[Immunohistochemical study on three peptidergic terminals in nasal mucosa in a rat AR model].

To study the density alterations of three peptidergic terminals in nasal mucosa of allergic rhinitis (AR), an exhaustive immunohistochemical sutdy on the changes of substance P (SP), neurokinin A (NKA) and neurokinin B (NKB) in nasal mucosa was carried out in a toluene-2, 4-isocyanate (TDI)-induced rat AR model. The densities of all three tachykininergic terminals in nasal mucosa were significantly increased (P < 0.01) in experimental group as compared with control group. Increased staining, thickening of peptidergic terminals as well as enlargement of varicosities were observed. The increased densities of three tachykininergic terminals (SP, NKA and NKB) in nasal mucosa in rat AR model indicates that tachykinins play an important role in the pathogenesis of AR.

Changes of cellular morphology and nuclear DNA content in different nasopharyngeal epithelium from different patients.

Cellular morphology and nuclear DNA content were measured with point-counting technique and flow cytometry (FCM) respectively in normal nasopharyngeal epithelium (NE), tissue adjacent to carcinoma (ATC) and nasopharyngeal carcinoma (NPC). The results showed that all 20 patients with NE showed diploid, whereas 38 (38%) of 101 patients with NPC, 7 (58%) of 12 patients with recurrent NPC, 6 (30%) of 20 patients with ATC and 2 (33%) of 6 patients with NE positive to EBVCA-IgA test showed nondiploid. A total of 19 morphometric parameters of the cell were measured and calculated. The results showed that with malignant transformation of NE, most of the cellular morphometric parameters gradually increased in numerical values while no marked difference was shown between normal NE and simple hyperplasia or metaplasia in terms of cytomorphometric characteristics. Most of the parameters in dysplasia changed significantly as compared with those normal NE, simple hyperplasia or metaplasia and NPC groups. Cytomorphometric analysis seems to give strong support to the concept that dysplasia is among the transitional stages of disease process between normal NE and NPC. Our results show that morphometric analysis and FCM may offer valuable and objective criteria in the early diagnosis of NPC and premalignant disease.

An immunocytochemical study on relations between mast cell and peptidergic terminals in nasal mucosa of chronic rhinitis patients.

Mast cell-nerve relation is a new topic explored deeply in different organs, but little documentation could be found in the literature on the relation in human nasal mucosa. We carried out this study using immunocytochemistry and found that substance P (SP) terminals were present in human nasal mucosa from six cases of chronic rhinitis. SP terminals were often found to be adjacent to or have a direct contact with mast cells (MCs). Electron-microscopic studies revealed that MCs could contact nonmyelinated nerve terminals. These results have important implications in the understanding of the pathogenesis of neurogenic inflammation seen in nasal mucosa and will probably cast new insight into the future treatment of such disease.

An association study between essential hypertension and HLA-DRB1 alleles.

It is well established that genetic and environmental factors are involved in the etiology of essential hypertension (EH). Previous studies have suggested that at least one of the HLA genes is responsible for the genetic susceptibility to EH. Our aim in the present study was to investigate this issue in China by the PCR-SSP HLA-DRB1 typing method. The results showed an increased frequency of HLA-DR2 and a decreased frequency of HLA-DR7 with EH patients compared with controls. We consider that HLA-DR2 may represent a marker for susceptibility to EH in the North Chinese population.

[Distribution of calcitonin gene-related peptidergic terminals in rat nasal mucosa].

The distribution of calcitonin gene-related peptidergic (CGRP) terminals in rat nasal mucosa was observed with immunocytochemistry technique (ABC method). The results showed that CGRP-ergic terminals had extensive localizations along the walls of arterioles and venules, around the acini of glands and between the epithelial cells, with the walls of arterioles having highiest densities of such terminals. The typical morphology of CGRP-ergic terminals was one of multistage branched fibers in the form of rosary varicosities. CGRP immunoreactive neurons were also present in trigeminal ganglion (TG). Based on our observations, it is suggested that CGRP-ergic terminals in rat nasal mucosa are the peripheral fibers originating from the ganglion cells in TG. These fibers may be involved in the perception of local mucosal stimuli and propagation of neural impulses to the central terminals and will then release CGRP in the brain stem. There is also possibility that CGRP may be peripherally released in nasal mucosa and exerts influences on mucosal functions.

