Effects of feed particle size and feed form on survival of Salmonella typhimurium in the alimentary tract and cecal S. typhimurium reduction in growing broilers.

An in vitro experiment and an in vivo experiment were conducted to investigate the effects of feed particle size (coarse and fine) and feed form (mash and pellet) on the survival of Salmonella typhimurium (ST) in the alimentary tract, and the reduction of cecal ST in growing broilers in a 2 x 2 factorial design. All diets in the 2 trials were corn and soybean meal-based diets that differed only in physical characteristics. Diets were provided for birds from d 1 to 22 of age. In the in vitro trial, the relative gizzard weight was lower in birds that were fed the pellet diet (P < 0.01), whereas the relative weight of the cecum was higher in these birds (P < 0.01). Broilers receiving the pellet diet had enhanced concentrations of volatile fatty acids in contents from both the gizzards and the ceca. In addition, there was an increase in gizzard pH (P < 0.01) with the pellet diet, but a reduction in cecal pH (P < 0.05). Pellet-fed birds showed a significantly decreased and increased in vitro death rate of ST in the contents from gizzards (P < 0.01) and ceca (P < 0.05), respectively. A higher in vitro ST death rate in the gizzard was observed in birds given the coarse mash diet compared with those given the fine mash diet. In the in vivo experiment, cecal volatile fatty acid concentrations were increased, whereas cecal pH was decreased significantly (P < 0.05) when birds were fed the pellet diet compared with the mash diet. Furthermore, cecal ST concentrations were higher (P < 0.05) in broilers fed the pellet diet than in those fed the mash diet. Results indicated that the pellet diet increases the incidence of ST in gizzards and ceca in growing broilers and provide evidence demonstrating that the gizzard may play a critical role in reducing ST contamination in growing broilers.

Synthesis of a divalent glycoside of an alpha-galactosyl disaccharide epitope involved in the hyperacute rejection of xenotransplantation.

3,6-dioxaoct-1,8-diyl di-(3-O-alpha-D-galactopyranosyl-beta-D-galactopyranoside) was synthesized for use in research on hyperacute rejection of xenotransplantation. The trichloroacetate method was successfully applied to form stereoselectively the alpha-D-galactosyl linkage under mild reaction conditions and a simple procedure. The divalent O-glycoside was formed from the corresponding trichloroacetimidate in one step with reasonable yield.

Changes in HIV-1 incidence in heroin users in Guangxi Province, China.

Guangxi Province, China recently experienced an outbreak of HIV-1 infection among heroin users. We studied HIV-1 incidence rates and associated risk factors for HIV-1 infection among heroin users residing in Pingxiang City. A total of 318 heroin users were followed from February 1998 through January 1999 (median follow-up: 8.1 months). Of these, 130 were prospectively followed from January through September 1999 (median follow-up: 8.3 months). HIV-1 and hepatitis C virus (HCV) incidence rates for each period were calculated. A generalized estimating equation approach was implemented to identify independent risk factors associated with HIV-1 infection across both periods. Among 318 study participants, 97.2% were men. The median age was 22 years. Approximately 60% reported sharing needles. HIV-1 prevalence at baseline was 15.4%. During the first follow-up period, HIV-1 incidence was 2.38 per 100 person years (py), and HCV incidence was 26.8 per 100 py. During the second follow-up period, HIV-1 incidence was 6.86 per 100 py, and HCV incidence was 28.9 per 100 py. After controlling for age and other factors, HCV seropositivity, history of sexually transmitted diseases, and sharing needles were independently associated with HIV-1 infection. These data suggest that HIV-1 incidence was rising over time in Pingxiang City, Guangxi Province. The high incidence of HCV heightens the importance of enhanced prevention programs to reduce injection and needle sharing among heroin users.

[Clinical analysis of 15 cases with tetramethylenedisulfotetramine].

