish, and zebrafish but not salmon, trout, or tilapia. Intraperitoneal injection of the recombinant gcGH enhanced the growth rates of juvenile common carp demonstrating biological activity of this GH preparation. Electron microscopic studies showed that the anti-gcGH-I antibody specifically reacted with GH localized in the secretory granules of the goldfish somatotroph. Using anti-gcGH-I in a dot-blot assay, it was found that in vivo implantation of solid silastic pellets containing E2, (100 micrograms/g body weight for 5 days) increased pituitary GH content by 150% in female goldfish. In a second, independent study employing a previously characterized anticommon carp GH antibody for radioimmunoassay, it was found that E2 increased pituitary GH content by 170% and serum GH levels by approximately 350%. The E2-induced hypersecretion of GH and increase in pituitary GH levels was not associated with changes in steady-state pituitary GH mRNA levels, suggesting that this sex steroid may enhance GH synthesis at the posttranscriptional or translational level. Previous observations indicate that GH can stimulate ovarian E2 production. The present results show that E2 can in turn stimulate GH production, indicating the existence of a novel pituitary GH-ovarian feedback system in goldfish.

First trial of home ECG and blood pressure telemonitoring system in Macau.

OBJECTIVE: To determine the feasibility of home monitoring of patients with cardiac disease or hypertension. METHODS: An improved home electrocardiographic and blood pressure telemonitoring system linked to a central workstation was tested in 10 patients in Macau for 3 months. RESULTS: The total number of connections was 1377. Of the automatic alarm connections, 32.5% were false positive, with the percentage of false positives ranging from 7.6 to 54.6 for different patients. Both patients and physicians found the system easy to use. CONCLUSIONS: Further investigation is required to match the number of patients with the system capacity. A more robust dysrhythmia detection algorithm is needed to reduce the number of false alarms. Nevertheless, the results were sufficiently good that the trial is being expanded.

A home electrocardiography and blood pressure telemonitoring system.

A home electrocardiography (ECG) and blood pressure telemonitoring system for cardiac patients was installed in the Macau region. The monitoring centre was established in the emergency unit at the Government Hospital of Macau. The first users were 10 cardiovascular patients selected by a physician. The average age of these users was 61 years (range 30-78). The results of a three-month trial showed that the system was easy to operate and technically reliable. It was found to be helpful for cardiac patients. The most significant problem during the trial was electrical noise from the ECG electrodes.

Studies on the transmission potential of surviving microfilaremias after basic control of filariasis.

After filariasis was basically controlled (the microfilarial rate was lower than 1%) in Henan Province in 1987, longitudinal observation of the disease has been carried out in all the province in order to study the regular pattern of growth and decline or the transmission potential of the disease. According to the distribution of filaria species and original microfilarial rate, 7 administrative villages in 7 counties were selected as surveillance sites. From 1988 to 1995, etiological and mosquito vector surveys were made continuously in all sites where no control measure was conducted. 10 surviving microfilaremic individuals became negative gradually over the first 6 years and no new microfilaremia was found. Since then, the microfilarial rate was zero. During the 8 years, 19 vector mosquitos were positive, with a total of 33 filarial larva. Culex pipiens pallens was the predominant mosquito species inside human dwelling in all sites. The man-biting rate of mosquitos for outdoor sleepers fluctuated greatly, the highest was 360.60 mosquitos per man per night and the lowest 7.20. The man-biting rate of mosquitos for sleepers inside mosquito-nets was approximately 1. The proportion of multiparous mosquitos also fluctuated greatly, the highest was 88.10% and the lowest 27.27%. According to the data described above, the man-biting rate of mosquitos which contained filaria L3 was less than 1 mosquito per man per transmission season. It is suggested that after the microfilarial rate was lower than 1%, the surviving microfilaremias became negative gradually in 3-5 years, and the transmission of the disease was blocked. Therefore, in the districts where filariasis was basically controlled, elimination of the disease was attainable.

Studies on the transmission potential of filariasis in controlled areas of Henan Province.

OBJECTIVE: To study the regular pattern of growth and declination or the transmission potential of filariasis after the disease was basically (the microfilarial rate was lower than 1%) in Henan Province in 1987. METHODS: According to the distribution of filaria species and original microfilarial rate, in 7 surveillance sites in 7 counties (cities) the etiology and mosquito vector surveys were carried out continuously during 1988-1995 and no control measurement for pathogen was taken. RESULTS: Ten residual microfilaremias became negative gradually in the first 6 years and no new microfilaremias occurred during 1988-1995. During 1993 to 1995, the microfilaremias rate of population in the sites was 0. The natural infection rates of filarial larvae in vector mosquito were 0.1%-0. During the 8 years, 15 vector mosquitos were positive with a total of 18 filarial larvae which were all of first or second-stage larvae. The C. pipiens pallens was the main vector, the second was A. sinensis, with a small number of C. fatigans and A. anthropophagus. The man-biting rates of mosquitoes for outdoor sleepers fluctuated greatly, the highest one was 360.6 mosquitoes/person per night and the lowest, 7.2. The man-biting rates of mosquitoes for sleepers inside mosquito-net was about 1 mosquito/person per night. The proportion of multiparous mosquitoes also fluctuated more greatly, the highest one was 88.1% and the lowest 27. 2% According to the data described above, the man-biting rate of vector mosquito which contained filarial L3 was 0. CONCLUSIONS: The results suggested that after the microfilarial rate was lower than 1%, the residual microfilaremias became negative gradually in 3-6 years, and the transmission of the disease was blocked. Therefore, in the areas where filariasis was basically controlled, elimination of the disease was attainable within sight.

