Intracisternally localized bacterial DNA containing CpG motifs induces meningitis.

Unmethylated CpG motifs are frequently found in bacterial DNA, and have recently been shown to exert immunostimulatory effects on leukocytes. Since bacterial infections in the CNS will lead to local release of prokaryotic DNA, we wanted to investigate whether such an event might trigger meningitis. To that end, we have intracisternally injected mice and rats with bacterial DNA and oligonucleotides containing CpG motifs. Histopathological signs of meningitis were evident within 12 h and lasted for at least 14 days, and were characterized by an influx of monocytic, Mac-3(+) cells and by a lack of T lymphocytes. To study the mechanisms whereby unmethylated CpG DNA gives rise to meningitis, we deleted the monocyte/macrophage population leading to abrogation of brain inflammation. Also, interaction with NF-kappaB using antisense technology led to down-regulation of proinflammatory cytokine production and frequency of meningitis. Furthermore, specific interactions with vascular selectin expression and inhibition of NO synthase led to a significant amelioration of meningitis, altogether indicating that this condition is dependent on macrophages and their products. In contrast, neutrophils, NK cells, T/B lymphocytes, IL-12, and complement system were not instrumental in meningitis triggered by bacterial DNA containing CpG motifs. This study proves that bacterial DNA containing unmethylated CpG motifs induces meningitis, and indicates that this condition is mediated in vivo by activated macrophages.

Staphylococcal peptidoglycans induce arthritis.

Staphylococcus aureus is one of the most important pathogens in septic arthritis. To analyse the arthritogenic properties of staphylococcal peptidoglycan (PGN), highly purified PGN from S. aureus was intra-articularly injected into murine joints. The results demonstrate that PGN will trigger arthritis in a dose-dependent manner. A single injection of this compound leads to massive infiltration of predominantly macrophages and polymorphonuclear cells with occasional signs of cartilage and/or bone destruction, lasting for at least 14 days. Further studies showed that this condition is mediated by the combined impact of acquired and innate immune systems. Our results indicate that PGN exerts a central role in joint inflammation triggered by S. aureus.

[Effect of NOS positive neurons in rats with learning-remembering barrier].

To study the variation of nitric oxide synthase (NOS) neurons in rats with learning-remembering barrier by immunohistochemistry and histochemistry. The number of NOS neurons decreased apparently in hippocampus CA1-4 of rats with learning-remembering barrier, there were lots of new NOS neurons in dentate gyrus granular cell grays and pear-shaped zone's cortex. It is suggested that NOS neurons of hippocampus exert very important action in rat's learning remembering; new NOS neurons in pear-shaped zone cortex and dentate gyrus granular cell grays may be a compensative reaction of the learning-remembering barrier, and they might exert important function in following recovery of learning-remembering function.

Electroencephalogram analysis using fast wavelet transform.

The continuous wavelet transform is a new approach to the problem of time-frequency analysis of signals such as electroencephalogram (EEG) and is a promising method for EEG analysis. However, it requires a convolution integral in the time domain, so the amount of computation is enormous. In this paper, we propose a fast wavelet transform (FWT) that the corrected basic fast algorithm (CBFA) and the fast wavelet transform for high accuracy (FWTH). As a result, our fast wavelet transform can achieve high computation speed and at the same time to improve the computational accuracy. The CBFA uses the mother wavelets whose frequencies are 2 octaves lower than the Nyquist frequency in the basic fast algorithm. The FWT for high accuracy is realized by using upsampling based on a L-Spline interpolation. The experimental results demonstrate advantages of our approach and show its effectiveness for EEG analysis.

Effect of the CYP2C19 oxidation polymorphism on fluoxetine metabolism in Chinese healthy subjects.