In our study, we observed the clinical manifestations of 15 tetramethylenedisulfotetramine (TEM) toxicosis emergency patients diagnosed by plasma toxin chemical analysis, and evaluated the effect of emergency treatments. All cases could not be differentiated from the other kinds of toxicosis by clinical manifestations, and toxin chemical analysis was the sole definite diagnosis method. The effect of gastric lavage in early stage was especially obvious, 13 cases with convulsions got immediate relief after plasma exchange, the use of large dosage of tranquilizer resulted in better control on convulsion in 4 cases treated with ventilator than that in the others. Gastric lavage and chemical toxin analysis in early stage are of vital importance, plasma exchange, tracheotomy and mechanical ventilation are the most effective reatment methods.

Synthesis, characterization, and magnetic properties of carbon- and boron-oxide-encapsulated iron nanocapsules.

Carbon- and boron-oxide-encapsulated iron nanocapsules have been synthesized by arc discharge in methane (CH4) and diborane (B2H6) atmospheres respectively. The characterization and magnetic properties of carbon- and boron-oxide-encapsulated iron nanocapsules [abbreviated as Fe(C) and Fe(B) respectively] were investigated and compared. The structure of the Fe(B) nanocapsules is different from that of the Fe(C) nanocapsules. The Fe(C) nanocapsules consist of a crystalline graphite shell and a core of alpha-Fe and/or Fe3C. The Fe(B) nanocapsules consist of an amorphous boron-oxide layer and a core of Fe(B) solid solution, alpha-Fe, gamma-Fe, FeB, and/or Fe3B phases. The saturation magnetizations of both the Fe(C) and the Fe(B) nanocapsules below 300 K decrease monotonically with increasing temperature. The coercivities of the Fe(C) and Fe(B) nanocapsules are almost 2 orders of magnitude higher than that of bulk Fe. The temperature dependence of magnetization at high temperatures indicates the existence of some phase transformations.

Molecular modeling study of chiral drug crystals: lattice energy calculations.

The lattice energies of a number of chiral drugs with known crystal structures were calculated using Dreiding II force field. The lattice energies, including van der Waals, Coulombic, and hydrogen-bonding energies, of homochiral and racemic crystals of some ephedrine derivatives and of several other chiral drugs, are compared. The calculated energies are correlated with experimental data to probe the underlying intermolecular forces responsible for the formation of racemic species, racemic conglomerates, or racemic compounds, termed chiral discrimination. Comparison of the calculated energies among ephedrine derivatives reveals that a greater Coulombic energy corresponds to a higher melting temperature, while a greater van der Waals energy corresponds to a larger enthalpy of fusion. For seven pairs of homochiral and racemic compounds, correlation of the differences between the two forms in the calculated energies and experimental enthalpy of fusion suggests that the van der Waals interactions play a key role in the chiral discrimination in the crystalline state. For salts of the chiral drugs, the counter ions diminish chiral discrimination by increasing the Coulombic interactions. This result may explain why salt forms favor the formation of racemic conglomerates, thereby facilitating the resolution of racemates.

[Separation of catechins and caffeine in tea polyphenol by isocratic elution high performance liquid chromatography].

The catechins, particularly in green tea, have been found to possess anti-mutagenic and anti-tumorigenic properties. As each catechin possesses distinct properties, a simple and rapid method that could be used for analysis of individual catechins in a complex mixture would be necessary. A relatively simple and rapid method for simultaneous separation of five catechins and caffeine in tea polyphenol by isocratic elution high performance liquid chromatography has been developed. The analysis could be finished within 30 min. They were measured using Resolve C18 column (at 43 degrees C) and UV detector (at 280 nm), water-85% phosphoric acid aqueous solution-acetonitrile-dimethyl formamide(DMF) (859:1:120:20, V/V) as mobile phase. There was a good linear relationship between the content of component and its peak area for catechins and caffeine, with the correlation coefficients of 0.9992-0.9999. The average recoveries (n = 5) were 83.33%-104.42%, and the relative standard deviations (n = 6) were 0.74%-1.43%. The effect of concentration of DMF in mobile phase was studied.

