The chemotherapy of rodent malaria, XXXIX. Ultrastructural changes following treatment with artemisinine of Plasmodium berghei infection in mice, with observations of the localization of [3H]-dihydroartemisinine in P. falciparum in vitro.

Ultrastructural changes were followed in Plasmodium berghei after the treatment of the mouse host with a single 10 mg kg-1 dose of artemisinine (qinghaosu). After 30 minutes, changes in the limiting and other membranes of the parasite were seen, together with alterations in ribosomal organization and endoplasmic reticulum. No changes were noted in digestive vacuoles or pigment, but nuclear membrane blebbing developed after one hour and segregation of the nucleoplasm after three hours. Further degenerative changes with disorganization and death occurred from eight hours onwards. The morphological changes in ribosomes and endoplasmic reticulum correlate in time with the depression in protein synthesis observed in P. falciparum in vitro. Similarly, the onset of nucleoplasmic segregation correlates with the development of nucleic acid synthesis inhibition. Tritiated reduced drug was shown to be localized in parasite membranes, indicating that changes in membrane integrity might precede the early depression of protein synthesis. Membrane association of artemisinine may be related to its amphipathic characteristics and similarity in some respects to a sterol.

The study of human transfer factor.

In this paper, the study of human transfer factor is reported. We established a negative pressure dialysis method instead of ordinary dialysis for treatment of the crude leukocyte extract. Dialysate is rapidly obtained in only 5 h so that a large volume of preparation is easier to handle and the chance of contamination avoided. When human TF was incubated with human cord blood T-lymphocytes and pig lymphocytes, a very high biological activity on SRBC rosette enhancement and an increase in electrophoresis rate appeared. It suggests that these assays may be used as in vitro method of evaluation of TF activity. In our clinic, TF has been in clinical trials for 5 years and has now been administered to a large number of patients with a variety of diseases, in which cell-mediated immune responses have been compromised. We observe that TF has served an efficient immunopotentiating or immunomodulation agent.

Cancer in the gastric remnant in China--a review of 33 cases in China.

A review of 33 cases of cancer in the gastric remnant (CGR) including 6 cases in Xian is presented. The detection rate of CGR consisting of different interval groups was 3.16%. The average age was 52.4 years, and Male: female ratio was 10:1. The average interval from the previous operation to the detection was 13.9 years. The interval of the subgroup over 30 years of age at the first operation was shorter than that of the subgroup below 30 years. The highest incidence was found in anastomotic stoma (58.1%) and cardia (32.3%). Borrmann I was the most common pathological type. Confirmed diagnostic rate with fibergastroscope was 81.8%. It is imperative that physician should follow up the patients with stomach remnant. No matter what the results of gastrointestinal barium study may be, the patients should undergo fibergastroscopic examination. The pathogenesis of CGR is also discussed.