AIMS: The study was designed to investigate whether genetically determined CYP2C19 activity affects the metabolism of fluoxetine in healthy subjects. METHODS: A single oral dose of fluoxetine (40 mg) was administrated successively to 14 healthy young men with high (extensive metabolizers, n=8) and low (poor metabolizers, n = 6) CYP2C19 activity. Blood samples were collected for 5-7 half-lives and fluoxetine, and norfluoxetine were determined by reversed-phase high performance liquid chromatography. RESULTS: Poor metabolizers (PMs) showed a mean 46% increase in fluoxetine peak plasma concentrations (Cmax, P < 0.001), 128% increase in area under the concentration vs time curve (AUC(0, infinity), P < 0.001), 113% increase in terminal elimination half-life (t(1/2)) (P < 0.001), and 55% decrease in CLo (P < 0.001) compared with extensive metabolizers (EMs). Mean +/- (s.d) norfluoxetine AUC(0, 192 h) was significantly lower in PMs than that in EMs (1343 +/- 277 vs 2935 +/- 311, P < 0.001). Mean fluoxetine Cmax and AUC(0, infinity) in wild-type homozygotes (CYP2C19*1/CYP2C19*1) were significantly lower than that in PMs (22.4 +/- 3.9 vs 36.7 +/- 8.9, P < 0.001; 732 +/- 42 vs 2152 +/- 492, P < 0.001, respectively). Mean oral clearance in individuals with the wild type homozygous genotype was significantly higher than that in heterozygotes and that in PMs (54.7 +/- 3.4 vs 36.0 +/- 8.7, P < 0.01; 54.7 +/- 3.4 vs 20.6 +/- 6.2, P < 0.001, respectively). Mean norfluoxetine AUC(0, 192 h) in PMs was significantly lower than that in wild type homozygotes (1343 +/- 277 vs 3163 +/- 121, P < 0.05) and that in heterozygotes (1343 +/- 277 vs 2706 +/- 273, P < 0.001), respectively. CONCLUSIONS: The results indicated that CYP2C19 appears to play a major role in the metabolism of fluoxetine, and in particular its N-demethylation among Chinese healthy subjects.

Interleukin-6 (IL6) and cellular response to facial nerve injury: effects on lymphocyte recruitment, early microglial activation and axonal outgrowth in IL6-deficient mice.

Nerve injury triggers numerous changes in the injured neurons and surrounding non-neuronal cells. Of particular interest are molecular signals that play a role in the overall orchestration of this multifaceted cellular response. Here we investigated the function of interleukin-6 (IL6), a multifunctional neurotrophin and cytokine rapidly expressed in the injured nervous system, using the facial axotomy model in IL6-deficient mice and wild-type controls. Transgenic deletion of IL6 caused a massive decrease in the recruitment of CD3-positive T-lymphocytes and early microglial activation during the first 4 days after injury in the axotomized facial nucleus. This was accompanied by a more moderate reduction in peripheral regeneration at day 4, lymphocyte recruitment (day 14) and enhanced perikaryal sprouting (day 14). Motoneuron cell death, phagocytosis by microglial cells and recruitment of granulocytes and macrophages into injured peripheral nerve were not affected. In summary, IL6 lead to a variety of effects on the cellular response to neural trauma. However, the particularly strong actions on lymphocytes and microglia suggest that this cytokine plays a central role in the initiation of immune surveillance in the injured central nervous system.

Pharmacokinetics of sertraline in relation to genetic polymorphism of CYP2C19.

OBJECTIVE: Our objective was to evaluate the relationship between the disposition of sertraline and the presence of the CYP2C19 gene and to define the contribution of cytochrome P450 2C19 (CYP2C19) to sertraline N-demethylation. METHODS: A single oral 100-mg dose of sertraline was administered to 6 subjects who were extensive metabolizers and 6 subjects who were poor metabolizers recruited from 77 healthy Chinese volunteers whose genotypes were predetermined by polymerase chain reaction-based amplification, followed by restriction fragment length polymorphism analysis. Phenotypes were determined by use of the omeprazole metabolic rate. The plasma concentrations of sertraline and desmethylsertraline were determined by gas chromatography with electron-capture detection. RESULTS: Six poor metabolizers with m1 mutation had area under the plasma concentration versus time curve (AUC(0-infinity)) values (983.6 +/- 199.3 microg x h/L versus 697.6 +/- 133.0 microg x h/L; P <.05) and terminal elimination half-life values of sertraline (35.5 +/- 5.6 hours versus 23.5 +/- 4.4 hours; P <.01) that were significantly higher than the values in 6 extensive metabolizers who were either homozygous or heterozygous for CYP2C19*1. The oral clearance of sertraline in poor metabolizers (105.3 +/- 19.4 L/h) was significantly lower than that of extensive metabolizers (148.4 +/- 28.6 L/h). The area under the concentration-time curve from 0 to 144 hours and the maximum plasma concentration of desmethylsertraline in poor metabolizers were significantly lower than the values of extensive metabolizers (627.6 +/- 203.8 microg x h/L versus 972.1 +/- 270.3 microg x h/L; P <.05; and 23.6 +/- 6.5 nmol/L versus 32.4 +/- 8.2 nmol/L; P <.01; respectively). CONCLUSIONS: The polymorphic CYP2C19 appears to be a major enzyme involved in the N-demethylation of sertraline, and both extensive and poor metabolizers had marked differences in the disposition of sertraline.

