Prediction of carbon-13 NMR chemical shift of alkanes with rooted path vector.

Systematic studies were further made on graph theory in quantitative structure-spectrum relationships (QSSR) for various areas of spectroscopies. Chemical shifts (CS) in alkanes for carbon-13 nuclear magnetic resonance (13C NMR) were well correlated with a set of novel molecular graph indices, called the rooted path vector of various lengths, as several multivariate regression equations as following:CS=3.022+5.336P1+7.356P2-1.648P3+0.83859P4+0.210P5-0.138P6-0.506P7+2.486P8-1.669P9; n=402, m=9, R=0.944, RCV=0.9413, S.D.=3.333, F=358.343, U=35833.211, Q=4355.422 for all types (primary, secondly, tertiary, quaternary as well as methane) of carbon atoms CS=0.983+6.811P1+7.584P2-2.029P3+0.809P4+0.106P5+0.043P6-0.124P7+1.715P8-1.101P9; n=374, m=9, R=0.975, RCV=0.9737, S.D.=2.303, F=773.372, U=36912.109, Q=1930.363 for primary, secondly, tertiary (including methane) carbon atoms; and CS=27.819+2.351P2+0.549P3-0.440P4+0.170P5-0.050P6; n=27, m=5, R=0.992, RCV=0.9674, S.D.=0.324, F=265.418, U=138.891, Q=2.198 for quaternary carbon atoms, respectively. Quite good estimation and prediction results were obtained from the quantitative molecular modeling and the performance of multiple linear regression (MLR) equations were tested to work well through cross-validation (CV) with the leave-one-out (LOO) procedure.

A combined bioprocess for integrated removal of copper and organic pollutant from copper-containing municipal wastewater.

Heavy metal-containing wastewater is difficult to treat by most of the bioprocess due to its toxicity to microorganism in activated sludge. In this study, a combined bioprocess consisting of biosorption section that contained magnetite immobilized Pseudomonas putida 5-x cell as biosorbent followed by sequence batch reactor (SBR) was developed to treat Cu(2+)-containing municipal wastewater. The production techniques of Pseudomonas putida 5-x cell as biosorbent, such as optimal cell harvest period and cell pretreatment techniques were studied. Experimental results showed, considering both cell biomass and cell adsorption capacity to Cu(2+), 36 h is the optimal harvest period in the course of culturing Pseudomonas putida 5-x cells as biosorbent, and 0.1-0.3 mol L(-1) HCl is an optimal cell pretreating eluant to improve Cu(2+) adsorption capacity. The performances of the combined bioprocess for treating Cu(2+) containing wastewater were assessed. Experimental results showed that after treatment by biosorption, the Cu(2+) level in wastewater was reduced to level that did not inhibit COD removal efficiency of subsequent SBR activated sludge process, although it still affected the COD adsorption capacity of activated sludge. In terms of COD removal, the biosorption section was efficient for reducing Cu(2+) concentration to provide biodegradable wastewater for subsequent SBR activated sludge process.

The biodegradation of 1,3-dichlorobenzene by an adapted strain Bacillus cereus PF-11 derived from town-gas industrial effluent.

In the present study, an adapted bacterium PF-11 with high 1,3-dichlorobenzene degradation capability was isolated from town-gas industrial effluent through continuous introducing of N-methyl-N'-nitro-N-nitrosoquanidine (NTG). In suitable condition, a degradation rate of 32 mg L(-1) d(-1) of 1,3-dichlorobenzene was obtained by strain PF-11 with effective chlorion release. Strain PF-11 was tentatively identified as gram-positive Bacillus cereus. The substrate specificity of the strain PF-11 was relatively low, and the degradation rate for different chlorobenzenes was in the order of monochlorobenzene > 1,3-dichlorobenzene > 1,2-dichlorobenzene. Initial oxidation step was molecular oxygen attacking chlorobenzene ring catalyzed by dioxygenase.

Identification of c-Jun as bcl-2 transcription factor in human uterine endometrium.

We describe the application of the biomolecular interaction (BIA) technique to detection of the interaction between protein (e.g., c-Jun) and DNA (e.g., two AP-1 motifs from bcl-2 promoter), compared with immunohistochemistry (IHC) of c-Jun. The specific binding assay for the interaction of c-Jun and activating protein-1 (AP-1) motifs was performed using a Biacore 2000 system. Intense immunoreactivity of c-Jun in glandular cells of the human uterine endometrium was observed in the proliferative phase, while c-Jun in stromal cells was expressed throughout the menstrual cycle. In contrast to the IHC of c-Jun, the specific binding of c-Jun to two separate AP-1 motifs in the bcl-2 promoter region was detected only in nuclear extracts of glandular cells, but not in stromal cells, during the proliferative phase. These results indicate that, while transmitting various signals, c-Jun enhances the transcription level of bcl-2, which in turn keeps glandular cells alive and proliferating in normal human endometrium during the proliferative phase. Moreover, the method involving real-time biomolecular interactions such as DNA-protein binding is novel for the study of transcription factors when combined with IHC.

