Experimental study of environmental tobacco smoke particles under actual indoor environment.

Environmental tobacco smoke (ETS) is a major source of human exposure to airborne particles. In order to provide more information necessary for human exposure investigations, the aim of the work presented here is to investigate experimentally the variation of the ETS particle concentration and size distribution under an actual indoor environment, in a room of 30 m3, using human smokers. The effect of number of cigarettes and brands of cigarettes, the effect of sampling location and the effect of ventilation rates were investigated. The results indicated little difference in the geometric mean diameter (GMD) of the ETS particles from those in background air. Under a ventilation rate of 0.03 m3/s, the concentration of the ETS particles reached a peak value at the sampling point shortly after completing the smoking process. The GMD first increased due to coagulation and diffusion deposition, and finalize decreased due to the effect of ventilation. Smoking two cigarettes at the same time would increase the initial concentration and led to an increase in GMD of the ETS particles. Two different brands of cigarette with different tar contents released ETS particles of different GMDs but similar particle concentrations. Spatial variation in particle concentration was obvious only in the first 600 s of the tests and tended to fade out subsequently. Stronger ventilation would reduce the concentration and GMD of the particles.

Reaction-controlled phase-transfer catalysis for propylene epoxidation to propylene oxide.

The epoxidation of olefins with H2O2 was performed with a tungsten-containing catalyst. This insoluble catalyst forms soluble active species by the action of H2O2, and when the H2O2 is used up, the catalyst precipitates for easy recycling. Thus, the advantages of both homogeneous and heterogeneous catalysts are combined in one system through reaction-controlled phase transfer of the catalyst. When coupled with the 2-ethylanthraquinone/2-ethylanthrahydroquinone redox process for H2O2 production, O2 can be used for the epoxidation of propylene to propylene oxide with 85% yield based on 2-ethylanthrahydroquinone without any co-products. This approach avoids the problematic co-products normally associated with the industrial production of propylene oxide.

A map of human genome sequence variation containing 1.42 million single nucleotide polymorphisms.

We describe a map of 1.42 million single nucleotide polymorphisms (SNPs) distributed throughout the human genome, providing an average density on available sequence of one SNP every 1.9 kilobases. These SNPs were primarily discovered by two projects: The SNP Consortium and the analysis of clone overlaps by the International Human Genome Sequencing Consortium. The map integrates all publicly available SNPs with described genes and other genomic features. We estimate that 60,000 SNPs fall within exon (coding and untranslated regions), and 85% of exons are within 5 kb of the nearest SNP. Nucleotide diversity varies greatly across the genome, in a manner broadly consistent with a standard population genetic model of human history. This high-density SNP map provides a public resource for defining haplotype variation across the genome, and should help to identify biomedically important genes for diagnosis and therapy.

SSAHA: a fast search method for large DNA databases.

We describe an algorithm, SSAHA (Sequence Search and Alignment by Hashing Algorithm), for performing fast searches on databases containing multiple gigabases of DNA. Sequences in the database are preprocessed by breaking them into consecutive k-tuples of k contiguous bases and then using a hash table to store the position of each occurrence of each k-tuple. Searching for a query sequence in the database is done by obtaining from the hash table the "hits" for each k-tuple in the query sequence and then performing a sort on the results. We discuss the effect of the tuple length k on the search speed, memory usage, and sensitivity of the algorithm and present the results of computational experiments which show that SSAHA can be three to four orders of magnitude faster than BLAST or FASTA, while requiring less memory than suffix tree methods. The SSAHA algorithm is used for high-throughput single nucleotide polymorphism (SNP) detection and very large scale sequence assembly. Also, it provides Web-based sequence search facilities for Ensembl projects.

Optical properties of Nannochloropsis sp and their application to remote estimation of cell mass.

The absorption and scattering coefficients and reflectance spectra of ultra-high density Nannochloropsis occulata cultures were investigated in detail to identify the optical properties of the cultures and devise algorithms for remote estimation of dry cell mass in ultra-high cell density cultures. High-spectral resolution measurements of apparent absorption and attenuation as well as reflectance from 400 to 900 nm were carried out in relation to the dry weight, cell count, and pigment concentration in outdoor cultures. Indices calculated as (R(NIR) - R(red))/(R(NIR) + R(red)) and R(NIR)/R(red), in which R(NIR) is reflectance in the range from 750 to 800 nm and R(red) is reflectance in the range 670-680 nm, were used for remote assessment of dry cell mass. Remote estimation in the range 1 to 8 g/L was accomplished with an error of less than 0.66 g/L. A different index, i.e., (R(NIR) - R(red)) was employed for estimation of cell-chlorophyll concentration. This is the first report of in vivo specific absorption coefficient of chlorophyll-a and specific scattering coefficient per dry algal weight of Nannochloropsis sp., providing a basis for remote monitoring of dense phytoplankton masses.

