Ultrasound-promoted preparation of disteryl ethers catalyzed by montmorillonite K 10.

In the presence of montmorillonite K 10, 5(6)-unsaturated sterols (1) were heated under ultrasound at 45 degrees C in dichloromethane for 2-5.5 h to provide 3 beta,3'beta-5(6)/5'(6')-unsaturated disteryl ethers (2) in 35-79% yield. Meanwhile, 3 alpha,3'alpha-diandrost-5-en-17-one-3-yl ether (3a) and 3 alpha,3'alpha-dipregn-5-en-20-one-3-yl ether (3b) were also obtained as by-products from 3 beta-hydroxyl-androst-5-en-17-one (1a) and 3 beta-hydroxyl-pregnan-5-en-20-one (1b) respectively.

Both N-terminal myosin-binding and C-terminal actin-binding sites on smooth muscle caldesmon are required for caldesmon-mediated inhibition of actin filament velocity.

It has been suggested that the tethering caused by binding of the N-terminal region of smooth muscle caldesmon (CaD) to myosin and its C-terminal region to actin contributes to the inhibition of actin-filament movement over myosin heads in an in vitro motility assay. However, direct evidence for this assumption has been lacking. In this study, analysis of baculovirus-generated N-terminal and C-terminal deletion mutants of chicken-gizzard CaD revealed that the major myosin-binding site on the CaD molecule resides in a 30-amino acid stretch between residues 24 and 53, based on the very low level of binding of CaDDelta24-53 lacking the residues 24-53 to myosin compared with the level of binding of CaDDelta54-85 missing the adjacent residues 54-85 or of the full-length CaD. As expected, deletion of the region between residues 24 and 53 or between residues 54 and 85 had no effect on either actin-binding or inhibition of actomyosin ATPase activity. Deletion of residues 24-53 nearly abolished the ability of CaD to inhibit actin filament velocity in the in vitro motility experiments, whereas CaDDelta54-85 strongly inhibited actin filament velocity in a manner similar to that of full-length CaD. Moreover, CaD1-597, which lacks the major actin-binding site(s), did not inhibit actin-filament velocity despite the presence of the major myosin-binding site. These data provide direct evidence for the inhibition of actin filament velocity in the in vitro motility assay caused by the tethering of myosin to actin through binding of both the CaD N-terminal region to myosin and the C-terminal region to actin.

Functional and structural relationship between the calmodulin-binding, actin-binding, and actomyosin-ATPase inhibitory domains on the C terminus of smooth muscle caldesmon.

Multiple functional domains responsible for calmodulin (CaM) binding and actin-binding/actomyosin ATPase inhibition are present in the region between residues 598-756 of the chicken gizzard smooth muscle caldesmon (CaD) molecule. To precisely localize these functional domains and to further elucidate the structural basis of these domains, we analyzed a series of purified mutants of chicken gizzard smooth muscle CaD generated by internal deletions of amino acid sequences and expression in a baculovirus expression system. Our results demonstrate that, in addition to a strong actin-binding site sequence between residues 718-723 (Wang, Z., and Chacko, S. (1996) J. Biol. Chem. 271, 25707-25714), two weak actin-binding motifs are present in the regions between residues 690-699 and 650-666. These weak actin-binding regions function independently and are associated with weak actomyosin inhibitory activity. Analysis of the CaM-binding sites A (residues 658-666) and B (residues 690-695), the major CaM-binding sites in the C-terminal region of CaD, provided direct evidence for the involvement of both CaM-binding sites in the CaM-mediated reversal of the inhibition of actomyosin ATPase activity by CaD and for the functional independence of the two CaM-binding sites. Furthermore, the sequences between residues 598-649, upstream of CaM-binding site A, and 700-717, downstream of CaM-binding site B, appear to have no effect on either actin-binding or CaM-binding. The data also suggest that both CaM-binding sites A and B structurally overlap or lie in close proximity to the adjacent weak actin-binding sites and weak actomyosin ATPase inhibitory determinants.

Assessment of an ectopic bone formation system with implantation of pelleted rat primary growth cartilage cells.

