[Removal of PEG and salt from recombinant human tumor necrosis factor-alpha(rhTNF-alpha) with serial operation of gel filtration chromatography].

A serial gel filtration operation with the combination of G25 and G100 in one column was developed to remove salt and PEG from upper phase rich in recombinant human tumor necrosis factor alpha after simultaneous cell disruption and two-phase aqueous extraction. Buffer exchanging and primary purification were also achieved at same time. The purification factor of 4.4 was obtained with an activity recovery of 97% by one single step of serial operation.

[The synthesis of phenyl-silica gel and its application in purification of taxol].

Phenyl-silica gels were synthesized with four methods of reaction on gamma-aminopropyltriethoxysilane (APTS) silica gel. The synthesized gels were used as stationary phase of normal pressure chromatography to separate and purify taxol. The comparison of these four silica gels was carried out. It was found that the gel synthesized by gas-phase reaction gave the best results. The purity of taxol was 77.4%, and the recovery was about 90%.

[Separation and purification of taxol using normal- and reversed-phase chromatography in tandem].

A method is described for separation and purification of taxol from the extract of Taxus cuspidate callus culture with Al2O3 normal-phase chromatography and C18 reversed-phase chromatography, The operating conditions of the two chromatographic steps were investigated and optimized. The content of taxol was greatly increased after Al2O3 chromatography. The purity of taxol reached 95% from less than 1.0% after the two chromatographic steps. The final purity of taxol was more than 98% after the process of crystallization. The 13CNMR spectrum of the product was identical to that of taxol reported in the literature.

[Studies of the mechanism of 7-epi-taxol converted to taxol from Taxus extract catalyzed with Al2O3].

Under the catalysis of Al2O3, 7-epi-taxol in Taxus yunnanensis extract could convert to taxol. The factors including the concentration of CH3OH in the mobile phase, the type of Al2O3 and the reaction time could influence the isomeric reaction. The catalysis mechanism fo Al2O3 was investigated. The isomerization of 7-epi-taxol was under the cooperation of the Lewis souci activity core and the basic activity core in the surface of Al2O3 at room temperature. The type and strength of activity core controlled the procedure of reaction, and the suitable content of CH3OH in the mobile phase would accelerate the reaction.

[Enhancement of fermentative glycerol yield with heat shock treatment].

A heat shock treatment was studied in glycerol fermentation of osmotolerant yeast Candida krusei. The experiment results showed that the optimal temperature and duration time for heat shock is 45 degrees C and 30 min respectively, and the optimal start time of the treatment is at the mid term in exponential growth stage. With such treatment, glycerol yield was enhanced greatly, while no significant effects on both cell growth and glucose consumption were observed.

Purification of serum albumin with dye-ligand adsorption chromatography.

Dye molecules were coupled to agarose gel beads to form blue, red and yellow chromatography media for the separation of serum albumin. The adsorption properties of five dye-ligands were compared and the effects of temperature, pH, protein concentration and salt concentration were investigated. Human serum albumin (HSA) was purified from human placental plasma by a single step of dye-ligand chromatography under suitable operating conditions. The product was electrophoretically pure, and the elution recovery was as high as 95%. This new technique is superior to the conventional salting-out process.