[Method and significance of removal of dermal epithelial components in the study of composite skin].

UNLABELLED: In order to investigate the possible re-epithelialization originated from the dermal epithelial components after trypsinization of the allo-dermis, microscopic observation, tissue culture study and histological observation after allo-grafting were adopted. The number of the hair follicles in dermal layer was counted in the study process. The results showed that a few of hair follicles in the structure of allo-dermis remained after trypsin digestion for 80 minutes. After culturing and grafting of such trypsinized dermis, none of the newly formed epithelium was observed. CONCLUSION: 1. Trypsinization of allo-dermis eliminates the antigenic epithelial components of dermis preventing the possibility of re-epithelization; 2. Strictly controled time is required while digesting the allo-dermis with trypsin.

A genetic defect in phosphatidylcholine biosynthesis triggers apoptosis in Chinese hamster ovary cells.

We have investigated the cell death of a Chinese hamster ovary mutant (MT-58) with a thermo-sensitive CTP:phosphocholine cytidylyltransferase, the rate-limiting enzyme of the CDP-choline pathway for phosphatidylcholine biosynthesis (Esko, J. D., Wermuth, M. M., and Raetz, C. R. H. (1981) J. Biol. Chem. 256, 7388-7393). After MT-58 cells were shifted to the restrictive temperature of 40 degrees C, the cytidylyltransferase was inactivated immediately leading to a decrease in phosphatidylcholine biosynthesis and cell death. DNA content and number of cells in the S phase decreased significantly in the dying MT-58 cells according to flow cytometrical analyses. The fragmentation of genomic DNA was detected by DNA ladders in agarose gel and release of the prelabeled genomic DNA into cytosolic fractions 14 h after the temperature shift. The dying cells underwent a dramatic reduction of cellular volume while maintaining the membrane containment of cellular contents. These events indicated that the inactivation of cytidylyltransferase triggered apoptosis in Chinese hamster ovary cells. This is the first report that apoptosis was induced in cultured cells, not by an added agent, but by a mutation in phospholipid biosynthesis.

The multidrug resistance-associated protein (MRP) subfamily (Yrs1/Yor1) of Saccharomyces cerevisiae is important for the tolerance to a broad range of organic anions.

We have cloned and characterized a Saccharomyces cerevisiae gene YRS1 that complements the phenotype of the mutant sensitive to the anionic drug reveromycin A. The YRS1 gene, which is identical to the recently identified YOR1 gene, encodes a protein with extensive homology to the human multidrug resistance-associated protein (MRP) and the yeast cadmium factor (Ycf1). A chromosomal deletion of YRS1 lead to viable Deltayrs1 cells, which exhibited hypersensitivity to reveromycin A. Elevation of the YRS1 gene dosage in wild type cells conferred increased resistance to reveromycin A. By analyzing the effect of YRS1 disruption and overexpression it was demonstrated that Yrs1 is involved in the detoxification of a wide range of the organic anions that contain carboxyl group(s) but none of the other type of toxic compounds examined. Fluorescence-activated cell sorter analysis indicated the increased accumulation of the anionic fluorescent compound rhodamine B in Deltayrs1 cells. The expression of YRS1 was induced strikingly by reveromycin A. These results suggest that Yrs1 is a multispecific organic anion transporter important for tolerance against toxic environmental organic anions. Yrs1 had an overlapping specificity with Ycf1 in the resistance to cadmium.

Expression of phosphatidylethanolamine N-methyltransferase-2 is markedly enhanced in long term choline-deficient rats.

When rats are fed a choline-deficient (CD) diet, acute fatty liver develops along with other biochemical changes. However, when choline deficiency is prolonged, the growth rate of CD rats is similar to that of control rats fed a choline-supplemented diet. Furthermore, CD rats maintain their levels of choline-containing lipids, such as phosphatidylcholine, lysophosphatidylcholine, and sphingomyelin. The mechanism for this compensation in CD rats was investigated. We screened the major tissues for the activities of two important enzymes involved in the biosynthesis of phosphatidylcholine, CTP:phosphocholine cytidylyltransferase (CT) and phosphatidylethanolamine N-methyltransferase (PEMT). Only the livers of CD rats had higher specific enzyme activities of PEMT and CT than control animals. The amount of PEMT2, one of two PEMTs in liver, increased 5-fold in CD rats after 6 weeks on the CD diet. A similar increase in the level of PEMT2 mRNA suggested that this activation was due to enhanced expression of the PEMT2 gene in CD livers. The labeling of phosphatidylcholine in isolated hepatocytes from CD rats was consistent with the conversion of PE to PC being increased as a result of a higher expression of liver PEMT. We conclude that activation of PE methylation at the level of gene expression may be the mechanism by which CD rats compensate for the lack of dietary choline.

The design and preliminary evaluation of a home electrocardiography and blood pressure monitoring network.

A home electrocardiography (ECG) and blood pressure (BP) monitoring network was established in Beijing. The network consisted of three parts: (1) the home ECG and BP monitoring devices; (2) the monitoring centre at the hospital; and (3) the data communication network, for which we chose the public telephone network. The system was tested in our university hospital for two months with 30 patients and six normal volunteers. A preliminary evaluation by the users indicated that the system was acceptable in terms of its efficiency and effectiveness. This paper describes the system design, the preliminary testing procedure, and results.
