A hybrid region-boundary model for cerebral cortical segmentation in MRI.

Automatic segmentation of cerebral cortex in magnetic resonance imaging (MRI) is a challenging problem in understanding brain anatomy and functions. The difficulty is mainly due to variable brain structures, various MRI artifacts and restrictive body scanning methods. This paper describes a hybrid model-based method for obtaining an accurate and topologically-preserving segmentation of the brain cortex. The approach is based on defining region and boundary information using, respectively, level set and Bayesian techniques, and fusing these two types of information to achieve cerebral cortex segmentation. It is automatic and robust to noise, intensity inhomogeneities, and partial volume effect. Another particularity of the proposed approach is that bias field is corrected during segmentation process and that the central layer of the cortex is accurately obtained through a topology correction step. The proposed method is evaluated on both simulated and real data, and compared with existing segmentation techniques. The obtained results have demonstrated its improved performance and robustness.

Intracerebral microdialysis in neurosurgical intensive care patients utilising catheters with different molecular cut-off (20 and 100 kD).

OBJECTIVE: To compare the properties of a new intracerebral micro-dialysis catheter with a high cut-off membrane (molecular cut-off 100 kDalton) with a standard catheter (CMA70, molecular cut-off 20 kDalton). METHODS: Paired intracerebral microdialysis catheters were inserted in fifteen comatose patients treated in a neurosurgical intensive care unit following subarachnoid haemorrhage or traumatic brain injury. The high-cut-off catheter (D(100)) differed from the CMA 70 catheter by the length (20 mm) and cut-off properties of the catheter membranes (100 kDalton) and the perfusion fluids used (Ringer-Dextran 60). Samples were collected every 4-6 hours, analyzed bedside (for glucose, glutamate, glycerol, lactate, pyruvate and urea) and later in the laboratory (for total protein). RESULTS: Fluid recovery was similar for the two types of catheters, but significantly more protein was recovered by the D(100) catheter. The recovery of glycerol and pyruvate did not differ, while minor differences in recovery of glutamate and glucose were observed. The recovery of lactate was considerably lower in the D(100) catheter (p < 0.01), influencing the lactate/pyruvate-ratio. The patterns of concentration changes over time were consistent for all metabolites, and independent of type of catheter. CONCLUSION: Microdialysis catheters with high cut-off membranes can be used in routine clinical practice in the NSICU, adding the possibility of macro-molecule sampling from the extracellular space during monitoring.

Increased body weight via injecting myogenic expression growth hormone-releasing hormone (GHRH) plasmid DNA into sheep.

The ectopic expression of a sheep growth hormone-releasing hormone (GHRH) from muscle tissues, using a myogenic plasmid expression vector pM-GHRH, has been shown to result in enhanced animal growth. Animal injected with pM-GHRH alone exhibited a daily weight gain rate of 0.84+/-0.02%/d (p < 0.05), while that of the control animal was only 0.73+/-0.04%/d. A more significant growth enhancement was observed in the group treated with pM-GHRH plus 0.25% bupivacaine. The animal displayed a daily weight gain rate of 0.87+/-0.03%/d (p<0.01). In addition, the group treated by pM-GHRH and bupivacaine had higher levels of growth hormone (GH) in their blood (1.18+/-0.33 ng/mL) compared with the group treated by pM-GHRH vector alone (p < 0.05). Evaluation of body composition by dissection and histochemical analysis indicated a significant increase in bulk with no organomegaly or associated pathology. PCR analysis demonstrated that there was little plasmid residue detected in the pM-GHRH treated sheep.

[The determination of porphyrin carbon isotope composition by gas chromatography-isotope ratio monitoring mass spectrometry technique].

The porphyrin carbon isotope composition can be used to explore the precursor of porphyrin, oil-oil and oil-source rock correction and calculation of paleo P CO2. The conventional method is limited because of its time consuming and large sample size (several mg of individual porphyrin) required. Therefore, it hampers the application of porphyrin carbon isotope composition into the chemistry and geoscience. The present paper describes a quantification method to prepare bis-(tert-butyldimethylsiloxy) silicon (IV) [(TBDMSO)2Si(IV)] porphyrin which is sufficiently volatile at 300 degrees C and can be used for GC-IRMS analysis. The analysis of carbon isotope composition of aetio I as the form of free base, nickel, demetalization derivative, silicon(IV) and (TBDMSO)2Si(IV) have shown that aetio I porphyrin has no obvious isotope fractionation in the whole synthesis procedure for (TBDMSO)2Si(IV) porphyrin. The carbon isotope study on the porphyrin mixtures of aetio I and OEP indicates that isotope exchange between porphyrins during the synthesis of (TBDMSO)2Si(IV) porphyrin is absent. The method can be applied to the determination of porphyrin carbon isotope compositions. The advantages of the method are time saving, less sample size and lower standard deviation.