The correlation between immune rejection and osteoinduction of allogeneic bone grafting.

OBJECTIVE: To evaluate the relationship between the immune rejection and the osteoinductive potential of bone allograft. METHODS: Allogeneic and syngeneic fresh bone, autolyzed antigen-extracted bone, bone matrix gelatin and demineralized bone matrix were implanted into the muscle of mice, and immunological tests, histological observation and alkaline phosphatase assay were performed. RESULTS: Three and 6 weeks after implantation, all kinds of allogeneic implants activated immune rejection, among them, fresh bone induced the most vigorous immune rejection and bone matrix gelatin caused the weakest response. Allogeneic autolyzed antigen-extracted bone, bone matrix gelatin and demineralized bone matrix inhibited proliferation of the lymphocytes in vitro and bone matrix gelatin had the most powerful inhibiting effect. Both allogeneic and syngeneic autolyzed antigen-extracted bone, bone matrix gelatin, and demineralized bone matrix induced heterotopic osteogenesis in vivo and bone matrix gelatin had the best osteoinductive capacity. CONCLUSION: There is a negative correlation between immune rejection to bone allograft and osteoinductive capacity of the graft.

[Immunological comparison of differently treated allografts of bone].

The immunnologic rejection induced by differently treated allografts of bone was compared. Methods Fresh bone (FB), autolyzed antigen-free bone (AAA), bone matrix gelatin (BMG), demineralized bone matrix (DBM) were implanted into the muscle pouch of mice, then, the immunological tests and alkaline phosphatase assay were conducted. Results Allogeneic FB induced the highest level of serum antibody in the host and stimulated lymphocytes proliferation remarkably in vitro; in contrast, AAA, BMG and DBM caused low titer of antibody and inhibited lymphocytes reproduction in vitro. Conclusions. Immunological rejection restrained osteogenesis of the bone implant, whereas the osteoinductive substance of bone suppressed immunological reaction.

Molecular characterization of cloned variants of Coxiella burnetii isolated in China.

To study the molecular properties of Coxiella burnetii phase variants we cloned the phase variants of C. burnetii Qiyi (CBQY) strain by the red plaque technique. Three cloned strains, CBQYIC3 (phase I), CBQYIIC7 (phase II) and CBQYIIC5 (semirough-phase) were analysed by SDS-PAGE, immunoblot assay, plasmid isolation and agarose gel electrophoresis of DNA restriction fragments. The results suggest that the unique phase-dependent substance is a lipopolysaccharide and that most protein components of phase I and phase II cells are shared. No significant differences of DNA restriction fragments were found between clonal isolates of phase I and phase II C. burnetii CBQY strains. A plasmid of approximately 56 Kb was isolated from both phase I and phase II variants indicating that phase variation probably could not be attributed to its presence or absence.

Human nails as a biomarker of arsenic exposure from well water in Inner Mongolia: comparing atomic fluorescence spectrometry and neutron activation analysis.

Arsenic (As) is found naturally in the geological strata within the Ba Men Region of Inner Mongolia, China. A study was conducted to compare the total As measurements from two analytical techniques: instrumental neutron activation analysis (INAA) and atomic fluorescence spectrometry (AFS), and to verify nails as an exposure biomarker in this population. In 1999, nail and water samples were collected in a pilot study. Fingernails and toenails were pooled from 32 participants and analysed for total As by both INAA and AFS. Mean nail As values were 14.8+/-2.4 and 19.4+/-2.8 microg g-1 (+/-SEM) for INAA and AFS, respectively. Results from these two methods were significantly correlated (r=0.93, p<0.0001). In 2000, a second study was conducted and INAA was used to measure total As in toenails from 314 Ba Men residents. Well water samples were collected from 121 households and analysed by AFS. A significant correlation was observed between toenail and well water As (r=0.84, p<0.0001). Based on the results, INAA was significantly correlated with AFS and proved to be a reliable measure of nail As levels. In this population, toenail samples are a useful internal As exposure biomarker from drinking water sources.