To assess the ectopic bone formation system, growth cartilage (GC) cells were implanted to ectopic sites, and the effect of the implanting sites and gender of animals on bone formation were examined. Each 5 x 10(5) batch of GC cells from young rat ribs was pelleted, cultured in calcification medium, and implanted to ectopic sites (in a peritoneal cavitiy or on subcutaneous muscle) in female or male rats. The implanted pellets formed bone after 3-4 weeks at the ectopic sites. Histological examination concluded that, first, a subcutaneous muscle site is more conductive to bone formation than a peritoneal cavity site and, second, the pelleted GC cells implanted in male rats formed bone more effectively than those in female rats. Therefore, at present, the ectopic bone formation system of pelleted primary GC cells onto subcutaneous muscle of male rats is an appropriate system for the evaluation of bone formation.

[Studies on clinical epidemiology of Tsutsugamushi disease of the autumn-winter type in the easten suburbs of Jinan].

This paper reported the epidemiological characteristics of 154 cases under investigation for tsutsugamushi disease of autumn-winter type in the eastern suburb of Jinan. Results showed that the characteristics differed greatly from those in the southern part of China. Majority of patients had toxic symptoms due to infections but specific characteristics were less obvious. Eschars and ulcers appeared only in 14.94% of the cases and occurred only in autumn-winter season. Clinically mild cases constituted the majority. Cases not only distributed sporadically, but also had multi-organ lesions (MOL). Young peasants seemed to be most vulnerable. The pathogen of the disease belonged to low virulent strain. Serological typing belonged to the Gilliam type. The diagnosis and the treatment of the disease were discussed. Specific serum IgG of the disease was detected 6 years after convalescence.

[Study on the mode of transmission of EHFV among host animals and its epidemiologic significance].

In our study, IFA was adopted to detect the epidemic haemorrhagic fever virus (EHFV) antigen and antibody. We studied on the mode of transmission of EHFV in mice and its epidemiologic significance. The results demonstrated that both horizontal and vertical modes of transmission existed in mice under natural circumstances. Horizontal transmission was noticed as the major mode of transmission of EHFV in mice which played an important role in the spread and development of EHF natural foci. Vertical transmission seemed to act merely on the maintenance of EHF natural foci in mice.

Effect of recombinant human transforming growth factor beta 1 on immune responses in patients with chronic hepatitis B.

Studies were undertaken to examine the effect of recombinant human transforming growth factor beta 1 (rTGF-beta 1) on cellular and humoral immune responses of peripheral blood mononuclear cells (PBMC) from patients with chronic hepatitis B. The addition of TGF-beta 1 caused a significant dose-dependent inhibition of hepatitis B (HB) core Ag-stimulated interferon-gamma and antibody to HB core Ag production and proliferation of PBMC from chronic hepatitis patients and HB-immune donors. TGF-beta 1 also induced a significant reduction in pokeweed mitogen-stimulated IgG and IgM production, as well as phytohemagglutinin p-stimulated proliferative response of PBMC. The degree of inhibition of TGF-beta 1 did not differ between antigen-specific and -nonspecific cellular and humoral immune responses, and between control individuals and patients. Pretreatment study with TGF-beta 1 showed that the activities of T cells, B cells and monocytes were similarly inhibited. Further, TGF-beta 1 inhibited activities of HLA class I antigen-matched cytotoxic T cells from patients with chronic hepatitis B for HBV DNA-transfected HepG2 cells in a 51Cr release assay. The results suggest that TGF-beta 1 may play a role in the regulation of antigen-dependent and -independent immune responses in patients with chronic hepatitis B.

Early analysis of viremia and clinical tests in patients with epidemic hemorrhagic fever.