Does aluminum inhibit pollen germination via extracellular calmodulin?

The effect of aluminum (Al) on pollen germination and its mechanism of action were investigated. Pollen germination and pollen tube elongation were inhibited by Al at pH 4.5. This inhibitory effect was reversed by the addition of purified calmodulin (CaM), whereas neither the calcium binding-protein S-100 nor Al chelator citric acid at the same concentrations had any obvious effect on Al-inhibited pollen germination. The presence of either the membrane-impermeable CaM inhibitor anti-CaM antiserum or Ca2+ chelator EGTA completely suppressed the effect of exogenous CaM. These results indicate the involvement of extracellular calmodulin in the short-term effects of Al on pollen germination and pollen tube elongation.

[A prevalence study on HBV and HCV infection status in patients with hemopathy].

A prevalence study on Hepatitis B virus (HBV) and Hepatitis C virus (HCV) infection in 144 patients with hemopathy was carried out, using an enzyme linked immunosorbent assay (ELISA). The HBsAg, HBcAb and anti-HCV positive rates in sera of these patients were 9.72%, 36.81% and 24.31% respectively, comparing with data from healthy control subjects, 12.03%, 46.30% and 0.93% respectively. No correlation between HCV infection and patient's age or sex was noticed. Nevertheless the incidence of HCV infection was closely related to the history and the frequency of blood transfusion. The ALT level in sera of patients with HCV infection was higher than that of patients without HCV infection. The ALT level in sera was the highest in patients superinfected with HBV and HCV. Our results suggested that blood transfusion and receiving blood products were important routes of HCV transmission in patients with hemopathy.

[Study of HCV infection and viremia in patients with posttransfusion hepatitis in China].

One hundred and sixty-nine patients with posttransfusion hepatitis (PTH) were investigated for etiology with serological and PCR methods. Our result showed that HCV was the major causative agent of PTH in this study. A part of patients (24/169) were also HBV markers positive. Three patients were negative for all detected viral markers, looking forwards to progressive study. Anti-HCV was detectable from 7 days to one year after onsetting of clinical symptoms, average time 54.62 days. HCV RNA could be detected in early time of HCV infection ( 6 to 20 days later), average time 8.72 days. A follow-up study of 84 patients showed that all patients remained anti-HCV positive during the observation for 1.5 -- 3 years, but their HCV viremia had three forms: transient, persistent or intermittent. The interaction between HCV and HBV by their superinfection was unknown, awaiting further study.

[The prognostic value of focusylated alpha-fetoprotein measurement in liver cancer patients after surgery].

Postoperative change of focusylation index (FI) of serum alpha-fetoprotein (F-AFP) and the result of surgery were studied in 34 patients of hepatocellular carcinoma. In 23 patients, serum F-AFP was disappeared with serum AFP turning negative and no tumor recurrence heppaned within the first 6 months postoperatively. In 4 patients, owing to hepatitis or active cirrohosis in the remained liver, serum AFP didn't turn negative, but FI of AFP was obviously decreased, follow-up found no signs of tumor recurrence within the first 6 months. In 5 of 7 patients, with decreased postoperative serum AFP levels and unchanged FI, tumor recurred. These results showed that postoperative F-AFP monitoring is helpful in evaluating surgical efficacy in liver tumor producing AFP.

Hepatitis C virus infection: its role in chronic liver diseases.

In order to find out the infectious rate of hepatitis C virus (HCV) among chronic liver diseases, we investigated antibodies against hepatitis C virus and HCVRNA by method of ELISA, RIBA and RT-PCR in 410 patients with chronic liver disease. The prevalence of HCV infection was found to be 4%. Whereas the positive rate of HBsAg and HBV-DNA of these cases was 69% and 58%, respectively. There is no statistical significance between HCV infectious rate of patients with positive and negative HBsAg. The relative low infectious rate of HCV infection among chronic hepatic diseases indicates that HBV infection plays a more important role in causing chronic hepatitis than that of HCV. Thus, special emphasis should be paid to the preventive and therapeutic measures against hepatitis B in China.

