Generation of mice with a 200-kb amyloid precursor protein gene deletion by Cre recombinase-mediated site-specific recombination in embryonic stem cells.

Gene disruptions and deletions of up to 20kb have been generated by homologous recombination with appropriate targeting vectors in murine embryonic stem (ES) cells. Because we could not obtain a deletion of about 200 kb in the mouse amyloid precursor protein gene by the classical technique, we employed strategies involving the insertion of loxP sites upstream and downstream of the region to be deleted by homologous recombination and elicited excision of the loxP-flanked region by introduction of a Cre expression vector into the ES cells. In the first approach, the loxP sequences were inserted in two successive steps and after each step, ES cell clones were isolated and characterized. Deletion of the loxP-flanked sequence was accomplished by introducing the cre gene in a third step. In the second approach, ES cells containing the upstream loxP cassette were electroporated simultaneously with the downstream loxP targeting vector and the Cre expression plasmid. ES cells were obtained that gave rise to chimeric mice capable of germ-line transmission of the deleted amyloid precursor protein allele.

Mice homozygous for a modified beta-amyloid precursor protein (beta APP) gene show impaired behavior and high incidence of agenesis of the corpus callosum.

The amyloid precursor protein (beta APP) gene of the mouse was disrupted by homologous recombination; however, contrary to expectation, brain and other tissues still contained beta APP-specific RNA, albeit at a level 5-10 fold lower than wild-type and lacking the disrupted exon, which had been spliced out. The brain contained shortened beta APP-specific protein at a low level. Mutant mice were severely impaired in spatial learning and exploratory behavior and showed increased incidence of agenesis of the corpus callosum.

Modulatory effects of gonadorelin on GABA-induced depolarization and GABA-activated current in rat spinal ganglion neurons.

AIM: To explore the modulatory effects of gonadorelin on GABA-induced depolarization and GABA-activated current in membrane of rat primary sensory neurons. METHODS: Intracellular recordings and whole-cell patch clamp techniques were performed on neurons in rat spinal ganglia (SG) preparation and neurons freshly isolated from rat SG, respectively. Drugs were applied by superfusion and/or by bath application. RESULTS: In the majority of neurons GABA (10 mumol.L-1 -1 mmol.L-1) induced a depolarization, which was blocked by bicucullin (100 mumol.L-1). Pretreatment with gonadorelin (50 mumol.L-1) decreased the GABA-induced depolarization by 79 +/- 22% (n = 29), while gonadorelin elicited no effect or slight depolarization alone. In 6 of 11 cells, GABA-activated currents were also inhibited by pretreatment with gonadorelin (50 mumol.L-1), while in 5 of 11 cells, there was no change or a slight potentiation. CONCLUSION: Gonadorelin exerts an inhibitory effect on GABA-induced depolarization and GABA-activated current in the primary sensory neurons.

[Effect of substance P on the somatic membrane of rat DRG neurons].

Intracellular recordings were performed on isolated rat DRG neurons to investigate the changes in the membrane potential in response to substance P and the involved ionic mechanisms. The resting membrane potential examined was -58.9 +/- 8.2 mV (X +/- SE) (n = 81). The conduction velocities estimated were: 20.4 +/- 4.8 m/s (X +/- SE) ranging from 14.1 to 28.7 m/s (47/60) in type A(alpha beta) cell, 9.8 +/- 5.2 m/s (X +/- SE) ranging from 1.2 to 13.7 m/s (13/60 in type A(delta) and type C cell. In majority of the neurons bath application of SP (10(-7) - 3 x 10(-4) mol/L) induced marked membrane potential depolarization (56/60). The membrane conductance increased 24.6% in average from control value of 2.72 x 10(-8) S during SP-induced depolarization (n = 3). The reversal potential was between +40 - %50 mV (n = 3). When NaCl in BSS was substituted with choline chloride or containing TTX (10(-5) mol/L), the amplitude of SP-induced depolarization attenuated markedly but not incompletion eventually. When high (20 mmol/L) and low (0 mmol/L) Ca2+ BSS was used, the amplitude of SP-induced depolarization increased and decreased respectively. However, when BSS containing 10(-4) mol/L Cd2+ or 10(-2) mol/L TEA was used SP-induced depolarization was reduced. The above results indicate that SP-induced depolarization involves a rather multiple changes in ionic conductance.

