Some extracellular matrix elements as markers of 'capillary tunnels' in hypertrophied rat heart.

We studied the distribution of the extracellular matrix proteins fibronectin (FN) and laminin (LM) in the hypertrophied hearts of spontaneously hypertensive rats (SHR), using an immunofluorescence method with specific antibodies. The immunohistochemical reaction was positive in the cytoplasm of some hypertrophied cardiomyocytes. The results showed that FN and LM can be used as markers for tunnels, i.e. intracardiocytic invaginations of the sarcolemma. The tunnels observed contained capillaries.

Effect of simultaneous administration of ibuprofen and ethanol on the alkaline phosphatase activity in the small intestine.

Using the interferometric technique the authors studied the effect of simultaneous administration of ibuprofen (Polfa) and ethanol on the activity of alkaline phosphatase in the proximal jejunum. Both ibuprofen and ethanol cause in the digestive tract functional and morphological changes. The used technique of quantitative determination of alkaline phosphatase activity at the site of its primary location made possible an assessment of changes in the activity of the enzyme caused by the administered agents. It was found that after 60 days of the experiment both agents caused a statistically greater changes were observed in the rats receiving both these agents simultaneously. The obtained results suggest the conclusion that simultaneous administration of ibuprofen and ethanol causes in the mucosal gland in the proximal small intestine development of an interaction of the additive.

The effect of jejunectomy on alkaline phosphatase activity in the ileum.

The activity of alkaline phosphatase activity in the small intestinal wall was studied in dogs after jejunectomy. The observations were made 3, 6, 8 or 9 and 12 weeks after operation. The studies aimed to obtain some information about the functional adaptation processes of the remaining intestinal segments. The enzyme activity in homogenates of duodenal and ileal mucosa was determined. Parallel interferometric measurements in the brush border and on the surface of the absorptive cells were performed. The results obtained indicate that after temporary reduction (especially 6 weeks postoperatively) a gradual rise of the alkaline phosphatase activity both in homogenates and in the brush border of the intestinal remnants took place. Several times repeated biopsies confirmed the ability of the intestinal segments (duodenum and distal ileum) significant increase in enzyme activity over the normal (control) level was observed.

The effect of doxycycline upon alkaline phosphatase activity in rat kidney (quantitative study using the interferometric technique).

The doxycycline effect on the activity of alkaline phosphatase in the brush border of the convoluted renal tubules in rat was examined, using the interferometric technique. On the basis of measurements taken, the antibiotic was shown to cause a statistically significant fall of the alkaline phosphatase activity and the enzyme activity was found to decrease with consecutive doses of the drug under investigation.

Hepatic lesions in dogs treated with horse antidog lymphocyte globulin (HADLG) after kidney transplantation.

Microscopical examination of the liver of dogs which after allogenic kidney transplantation were transiently treated with antilymphocyte globulin revealed infiltrates of mononuclear cells (lymphocytes, plasma and reticulum cells). These infiltrates were usually situated in perivascular spaces (around the central veins or on the portobiliary tract). A proportion of capillaries showed endothelial proliferation and the narrowing of their lumen. The authors were unable to determine whether these lesions resulted from the direct toxic effect of antilymphocyte globulin or represented a nonspecific reaction to the used preparation.

Application of interferometry in quantitative histoenzymatic investigations.

The interferometric technique has been used to measure the alkaline phosphatase activity in the kidney and small intestine. The effect of post-mortem autolysis on the activity of this enzyme was checked in specimens taken from five normal rabbits. The results were entirely reproducible. The limitations of the method are discussed and the possible sources of error analysed.

Activity of alkaline phosphatase in the healing rat liver after hepatectomy.

Changes of alkaline phosphatase activity in the process of healing liver injuries of rats on which partial hepatectomy was performed were the subject of our researches. Reaction on alkaline phosphatase was performed on cryostat sections according to the method of Gomori-Takamatsu. It has been found that granulated precipitate of cobalt sulphide was localized in the cytoplasm of granulocytes, macrophages and fibroblasts. A diffusive reaction comprized clusters of newly created collagen fibres. In early stages in the centres of necrosis, the activity of alkaline phosphatase increased, in later periods a lack of the activity was observed. Steatosis of liver tissue does not correlate with the intensity of the reaction. Lack of alkaline phosphatase activity is characteristic for old cicatrices. The authors discuss the effects of the histogenetical process during healing of liver injury on the results of the histoenzymatic reactions. The authors suggest that alkaline phosphatase is involved in the mechanism of re-building collagen fibres. It is stressed in the conclusion that the localization and intensity of alkaline phosphatase in the area of healing liver lesions are variable depending on the period of healing wound of the liver and the influence of other complications.

The activity of alkaline phosphatase in the process of regeneration of rat liver.

Using GOMORI'S technique, the present authors investigated the dynamics of the alkaline phosphatase activity in the process of liver regeneration after partial hepatectomy. In all 80 rats of the Wistar strain were subjected to experiment, 60 to 75% of the liver parenchyma being removed from each of them. In the course of regeneration a gradual increase in the enzyme activity was observed within the first 48 hours following the operation. This was succeeded by a slow decline of the activity, and after the 25th day after the operation the reaction intensity resembled that recorded for the control animals. It was also ascertained that the fatty degeneration of the liver noted in the initial period of regeneration does not inhibit the activity of alkaline phosphatase.
