Suppression of IgE antibody production in SJL mice. V. Effect of irradiation and adult thymectomy on the suppression of IgE antibody production in SJL mice.

Anti-DNP IgE antibody production was low and transient in SJL mice which were immunized with 1 microgram DNP-Nb and 1 mg A1(OH)3. The immunized SJL mice were irradiated (60-540 R) 1 day after challenge. A dose higher than 180 R induced enhancement of anti-DNP IgE antibody production as compared to nonirradiated control mice, suggesting the existence of irradiation-sensitive suppressor cells. Anti-DNP IgE antibody production was suppressed when immunized and irradiated SJL mice were injected with spleen cells from adult-thymectomized SJL mice. The donors of the spleen cells were thymectomized 2 or 4 months previously, and this suggests that the suppressor cells from unprimed mice are long-living T cells.

Studies on IgE production in mice. I. Spontaneous suppression of IgE production in SJA/9 mice.

IgE levels after Nippostrongylus brasiliensis infestation were high in all strains of mice examined except in the SJA/9 strain. However, surface IgE levels on B cells were not different from those observed in the SJL strain. Most surface IgE-bearing cells also had surface IgM molecules. We suggest that the surface IgE-bearing cells are precursors of IgE-producing cells. In adoptive transfer, hapten-specific IgE antibody was produced when hapten-primed cells from SJA/9 mice were transferred together with cells from SJL donors infested with N. brasiliensis but not when the cells were from SJA/9 donors. It is suggested that the lack of collaboration between hapten-primed B cells and cells from infested mice is a genetic trait of the SJA/9 strain.

Regulation of murine IgE production: importance of a not-yet-described T cell for IgE secretion demonstrated in SJA9 mice.

To investigate why SJA9 mice do not produce IgE antibody, we took advantage of the adoptive transfer method and the carrier effect. Antibody-producing B cells (donor I) were primed with 2,4-dinitrophenylated ovalbumin (DNP-OVA) and carrier-specific T cells (donor II) were primed with keyhole limpet hemocyanin (KLH). The recipients were challenged with DNP-KLH. When 0.2 micrograms of DNP-OVA was used and the donor II cells were from SJL mice the recipient produced anti-DNP IgE. When donor I cells were from the SJL strain and donor II cells were from the SJA9 strain the recipient produced anti-DNP IgE. This pointed to a T-cell defect (absent or functionally inactive T epsilon o cell) in the SJA9 strain, which could be corrected either by injecting unprimed T cells from the SJL strain into the SJA9 donor I or by infection with Nippostrongylus brasiliensis, a hookworm, after priming. It seems that in the SJA9 strain in the absence of functioning T epsilon o cells isotype-specific (IgE) suppression, described previously in the SJL strain, is more pronounced, because when the SJA9 donor I was primed with 1 microgram of DNP-OVA only trace amounts of anti-DNP IgE were produced with SJL donor II cells.

Half-life of murine IgE antibodies in the mouse.

The half-life of intravenously injected murine anti-DNP IgE antibody was determined in recipient mice by sequential bleeding and was found to be 12 h. Intradermally injected IgE antibody persisted much longer, the half-life was 6 days.

Immune response to Trichomonas vaginalis. IV. Immunochemical and immunobiological analyses of T. vaginalis antigen.

The immune response of patients with trichomoniasis vaginitis to Trichomonas vaginalis was examined by using passive hemagglutination assay and skin reactions. Trichomonas-specific immune responses could hardly be detected in the patients by these assay methods. In order to develop a simple and reliable assay for demonstration of T. vaginalis-specific immune responses in the patients, proliferation responses of peripheral mononuclear cells from the patients were examined. Lymphocytes obtained from the patients were shown to incorporate 3H-methyl-thymidine when stimulated with T. vaginalis antigen in vitro. Furthermore, immunochemical analysis of T. vaginalis antigen has been done by Sephadex chromatography and sodium dodecyl sulfate gel electrophoresis. The data revealed that the murine and human IgM antibodies specific to T. vaginalis recognize various antigens with a wide range of molecular weights (between 13,000 and 100,000 daltons), while human and murine IgG antibodies recognize molecules with a much narrower range of molecular weights (50,000-100,000 daltons).

Presence of an intrastrain cross-reactive idiotype on A/J antibodies of the IgE class specific for the p-azophenylarsonate group.

An intrastrain cross-reactive idiotype, CRIA, is associated with a large proportion of the anti-p-azophenylarsonate (anti-Ar) antibodies of A/J mice. The present experiments, in which the methods of direct and reverse passive cutaneous anaphylaxis (RPCA) were used, indicate that IgE anti-Ar antibodies are produced in A/J mice upon stimulation with a protein-Ar conjugate and that a large proportion of these antibodies express CRIA. The use of monoclonal anti-CRIA for RPCA eliminated the strong nonspecific reactions previously observed. The results provide a basis for studying factors that regulate the switch to IgE biosynthesis during an immune response.

