Pharmacokinetic investigation with 125I labeled T-actemodulin (TAM).

The aim of the study was to label TAM with radioactive iodine and to follow its distribution in different organs of rats. T-actemodulin is a thymic hormone isolated from calf thymus by a new technology. TAM was labeled according to the method of Greenwood and Hunter. Two major peaks were observed on the gel chromatography: The first peak contained the labeled TAM; the second peak contained 125I which was not bound to the organic compound. Additional purification of the first peak was performed on a Sephadex G-75 column, and the eluate was designated as purified peak I. After a radioimmune testing it could be concluded that TAM was labeled successfully and the peak I contained 125I-TAM. Peak I, purified peak I and the peak II were applied to young male Wistar rats. The radioactivity of pancreas, kidney, spleen, adrenals, liver, thymus, bone marrow, testis and thyroid were measured after killing the animals 10 min, 60 min, and 24 h after the injections. The pharmacokinetic study showed that all organs, except for the thyroid have a higher affinity for labeled TAM than for the free 125I. Higher accumulation of labeled TAM was observed in the pancreas, kidney, adrenals and spleen. A specific uptake of labeled TAM in these organs can be suggested. Functional interrelations between thymus and pancreas, spleen, adrenals are discussed.

Influence of T-actemodulin on herpes simplex virus infection.

The aim of the investigation was to study the direct influence of T-actemodulin (TAM) on the Herpes Simplex Virus (HSV) infection in vitro and in vivo. HSV-1 (strain Vic) and HSV-2 (strain Nissa) were used in the experiments in vitro. The experiments were performed using multicycle and singlecycle growth tests. Seventy-five patients suffering from HSV infections (labialis and genitalis) were locally treated by 0.04% TAM-containing hydrophilic gel or water-washable cream (applied every 3 h). The results showed that TAM: decreased the infectious virus titre; reduced the infectious virus yield at the end of the single cycle of the virus replication; prolonged the latent period; and exerted an irreversible inhibitory effect on the infectious virus titre when applied at immediate early intervals after the infection. No relapses were observed in 84% of the patients treated with TAM. Eight percent of the patients were with only reduced relapses and in 8% of the patients TAM had a mild effect. TAM exerted a preventive effect against the disease when the treatment was initiated in the period of paresthesia.

Time course changes in blood glucose and insulin levels of thymectomized rats.

The endocrine function of the thymus and its relations to other endocrine glands have been discussed in the scientific literature. Blood glucose contents (assayed on a 'Beckman' glucoanalyzer by the glucoseoxidase method) and serum insulin level (using radioimmunoassay) were studied in male Wistar rats--1, 2, 3, 4, 5, 6, 9, and 12 months after adult thymectomy. The results obtained demonstrated that the thymectomy provoked increase of insulin level accompanied by phasic decrease of blood glucose in comparison to age matched controls. Correlation between insulin and glucose levels in the thymectomized rats was not found. The results support the hypothesis of functional interrelations between the thymus and the endocrine pancreas.

The influence of neonatal thymectomy on the blood serum level of total lipids, NEFA, triglycerides, total cholesterol and beta-lipoproteins in rats.

The time course of changes in the blood serum content of total lipids (TL), nonesterified fatty acids (NEFA), triglycerides (TG), total cholesterol (TC) and beta-lipoproteins (beta-LP), caused by neonatal thymectomy (NTE), was investigated in neonatally thymectomized and in sham-operated rats aged 15, 30, 45 days, 2, 3 and 5 months. TL were significantly increased on days 15, 30, 45 and 60 after operation; their level in 3- and 5-month-old rats was already normal. In control rats, NEFA level increased with age; their level in NTE rats was significantly higher on days 15, 30, 45 and 60 after thymectomy. Similar changes were registered in TG level. The effect of NTE was most pronounced in TC whose level was significantly higher in thymectomized rats than in control rats till the 150th day after operation. Serum concentration of beta-LP in NTE rats exceeded several times that of sham-operated rats in all age groups. The role of the thymus in the control of lipid metabolism in rats is discussed.

Changes in the connective tissue components of the aorta in thymectomized rats.

The character and essence of changes taking place in the connective tissue matrix of the vascular wall under the effect of thymus deficiency are not well enough clarified. The aim of the study was to assess the content of collagen, elastin, total glucosamines, total glycoproteins, total nonfibrillar protein and total protein in the aorta total glycoproteins, total nonfibrillar protein and total protein in the aorta of adult thymectomized rats of the Wistar strain and in control animals over a 12-month post-thymectomy period. The amount of collagen and elastin augments both in connection with the aging process (after 2-4 months), and as a result of thymectomy. Glycoproteins show an increase starting from the third month in the controls, and from the fifth month in thymectomized animals. Glucosamines display a fluctuating decrease parallel to aging in either of the groups under study. The content of total protein and nonfibrillar protein reveals no differences worthy of notice. It is reasonable to assume the existence of a correlation between thymus activity and metabolism of the connective tissue components in the vascular wall of rats.

[Morphological changes in the myocardium after thymectomy].

The authors followed in dynamics the systolic arterial blood pressure and morphometric changes in the myocardium (cardiac, index, mean number of myocardial cells per unit of area c I mm2, mean diameter of the myocardial cells of the papillary muscles and subendocardial laer of the left ventricle) in thymectomized and sham thymectomized rats. The studies were made after thymectomy and sham thymectomy on the second, sixth, twelfth and sixteenth week. The systolic arterial blood pressure in thymectomized rats was elevated gradually and reached up to 170 mmHg on the sixteenth week, but it was in normal ranges in sham thymectomized animals. The cardiac index and mean diameter of the myocardial cells were larger in thymectomized rats, but the mean number of cells was smaller in comparison with sham thymectomized animals-results showing development of myocardial hyperthrophy after thymectomy. The induced myocardial changes were probably of adaptive character and could be due to neurohormonal mechanisms and hemodynamic loading of the heart becuase of the elevated arterial blood pressure after thymectomy.

[Pregnanediol and pregnanetriol levels in the urine of rats after thymectomy].

The authors examined arterial blood pressure and excretion of pregnanetriol and pregnanediol in urine of adult male rats of Westar strain under normal conditions and after thymectomy. There was an elevation of arterial blood pressure after 45th day of thymectomy. Excretion of pregnanediol was increased on the 30th day, but that of pregnanetriol on the 60th day after removal of the gland. The increased excretion of both progesterone metabolites was connected with increased renal function, which probably played pathogenic role in postthymectomized hypertensive reaction.

[Changes in adrenocortical activity in grown rats following thymectomy].

The authors measured arterial blood pressure by direct and indirect method in 80 adult rats and examined: content of ascorbic acid and cholesterol and the activity of alkaline and acid phosphatase in adrenals after thymectomy. The indices were followed in dynamics on the 15, 30, 45, 60 and 90th day after thymectomy. The established changes, manifested by a reduced concentration of the ascorbic acid and cholesterol in the early period of the experiment and the increased phosphatase activity during the whole period were interpreted as a manifestation of elevated adrenal activity after thymectomy. Activation of the adrenal cortex preceded the hypertensive reaction.