Establishment and application of SPA-co-operated ELISA for detection of anti-HCV-IgM.

A staphylococcus aureus protein A co-operated ELISA (SPA-ELISA) for the detection of anti-HCV-IgM has been established using HCV antigenic polypeptide, SPA-bearing germs and horseradish peroxidase labelled anti-human IgM. The specificity of SPA-ELISA has been confirmed by some substitution tests, blocking tests and destroying test with 2-mercaptoethanol. The results showed that the rate of anti-HCV-IgG in a group of patients with acute hepatitis and there were significant difference in anti-HCV-IgM was higher than that of anti-HCV-IgM detected rates between patients with acute hepatitis and those with chronic hepatitis (32.26%, P < 0.01). On the other hand, the positive rates of anti-HCV-IgM were 53.66% and 63.41% in transfusion associated hepatitis, 38.10% and 42.86% in sporadic hepatitis, 6.11% and 16.33% in people who have had active social activities, 40.00% and 10.00% in a group of blood donors respectively. Furthermore, taking into account the characteristics of HCV polypeptide used, its easiness of manipulation, and elimination of the interference of anti-HCV-IgG in sera, the new SPA-ELISA is believed to be of practical value in clinical and epidemiological studies of hepatitis C.

Analysis of intrahepatic HBVDNA in serologic HBV markers-negative or HBsAg-negative patients with chronic hepatitis.

To explore HBV infection status in a group of serologic HBV markers-negative or HBsAg-negative patients with chronic hepatitis, in situ hybridization to hepatocellular HBVDNA was carried out in combination with detection of HBsAg and HBcAg in the liver tissues. It was found that prevalence of intrahepatic HBVDNA, HBsAg and HBcAg was 43%, 39% and 17% respectively, and 15 out of 23 cases studied were sure to bear one or more positive marker(s) of intrahepatic HBVDNA, HBsAg and HBcAg. These findings suggest that more than half of the patients with chronic hepatitis were still undergoing HBV infection, despite serologic HBV markers or HBsAg negative. Furthermore, we found that hepatocytes containing HBVDNA or surface or core antigen were often close to hepatic necrosis foci, indicating that HBV replication and its antigen(s) expression in hepatocytes could account for chronic, active and necrotic inflammation occurring in the liver of the patients mentioned above.

The cytoplasmic HBVDNA implication: HBV replication or non-replicative status.

Intrahepatocellular HBVDNA was investigated in patients with chronic hepatitis B by in situ hybridization assay, employing biotin-labelled HBVDNA as the specific probe. It was found that HBVDNA was predominantly present in the cytoplasm of hepatocytes. In general the cases with cytoplasmic HBVDNA could be grouped as follows: 1) cytoplasmic HBVDNA coinciding with intrahepatic envelope protein and nucleocapsid component expression, this pattern of the cytoplasmic HBVDNA would be replicative; 2) cytoplasmic HBVDNA accompanied by intrahepatic single HBsAg expression, common to HBVDNA integrated into host genome with only HBsAg being found frequently in liver; 3) with only cytoplasmic HBVDNA, with neither HBsAg nor HBcAg expression, representative for non-replicative status. It is speculated that the reason for the occurrence of the cytoplasmic hybridization signal in cases with non-replicative HBVDNA may be due to DNA-RNA transcript hybrid. The prudent way to evaluate the implication of the cytoplasmic HBVDNA is to consider the combination of HBVDNA location, intrahepatic gene product expression, and HBV markers in the serum.

Comparison of pre-S1 and pre-S2 proteins in hepatocytes with replication by in situ hybridization assay.

To compare the relationship of pre-S1 and pre-S2 proteins in hepatocytes with HBV replication, HBVDNA in situ hybridization was performed for a group of patients with chronic hepatitis by bio-probe, in conjunction with detection of intrahepatic HBsAg, HBcAg, pre-S1 and-S2 antigens. It was found that the expression of intrahepatic pre-s1 and pre-S2 proteins was visualized as cytoplasmic homogeneous and inclusion types, on some occasions as membranous localization. However, the positive rate for pre-S1 protein was up to 75% (15/20), being significantly higher than that of pre-S2 protein (35%; 7/20) in the same group (P less than 0.05). What in more important, the pre-S1 expression, rather than pre-s2, was in parallel with the presence of intrahepatic HBVDNA and HbcAg. All these findings suggest that the pre-S1 protein might be mainly expressed at the HBV replication phase, thus, to some extent, the detection of pre-S1 protein might complement HBe system and HBVDNA clinical routine.