Rapid HLA-DRB1 generic typing by PCR-SSP method.

We report a simple and rapid HLA-DRB1 generic typing method, PCR-SSP, which is practical and inexpensive. We use 9 sequence-specific primers and 2 group specific primers to define the HLA-DRB1 specificities DR1, DR2, DR3, DR4, DR5, DR6, DR7, DR8, DR9 and DR10. The HLA DR3, DR5, DR6 and DR8 can be amplified by the two primers of DR3568 and DRB1. The DR6 specificity can be identified by excluding the DR3, DR5 and DR8 when the DR3568 are positive. Any individuals can be typed with some exception: the three pairs of phenotype DR3/DR3 and DR3/DR6, DR5/DR5 and DR5/DR6, DR8/DR8 and DR8/DR6 cannot be discriminated from each other. We typed 106 unrelated healthy people from Beijing locations in two weeks. We think this typing method is suitable to replace the error-prone serologic HLA-DR tests in routine clinical practice, including the prospective typing of cadaveric organ donors.

[Analysis of misdiagnostic acoustic neuroma].

72 cases (74 tumors) of acoustic neuroma pathologically confirmed after operation were analysed with special regard to the misdiagnosis made in their initial seeking for medical advice from ENT doctors. The average period of misdiagnosis was 5.48 years. The disease entities that the acoustic neuroma had been mistaken for were sensorineural hearing loss of unknown origin, idiopathic sudden deafness, tinnitus of unknown origin, cervical osteophytosis, nasopharyngeal carcinoma, etc. Emphasis was placed on the discussion of various causes of misdiagnosis.

[The segmental distribution of afferent neurons of liver, and "qi men" point, "tai chong" point in the spinal ganglion].

Experimental animals were 38 cats, 10% HRP 50-60 microliters was injected into "Tai Chong", "Qi Men" points respectively and 10 cats were used for each point. Another 18 cats were injected with 10% HRP 100 microliters into the liver. The segmental distribution of afferent neurons of points and liver was observed under light microscope. We observed the majority of labeled cells in the spinal ganglion. The segmental distribution of afferent neurons of "Tai Chong" point was T12, L1-7 and S1-3. The segmental distribution of afferent neurons of "Qi Men" point was T5-8. The segmental distribution of afferent neurons of liver was T3-12 and L1. The results show that the afferent of "Tai Chong, Qi Men" points and the liver are mutual intersected and overlapped for 2-4 segments. The mutual intersection and overlapping were probably the morphological foundation of treatment of liver disease by needing "Tai Chong", "Qi Men" point, at the same time, the results may elucidate the underlying mechanism of correlation between acupoints and viscera, too.

[The segmental distribution of sympathetic afferent neurons of the heart, cardiac nerve, and projection of the cardiac nerve to the central nervous system].

1. The HRP labeled method was used to study the segmental distribution of sympathetic afferent neurons of the heart. 10%-20% HRP 150 microliters solution was injected around the left coronary artery of 18 cats. The HRP labeled cells were observed in the C8-T10 spinal ganglions, therefore the segmental distribution of sympathetic afferent neurons of the heart were in C8-T10 spinal cord. 2. The segmental distribution of cardiac nerve's afferent neurons was studied, 30% HRP 15 microliters was injected into the cardiac nerve of 18 cats. The HRP labeled cells were observed in the C8-T9 spinal ganglions. 3. CB-HRP 10 microliters was injected into the medium and inferior cardiac nerves. The cardiac nerve's fibers project into T1-5 spinal cord, the HRP labeled fibers were observed in III-VII lamina of spinal cord, and there were connections with III, IV, V, VI, VII laminas. 4. The segmental distribution of sympathetic afferent neurons of heart and cardiac nerve, projection of the cardiac nerve the III-VII laminae of spinal cord and the connection of fibers in the III-VII laminae were the morphologic foundations of referred pain and the heart disease. (tachycordia, angina etc.) treated with acupuncture.