Biological activities of antitrinitrophenyl and antidinitrophenyl mouse monoclonal antibodies.

The biological activities of mouse IgG1, IgG2a, IgG2b, and IgE monoclonal were investigated. As has been shown with conventional antibodies, monoclonal IgG1 and IgE are the two classes capable of sensitizing the mouse for anaphylactic reactions, IgE sensitizes the rat, and IgG2a sensitizes the guinea pig. What has not been seen with conventional antibodies is that large amounts of monoclonal IgG2a or IgG2b can sensitize the mouse, very large amounts of monoclonal IgE can sensitize the guinea pig, and very large amounts of IgG1 and IgG2a can sensitize the rat. One of the IgG1 monoclonal antibodies was capable of fixing complement by the classical pathway.

Recent insight into class-specific properties of murine immunoglobulins obtained with the help of monoclonal antibodies.

BALB/c and C57BL/6 mice were immunized with monoclonal anti-DNP IgE, obtained by fusion of PX63AG8-6-5-3 cells with spleen cells from immunized C57BL/6 or BALB/c mice, respectively. With the antiallotype sera thus produced the allotype of IgE (i.e. 7) could be defined since BALB/c is of the Igh-a and C57BL/6 of the Igh-b allotype. The antiallotype 7b does not cross-react with other allotypes. Antiallotype 7a recognizes in addition to allotype 7a also allotypes 7j, 7d, and 7e as expected. Both the complement fixing and the sensitizing activities of murine anti-TNP and anti-DNP monoclonal antibodies were examined. The minimal amount/ml for complement fixation was 0.46 micrograms from one of the IgG1 antibodies, 0.46 from IgG2a, 12.5 ng from IgM, 0.7 micrograms from IgG3. Complement fixation by IgG1 was unexpected. The minimal amount/ml for mouse PCA was 0.5-0.7 micrograms from IgG1, 1.44-2.3 micrograms from IgG2a, 5.6 micrograms from purified IgG2b and about 15 ng from IgE. Sensitization by IgG2a and IgG2b were new findings. Minimal amount/ml for guinea pig PCA was 0.23 micrograms from IgG2a. Minimal amount/ml for rat PCA was 2 ng from IgE.

Immune response to Trichomonas vaginalis. I. Genetic regulation of IgE antibody response to T. vaginalis in mice.

The antigenicity of Trichomonas vaginalis to elicit IgE antibody was examined in mice. The soluble antigen was prepared by sonication of in vitro cultured T. vaginalis lines. BALB/c mice were immunized by various doses of the antigen with alum followed by a booster injection on day 14 after primary injection of the protozoan antigen. The highest IgE antibody titer was found in mice immunized with 30 micrograms of the antigen. The IgE antibody response to Trichomonas antigen is regulated by at least two immune response genes: one localized in the I region (K to I-B subregions) of chromosome 17, the other in a not yet identified chromosome.

Allelic polymorphism of murine IgE controlled by the seventh immunoglobulin heavy chain allotype locus.

Two alloantisera against hybridoma-derived IgE detected allotypic determinants expressed on the murine epsilon chain. An antiserum raised in BALB/c mice against monoclonal IgE of C57BL/6 origin reacted exclusively with IgE of strains having Igh-1b (IgG2 a) allotype. The second antiserum, C57BL/6 anti-BALB/c monoclonal IgE, reacted with IgE of strains having Igh-1a, Igh-1d, Igh-1e and Igh-1j allotypes. The genetic studies of (BALB/c x C57BL/6)F1 and backcross F2 animals indicated that the locus controlling the IgE allotype is linked to the Igh-1 locus. This was further confirmed by the possession of respective IgE allotypes by Igh-C congenic mice, BALB/c and BAB-14, C3H.SW/Hz and CWB/Hz. Thus, the allotype detected on the epsilon chain is controlled by the seventh murine immunoglobulin allotype locus, and should be designated as the Igh-7 allotype.

Production of monoclonal mouse IgE antibodies with DNP specificity by hybrid cell lines.

IgE anti-DNP antibody-secreting hybridomas were obtained by polyethylene glycol fusion of murine myeloma cells with spleen cells from mice, primed with DNP-KLH and challenged with DNP-Nippostrongylus brasiliensis extract. The in vivo and in vitro continuously growing hybridomas are producing high amounts of monoclonal IgE anti-DNP antibodies, which are heat-labile, hapten-specific and have rat mast cell sensitizing capacity.
