[The use of the serum reference sample containing the antinuclear factor (homogeneous) to determine the working dilution of diagnostic fluorescing immunoglobulins against human IgG(H)].

A previously developed serum reference sample containing the antinuclear factor (homogeneous) in a standard dilution 1:40 was used to estimate the working dilution of commercial lots of diagnostic fluorescing immunoglobulins against human IgG (H) (IGF). Use of a working dilution of IGF in the diagnostic titer helps optimize the immunofluorescence test and save IGF.

[Experience in preserving the viability of Pseudomonas aeruginosa in the laboratory].

Experiments with 39 type collection strains and 25 newly isolated strains of P. aeruginosa have demonstrated the possibility of preserving the culture viability for a long time in sterile tap water at 6-8 degrees C. The initial morphologic, cultural, biochemical, and antigenic characteristics of the cultures are unchanged in sealed ampules for up to 1 year and in glass tubes corked with rubber corks for up to 8 months.

[The spectrum of extracellular proteins of staphylococci--the passport of a strain].

The possibility of detecting identical strains was analyzed by comparing extracellular protein spectra of Staphylococcus aureus strains from the collection of the L. A. Tarasevich Institute for Standardization and Control of Medical Preparations. Analysis of the findings confirms that S. aureus and other Staphylococci should be characterized by their protein spectra and this marker should be present in the certificate of the strain in collections. Protein spectrum determination is obligatory for production strains.

[The identification of lactose-negative enterobacteria].

Study of the morphologic and enzymatic characteristics of 10 strains of lactose-negative enterobacteria, sent to the L. A. Tarasevich State Institute for Control and Standardization as B. paracoli in order to determine their taxonomic status in accordance with the 1974 and 1984 Bergey classifications has referred 9 of these strains to Hafnia alvei.

[Tinctorial properties of microorganisms isolated from blood].

Cultures isolated from human and animal blood have been examined in tests with 3% KOH solution; their sensitivities to various concentrations of sulfanol have been tested in order to specify the tinctorial characteristics of these cultures; antibiotic sensitivity of this group of microorganisms has been studied with the use of discs. The findings evidence that gram-variable nonspore-carrying aerobic bacilli isolated from human and animal blood should be referred to gram-positive microorganisms; their antibiotic sensitivity has been detected. The tests employed in the study may be rationally used for the identification of polychromic bacteria isolated from the blood.

[Diagnostic meningococcal sera and their quality].

The findings evidence that preparation of highly active specific meningococcal sera depends on the selection of production strains and reference cultures for the assessment of their conformity to technological requirements. Meningococcal cultures selection conditions may be unified if standard immune sera are developed, characterized by stable titres of antibodies to group capsule antigens. Employment of such sera will improve the quality of commercial agglutinating sera.

[A comparative study of nutrient media for isolating Corynebacterium diphtheriae].

The diagnostic characteristics of 8 nutrient media most frequently employed at practical bacteriologic laboratories for the isolation of C. diphtheriae have been under study. Clauberg-2 medium and blood-tellurite agar have been found the most suitable for the diagnostic studies. Addition of extra ingredients to the standard recipe deteriorates the diagnostic potentialities of the media. Tinsdal's medium is characterized by the lowest diagnostic efficacy. The schemes of using these media are presented, with due consideration for their diagnostic potentialities.

[The therapeutic efficacy of a gonococcal vaccine].

The therapeutic efficacies of 2 gonococcal vaccines are compared. One of them is made from gonococcal strains selected for their antigenic characteristics, the other is a commercial vaccine prepared from a random sample of gonococcal strains. The studies have demonstrated a higher therapeutic efficacy of the former vaccine. This fact calls for the creation at production centers of permanent collections of antigenically full-value gonococcal cultures; such collections will help improve the gonococcal vaccine, standardize it, and enhance its biological activity.

[The fatty acid composition of Haemophilus and Pasteurella multocida cells as a phylogenetic marker].

When grown on meat-peptone agar with heated blood, different Haemophilus species (H. influenzae, H. parahaemolyticus, H. parasuis, H. pleuropneumoniae), including different H. influenzae serovars (a, b, c, d, e, f), and Pasteurella multocida have identical fatty acid composition, characterized by the prevalence of fatty acids with 16 carbon atoms, constituting about 70% and more of the total number of fatty acids, and a low level of fatty acids with 18 carbon atoms. P. multocida strains cultivated on meat-peptone agar with unheated blood have a greatly increased content of fatty acids with 18 carbon atoms, while the content of fatty acids with 16 carbon atoms is much lower. The identity of fatty acid composition under similar cultivation conditions, together with their similarity in other phenetic signs, is indicative of close phylogenic relationship between bacteria belonging to the genus Haemophilus and P. multocida.

[Lytic ability, in relation to gram-positive microorganisms, of a preparation isolated from a Pseudomonas lytica culture].

A lytic enzyme isolated from P. lytica was studied with respect to its effect on pathogenic grampositive and gramnegative organisms. All the grampositive organisms were lyzed by the enzyme to this or that extent. The cells of staphylococci were the most sensitive. The gramnegative organisms were resistant. It was suggested that the lytic enzyme could be used in preparing drugs for treating certain skin diseases caused by pathogenic staphylococci.

[Determination of the biological activity of Clostridium difficile toxins in in vivo and in vitro experiments].

The biological activity of the filtrates of 29 C. difficile strains was studied in vivo (suckling white mice) and in vitro (cell cultures of different species and origin). The action of the filtrates on the experimental models in vivo was evaluated from the cytotoxic effect index, while in vitro the intensity of the cytotoxic effect was evaluated from the percentage of dead cells in the monolayer. The results of the comparative determination of toxicity characteristics in vivo and in vitro demonstrated that cell cultures were more sensitive experimental models than suckling white mice. The use of cell cultures permitted the quantitative evaluation of the cytotoxic activity of the filtrates under study, as well as the detection of their cell-directed action at minimal concentrations.

[Rapid immunoenzyme analysis].

A variant of the immunoenzyme assay, reducing the duration of the assay procedure by 3 hours in comparison with the standard technique, is presented. This rapid variant is sufficiently sensitive and reproducible.

[Nonpathogenic Neisseria on the mucosa of the respiratory tract in healthy persons and patients with infectious-allergic bronchial asthma].

The comparative study of Neisseria strains, isolated from the bronchi and the pharynx of healthy persons and infectious asthma patients, was made. Nonpathogenic Neisseria isolated from bronchial asthma patients were represented by the same species as those in the pharyngeal flora of healthy persons. Only N. perflava occurred in the pharynx and the bronchi of infectious asthma patients twice as often as in healthy persons. Neisseria colonies in the R-form were shown to prevail in the cultures obtained by the inoculation of the smears taken from asthma patients.