[The role of immune enhanced enteral nutrition on plasma amino acid, gut permeability and clinical outcome (a randomized, double blind, controlled, multi-center clinical trail with 120 cases)].

OBJECTIVE: To evaluate the role of arginine, RNA and omega 3 fatty acid enriched enteral nutrition. METHODS: The study was designed as a prospective, randomized, double blind, multi-central trial. It was an isocaloric and isonitrogenous intake in both groups. The protocol was approved by the Ethic Committee and, written informed consents were obtained. RESULTS: There were 120 patients enrolled in this protocol. After data were input to computer, open the code. 118 out of 120 patients completed the study and, 2 of them were dropped out. One is because the nasal jejunum tubes dropped and not willing to be replaced. Second patient had fistula of anastomosis on 4th days after operation. There were finally 60 patients in the study group and 58 in the control group. There were no liver or renal functions damage and, obvious adverse in both groups. Plasma amino acid profile: There was significant difference (delta) of plasma arginine levels pre- and after study [(33.7 +/- 58.5) mumol/L vs (-2.4 +/- 30.7) mumol/L] (P = 0.004). Intestinal Permeability (lactulose/mannitol ratio): The differences (delta) of lactulose/mannitol ratio pre- and after the study were 0.017 +/- 0.012 in study group and, 0.027 +/- 0.016 in control group. (P = 0.047). Immunological markers: Humoral immunity: The differences of IgM levels pre- and after the study were (0.6 +/- 0.4) g/L in study group and, (0.2 +/- 0.4) g/L in control group(P = 0.006). Cellular immunity: The differences (delta) of CD3 levels pre- and after the study were (3.8 +/- 5.2)% in study group and (0.3 +/- 6.5)% in control group (P = 0.01). In CD4, (3.4 +/- 5.3)% in study group and, (-0.3 +/- 5.7)% in control group (P = 0.032). Clinical Outcomes: There was no infection-related in study group and, 2 abdominal infection patients in control group. No significant difference was found between groups (P = 0.46). The hospital stays were (13 +/- 2.5) days in study group and, (14.5 +/- 3.0) days in control group (P = 0.004). The cost for full hospitalization was (15,122 +/- 6,279) Yuan in study group and, (17,403 +/- 7,091) Yuan in control group. There was 2,281 Yuan lower in study group (P = 0.07). The costs for nutritional drugs were (1,383 +/- 242) Yuan in study group and, (707 +/- 111) Yuan in control group. The difference was 676 Yuan higher in study (P = 0.001). CONCLUSION: Immune enhanced enteral nutrition had better plasma arginine level, intestinal permeability marker, IgM, CD3 and CD4. Also had less hospital stay and, less totaled hospital cost in study group.

Fungal morphology and fragmentation behavior in a fed-batch Aspergillus oryzae fermentation at the production scale.

It is well known that high-viscosity fermentation broth can lead to mixing and oxygen mass transfer limitations. The seemingly obvious solution for this problem is to increase agitation intensity. In some processes, this has been shown to damage mycelia, affect morphology, and decrease product expression. However, in other processes increased agitation shows no effect on productivity. While a number of studies discuss morphology and fragmentation at the laboratory and pilot scale, there are relatively few publications available for production-scale fungal fermentations. The goal of this study was to assess morphology and fragmentation behavior in large-scale, fed-batch, fungal fermentations used for the production of protein. To accomplish this, a recombinant strain of Aspergillus oryzae was grown in 80 m(3) fermentors at two different gassed, impeller power-levels (one 50% greater than the other). Impeller power is reported as energy dissipation/circulation function (EDCF) and was found to have average values of 29.3 +/- 1.0 and 22.0 +/- 0.3 kW m(-3) s(-1) at high and low power levels, respectively. In all batches, biomass concentration profiles were similar and specific growth rate was < 0.03 h(-1). Morphological data show hyphal fragmentation occurred by both shaving-off of external clump hyphae and breakage of free hyphae. The fragmentation rate constant (k(frag)), determined using a first-order model, was 5.90 and 5.80 h(-1) for high and low power batches, respectively. At the end of each batch, clumps accounted for only 25% of fungal biomass, most of which existed as small, sparsely branched, free hyphal elements. In all batches, fragmentation was found to dominate fungal growth and branching. We speculate that this behavior was due to slow growth of the culture during this fed-batch process.