Cell death and proliferation in Nd-YAG laser, electrocautery, and scalpel wounds on mice skin.

The purpose of this study is to compare cell death and proliferation in laser, electrocautery and scalpel wounds on the mice epidermis. Wounds were examined by transmission electron microscopy, the detection of free 3'-OH DNA ends and immunohistochemistry of proliferating cell nuclear antigen (PCNA), inducible nitric oxide synthase (iNOS), keratinocyte growth factor (KGF) and keratinocyte growth factor receptor (KGFR). Reepithelization was first observed 5 days after scalpel and laser incisions and 7 days after electrocautery incision. Ultrastructurally, keratinocytes in both electrocautery and laser wounds showed similar post-apoptotic necrotic changes. Interestingly, dividing cells were often observed 3 days after laser incision. Apoptotic index in electrocautery wounds was higher than in laser wounds, although there was no significant difference in the PCNA expression level between them. The expression of iNOS, KGF and KGFR in laser wounds was more intense than in electrocautery wounds. In scalpel wounds, keratinocytes did not show significant changes in morphology or of markers of cell death and proliferation during the observation period. Therefore, the increase in the number of dividing cells and in the expression level of iNOS, KGF and KGFR may induce earlier and thicker reepithelization in laser wounds than in electrocautery and scalpel wounds.

Diagnosis and treatment of epithelial salivary gland tumours in children and adolescents.

In a series of 2,871 epithelial salivary gland neoplasms managed in the Peking University School of Stomatology between 1974 and 1999, 86 arose in children <16 years of age (52 parotid, 12 submandibular gland, 2 sublingual gland, and 20 minor salivary gland). Considerable delay was encountered in diagnosis (benign 24 months and malignant 16 months). In this group of children, 46 tumours (53%) proved to be malignant, with an incidence in the parotid, submandibular, sublingual, and minor salivary glands of 31/52 (60%), 2/12, 0/2, and 13/20 (65%), respectively. Sixty-six of 86 neoplasms (77%) occurred in children between 10 and 16 years of age. Only six neoplasms were encountered in children of 5 years or younger, four of which were high-grade malignant tumours. Benign tumours were successfully treated by local excision with only one recurrence. Of 46 malignant neoplasms, 8 were treated palliatively; of the remainder 8 were lost to follow-up and 2 patients died of their disease.

Mutations of arginine 64 within the putative Ca(2+)-binding lumenal interhelical a-b loop of the photosystem II D1 protein disrupt binding of the manganese stabilizing protein and cytochrome c(550) in Synechocystis sp. PCC6803.

Mutations D1-R64E, D1-R64Q, and D1-R64V in the putative calcium-binding lumenal interhelical a-b loop of the photosystem II (PSII) D1 protein were characterized in terms of impact on growth, extrinsic protein binding, photoactivation, and properties of the H(2)O-oxidation complex. The D1-R64E charge reversal mutation greatly weakened the binding of the extrinsic manganese-stabilizing protein (MSP) and, to a considerably lesser extent, weakened the binding of cytochrome c(550) (c550). Both D1-R64Q and D1-R64E exhibited an increased requirement for Ca(2+) in the cell growth medium. Bare platinum electrode measurements of O(2)-evolving membranes showed a retarded appearance of O(2) following single turn-over flashes, especially in the case of the D1-R64E mutant. The D1-R64E mutant also had a pronounced tendency to lose O(2) evolution activity in the dark and exhibited an increased relative quantum yield of photoactivation, which are characteristics shared by mutants that lack extrinsic proteins. S(2) and S(3) decay measurements in the isolated membranes indicate that D1-R64E and D1-R64Q have faster decays of these higher S-states as compared to the wild-type. However, fluorescence decay in the presence of DCMU, which monitors primarily Q(A)(-) charge recombination with PSII donors, showed somewhat slower decays. Taken together, the fluorescence and S-state decay indicate that the midpoint of either Q(B)(-) has been modified to be more negative in the mutants or that a recombination path presumably involving either Q(B)(-) or Y(D) has become kinetically more accessible.

Does histidine 332 of the D1 polypeptide ligate the manganese cluster in photosystem II? An electron spin echo envelope modulation study.