A study of the binding ability of pre-S1 and S2 proteins of hepatitis B virus to human serum albumin.

To assess the relationship between pre-S proteins of HBV and polymerized human serum albumin (PHSA), a labeled avidin biotin ELISA was used to detect pre-S1, S2 and PHSA receptor (PHSAR) activity. PHSAR activity was only present in the samples positive for pre-S1 and/or S2, but not in the samples positive only for HBsAg. Pre-S1, S2 and PHSAR could in some degree be blocked by preincubating serum with PHSA, and the blocking efficiency of PHSA with respect to pre-S2 and PHSAR was similar, suggesting that pre-S2 is the dominant site for binding PHSA in vitro. We also found that PHSAR activity was detectable in 2 cases positive only for pre-S1, but not pre-S2. Furthermore, PHSA could selectively block pre-S1 and PHSAR activity in 2 cases negative for pre-S2, revealing that pre-S1 also possesses binding ability to PHSA, at least in a small number of cases. Using sandwich ELISA, we demonstrated the existence of complexes of HBV envelope proteins and human serum albumin (HSA) in some HBV infected serum samples. The possible significance of these complexes is discussed.

Expression and clinical significance of pre-S1 and S2 proteins of HBV in sera of patients with chronic liver disease.

To assess the significance of pre-S proteins expression during chronic hepatitis B virus (HBV) infection, we developed a sensitive labeled avidin biotin ELISA to detect pre-S1 and pre-S2 proteins. In serum specimens from 80 patients with chronic liver disease, the frequency of pre-S1 and S2 proteins was 53.7% and 47.5%, respectively. Furthermore, it reached 87.8% and 77.6%, respectively, in cases with chronic HBV infection, indicating that pre-S proteins are usually expressed in sera with chronic HBV infection. We found that the expression of pre-S proteins is closely associated with HBV replicating markers, such as HBV DNA, HBcAg and HBeAg, in sera of patients with chronic HBV infection. Both pre-S1 and S2 proteins were often concurrently expressed in liver and serum with chronic HBV infection. However, the frequency was slightly higher in liver than in serum, suggesting that it may be clinically valuable to detect pre-S proteins in serum and liver simultaneously to determine the status of HBV infection. Our results also indicated that pre-S proteins expression in serum can serve as markers of HBV infection, but cannot be used to estimate the severity and activity of hepatic pathology.

[The experiment studies of semiconductor GaAs-laser points irradiation the analgesic effect].

The article reported that Rabbits threshold of Pain were made a target of observing analgesic effect of the Semiconductor GaAs laser irradiation Znsanli (V36) points. The relations between meridian with peripheral nervous and analgesic mechanism had been explored. The result showed that GaAs laser irradiation at Znsanli (V36) points not only increase basic threshold of pain for Rabbits, but also had all over analgesic effect. After irradiation fifteen minutes obviousness effect in particular. That both the low-frequency electropulse and the laser affected points can raised analgesic effect, but it showed obvious analgesic-back-effect. GaAs laser irradiation nervis fibularis profundus were similar analgesic effect to the points irradiation indicated that points irradiation stimulus conduction had relation to peripheral nervous. After intravenous injection naloxone analgesic effect that GaAs laser irradiation points arose reversion were similar acupuncture analgesia. It indicated that analgesic effect had relation release of the internal source opiate-like substance (OLS).

Element concentrations in the air of an indoor shooting range.

Elemental emissions during firing in a shooting range were measured for different types of ammunition. When using Hirtenberger bullets, lead, barium, antimony and to a lesser extent copper and arsenic were the primary metal pollutants. Stationary sampling at three locations in the range did not reveal large concentration gradients. Large concentration variations were observed by sampling before, during and after shooting. Lead and antimony concentrations peak at 5060 and 119 micrograms m-3, respectively. Soil elements such as aluminium, sodium and calcium are enriched during shooting, probably due to soil resuspension by the shooters and the bullets hitting the sand backstop. After shooting has ceased the concentrations fall to within pre-shooting levels within a couple of hours. Measurement of the aerodynamic particle size shows low mass median diameters for the elements emitted during firing and larger diameters for the soil-associated elements. The peak airborne concentrations measured by stationary sampling, and human exposure measured by a personal sampler carried by an instructor were compared with threshold limit values. During the shooting the TLV is significantly exceeded for lead.