Synthesis of galactosyl and lactosyl derivatives as potential anti-metastasis compounds.

Based on the known anti-metastasis activities of lactosides and galactosides, a galactosyl and a lactosyl trimannoside were prepared via the conventional Koenigs-Knorr and trichloroacetimidate methods, respectively. Through typical deblocking procedures, a tetrasaccharide alpha-D-Galp-(1 --> 2)-alpha-D-Manp-(1 --> 2)-alpha-D-Manp-(1 --> 6)-alpha-D-ManpOCH3 and a pentasaccharide beta-D-Galp-(1 --> 4)-beta-D-Glcp-(1 --> 2)-alpha-D-Manp-(1 --> 2)-alpha-D-Manp-(1 --> 6)-alpha-D-ManpOCH3 were obtained.

Highly stereoselective alpha-O-glucosylation of heterocyclic ketene aminals.

A new series of alpha-glucosides of heterocyclic ketene animals have been synthesized. The double bonds of the glucosides resulted in Z or E configuration by using CaH2 or a Lewis acid as catalyst, respectively. A special complex of mercuric cyanide with glucopyranosides was the result when Hg(CN)2 was used to catalyze the reaction.

Effect of nitric oxide on electric and mechanical activities of gastric antral circular muscles in guinea pigs.

AIM: To study the effect of exogenous nitric oxide (NO) on electric and mechanical activities of gastric antral circular muscle in guinea pigs in vitro. METHODS: Mechanical and electric activities of gastric antral circular muscle in guinea pigs were recorded simultaneously. RESULTS: Sodium nitroprusside (SNP, 0.5 mumol.L-1), an NO donor, inhibited the frequency and amplitude of fast wave and spontaneous contraction of the strips (P < 0.01). SNP-induced inhibition was not blocked by tetrodotoxin, atropine, phentolamine, and propranolol (P > 0.05), but diminished by methylene blue (P < 0.01) and oxyhemoglobin (P < 0.01). CONCLUSION: Exogenous NO inhibits gastric antral myoelectric and mechanical activities in guinea pigs. The inhibitions are produced by NO acting on extracellular membrane and enhancing the level of cGMP.

[The enlarged translabyrinthine removal of large acoustic neuromas: 18 cases report].

OBJECTIVE: To investigate the surgical methods and results of enlarged translabyrinthine approach in the removal of large acoustic neuromas. METHOD: A large mastoidectomy involved complete exposure of sigmoid sinus, the dura behind the sinus for at least 1 cm, the superior petrosal sinus and the middle fossa dura. The jugular bulb was exposed and pressed downwards if necessary. The internal auditory meatus was skeletonized and uncovered for at least 270 degrees. The debulking of the tumor began inside the anterior and inferior poles in order to find the brainstem and the facial nerve root as early as possible, then the dissection of the nerve was performed medially to laterally. Intraoperative facial nerve monitoring and postoperative CT and MRI were performed in all cases. RESULT: Total removal was achieved in all 18 patients with tumors larger than 3 cm (mean size: 4.2 cm). There were no death as well as other complications such as intracranial infection and persistent cerebrospinal fluid leakage. There were no obvious cerebral sequela. The facial nerve was preserved both anatomically and functionally in 14 cases, with Grade I or II in 8 cases, Grade III or IV in 6 cases. Nerve interruption occurred in 4 cases who all had severe facial paralysis or nerve interruption before operation. 16 patients returned to work within 1-3 months. CONCLUSION: Total removal of large acoustic neuroma could be accomplished via the translabyrinthine approach, with good result of facial nerve function and minimum incidence of morbidity.