The tetranuclear manganese cluster in photosystem II is ligated by one or more histidine residues, as shown by an electron spin echo envelope modulation (ESEEM) study conducted with [(15)N]histidine-labeled photosystem II particles isolated from the cyanobacterium Synechocystis sp. strain PCC 6803 [Tang, X.-S., Diner, B. A., Larsen, B. S., Gilchrist, M. L., Jr., Lorigan, G. A., and Britt, R. D. (1994) Proc. Natl. Acad. Sci. U.S.A. 91, 704-708]. One of these residues may be His332 of the D1 polypeptide. Photosystem II particles isolated from the Synechocystis mutant D1-H332E exhibit an altered S(2) state multiline EPR signal that has more hyperfine lines and narrower splittings than the corresponding signal in wild-type PSII particles [Debus, R. J., Campbell, K. A., Peloquin, J. M., Pham, D. P., and Britt, R. D. (2000) Biochemistry 39, 470-478]. These D1-H332E PSII particles are also unable to advance beyond an altered S(2)Y(Z)(*) state, and the quantum yield for forming the S(2) state is very low, corresponding to an 8000-fold slowing of the rate of Mn oxidation by Y(Z)(*). These observations are consistent with His332 being close to the Mn cluster and modulating the redox properties of both the Mn cluster and tyrosine Y(Z). To determine if D1-His332 ligates the Mn cluster, we have conducted an ESEEM study of D1-H332E PSII particles. The histidyl nitrogen modulation observed near 5 MHz in ESEEM spectra of the S(2) state multiline EPR signal of wild-type PSII particles is substantially diminished in D1-H332E PSII particles. This result is consistent with ligation of the Mn cluster by D1-His332. However, alternate explanations are possible. These are presented and discussed.

The involvement of calcium mobilization in the calcium-activated potassium currents activated by hyposmotic swelling in gastric antral circular myocytes of the guinea-pig.

In our study of the effects of hyposmotic swelling on the Ca(2+)-activated potassium currents [I(K(Ca))] and its mechanism, we employed the whole-cell patch clamp technique using the gastric antral circular myocytes of the guinea-pig. Hyposmotic swelling efficiently increased I(K(Ca)), and the extent of changes in I(K(Ca)) was sharply dependent on the osmolarity of the perfusion solutions. When the calcium-free solution (EGTA 10 microM added in calcium-free solution) was superfused, I(K(Ca)) was not increased by the hyposmotic swelling. Gadolinium (Gd(3+)) 100 nM, a blocker of the stretch-activated nonselective cation channel, blocked the activation of I(K(Ca)) induced by hyposmotic swelling, but nicardipine 5 microM (the L-type calcium channel blocker) did not. Heparin 3 mg/ml, a potent inhibitor of inositol triphosphate receptor (InsP(3)R), did not inhibit the response, and caffeine 1 mM (the agonist for calcium-induced calcium release [CICR]) imitated the effect of hyposmotic swelling. Ryanodine (15 microM), markedly inhibited the effect. These results suggest that hyposmotic swelling activates I(K(Ca)), and the activation is associated with CICR, which is triggered by extracellular calcium influx through the stretch-activated channel (SA channel).

[Quantitative structure-retention relationship (QSRR) studies of polychlorinated dibenzofurans (PCDFs) on diverse gas chromatographic stationary phases on a set of novel molecular distance edge vector].

By taking phenyl as a pseudo atom, there are three types of atom in the polychlorinated dibenzofuran (PCDF) molecule, being chlorine atom, oxygen atom and pseudo atom. The chemical structure of PCDF congeners is depicted by a novel molecular distance-edge vector(VMDE, mu in short), developed in our laboratory, which consists of the modified VMDE parameters based on the identical group as a pseudo atom instead of a traditional atom. Furthermore quantitative structure-retention relationships (QSRR) between the new mu vector and gas chromatographic (GC) retention behavior of PCDFs are generated by multiple linear regression method for various stationary phases. Four models, each of which is constructed by using all sample sets, with high correlation coefficient, r > 0.98, are developed for three columns (DB-5, SE-54, OV-101). In order to test the equation stability and prediction ability of each model, it is essential to perform a cross validation (CV) procedure. Satisfactory CV results have been obtained by using one external predicted sample every time with high correlation coefficients, r > 0.97. These results show that the new mu vector has high structural selectivity and good property relativity, and it is easy to calculate the mu vector. And the QSRR models have high relative coefficients, good stability and good predictability.

[Estimation of gas chromatographic retention index for polycyclic aromatic hydrocarbons using VMDE].

A novel molecular distance-edge vector with 6 parameters was calculated for 100 polycyclic aromatic hydrocarbons (PAHs) based on the molecular interaction theory. It was found that there was a good linear relationship between the molecular distance-edge vector (VMDE) and the gas chromatographic retention index for the 100 PAHs, and the molecular modelling equation for the quantitative structure-retention relationship (QSRR) can be expressed as follows: IR = a + b VMDE, n = 100, r = 0.988, where a and b represent regression coefficients. The regression results showed that the molecular distance-edge vector is in good agreement with the results of molecular polarizability index to the gas chromatographic retention index for the polycyclic aromatic hydrocarbons.