[Optional growth medium and conditions for mass production of Pandora delphacis mycelia in submerged culture].

The entomophthoraceous fungi are important microbial control agents for insect control but their mass production is usually difficult and expensive. To produce in large quantity the mycelia of the entomophthoraceous fungus, Pandora delphacis (isolate F95129), this study was aimed at replacing expensive components of liquid medium that is usually used in laboratory only with cheap materials easily available. Based on comparative experiments with primary components of several media designed and optional conditions for submerged culture, an appropriate medium was recognized that included(per liter) 10 g of homemade yeast extract, 10 g industrial fish meal, 10 g wheat bran, 15 g corn meal, 1.0 g KH2PO4, 3.0 g NH4NO3, and 0.25 g MgSO(4).7H2O. The optional conditions for submerged culture was 25-30 degrees C, initial pH 6.5, and 40% (V/V) of flask filled with the medium and 10% of initial inoculum (liquid culture containing mycelia) for inoculation. Using the selected medium and the conditions considered, 48 h culture resulted in a considerably high yield of dry mycelia(> 25 mg/mL) with desirable capacity of spore production.

[Relationship between EGG and the dynamic process of motion sickness induced by optokinetic vection].

OBJECTIVE: To investigate the correlation between Electrogastrography (EGG) and the occurrence of nausea or/and vomit as well as the development of motion sickness (MS) induced by optokinetic stimulation (OPS) and to provide a clue and method for prediction and prevention of MS. METHOD: Twenty male subjects, aged from 18 to 25 years, performed the test only once with the optokinetic drum rotated around a vertical axis at the speed of 60 degrees/s for 15 min. Nausea and EGG signals were observed and recorded. RESULT: (1)Both the increment of tachygastric percentage and the decrement of primary wave percentage of EGG in eleven subjects with nausea were significantly larger than those in nine subjects with no nausea (both P < 0.001). (2)The circular vection preceded the changes of EGG in eleven subjects with nausea while the MS scores in eleven subjects with nausea were higher than those in nine subjects with no nausea (P < 0.001). (3)During the rotation of drum, the decrement of primary wave percentage or the increment of tachygastria percentage also preceded the occurrence of nausea in eleven subjects with nausea (both P < 0.01). CONCLUSION: (l)The changes of EGG in subjects with nausea were significantly greater than those of EGG in subjects with no nausea. (2)There was a close relationship between circular vection or illusory self-rotation and EGG changes as well as the occurring of nausea induced by OPS. (3)Changes of EGG preceded the nausea of MS induced by OPS.

Effects of CYP2C19 genotype and CYP2C9 on fluoxetine N-demethylation in human liver microsomes.