We analysed the early viremia and clinical tests in 82 patients with epidemic hemorrhagic fever (EHF). The results showed that the changes in viremia and clinical tests are related to the severity of the disease and prognosis. Higher concentrations of the virus in infected patients might cause a more unfavourable prognosis and more abnormalities in clinical tests. CK-MB, SGOT, SGPT, serum creatinine and urea nitrogen contents increased markedly, while serum total protein, albumin and calcium contents decreased markedly, indicating that the heart, liver and kidney in EHF patients were severely damaged. Markedly increased WBC and monocytes showed that the patients were seriously infected. Platelet count, antithrombin-III and plasminogen decreased markedly, demonstrating that there were marked changes in the coagulation-anticoagulation and fibrinolytic system of the EHF patients. Changes in RBC, Hb and HCT contents indicated that the blood in the EHF patients had a higher concentration. This study gives further evidence that EHFV plays an important role in the pathogenesis of EHF.

[Alternanthera philoxeroides (Mavt.) Griseb protection against fetal epidemic hemorrhagic fever virus infection in suckling mice].

It has been confirmed that Alternanthera philoxeroides (APG) can markedly protect suckling mice from being infected by epidemic hemorrhagic fever (EHF) virus. After the infected mice were treated with the effective components their survival rate increased, pathological lesion and virus antigen in the tissues mitigated as compared with the controls. Therapeutical dose of APG caused only slight deformation of the hepatic cells.

Effects of transforming growth factor-beta 1 against the inhibitory action of interferon on DNA synthesis and viral replication in hepatitis B virus DNA-transfected cell.

Studies were undertaken to examine the effects of recombinant human transforming growth factor beta 1 (TGF-beta 1) on DNA synthesis and antiviral actions of interferons (IFNs) in HepG2 cell, a hepatoma cell line, transfected with hepatitis B virus (HBV) DNA. The inhibitory effects of IFN-alpha and -gamma on DNA synthesis of HepG2 cells were enhanced in a dose-dependent manner by a simultaneous addition of TGF-beta. The degree of suppression by the reagents was greater in HBV-nontransfected cells than in transfected cells. Inhibition of DNA synthesis was not due to direct cytotoxic effects of the additives, since the viability of HepG2 cells was comparable in the control and treated cultures as determined by trypan blue exclusion. Treatment of HBV DNA-transfected HepG2 cells with IFNs resulted in decrease in production of HB surface and e antigens, and in the level of HBV DNA, but TGF-beta reversed the IFN-induced antiviral state in HBV DNA-transfected HepG2 cells. TGF-beta had no direct effect on HBV replication. These results indicate that rTGF-beta 1 exerts a differential effect against the inhibitory actions of IFN on DNA synthesis and viral replication in HepG2 cells.

Interruption study of viremia of patients with hemorrhagic fever with renal syndrome in the febrile phase.

Kinetic changes of viremia were observed in 287 cases of hemorrhagic fever with renal syndrome (HFRS) in whom ribavirin was administered with double blind random control studied by means of virus isolation, indirect immunofluorescence assay and enzyme-linked immunosorbent assay. The positive rate of viremia was 79.7% (Sp = 3%) and positive rate of HERS IgM was 85% (Sp = 3.1%) before treatment. Viremia could be interrupted by ribavirin as in the ribavirin treated group, the viremia positive rate decreased, duration of viremia was shortened, viral antigen products, virus titer and HFRS IgG antibody level were reduced as compared with the control group. This showed that viremia was very frequent in patients in the febrile phase and ribavirin is an effective antiviral drug in HFRS during the febrile phase. Dosage and course of treatment of this drug are discussed.

Nucleotide sequence and molecular characterization of plasmid-like DNAs from mitochondria of cytoplasmic male-sterile rice.

Two plasmid-like DNAs, B2 and B3, were isolated from mitochondria of the cytoplasmic male-sterile rice, A-58 CMS. Molecular clones having their complete sequences were constructed and used as probes of mitochondrial and nuclear genomes by Southern hybridization. No evidence was found that integrated copies of either one exist in the main mitochondrial genome, but sequences homologous to both were present in the nuclear genome. The complete nucleotide sequences of B2 and B3 were established and compared to those of rice B1 and B4 and to the 1.9- and the 1.4-kbp plasmid-like DNAs of maize. Many of the sequences were common to both plant species.