[Subtypes of glutamate receptor and their distribution in the somatic membrane of neurons freshly isolated from rat DRG].

The aim of the present study was to investigate the distribution and coexistence of glutamate (Glu) receptor subtypes in the isolated rat DRG neurons as identified by the recorded NMDA-, KA- or QA/AMPA-activated current using whole-cell patch-clamp technique. Of the 37 identified neurons, 15 contain only a single subtype receptor, 13 with two kind subtype receptors, only 4 with all subtype receptors. With regard the frequency of occurrence of the three types of receptors, the NMDA, KA and QA/AMPA subtype were found present respectively in 26 (70.3%), 7 (18.9%) and 21 (56.8%) of the recorded cells.

[Influence of peripheral process branching in nerve of spinal ganglion on the primary afferent impulse].

In order to observe the electrical and morphological characteristics of neurons in toad spinal ganglion (SG) preparation, microelectrode filled with 0.75% horseradish peroxidase (HRP) was used both for intracellular recording and for intracellular labelling. Histological result showed that there were apparent branchings in peripheral process of A type cells. Intra-axonal recording of the discharges of dorsal root fiber could be evoked by simultaneous stimulation of two separate fine bundles in spinal nerve. The discharging event exhibited the patterns of interruption, default and addition. The above results suggest that the peripheral process branching might alter the array and frequency of primary sensory afferent input.

Calbindin D28k mRNA in hippocampus, superior temporal gyrus and cerebellum: comparison between control and Alzheimer disease subjects.

To further investigate the role of calbindin D28k in Alzheimer's disease (AD); hippocampus, superior temporal gyrus and cerebellum from control and AD cases were examined by quantitative in situ hybridization. We report here a decrease in CaBD28k mRNA in the CA2 region of AD hippocampus compared to control subjects. There were no significant differences between AD and control subjects in the other regions studied.

Behavioral and anatomical deficits in mice homozygous for a modified beta-amyloid precursor protein gene.

The beta-amyloid precursor protein (beta APP) gene of the mouse was disrupted by inserting into exon 2 a cassette containing a neomycin resistance gene and a putative transcription termination sequence. Contrary to expectation, brain and other tissues from mice homozygous for the insertion still contained beta APP-specific RNA, albeit at a level 5- to 10-fold lower than wild type and lacking the disrupted exon, which had been spliced out. The brain contained shortened beta APP-specific protein at a low level. Mutant mice were severely impaired in spatial learning and exploratory behavior and showed increased incidence of agenesis of the corpus callosum.

A modified product inhibition model describes the nonlinear pharmacokinetics of nicorandil in rats.

Nicorandil, a vasodilator which acts through both cyclic GMP accumulation and K+ channel opening, has been used in the treatment of various cardiovascular diseases. We have examined the pharmacokinetics of nicorandil in the rat as a function of dose, as both i.v. boluses (9 doses, 0.75-12 mg, n = 1.4 per dose), and as a 5-hr infusion followed by a 5-hr washout (6 doses, 10-500 micrograms/kg/min, n = 3 per dose). Plasma nicorandil concentrations were determined by HPLC. Nicorandil plasma concentrations increased disproportionately with dose, but nicorandil elimination obeyed apparent monoexponential kinetics, and the apparent half-life (t1/2) increased with dose. In addition, the approach to apparent steady-state during the infusion phase was not overtly sensitive to the drastic changes in t1/2 observed. Pharmacokinetic modelling with several nonlinear models, viz: Michaelis-Menten with parallel first-order, cosubstrate depletion and competitive product inhibition, were carried out. Addition of the sulfhydryl donor, N-acetyl-L-cysteine, did not change the pharmacokinetics of nicorandil, providing experimental indication that a cosubstrate depletion model might not be applicable. To described the unique pharmacokinetics, a modified product inhibition model was developed. This new model includes the classic competitive product inhibition equation, describing both parent and product kinetics, and it incorporates, in addition, separate first-order elimination rate constants for both nicorandil and the inhibition metabolite. Experimental evidence showed that N-(2-hydroxyethyl) nicotinamide, the major metabolite of nicorandil in rats, and nicotinamide (niacinamide) itself, indeed inhibited nicorandil elimination.