Evidence for thiol-dependent metallo-endopeptidase involved in degradation of luteinizing hormone-releasing hormone in glioma cells.

An endopeptidase generating a peptide comprised of the first five amino acids of luteinizing hormone-releasing hormone (LHRH) was isolated form the membranes of glioma C6 cells by a procedure including Brij extraction, p-mercuribenzoate-Sepharose chromatography, and Mono Q high-performance liquid chromatography. The molecular weight of the enzyme was estimated to be 64,000. The glial enzyme shows a similar inhibitor susceptibility to that of the enzyme of neuronal origin, and also to that of endopeptidase-24.15 (EC 3.4.24.15). Angiotensin-converting enzyme (EC 3.4.15.11) is suggested to be involved in the secondary cleavage of LHRH by the glioma cells.

[Segmental distributions of sensory neurons of the "ganshu", "pishu", "liangmen", "qimen" points and the gallbladder].

One hundred guinea pigs were used. The 10% HRP 50-60 microliters solution was injected into the acupoints of "Gansu", "Pisu", "Liangmen" and "Qimen" of every animal. Moreover, the 10% HRP 80-100 solution was injected into the gallbladder of the another animals too. Under the microscope, observed the segmental distributions of sensory neurons of the four acupoints and the gallbladder. It could be found much clearly that HRP labelled cells were appearing in the spinal ganglions. The above experimental studies showed that there are mutual intersect 5-7 segments between the gallbladder and the "Gansu", "Pisu", "Liangmen" and "Qimen" four acupoints. It is probable that the mutual intersect is the neuroanatomic foundation of acupuncture and moxibustion to inhibit the formation of bilirubin gallstones.

IGF-II-like immunoreactivity in human tissues, neuroendocrine tumors, and PC12 cells.

We looked for cells that reacted with monoclonal antibodies against rat insulin-like growth factor II (IGF-II). We found them in human fetal kidneys in the 14th week of gestation and in adult adrenal medullas but not in human liver, pancreas, pituitary gland, or thymus. Every pheochromocytoma in a group of 20 had IGF-II-like immunoreactive cells, as did three out of four ganglioneuroblastomas (one of which was heterotransplantable in athymic nude mice) and one carotid body tumor. Using the electron microscope we localized the immunoreactivity in pheochromocytoma cells to their neurosecretory granules of the non-catecholamine type. We failed to find any IGF-II-reactive cells among the pancreatic islet cell tumors, neuroblastomas, medullary carcinomas of the thyroid, retinoblastomas, and Wilms' tumor we studied. One line of human neuroblastoma cells did show some immunoreactivity after treatment with dbcAMP. Additionally, the rat pheochromocytoma cell line PC12 and its subline PC12h occasionally produced a few immunostained cells. These results strongly indicate that human IGF-II is primarily produced in paraganglionic tissues and tumors.

[Percutaneous transhepatic and transileocolic obliteration with absolute ethanol for the treatment of bleeding gastroesophageal varices].

Embolization of bleeding gastroesophageal varices with absolute ethanol performed in the recent two years was reported. The treatment was carried out either by means of percutaneous transhepatic or transileocolic obliteration. Obliteration treatment was attempted in 42 cases, among which 20 were transhepatic and 22 transileocolic. 16 of the 20 and 17 of the 22 were effectively obliterated with a total success rate of 78.6%. Two control groups of bleeding varices, one of 45 cases treated with conventional medical methods and the other of 53 cases treated with endoscopic sclerotherapy, were studied to compare the results. The efficacy for control of active bleeding in the obliteration, conventional and sclerotherapy groups were 100%, 84.1%, and 91% respectively, while the rebleeding rate was 27.2%, 31.5% and 23.2% and mortality rate (due to rebleeding) 12.1%, 31.5% and 11.6% respectively. The control of active bleeding, the results in the three groups were more or less similar, but the rebleeding rate was much lower in the obliteration and sclerotherapy groups than in the conventional group. However, sclerotherapy requires longer time and yields more complications than obliteration is a valuable therapeutic measure in controlling variceal bleeding and preventing rebleeding.