AIM: The present study was designed to define the kinetic behavior of fluoxetine N-demethylation in human liver microsomes and to identify the isoforms of cytochrome P-450 (CYP) involved in this metabolic pathway. METHODS: The kinetics of Ne formation of norfluoxetine was determined in human liver microsomes from six genotyped CYP2C19 extensive metabolizers (EM). The correlation studies between the fluoxetine N-demethylase activity and various CYP enzyme activities were performed. Selective inhibitors or chemical probes of various cytochrome P-450 isoforms were also employed. RESULTS: The kinetics of norfluoxetine formation in all liver microsomes were fitted by a single-enzyme Michaelis-Menten equation (mean Km=32 micromol/L+/-7 micromol/L). Significant correlations were found between N-demethylation of fluoxetine at both 25 micromol/L and 100 micromol/L and 3-hydroxylation of tolbutamide at 250 micromol/L (r1=0.821, P1=0.001; r2=0.668, P2=0.013), respectively, and S-mephenytoin 4'-hydroxylase activity (r=0.717, P=0.006) at high substrate concentration of 100 micromol/L. S-mephenytoin (SMP) (a CYP2C19 substrate) at high concentration and sulfaphenazole (SUL) (a selective inhibitor of CYP2C9) substantially inhibited norfluoxetine formation. The reaction was minimally inhibited by coincubation with chemical probe, inhibitor of CYP3A4 (triacetyloleandomycin, TAO). The inhibition of fluoxetine N-demethylation at high substrate concentration (100 micromol/L) was greater in PM livers than in EM livers (73 % vs 45 %, P < 0.01) when the microsomes were precoincubated with SUL plus TAO. CONCLUSION: Cytochrome P-450 CYP2C9 is likely to be a major CYP isoform catalyzing fluoxetine N-demethylation in human liver microsomes at a substrate concentration close to the therapeutic level, while polymorphic CYP2C19 may play a more important role in this metabolic pathway at high substrate concentration.

Role of cytochrome P450 in estradiol metabolism in vitro.

AIM: Catechol estrogens and 16alpha-hydroxy estrogen are important metabolites that cause carcinogenesis. This study was aimed to stud y the role of cytochrome P450 in estradiol metabolism. METHODS: The estradiol metabolites were determined with HPLC-ECD. Correlation of estradiol metabolites production between cytochrome P450 activity, the inhibitory effect of specific inhibitors and enzyme catalyzing kinetics were studied in cDNA-expressed P450 or human liver microsomes. RESULT: CYP1A2, CYP3A4, and CYP2C9 catalyze the estradiol 2-hydroxylation. CYP2C9, CYP2C19, and CYP2C8 have high activity in catalyzing 17beta-hydroxy dehydrogenation in cDNA expressed P450, but CYP1A2 is the most important enzyme in catalyzing estradiol 2-hydroxylation. Using furafyllin and troleandomycin to inhibit CYP1A2 and CYP3A4 in liver microsomes, it was found that the 2-hydroxylation had been inhibited about the same amount. This result suggests that in human liver microsomes CYP1A2 and CYP3A4 play an important role in 2-hydroxy estradiol formation. At low substrate concentration, 17beta -hydroxy dehydrogenation dominated the estradiol metabolism, but at high substrate concentration, 2-hydroxylation exceeded 17beta-hydroxy dehydrogenation to become the important mechanism. CONCLUSION: CYP1A2 and CYP3A4 are two important enzymes catalyzing the main estradiol 2-hydroxylation metabolism pathway at high substrate concentrations. 17beta-hydroxy dehydrogenation is the main metabolism pathway at low concentrations, and CYP2C9, CYP2C19, and CYP2C8 may have high catalyzing activity.

Interindividual variations in levels and activities of cytochrome P-450 in liver microsomes of Chinese subjects.

AIM: To compare the level and/or activity of several important cytochrome P-450 (CYP) enzymes in liver microsomes prepared from different Chinese subjects. METHODS: Individual CYP contents, including CYP1A2, CYP2C9, and CYP3A4, in liver microsomes of 17 Han, 17 Zhuang, and 8 Miao subjects were determined by using Western blot analysis and densitometric scanning. The substrates for measuring the activity of individual CYP in vitro included phenacetin, tolbutamide, debrisoquine, and omeprazole. RESULTS: There was a large interindividual variability in the content and activity of CYP1A2, 2C9 and 3A4. And the activity of CYP2D6 also varied greatly between individual samples. CYP3A4 (32 %) is the most abundant CYP in Chinese liver microsomes, and the levels of CYP2C9 (19 %) and CYP1A2 (16 %) were also considerable. No clear ethnic, sex- and age-related differences in individual CYP content and catalytic activity were detected in 42 Chinese liver samples, except that there were somewhat ethnic and sex-related differences in the content and activity of CYP1A2. Good correlation between enzyme protein content and activity was found for CYP1A2, 2C9 and 3A4. CONCLUSION: Our results may provide useful information for the study of drug metabolism by liver microsomes in Chinese.