Differences in the distribution of synemin, paranemin, and plectin in skeletal muscles of wild-type and desmin knock-out mice.

Mice lacking the gene encoding for the intermediate filament protein desmin have a surprisingly normal myofibrillar organization in skeletal muscle fibers, although myopathy develops in highly used muscles. In the present study we examined how synemin, paranemin, and plectin, three key cytoskeletal proteins related to desmin, are organized in normal and desmin knock-out (K/O) mice. We show that in wild-type mice, synemin, paranemin, and plectin were colocalized with desmin in Z-disc-associated striations and at the sarcolemma. All three proteins were also present at the myotendinous junctions and in the postsynaptic area of motor endplates. In the desmin K/O mice the distribution of plectin was unaffected, whereas synemin and paranemin were partly affected. The Z-disc-associated striations were in general no longer present in between the myofibrils. In contrast, at the myotendinous and neuromuscular junctions synemin and paranemin were still present. Our study shows that plectin differs from synemin and paranemin in its binding properties to the myofibrillar Z-discs and that the cytoskeleton in junctional areas is particularly complex in its organization.

Effect of nitric oxide on electric and mechanical activities of gastric antral circular muscles in guinea pigs.

AIM: To study the effect of exogenous nitric oxide (NO) on electric and mechanical activities of gastric antral circular muscle in guinea pigs in vitro. METHODS: Mechanical and electric activities of gastric antral circular muscle in guinea pigs were recorded simultaneously. RESULTS: Sodium nitroprusside (SNP, 0.5 mumol.L-1), an NO donor, inhibited the frequency and amplitude of fast wave and spontaneous contraction of the strips (P < 0.01). SNP-induced inhibition was not blocked by tetrodotoxin, atropine, phentolamine, and propranolol (P > 0.05), but diminished by methylene blue (P < 0.01) and oxyhemoglobin (P < 0.01). CONCLUSION: Exogenous NO inhibits gastric antral myoelectric and mechanical activities in guinea pigs. The inhibitions are produced by NO acting on extracellular membrane and enhancing the level of cGMP.

Effects of nitroprusside, 3-morpholino-sydnonimine, and spermine on calcium-sensitive potassium currents in gastric antral circular myocytes of guinea pig.

AIM: To determine the effect of nitric oxide (NO) on calcium-activated potassium currents in antral circular myocytes of the guinea pig stomach. METHODS: The whole-cell patch-clamp techniques were used, and the cells were isolated by collagenase. Sodium nitroprusside (SNP), spermine, and 3-morpholino-sydnonimine (SIN-1) were used as nitric oxide donors. RESULTS: Outward potassium currents were remarkably inhibited by tetraethylammonium (TEA) 1 mmol.L-1. Charybdotoxin 200 nmol.L-1, a specific inhibitor of KCa channel, also greatly inhibited IK(Ca). NO donors remarkably increased IK(Ca) in guinea pig gastric antral circular myocytes with perforated patch-clamp techniques but not conventional whole-cell patch-clamp techniques. Spermine 100 mumol.L-1 increased IK(Ca) by 14% +/- 10% (at 60 mV, P < 0.01), SIN-1 200 mumol.L-1 increased IK(Ca) by 19% +/- 14% (at 60 mV, P < 0.01), and SNP 100 mumol.L-1 increased IK(Ca) by 24% +/- 13% (at 60 mV, P < 0.01) respectively. SIN-1- and SNP-induced increase of IK(Ca) was blocked by methylene blue 1 mumol.L-1. CONCLUSION: NO increases calcium-activated potassium currents in gastric antral circular myocytes of guinea pig, and the effect of NO on IK(Ca) may be mediated by cyclic GMP.

Effects of different kinds of stretch on voltage-dependent calcium current in antrial circular smooth muscle cells of the guinea-pig.

In order to elucidate the effect of membrane stretch on ionic currents, we employed the whole-cell patch-clamp technique to investigate the effects of different kinds of stretch on voltage-dependent calcium currents in antrial circular smooth muscle cells of the guinea-pig. The membrane stretch induced by superfusing the smooth muscle cells with hyposmotic bath solution enhanced voltage-operated calcium current and activated inward holding current. The increase in calcium current occurred within 1 minute of superfusion and the sustained inward holding current was slowly activated after prominent cell swelling. Voltage-dependent calcium currents (Ica) were significantly increased by membrane stretch which was induced by cell swelling and cell inflation, but was not affected by direct longitudinal stretch (110 130%) using two electrodes. The results suggest that the cell membrane stretch can increase voltage-dependent calcium channel activity and the effect of stretch on calcium channels was related to the membrane tension and/or the direction of membrane stretch.