[The semiconductor diode laser application in tympanoplasty].

OBJECTIVE: To investigate the method, indications and initial results of semiconductor diode laser application in tympanoplasty. METHOD: The diode laser was used in 14 tympanoplasty procedures. The laser manipulation was mainly used in cases of malleus and incus bony fixation with the tympanum, the fibrous scar tissue around the ossicular chain and the malleus head resection. RESULT: There was no postoperative vertigo and facial paralysis. The average hearing improvement was 24 dB. Seven cases (50%) reached an air-bone gap within 20 dB. CONCLUSION: The application of diode laser in tympanoplasty could effectively reduce the manipulation trauma of ossicular chain and control the bleeding of operative field, especially in managing the bony adhesion of the ossicular chain.

[Evaluation of Schistosoma japonicum tegumental membrane antigen (SjMAg) in immunodiagnosis and efficacy assessment].

OBJECTIVE: To evaluate the specificity and sensitivity of SjMAg in detecting specific antibodies in sera of patients with schistosomiasis japonica and in assessing therapeutic efficacy. METHODS: SjMAg-ELISA was used to determine the specific IgG and IgG4 in sera of patients with schistosomiasis japonica at different time points after chemotherapy. RESULTS: SjMAg-ELISA was as sensitive and specific as SEA-ELISA. The negative conversion rate of IgG detected by SjMAg-ELISA 12 months after treatment was 80.0%, being significantly higher than 43.3% by SEA-ELISA. The negative conversion rate of IgG4 detected by SjMAg-ELISA 12 months after treatment was 93.3%, being significantly higher than 60.0% by SEA-ELISA. The negative conversion rates of IgG and IgG4 2 yr after treatment were 92.9% and 97.6% respectively. CONCLUSION: SjMAg-ELISA is comparable to SEA-ELISA in diagnosing schistosomiasis japonica and is more effective in assessing therapeutic efficacy.

Synthesis of glucosylated 1,2,3-triazole derivatives.

A series of potential bioactive compounds, 1-glucosyl-4-heterocyclyl-5-(p-substituted-phenyl)-1,2,3-triazoles , were synthesized. Highly stereoselective products were obtained in good yield. Primary activity screening showed that this type of N-glucosylic compound possessed antitumour and antiviral activities.

Characterization of racemic species of chiral drugs using thermal analysis, thermodynamic calculation, and structural studies.

The identification of the racemic species, as a racemic compound, a racemic conglomerate, or a racemic solid solution (pseudoracemate), is crucial for rationalizing the potential for resolution of racemates by crystallization. The melting points and enthalpies of fusion of a number of chiral drugs and their salts were measured by differential scanning calorimetry. Based on a thermodynamic cycle involving the solid and liquid phases of the enantiomers and racemic species, the enthalpy, entropy and Gibbs free energy of the racemic species were derived from the thermal data. The Gibbs free energy of formation, is always negative for a racemic compound, if it can exist, and the contribution from the entropy of mixing in the liquid state to the free energy of formation is the driving force for the process. For a racemic conglomerate, the entropy of mixing in the liquid state is close to the ideal value of R ln 2 (1.38 cal.mol-1. K-1). Pseudoracemates behave differently from the other two types of racemic species. When the melting points of the racemic species is about 30 K below that of the homochiral species, is approximately zero, indicating that the racemic compound and racemic conglomerate possess similar relative stabilities. The powder X-ray diffraction patterns and 13C solid-state nuclear magnetic resonance spectra are valuable for revealing structural differences between a racemic compound and a racemic conglomerate. Thermodynamic prediction, thermal analysis, and structural study are in excellent agreement for identifying the nature of the racemic species.