[Effects of adaptive changes of vestibular system on cardiovascular regulation and orthostatic tolerance].

Astronauts may experience Space Adaptation Syndrome at the primary period in spaceflight. It is widely accepted that it is mainly caused by vestibular adaptive changes. The reverse changes of adaptation present after the return from weightlessness to normal 1 G. It may cause astronauts' postural instability and orthostatic intolerance. Evidences from animal model and some data from humans suggest that the vestibular system have influences on the sympathetic nervous system and cardiovascular control. The inputs that appear to be critical in producing these responses come from otolith receptors. The important medical problem--orthostatic intolerance in space medicine is mainly caused by cardiovascular abnormal control. It is possible that vestibular adaptive changes under microgravity may have its influence on the cardiovascular function and orthostatic intolerance.

[Effect of Astragalus injection on platelet function and plasma endothelin in patients with early stage diabetic nephropathy].

OBJECTIVE: To study the therapeutic effect of Astragalus injection (AI) in treating early stage diabetic nephropathy (DN) patients. METHODS: The total of 136 early diabetic nephropathy patients were randomly divided into two groups, 50 cases in the conventional treated group and 86 in the AI treated group, the therapeutic course being 3 weeks. Levels of plasma endothelin-1 (ET-1), 24 hrs urinary albumin excretion rate (uAER), and platelet granule membrane protein (GMP-140), 6-keto-prostaglandin F1 alpha(6-keto-PGF1 alpha), and thromboxane B2(TXB2) before and after treatment were determined by radioimmunoassay (RIA) and enzyme-linked immunosorbent assay (ELISA) respectively. Moreover, the above-mentioned criteria in 26 healthy subjects were also measured for control. RESULTS: The plasma ET-1, GMP-140, TXB2 and uAER levels in DN patients were higher, but 6-keto-PGF1 alpha level was lower than those in healthy subjects. The above elevated criteria in DN patients could be lowered by AI treatment. CONCLUSION: The pathogenesis and development of DN might be closely associated with the changes of plasma ET-1 level and platelet function. Astragalus could improve the above-mentioned changes in patients of early stage DN.

[Study on allele-specific diagnostic PCR of the traditional Chinese medicines of the deers].

AIM: To develop a convenient and accurate method of DNA molecular marker for the identification of traditional Chinese medicines made of deers, consisting of pilose antler, penis and testis, tendon and foetus. METHODS: Based on the analysis of DNA sequence of mitochondrial Cyt b gene from original animals of both genuine crude drugs, Cervus nippon and Cervus elaphus, and adulterants, a pair of allele-specific primers named as ILu01-L and ILu01-H were designed for distinguishing geniune crude drugs of deers from their adulterants. RESULTS: The results of diagnostic PCR annealing at 64 degrees C for original animals showed that a 365 bp fragment was only amplified from DNA templates of Cervus nippon and Cervus elaphus. For the identification of medicinal materials total of 43 samples from 6 packages were tested under the same reaction conditions except for DNA templates extracted from these crude drugs. Only 9 samples mentioned above was shown to generate positive amplificon. The result indicate that of 8 samples from 1 package of pilose antler and only 1 sample of deer tendon was genuine crude drug. After that, 3 amplified fragments selected randomly were performed with sequencing analysis with the purpose of verifying the results from diagnostic PCR. Data from sequencing confirmed the reliability of diagnostic PCR identification. CONCLUSION: The diagnostic primers designed in the present study were highly specific for Cervus nippon and Cervus elaphus, and they could be used for the authentication of traditional Chinese medicines made from the deer. The quality of the crude drugs of the deer in the current market is a problem and more effective quality control for these traditional Chinese medicines is urgently needed.