[Modulatory effects of substance P on the membrane responses mediated by GABAA and GABAB receptors in DRG neurons].

Intracellular recordings were performed on the neurons of young rat DRG to study the modulatory effects of SP on the responses mediated by GABAA and GABAB receptors. In the majority of the neurons (20/30) the GABA induced depolarization was suppressed to 50.8 +/- 20.2% by preapplication of SP (5 x 10(-6)-4 x 10(-5) mol/L), which applied alone had no effect or only caused a slight depolarization. In addition, SP could increase APD50 by 28.7 +/- 9.1% in many neurons (10/18), and the baclofen-induced shortening of 20.6 +/- 2.9% in APD50 could abolished (4/12) or even reverse to a lengthening of 19.3 +/- 8.9% in APD50 (8/12) by the preapplication of SP. Moreover, it was found that the activation of GABAB receptors could inhibit the succeeding response mediated by GABAA receptors. Since the soma of the DRG neuron is generally considered as an accessible model for the primary afferent terminals, the results suggest that SP which is released in the dorsal horn during nociceptive stimulation can antagonize the responses mediated by GABAA and GABAB receptors.

[Inhibitory effect of SP on GABA-activated currents in freshly isolated rat DRG neurons].

The whole-cell patch-clamp technique was used to record GABA-activated currents in freshly isolated DRG neurons. It was found that application of SP (10(-7)-10(-5) mol/L) could induce concentration dependent inward currents in some cells (4/26), while no such current was detectable in the other neurons. However, preapplication of SP could inhibit the inward currents mediated by GABAA receptor (18/22). The inhibitory effect of SP was concentration dependent and reversible, and the desensitization of GABA-activated current could also be enhanced by SP.

Determination of fluconazole by direct injection of plasma and high performance liquid chromatography with column switching.

High performance liquid chromatography with column switching has been developed for the determination of fluconazole in plasma. The plasma samples were injected onto a precolumn packed with LiChromprep RP2(25-40 microns) after simple dilution with a solution of 0.2 mol.L-1 acetic acid. Distilled water was used to wash out protein and other polar components in plasma. After switching, the concentrated fluconazole was eluted in the back-flush mode onto a Shimpack CLC-ODS column with methanol--0.2 mol.L-1 ammonium acetate (pH 2.7) (50:50) as mobile phase. Purge solutions were used for clean-up and for regenerating the precolumn. The method showed a good precision and excellent recovery. The detection limit was 0.12 micrograms.ml-1 plasma. The RSD's (intra- and interday) were less than 6.3%. The method has been successfully used to determine bioavailability and pharmacokinetics of fluconazole in Chinese volunteers.

Electrophysiological and relevant morphological properties of neurons in toad dorsal root ganglion.

Intracellular recordings were performed on isolated preparations of toad dorsal root ganglion (DRG). Of the 87 neurons examined, 83 were of type A cells and 4 of type C cells. The conduction velocity (CV) of type A cells ranged from 2.55 to 35.19 m/s with a mean of 11.18 +/- 0.78 m/s (mean +/- s mean), while that of type C cells ranged from 1.34 to 2.43 m/s with a mean of 2.11 +/- 0.22 m/s (mean +/- s mean). On the basis of the characteristics of Ap configuration, these 87 cells can be classified into three groups: H neuron (24/87); F neuron (50/87) and F-H neuron (13/87). The Sizes of DRG cells were examined by measuring of HRP labelled cells in part of the experiment. By comparing the size of F and H cell with their CV, we proposed a revised view which is against the public opinion concerning the relation between the size and CV of DRG cells.

[Protective effects of nitrendipine against myocardial infarction in rabbits].

