Effects of ethanol intake on the immune system of guinea pigs.

Groups of guinea pigs were maintained with alcohol solutions not sufficient to result in liver dysfunction prior to and during investigations designed to determine whether ethanol would nonetheless impair cell-mediated or humoral immunity. No impairment of either was observed in any of the experimental groups.

Biologic activities of rabbit immunoglobulin G in relation to domains of the Fc region.

Biologic properties of antibodies are known to be mediated by the Fc portion of the H chains. In the present study such properties as complement fixation, cutaneous anaphylaxis, and macrophage cytophilia were examined in relation to the CH2 and CH3 domains of rabbit IgG. Fragments containing but one of these domains were prepared from plasmin and papain digests. Facb fragments of anti-DNP antibodies, together with the antigen DNP-BSA, were able to fix complement by the classical pathway, a result which implicates the CH2 domain; however, guinea pig Fab fragments directed to regions of the rabbit antibody molecule other than CH2 were able to inhibit complement fixation. Facb fragments were unable to mediate PCA or reverse PCA reactions in guinea pigs, nor were CH3 fragments active in tests of reverse PCA or inhibition of PCA. These results suggest that the entire Fc region is needed for cutaneous anaphylaxis. The ability to bind to guinea pig lung macrophages was studied with a rosette technique. Facb fragments were active whereas CH3 fragments failed to inhibit. It is suggested that although some effector functions of antibodies can be assigned to individual domains, others require the entire Fc region.

Further studies on the inheritance of responsiveness to pertussis HSF in mice.

Inheritance of responsiveness to histamine-sensitizing factor of pertussis bacilli (HSF) was investigated in (C3H/HeJ times C57BL/6J)F1 hybrids, backcross progeny of this hybrid to C57BJ/6J parent (C3H/HeJ times DBA/2J)F1 hybrids, and in backcross progeny of this hybrid to DBA/2J parent. It was found that transmission is not by virtue of a single autosomal dominant gene, as has been postulated. Rather, inheritance of responsiveness to HSF is far more complex, probably involving polygenic transmission.

PCA reactions with mouse antibodies in mice and rats.

Mouse IgE was titrated in rats. The sensitization period was 2 h. The results were consistent and corresponded to the titers obtained in young adults SJL mice using a sensitization period of 48 h. With longer sensitization periods in rats, higher antibody titers were obtained. The optimum sensitization period in rats was found to be 48 h. Old CFW mice are inadequate for mouse IgE titration. IgG1 will not give a PCA reaction in rats. IgG1 titers are higher in SJL or A/J mice than in young CFW mice, and markedly higher than in old CFW mice.

The humoral response to DNCB: its detection by different techniques.

Antibodies in the sera of guinea pigs immunized with DNCB were detected by Ouchterlony-type analyses and were estimated in weight units by the mABC procedure. Assays of these sera by means of passive cutaneous anaphylaxis, passive hemolysis, or the ABC-33 antigen-binding technique were essentially negative. The validity of the mABC analyses was established by comparison of results obtained in quantitative precipitation studies with sera of guinea pigs immunized with DNP-BGG. In these instances, also, the ABC-33 assays failed to detect all the anti-DNP immunoglobulins.

Enhancement of tumor growth by graft-versus-host reaction.

A graft-versus-host reaction was induced in (BALB/c x C5 7BL/6)F(1) mice by intravenous injection of BALB/c spleen cells. Six days later, the mice received subcutaneous grafts of a chemically induced BALB/c sarcoma. The growth of the tumor grafts was accelerated.

Independent segregation of the H-2 locus and the locus for responsiveness to histamine-sensitizing factor.

The inheritance of responsiveness to histamine-sensitizing factor of pertussis bacilli was examined in (SWR/J x CBA/J)F(1) x CBA/J backcross mice. We found that about half of the backcross mice inherited this responsiveness in both sexes. There was no linkage between the inheritance of responsiveness to histamine-sensitizing factor and the H-2(q) locus from the sensitive SW/J progenitor.