In rabbits the left circumflex coronary artery was ligated for 24 h. Nitrendipine (Nit) 10 mg.kg-1 ig were given 1 h before ligation and 1, 4, and 7 h after ligation. Nit decreased myocardial infarctive size (MIS) from 25.2 +/- 1.7% to 15.5 +/- 1.6% (P < 0.01), lowered serum creatine kinase (CK) and lactate dehydrogenase isoenzyme 1 (LDH1) from 381 +/- 69 IU.L-1 and 42 +/- 6% to 281 +/- 49 IU.L-1 (P < 0.01) and 31 +/- 8% (P < 0.05), diminished injury of myocyte ultrastructure, especially mitochondria and myofibrillae, in ischemic and infarctive zone. Nit exerted no significant effect on plasma level of norepinephrine (NE). The results indicate that Nit has protective effects against the acute myocardial infarction.

[Responses mediated by GABAA and GABAB receptors on the somatic membrane of toad dorsal root ganglion neurones].

Intracellular recordings were made from neurons in toad dorsal root ganglion in vitro. Bath application of GABA (10(-4)-10(-3) mol/L) elicited either depolarization or depolarization followed by hyperpolarization respectively in 79% and 10% of the neurons. The remaining 11% did not show any response. The GABA-induced depolarization, which was accompanied by an increase of membrane conductance and usually a shortening of action potential duration (ApD), could be blocked by bicuculline. The GABA-depolarization was enhanced by low Cl- Ringer or reduced by high Cl- Ringer The membrane potential was not affected by 10(-4) mol/L baclofen. On the other hand, shortening of ApD could be produced by baclofen, but not blocked by bicuculline. Our results suggested that GABAA and GABAB receptors coexist on the somatic membrane of toad dorsal root ganglion neurons and mediate the changes respectively in membrane potential and shortening of ApD. The mechanism underlying presynaptic inhibition in the terminals of the primary afferents was discussed.

Use of a dihydrogen osmium complex as a versatile 1H NMR recognition probe.

A new recognition probe for biomolecules, [en2Os(eta 2-H2)]2+ (1; en, ethylenediamine), is reported. In aqueous solution, 1 binds readily to a variety of biomolecules, including nucleotides, RNA, amino acids, peptides, and phospholipids. In each case, binding leads to a characteristic proton nuclear magnetic resonance (1H NMR) for the dihydrogen that appears in a spectral window in the range delta = 0 to -20 parts per million, and as well to characteristic values of the coupling JHD and of the relaxation time T1. Small structural differences in molecules such as DGMP (2'-deoxyguanosine 5'-monophosphate) and IMP (inosine 5'-monophosphate) or Asp and Glu can readily be distinguished, such as when 1 binds to the N-7 position of the nucleobase of DGMP or IMP and when 1 binds to the carboxylate of Asp or Glu. Upon one-electron oxidation of the metal center, diamagnetic 1 is converted to a paramagnetic probe.

[Direct injection of plasma to determine gliquidone in plasma using HPLC column switching technique].

An automated clean up and concentration method by column switching is described for the assay of gliquidone, a hypoglycemic agent, in plasma. The system uses Merck Lichroprep RP2 (25-40 microns) as short precolumn packing material for on--line clean up and concentration. A Shimpack CLC--ODS (5 microns) is used as analytical column. Water is used as the pretreatment mobile phase and a mixture of methanol--isopropanol--0.2 mol/L ammonium acetate solution (61:5:34) is used as the analytical mobile phase. The plasma is diluted with a solution of 0.2 mol/L acetic acid and injected directly on the precolumn. The low speed of 0.2 ml/min of pretreatment mobile phase during transfer of plasma sample on the precolumn can get recovery of 80.7%. The detection is at 310 nm and the detection limit is 30 ng/ml plasma. The procedure is simple and automatic.

Techniques for intra-axonal recording of electrical activity from single nerve fiber in vitro and in situ.

The method usually applied for recording the electrical activity of single nerve fiber is extracellular recording on isolated single nerve fiber. It is difficult to obtain stable and satisfactory results from direct intracellular recording of single nerve fiber, except for some special biological materials, such as Loligo giant axon. We present in this paper a technique for recording intracellular resting membrane potential and action potential from single nerve fiber both in vitro and in situ, using glass microelectrode and a special mirror-base plate for fixing the preparation. Besides, we also report a method for labelling single nerve fiber by means of injection of HRP into the axon.
